Correlation networks are widely used to uncover latent interaction structures within biological datasets. These networks are typically constructed using Pearson or partial correlation coefficients. Pearson correlation assesses pairwise associations without accounting for other variables, whereas partial correlation measures associations while adjusting for potential confounders. In both approaches, network construction relies on statistical significance testing. Because statistical significance depends heavily on sample size, these methods may result in under- or over-estimate the network stru

Systemic mastocytosis (SM) is a rare hematological neoplasm characterized by the proliferation and accumulation of clonal mast cells (MCs) in the skin, bone marrow, and other extracutaneous organs. Mast cell leukemia (MCL) is the rarest and more aggressive subtype of SM, with very poor prognosis and abysmal survival. Treatment strategies are poorly defined and non-standardized, ranging from symptomatic management to KIT-targeted tyrosine kinase inhibitors and, in selected cases, allogeneic stem cell transplantation, currently the only potentially curative chance for MCL patients. Here, we repo

Polycythaemia vera (PV) may rarely evolve into acute erythroid leukaemia (AEL). We herein report a known case of JAK2 p. V617F mutated PV presenting with progressive hepatosplenomegaly, abdominothoracic lymphadenopathy, ascites and various haematological derangements. Rapid diagnostic work-up revealed extensive marrow, hepatic and ascitic fluid infiltration by many CD71+, LMO2+ and p53+ proerythroblasts. Next generation sequencing showed TP53, ASXL1 and TET2 gene mutations. A chemotherapeutic regimen of ruxolitinib, azacitidine and venetoclax was initiated. A comprehensive review of the Englis

AIMS: Chronic myelomonocytic leukaemia (CMML) is a rare overlap syndrome between myelodysplastic neoplasms and myeloproliferative neoplasms. One distinct associated condition is systemic inflammatory and autoimmune disorders (SIADs).
METHODS: We conducted a retrospective cohort study of newly diagnosed CMML patients at our institution between January 2006 and December 2020, comparing those with and without SIADs. Clinical characteristics, laboratory findings, molecular profiles, survival outcomes and leukaemic transformation rates were analysed.
RESULTS: A total of 108 patients were included

Allogeneic haematopoietic stem cell transplantation (allo-HSCT) offers a curative potential for myelodysplastic syndrome (MDS) and myelodysplastic/myeloproliferative neoplasm (MDS/MPN). We examined survival trends using a nationwide database of 7175 patients who underwent their first allo-HSCT between 1998 and 2022. Overall mortality decreased over time in patients with early MDS (hazard ratio [HR] 0.79, 95% confidence interval [CI] 0.65-0.97, p = 0.026 in 2013-2017; HR 0.54, 95% CI 0.42-0.70, p < 0.001 in 2018-2022), advanced MDS (HR 0.80, 95% CI 0.71-0.90, p < 0.001 in 2013-2017; HR 0.74, 95

NUP214::ABL1 generates a fusion protein with constitutively activated tyrosine kinase activity and is present in rare T-lymphoblastic leukemia (ALL), B-ALL, and in rare cases of myelodysplastic neoplasm/acute myeloid leukemia (MDS/AML). Here, we for the first time reported NUP214::ABL1 in a case of erythroblastic sarcoma (ES), a rare extramedullary form of acute erythroid leukemia (AEL). A 55-year-old female with a reported three-year history of MDS presented with worsening lower extremity paresthesia. Magnetic resonance imaging revealed an intraspinal T2-T4 extradural mass. RATIONALE: Aplastic anemia (AA) rarely coexists with intestinal Behçet disease (iBD), and the post-partum period may further complicate its expression. Managing refractory iBD with recurrent perforation on a background of long-standing AA is challenging, and evidence on optimal treatment is limited.
PATIENT CONCERNS: A 42-year-old woman with non-severe aplastic anemia (NSAA) diagnosed in 2005 (AA duration ≈ 20 years) developed recurrent fever, oral ulcers, and penetrating ulcers of the ileocaecum and terminal ileum shortly after cesarean delivery in June 2019, with one episode of vulvar ulcer

Pembrolizumab can be associated with immune-related cutaneous adverse events, including rare purpuric eruptions that may pose a diagnostic challenge when clinicopathologic findings are inconclusive. While pembrolizumab is generally well tolerated, immune-related adverse events (irAEs) can affect multiple organ systems. Vasculitis is a rare irAE, with heterogeneous clinical presentations ranging from cutaneous purpura to life-threatening systemic involvement. We report the case of a 75-year-old male with a history of myelodysplastic/myeloproliferative neoplasm (MDS/MPN) overlap syndrome and sta

BACKGROUND/OBJECTIVES: Juvenile myelomonocytic leukemia (JMML) is a rare and very aggressive pediatric myelodysplastic/myeloproliferative neoplasm with molecular heterogeneity and constitutive activation of the RAS signaling pathway. The aim of this study was to identify the mutational landscape, driver genes, mutational signatures, functional pathways, and therapeutic targets of mutations that affect receptor tyrosine kinases (RTKs) and RAS pathways in JMML.
METHODS: We collected tumor and matched buccal swab samples from 35 patients with JMML and performed whole-exome sequencing. Variants w

OBJECTIVES: To characterize the genetic profiles of chronic myelomonocytic leukemia (CMML) cases that do not exhibit increased classical monocyte (CM) subsets in -peripheral blood.
METHODS: A total of 174 patients with CMML who were treatment‑naive for hypomethylating agents and had undergone next‑generation sequencing (NGS) testing were analyzed. Among these, 152 patients (87.4%) demonstrated increased CM subsets (>94%) by flow cytometry (CM group), while 22 patients (12.6%) did not show increased CM subsets (≤94%) (non‑CM group).
RESULTS: Patients in the CM and non‑CM groups exhibited comp

BACKGROUND: Myelodysplastic syndrome and myeloproliferative neoplasm (MDS/MPN) overlap syndromes are characterized by the presence of features of both MDS and MPN and have a poor prognosis. We conducted a phase 2 clinical trial evaluating the combination of Azacitidine (AZA) and Ruxolitinib (RUX) in MDS/MPN, with interim analysis showing an objective response rate of 57%. Herein, we report the final analysis from this clinical trial, including long-term survival data.
METHODS: This was an open-label phase 2 clinical trial including adult patients in 2 diagnostic arms: myelofibrosis (arm 1) an

Aging is associated with both a chronic pro-inflammatory state ("inflammaging") and the accumulation of somatic mutations in hematopoietic stem cells leading to clonal hematopoiesis. Clonal hematopoiesis of indeterminate potential (CHIP) is now recognized as a common phenomenon in older adults, defined by the expansion of blood cell clones bearing leukemia-associated mutations in the absence of overt malignancy. Mounting evidence links CHIP to an increased risk of cardiovascular disease (CVD) in older adults, particularly atherosclerotic disease, through inflammatory mechanisms that intersect

Several recent trials of new chronic lymphocytic leukemia (CLL) therapies used results of measurable residual disease (MRD)-testing as a primary endpoint assuming it is an accurate surrogate for progression-free survival (PFS) which is, itself, a surrogate endpoint for survival. But whether MRD-testing results accurately correlates with PFS following Bruton tyrosine kinase-inhibitor (BTK-i) therapy with or without venetoclax or for fixed duration therapy regimens is controversial. We searched PubMed, Web of Science, Embase, and Cochrane Library up to February 8, 2025, identifying 43 trials inv

Background Bone marrow examination (BME), comprising bone marrow aspiration (BMA) and bone marrow biopsy (BMB), remains vital for diagnosing hematological disorders, especially when peripheral blood findings are inconclusive. Data on marrow pathology among India's tribal population are limited. Hence, this study aimed to assess clinical indications, diagnostic utility, and association between peripheral blood smear (PBS) and bone marrow findings in the tribal population of northern Maharashtra. Methodology This retrospective, descriptive study included 60 tribal patients who underwent BMA and

IMPORTANCE: Adult T-cell leukemia/lymphoma (ATLL) is a rare but highly aggressive malignant disease caused by lifelong infection with human T-cell leukemia virus type 1 (HTLV-1), typically acquired through mother-to-child transmission. In southwestern Japan, the world's highly documented HTLV-1 endemic region, maternal HTLV-1 screening and breastfeeding counseling have reduced vertical transmission. Because of the long latency between infection and disease, reductions in ATLL burden are expected to occur over time. In the US, HTLV-1 screening is limited to blood donors, while mother-to-child t

BACKGROUND: High-risk multiple myeloma, defined by two or more high-risk cytogenetic abnormalities (HRCAs), high-risk gene expression profiling (GEP), or plasma cell leukaemia, remains associated with poor long-term outcomes despite quadruplet induction therapy. OPTIMUM showed that intensified induction, extended consolidation, and maintenance therapy after autologous stem-cell transplantation (ASCT) improved progression-free survival for patients with high-risk multiple myeloma. Here we report the 5-year follow-up of survival outcomes across molecular subgroups. METHODS: The multicentre, exte

BACKGROUND: Use of the ergot-derived dopamine receptor agonists (cabergoline and pergolide) is associated with an increased risk of cardiac valvulopathy. Pergolide was withdrawn from the US market in 2007 because of the risk of valvular heart disease, while the European Medicines Agency (EMA) required a reduction in the maximum daily dosage of cabergoline and pergolide from 6 mg/day to 3 mg/day in 2008. In Japan, the package inserts of both drugs were revised in April 2007 to request that physicians conduct periodic ultrasonic cardiography (UCG) examinations for patients taking cabergoline or

Transcription factors from the family of Signal Transducers and Activators of Transcription (STAT) are activated by numerous cytokines. Two members of this family, STAT5A and STAT5B (collectively called STAT5), have gained prominence in that they are activated by a wide variety of cytokines such as interleukins, erythropoietin, growth hormone, and prolactin. Furthermore, constitutive STAT5 activation is observed in the majority of leukemias and many solid tumors. Inactivation studies in mice as well as human mutations have provided insight into many of STAT5's functions. Disruption of cytokine

SRSF2/TET2 is a common co-mutation characterized by monocytosis across many myeloid neoplasms, including CMML. Myelodysplasia-related gene (MRG) mutations, including SRSF2, SF3B1, U2AF1, ZRSR2, ASXL1, EZH2, BCOR, and STAG2, are key contributors to leukemic transformation, though their interactions with other mutations are understudied. In this retrospective cohort study, we examined the clinical and prognostic impact of additional mutated MRGs in 412 patients with SRSF2/TET2 co-mutated neoplasms, identified from the Moffitt Next Gen Sequencing Database. The majority of patients (55%) had at le
