Restless legs syndrome (RLS) is a common but often overlooked cause of sleep disturbance and discomfort during rest. The prevalence of RLS remains unknown in chronic myeloid leukemia (CML). This study explores its frequency, clinical relevance, and impact on quality of life in patients with CML. We consecutively evaluated CML patients who visited our outpatient clinic between January and August 2023. Patients with iron deficiency (ID) and healthy subjects (HS) were also included as controls. Standardized psychometric assessments were administered to CML patients, including IRLSSG Rating Scale,

This investigation proposes and analyzes a novel mathematical framework for understanding the complex tumor-immune system dynamics in Chronic Myelogenous Leukemia (CML). We adapt the established Moore and Li three-population model (naive T-cells, effector T-cells, and CML cells) by reformulating it using fractal-fractional differential operators of the Atangana–Baleanu type. This advanced approach offers a significant enhancement over standard models, as it simultaneously captures the fractal geometry of tumor structures and the non-local memory effects inherent in biological immune responses.

Cellular stresses regulate transcriptional readthrough, whereby RNA polymerase II elongates past a gene's polyadenylation cleavage site without RNA cleavage. Readthrough has been reported in several cancer types. Here, we use long-read sequencing of nascent RNA to quantify transcriptional readthrough in chronic myeloid leukemia (CML) cells and characterize early responses to the targeted therapeutic, imatinib. We show that the amount, length, and gene specificity of readthrough increase within 1 hour, before gene expression and alternative splicing alterations emerge. Notably, imatinib-depende

Chronic myeloid leukemia (CML) is a myeloproliferative neoplasm characterized by the presence of the Philadelphia chromosome and the resulting fusion proteins with abnormal tyrosine kinase activity. The treatment of CML includes allogeneic hematopoietic stem cell transplantation (allo-HSCT) and TKI drug therapy. The BCR::ABL1 T315I mutation in CML leads to resistance to first- and second-generation tyrosine kinase inhibitors (TKIs), but is sensitive to two third-generation TKIs, olverembatinib and ponatinib. Olverembatinib, an oral third-generation BCR::ABL1 TKI, has preclinical activity again

BCR-ABL tyrosine kinase inhibitors (TKIs) effectively treat chronic myeloid leukemia (CML), but drug resistance remains a challenge. Inducing erythroid differentiation in CML cells to enhance their sensitivity to imatinib is a potential approach, but the key regulators are unclear. Imatinib-induced apoptosis and erythroid differentiation in CML cells are distinct processes, suggesting specific genes are involved in differentiation. Analysis of the Gene Expression Omnibus revealed that imatinib highly upregulated dual-specificity phosphatase 21 (DUSP21). A recent study showed that DUSP21 can in

INTRODUCTION: The annual incidence of Philadelphia chromosome-positive chronic myeloid leukemia (CML) is 1 per 100,000 pregnancies. The management of CML during pregnancy poses challenges due to the teratogenicity risk associated with the BCR:ABL1 tyrosine kinase inhibitors (TKIs).
AREAS COVERED: In this review, we discuss the possible fetotoxicity of CML therapies during pregnancy, including TKIs and non TKI-based regimens and the treatment strategies for pregnant patients.
EXPERT OPINION: The diagnosis of CML during pregnancy is rare, and evidence guiding optimal management remains limited

BACKGROUND: Chronic myeloid leukemia (CML) is a myeloproliferative neoplasm caused by the t(9;22)(q34;q11.2) translocation, known as the Philadelphia chromosome (Ph+), which generates the BCR::ABL1 fusion oncogene central to CML pathogenesis. The disease accounts for approximately 15% of adult leukemias, predominantly affects males, and has a median age at diagnosis of around 57 years. CML typically presents in the chronic phase and may progress to advanced disease phases, with prognostic risk stratification guiding treatment decisions. Tyrosine kinase inhibitors (TKIs) are the cornerstone of

Chronic myeloid leukemia (CML) is effectively treated with imatinib, but a fraction of patients develop resistance. Although genetic alterations are known contributors, metabolic reprogramming has emerged as an additional driver. In this study, we have conducted untargeted metabolomics and lipidomics using high-resolution (LC-MS/MS) to profile metabolic changes in K562-derived imatinib-resistant cell lines (IR1 and IR2). Significant alterations in glycerophospholipids, sphingolipids, and fatty acyls were observed in imatinib-resistant cells when compared to wild-type cells. Our findings highli

Radotinib is a second-generation tyrosine kinase inhibitor that selectively targets BCR-ABL1 and is used for the treatment of chronic myeloid leukemia (CML). This study aimed to evaluate the clinical pharmacokinetic characteristics and the food effect of radotinib. Healthy volunteers received a single oral dose of radotinib 400 mg under fasted or fed conditions in a crossover manner, and CML patients received repeated oral doses of 300 mg twice daily. The pharmacokinetic characteristics were evaluated by a noncompartmental method, using plasma concentration-time data following a single adminis

Chronic myeloid leukemia (CML) and Philadelphia (Ph)-negative myeloproliferative neoplasms (MPN) are generally distinct clonal disorders, with the co-occurrence of BCR::ABL1 rearrangement with concomitant Ph-negative MPN rarely reported. Here we describe the largest known international cohort of Ph-negative MPN and coexisting CML providing important insights into this rare clinical scenario. We performed an international, multicenter, retrospective analysis of patients with concomitant BCR::ABL1 rearrangement and Ph-negative MPN, identifying 61 cases from 30 centers in 7 countries, over a 29-y

We prospectively evaluated whether cytotoxic T-lymphocyte (CTL) activation and T-cell receptor (TCR) Vβ clonality predict treatment-free remission (TFR) after tyrosine kinase inhibitor (TKI) cessation in chronic-phase chronic myeloid leukemia (CML). Forty-five patients with sustained deep molecular response (DMR) were enrolled (On-TKI, n = 38; Off-TKI, n = 7) and underwent one-year immuno-monitoring from consent. The primary endpoint was 12-month TFR, defined as retention of MR4. Overall, 32/45 patients (71%) maintained TFR at 12 months. Longer TKI exposure and stable DMR were associated with

Ribosome collisions act as molecular sensors of cellular stress, yet their role in disease physiology remains unclear. Here, we demonstrate that inhibition of the oncogenic kinase BCR::ABL1 in chronic myeloid leukemia (CML) cells induces ribosome collisions and activates the ribotoxic stress response (RSR). Clinical analyses revealed that CML progression from the chronic phase to the aggressive blast phase correlated with elevated expression of the RSR-initiating kinase ZAK. Although ZAK sustained CML cell proliferation by promoting AKT activity, loss of ZAK function paradoxically reduced the

Following asciminib's initial approval in Japan for chronic myeloid leukemia (CML) resistant or intolerant to previous therapy, this all-patient, postmarketing surveillance study was initiated. Predefined safety specifications, overall safety, and effectiveness were assessed for 48 weeks from asciminib initiation (safety analysis set, n = 523). The approved daily dose of asciminib (80 mg [40 mg twice daily]) was most frequently used (63.9%) and not exceeded. Discontinuations (28.5%) were primarily due to adverse events (18.2%), including disease progression. Incidences of the safety specificat

INTRODUCTION: Chronic myeloid leukemia (CML) management has been revolutionized by tyrosine kinase inhibitors (TKIs), leading to near-normal life expectancy for most patients diagnosed in the chronic phase. The European LeukemiaNet (ELN) recommendations provide the internationally recognized framework for the diagnosis, monitoring, and treatment of CML.
AREAS COVERED: This review provides a chronological overview of the evolution of ELN recommendations for CML across five updates (2006, 2009, 2013, 2020, and 2025). Relevant literature published up to January 2026 was identified through a sear

PURPOSE: Tyrosine kinase inhibitors are available in the Brazilian public health care system. However, reimbursement challenges limit access to molecular testing of BCR::ABL-1, which is essential for diagnosing and monitoring measurable residual disease in chronic myeloid leukemia (CML). This study estimated the costs associated with delivering the BCR::ABL-1 test in a tertiary academic reference care center in Brazil and simulated alternatives to increase patient access to the test.
METHODS: This is an economic analysis that estimates the costs based on resource consumption from a hospital t

Dose-optimization of dasatinib is an important strategy in chronic myeloid leukaemia (CML) to balance efficacy, safety, and cost. While Western studies have explored low-dose approaches, data from India remain limited. We conducted a comparative study of 143 newly diagnosed chronic-phase CML patients, allocated to dasatinib de-escalation (DD, n = 80) after an initial 100 mg standard dose or upfront low-dose (50 mg) dasatinib (LD, n = 63). At 3 months, a significantly higher proportion of patients in the DD cohort achieved BCR-ABL1 ≤ 10% compared to LD cohort (98.4% vs. 72.1%, p < 0.001). At 6

BACKGROUND: Chronic Myeloid Leukemia (CML) progresses through chronic, accelerated, and blast crisis phases, posing challenges for disease stratification and predicting therapeutic response. IGF2BP3 (Insulin-like Growth Factor 2 mRNA Binding Protein 3) has recently gained attention as a potential prognostic biomarker due to its role in RNA stabilization and oncogenic signaling.
METHODS: This study employed a multi-platform approach, utilizing immunohistochemistry (IHC), ELISA, qRT-PCR, and Western blotting to assess IGF2BP3 expression in 121 CML patient samples across various disease phases.

BACKGROUND: Alpha-gal syndrome (AGS), a distinct form of IgE-mediated hypersensitivity to the carbohydrate galactose-α-1,3-galactose (α-Gal), typically occurs after repeated tick bites and leads to allergic reactions after ingestion of mammalian meat. Patients with AGS may experience a broad spectrum of allergic reactions, from pruritus and urticaria to angioedema and anaphylaxis. Although classically associated with food-triggered reactions, emerging reports describe AGS-related anaphylaxis following exposure to group B blood products, including cases without accompanying gastrointestinal sym
