Clinical Flow Cytometric Testing in Chronic Lymphocytic Leukemia.

Flow cytometry is the cornerstone for establishing the diagnosis of chronic lymphocytic leukemia (CLL), owing to its characteristic and well-defined immunophenotype that enables accurate distinction from other leukemias and lymphomas. Beyond diagnosis, flow cytometry provides essential prognostic information and allows sensitive detection of minimal residual disease (MRD), a strong predictor of clinical outcome. CLL MRD assessment is increasingly used to guide risk stratification, therapeutic decision-making, and treatment duration in the era of targeted therapies and immunotherapies. This cha

Survival Outcomes in Chronic Lymphocytic Leukemia During the Novel Agent Era: A Population-Based Analysis.

Chemoimmunotherapy (CIT) had long been used to treat chronic lymphocytic leukemia/small lymphocytic lymphoma (CLL/SLL) until 2014 when the first novel agent, ibrutinib, was approved in the United States. Numerous novel therapies have since become available. Nevertheless, as novel therapies have largely replaced CIT in CLL/SLL, its impact on survival has yet to be examined on the population level. In this study, we used the SEER (Surveillance, Epidemiology, and End Results) database to compare survival in patients with CLL/SLL before and after the approval of the first novel therapy. We include

Impact of extramedullary plasmacytoma on outcomes in patients with relapsed refractory multiple myeloma treated with talquetamab: U.S. Myeloma Immunotherapy Consortium.

Talquetamab, a GPRC5D-targeting bispecific antibody, shows promising activity in relapsed-refractory multiple myeloma. However, real-world outcomes in patients with extramedullary disease (EMD) remain poorly described. We studied 360 patients treated with talquetamab at 15 U.S. centers by Dec 2024. Soft tissue plasmacytomas (STP) were classified as EMD (not contiguous with bone) or paraskeletal disease (PSD; contiguous with bone). If both were present, patients were assigned to the EMD category. Of those, 97 (27%) had EMD, 22 (6%) had PSD, and 241 (67%) had No-STP. Median follow-up was 12.8 mo

Real-world effectiveness and safety of zanubrutinib in Spanish patients with relapsed/refractory marginal zone lymphoma.

There is no established standard treatment of relapsed/refractory (R/R) marginal zone lymphoma (MZL). Zanubrutinib was approved in this setting based on the phase 2 MAGNOLIA trial. We retrospectively evaluated the characteristics and outcomes of 118 real-world patients in Spain with R/R MZL treated with zanubrutinib after at least 1 previous anti-CD20-based regimen. The median age at zanubrutinib initiation was 75 years, and 56% were female. MZL subtypes included splenic (55.3%), mucosa-associated lymphoid tissue (18.9%), nodal (19.8%), and nonclassifiable (6%). The median number of previous l

The role of allogeneic hematopoietic stem cell transplantation in adults with mature T-cell lymphomas.

RATIONALE: Mature T-cell lymphomas (MTCL) are a rare, heterogeneous group of malignancies that arise from post-thymic T-lymphocytes, with a generally poor prognosis. For a long time, the standard therapy for most MTCL subtypes has been polychemotherapies. Hopes to prolong overall survival in MTCL have been associated with transplant procedures such as high-dose therapy with autologous stem cell transplantation (autoSCT) or allogeneic stem cell transplantation (alloSCT). OBJECTIVES: To assess the benefits and harms of alloSCT in the treatment of MTCL in adults (≥ 18 years old) compared to any

Pathology, Clinical Features, and Natural History of Low-Grade B-Cell Lymphomas With Primary Osseous Presentation.

Primary bone lymphomas are rare, and most reported cases represent diffuse large B-cell lymphoma. Low-grade B-cell lymphomas presenting with primary osseous involvement (PB-LGBCLs) are far less common, and their clinicopathologic features and clinical course remain poorly defined. We retrospectively analyzed 32 PB-LGBCLs diagnosed at a single tertiary cancer center. Cases were included when the disease presented with bone-only involvement, with or without contiguous soft tissue extension, and without lymph node, spleen, or noncontiguous extranodal disease at initial diagnosis. Staging marrow a

Primary ocular adnexal MALT lymphoma involving the upper eyelid in a young woman: a case report and literature review.

Primary ocular adnexal MALT lymphoma (POAML) is a rare indolent small B-cell lymphoma that primarily involves the orbit, conjunctiva, and lacrimal gland, with a high risk of early misdiagnosis. Herein, we report a rare case of POAML arising in the right upper eyelid of a 35-year-old woman, who presented with progressive eyelid swelling. The diagnosis was established by surgical excision, histopathological examination, and immunoglobulin gene rearrangement analysis. No systemic involvement was identified, and the disease was staged as Ann Arbor IE. The patient received local radiotherapy (28 Gy

Efficacy and safety of ifupinostat in relapsed or refractory diffuse large B-cell lymphoma: a multicenter, single-arm phase 2 study.

Ifupinostat (BEBT-908) is a novel dual-targeted inhibitor of histone deacetylase (HDACs) and phosphatidylinositol 3-kinase alpha (PI3Kα). We evaluated the efficacy and safety of ifupinostat monotherapy in patients with relapsed or refractory (r/r) diffuse large B-cell lymphoma (DLBCL) after at least 2 prior lines of systemic therapy. Eligible participants received intravenous ifupinostat at 22.5 mg/m² on days 1, 3, 5, 8, 10, and 12 of a 21-day cycle. The primary endpoint was objective response rate (ORR) assessed by blinded independent central review (BICR). Secondary endpoints included diseas

Cell Death Induction by MO2455, a Novel Potential Anti-Cancer Agent in Monocytic and T-Cell Leukemia Cells.

Despite advances in targeted therapies, treatment resistance and disease recurrence remain major obstacles in leukemia management. Structural modification of clinically successful kinase inhibitors represents a promising approach for discovering novel anti-leukemic agents with improved biological activities. In the present study, we investigated the anti-leukemic activity and mechanism of action of MO2455, a newly synthesized imatinib-derived compound, in monocytic leukemia U937, THP-1, and T-cell leukemia CCRF-CEM cells. MO2455 showed substantial cytotoxic activity against all three cell line

Primary Hepatic Mucosa-Associated Lymphoid Tissue Lymphoma and Primary Biliary Cholangitis: A Case Report of a Rare Association.

INTRODUCTION: Primary hepatic mucosa-associated lymphoid tissue (MALT) lymphoma is an exceedingly rare subtype of extranodal marginal zone B-cell lymphoma within the spectrum of non-Hodgkin lymphomas. Chronic inflammatory and autoimmune conditions are recognized risk factors for lymphomagenesis; however, the association between primary hepatic MALT lymphoma and primary biliary cholangitis (PBC) has been rarely reported. CASE PRESENTATION: We report the case of a 50-year-old woman with a 9-year history of non-cirrhotic PBC, treated with ursodeoxycholic acid and with only partial biochemical re

The Impact of Surgical Intervention on the Survival of Primary Head and Neck MALT Lymphoma.

INTRODUCTION: Primary head and neck mucosa-associated lymphoid tissue lymphoma (HN-MALT) is a rare B-cell lymphoma whose epidemiology, clinical features, and treatment strategies remain unclear. METHODS: We conducted a retrospective analysis of HN-MALT data from the Surveillance, Epidemiology, and End Results (SEER) database (2000-2021) and compared the incidence and mortality rates among age groups. The Kaplan-Meier was used to determine the overall survival (OS) and lymphoma-specific survival (LSS). Prognostic factors were identified using univariate and multivariate Cox regression models.

Clinicopathological analysis of oral-cavity B-cell neoplasms: Comparison between non-immunosuppressive and immunosuppressive patients.

No comparative analysis of the infrequently occurring B-cell neoplasms (BCNs) in the oral cavity has yet been reported in patients with non-immunosuppressiveness or immunosuppressiveness. To identify any differences in clinicopathological findings between patients with non-immunosuppressiveness and those with immunosuppressiveness, we compared the clinicopathological characteristics of oral-cavity BCNs in these patient groups. The clinical characteristics of 39 patients were analyzed from hospital records, including 24 with non-immunosuppressiveness and 15 with immunosuppressiveness. Histologi

Adverse Family Outcomes in Children With B-Cell Acute Lymphoblastic Leukemia From the Multicenter Trial Children's Oncology Group AALL0932.

BACKGROUND: Treatment for B-cell acute lymphoblastic leukemia (B-ALL) lasts >2 years and can impact families. The Children's Oncology Group multicenter therapeutic trial AALL0932 randomized patients with NCI standard-risk, average-risk B-ALL (NCI-SR AR B-ALL) to receive maintenance vincristine/dexamethasone every 4 or 12 weeks (VCR/DEX4 or VCR/DEX12, respectively). Embedded in AALL0932, this study aimed to (1) longitudinally measure adverse family outcomes overall, and (2) determine whether patients on the VCR/DEX4 regimen (vs VCR/DEX12) had fewer adverse family outcomes. METHODS: Patients wi

Non-epitheliotropic cutaneous T-cell lymphoma in a 5-week-old golden retriever puppy.

A 5-week-old male golden retriever puppy was presented with multiple, rapidly progressive skin masses. One of the masses had been present since birth; the duration of the other 2 masses was unknown. Fine-needle aspiration cytology of the lesions revealed a monomorphic population of round cells, raising suspicion for a round cell neoplasm. Two representative masses were submitted for histologic examination and immunohistochemical analysis. Histologically, the neoplasm was composed of dense, non-epitheliotropic infiltrates of atypical lymphoid cells within the dermis and subcutis. Immunophenotyp

Inhibition and Knockdown of Monocarboxylate Transporter 1 Induce L-Lactate Accumulation in T-Cell Acute Lymphoblastic Leukemia Cells.

Tumors exhibit the Warburg effect, characterized by elevated glucose metabolism through both oxidative phosphorylation and glycolysis to produce energy. To maintain intracellular pH and metabolic homeostasis, cancer cells depend on monocarboxylate transporters (MCTs) to export the glycolytic end product, l-lactate. Similar to other cancers, glucose metabolism is enhanced in T-cell acute lymphoblastic leukemia (T-ALL), supporting rapid cell proliferation. Although MCT1 expression and its sensitivity to inhibition have been reported in certain T-ALL models, the relative contributions of other co

Understanding the Immune Response Changes to Clinical Interventions for Epstein-Barr Virus Infection Prior to Lymphoma Development in Children After Organ Transplants (UNEARTH).

BACKGROUND: Post-transplant lymphoproliferative disorders (PTLD) remain a major complication of solid organ transplants (SOT), occurring in 1%-10% of patients, but in greater proportions in children. About 50%-80% of cases are Epstein-Barr Virus (EBV) associated, through partially known and still unknown mechanisms. Even in EBV seronaive SOT recipients, transplant of an SOT from an EBV seropositive donor does not always lead to peripheral blood EBV replication. Longitudinal peripheral blood EBV DNA nucleic acid testing (NAT) after SOT has not improved the individual prediction of PTLD occurren
