INTRODUCTION: Acute leukaemia is a rapidly progressive haematopoietic disorder affecting bone marrow, leading to excessive production of immature blast cells. Acute myeloid leukaemia (AML) in adults accounts for 80-90% of cases of acute leukaemia. Acute lymphoblastic leukaemia (ALL), common in childhood, comprises 12% of all leukaemia. Immunophenotyping using flow cytometry is crucial for classifying leukaemia into AML and acute lymphoblastic leukaemia (ALL) subtypes, aiding in accurate diagnosis and treatment.
OBJECTIVE: The aim of the study was to analyze the different immunophenotypic patt

Extensive genetic and epigenetic variegation has been demonstrated in many malignancies. Importantly, their interplay has the potential to contribute to disease progression and treatment resistance. To shed light on the complex relationships between these different sources of intra-tumour heterogeneity, we explored their relative contributions to the evolutionary dynamics of Acute Lymphoblastic Leukaemia (ALL) in children with Down syndrome, which has particularly poor prognosis. We quantified the tumour propagating potential of genetically distinct sub-clones using serial transplantation assa

Acute lymphoblastic leukaemia (ALL) represents the most common childhood malignancy, and emerging evidence underscores the impact of the gut microbiome (GM) on its pathogenesis. In this study, we used shotgun metagenomics to investigate the GM of 30 ALL patients at diagnosis-19 with B-ALL and 11 with T-ALL-and compared them to 176 healthy controls (HCs). When considered as a single ALL group versus HCs, clear compositional differences emerged: ALL patients exhibited higher relative abundances of Enterococcus faecium, oral commensals such as Rothia dentocariosa, and multiple opportunistic speci

BACKGROUND: Childhood acute lymphoblastic leukaemia (ALL) survival rates have improved, reaching 95%. However, 25-50% of survivors experience significant treatment-related toxicities. This study investigated late pulmonary adverse effects, including pulmonary symptoms and pulmonary function, in childhood ALL survivors compared to controls.
METHODS: This Danish national cross-sectional study (February 2019 to May 2024) recruited ALL survivors (n=295 eligible) and matched controls 1:1 for pulmonary symptom questionnaire and pulmonary function tests (PFTs): nitrogen multiple breath washout, spir

AIM: Children with acute lymphoblastic leukaemia (ALL) are at risk of metabolic and cardiovascular complications. We evaluated the development of overweight and obesity for 5 years after diagnosis in children and adolescents treated for ALL.
METHODS: The medical records of children diagnosed with ALL at one centre during 2000-2018 were assessed. Weight and height measurements were retrieved from medical records and were used to calculate age- and sex-adjusted International Obesity Task Force-Body Mass Index (ISO-BMI). ISO-BMI was determined at selected time points during treatment and up to 5

The British Osteonecrosis Study (BONES) is the first multicentre prospective study assessing bone health and vertebral fractures in patients aged 10-24 in the United Kingdom undergoing treatment for acute lymphoblastic leukaemia (ALL) or lymphoblastic lymphoma (LBL). Sixty-one patients were recruited from three tertiary centres in the United Kingdom. Dual-energy X-ray absorptiometry (DXA) scans with vertebral fracture analysis were performed within 4 weeks of diagnosis and annually for 3 years. Subjective pain assessments were performed at the same time points. Bone mineral density (assessing

BACKGROUND: Individuals with Down syndrome have an elevated risk of childhood leukaemia and are suggested to have a reduced risk of solid tumours in adulthood. However, it remains unclear which cancer subtypes contribute to this pattern and the lifetime cancer risk.
METHODS: This Swedish population-based matched cohort study investigated age- and subtype-specific cancer risks in Down syndrome. National healthcare registers were used to include 9742 individuals with Down syndrome, born in Sweden between 1930-2017. Each individual was matched by birth year, sex, and birth county to 50 compariso

Measurable residual disease (MRD) testing in acute lymphoblastic leukaemia (ALL) is considered standard of care as it is the strongest prognostic factor predicting disease outcome and is critical in therapeutic decision-making and assessing response to therapy. Across many centres, allele-specific oligonucleotide real-time quantitative polymerase chain reaction (ASO-RQPCR) has been the gold standard for assessing MRD in ALL patients for over 20 years. More recently, next-generation sequencing (NGS) technology has rapidly developed, with several platforms currently available for ALL MRD assess

Background: Chronic inflammation and the development of cancer are closely linked, with components that comprise the tumour microenvironment-including proinflammatory cytokines-exerting essential tumourigenic effects. These proinflammatory cytokines include IL-1β, which has been reported to be overexpressed in several cancers and shown to activate several signalling pathways. These pathways may involve kinases such as AKT (serine/threonine kinase) and Src (Proto-oncogene tyrosine-protein kinase), and have a broad capacity to activate nuclear factors, including NF-κB (Nuclear Factor kappa-light

UNLABELLED: This study investigates the anticancer effects of rosmarinic acid (RA) on colorectal cancer and elucidates its underlying mechanisms. A range of in vitro assays-including CCK-8, EdU incorporation, colony formation, wound healing, and Transwell invasion-demonstrated that RA significantly inhibited the proliferation, migration, and invasion of HCT-116 cells. Ferroptosis induction was confirmed by increased intracellular Fe2+, ROS, and MDA levels, decreased glutathione, and characteristic mitochondrial damage observed via transmission electron microscopy. Bioinformatics analysis and m

Hypogammaglobulinaemia is a potential complication in children with acute lymphoblastic leukaemia (ALL) receiving chemotherapy. However, real-world data on its prevalence and clinical impact remain limited. This study retrospectively reviewed 85 paediatric ALL patients completing Taiwan Pediatric Oncology Group (TPOG)-ALL-2013 maintenance therapy to investigate secondary hypogammaglobulinaemia. At diagnosis, 6% (5/85) of patients had hypogammaglobulinaemia, increasing to 49.2% (32/65) during maintenance chemotherapy. Patients with very-high-risk disease, poor cytogenetics or baseline immunoglo

BACKGROUND: Childhood cancers are an important cause of premature mortality and long-term disability worldwide, yet most global estimates stop at broad diagnostic groups. A clearer picture by subtype and its social determinants is needed to guide the World Health Organization (WHO) target of at least sixty percent survival for all children with cancer by 2030.
METHODS: In this population-based registry study, we analysed incidence and mortality data for 185 countries in 2022 using the GLOBOCAN database, and redistributed unspecified tumours according to subtype proportions derived from 670 hi

In acute lymphoblastic leukaemia (ALL), cytoplasmic CD3 (cCD3) is a defining marker for T-lineage, and CD19 plus additional B-cell marker(s) for B-lineage. We identified 23 ALL cases in which the lymphoblasts expressed both cCD3 and CD19, making lineage assignment challenging. These cases represented approximately 10% of cCD3+ ALL and expressed a median of two additional B-cell markers other than CD19, including CD79a (76%), CD22 (22%), PAX5 (57%) and CD10 (44%). Two cases were mixed for T/B-lineage ALL, both positive for BCR::ABL1 rearrangement. In the remaining 21 cases, IgH and/or IgK/L rea

Genetic findings are important independent prognostic factors in childhood acute lymphoblastic leukaemia (ALL). This study presents cytogenetic data correlated with clinical factors of 1337 patients aged 1-18 years with newly diagnosed ALL treated between 2011 and 2018 under the Polish ALL IC-BFM 2009 therapeutic protocol. Overall survival (OS) for children with B-cell ALL was 95.58% at 5 years, while OS for children with T-cell ALL was 80.43% (p < 0.001). The event-free survival (EFS) rates were 86.69% and 72.92%, respectively, and the difference was also statistically significant (p < 0.001)

Ophthalmic manifestations are a common occurrence in leukaemia cases and may result from a number of factors, including direct leukemic infiltration, haematologic abnormalities, central nervous system (CNS) involvement, or treatment-related effects. This case report discusses a paediatric patient with acute lymphoblastic leukaemia (ALL) who developed recurrent ocular symptoms, including retinitis pigmentosa-like findings, which is degeneration with pigment neopigmentation in the mid- to peripheral fundus, during remission, underscoring the necessity of continuous monitoring. Protein tyrosine phosphatase non-receptor type 13 (PTPN13) is a non-receptor protein tyrosine phosphatase with context-dependent roles as tumour suppressor or promoter. Its modular structure supports multiple molecular interactions, including a critical one with β-catenin, a regulator of the haematopoietic system. We previously identified three pathogenic PTPN13 mutations in families with acute lymphoblastic leukaemia (ALL), anaemia, and/or inherited bone marrow failure (IBMF). Our current findings reveal that these mutations impair the PTPN13-β-catenin interaction. β-catenin and PTPN13 are st
