Non-langerhans cell histiocytosis: A growing category of emerging entities.

Histiocytoses are uncommon and comprise a diverse group of disorders of myeloid-derived cells with a macrophage or dendritic cell phenotype. They encompass a large number of subtypes, including Langerhans cell histiocytosis (LCH) and non-LCH. LCH is most common and perhaps best understood. The non-LCH in the current World Health Organization Classification of Hematolymphoid Tumors (WHO-HAEM5) includes Erdheim-Chester disease, juvenile xanthogranuloma, Rosai-Dorfman disease, ALK+ histiocytosis, and histiocytic sarcoma. Due to the rarity of non-LCH, there is a lack of awareness, knowledge and ex

A canine macrophage cell line that can be polarized provides a model for canine macrophage differentiation.

Monocyte-like cell lines are valuable models for studying macrophage biology, yet a well-characterized canine macrophage model has been lacking. The canine histiocytic sarcoma-derived cell line 030D was assessed for its macrophage polarization potential by stimulation with IFN-γ + LPS (M1) or IL-4 (M2), or no additives (M0) and characterized for morphology, immunophenotype, marker gene expression, and phagocytic capacity. 030D M1 macrophages expressed higher levels of the surface markers CD32, CD40, CD80, CD83, CD86 and MHC II than 030D M2 macrophages. qPCR analysis showed higher expression of

Rare case of intracranial histiocytic sarcoma: A case report and literature review.

BACKGROUND: Histiocytic sarcoma (HS) is a rare malignant neoplasm derived from mature histiocytes, typically affecting lymph nodes, the gastrointestinal tract, spleen, soft tissues, and skin. Intracranial occurrence is extremely rare and poses significant diagnostic challenges due to its histopathological resemblance to other aggressive tumors such as anaplastic meningioma. CASE DESCRIPTION: A 70-year-old man presented with acute right hemiplegia and was found to have two extra-axial lesions with intracerebral hemorrhage in the left frontotemporal region. Initial imaging and immunohistochemis

Multifocal jejunal histiocytic sarcoma presenting with dual intussusception.

Histiocytic sarcoma (HS) of the small intestine is exceedingly rare. We report a woman in her early 50s who presented with acute abdominal pain and was found to have two synchronous jejunal intussusceptions caused by multifocal HS. Emergency laparoscopy converted to open resection revealed multiple jejunal tumours; pathology confirmed HS with mesenteric nodal metastasis. Although 13 tumours were identified, only two lesions served as lead points for intussusception. All tumorous lesions were resected. The patient was recommended adjuvant therapy but declined and died 12 months later. This case

Histiocytic Sarcoma of Palatine Tonsil in a Pediatric Patient: A Case Report of a Rare and Aggressive Malignancy.

A 7-year-old boy with a history of recurrent tonsillitis presented with progressive left-sided throat pain, foreign body sensation, and cervical lymphadenopathy. During the examination, doctors found an ulcerated mass on the left palatine tonsil. A contrast-enhanced CT scan showed an enlarged tonsil that had uneven growth and narrowing of the opening. The tissue sample revealed large, abnormal cells with a lot of pinkish cytoplasm, irregular nuclei, and a high Ki-67 index (around 80%). Immunohistochemistry tests confirmed the cells were of histiocytic origin, strongly expressing CD68, CD163, L

Hypertrophic Osteopathy Associated With Primary Pulmonary Histiocytic Sarcoma in a Dog.

Hypertrophic osteopathy (HO) is a paraneoplastic or parainflammatory syndrome characterised by painful periosteal new bone formation and soft tissue swelling, most commonly associated with thoracic neoplasia. The pathogenesis of HO remains incompletely understood and may involve neurovascular mechanisms, altered pulmonary filtration of megakaryocytes and platelets and increased release of growth factors such as vascular endothelial growth factor (VEGF) and platelet-derived growth factor (PDGF). This report describes the first documented case of HO associated with primary pulmonary histiocytic

Disseminated histiocytic sarcoma in 2 FeLV-positive cats in Brazil.

Histiocytic sarcomas (HSs) are uncommon malignant neoplasms in cats, typically affecting older animals and not previously associated with feline leukemia virus (FeLV). Here, we describe 2 cases of disseminated HS in young FeLV-positive domestic shorthair cats that clinically and grossly mimicked lymphoma. Case 1, a 1-y-old female cat, was presented with hemiplegia and dysuria and was euthanized because of the clinical suspicion of spinal lymphoma. Case 2, a 4-y-old male cat, had hyporexia, ocular abnormalities, and bilateral renal enlargement and was euthanized because of the clinical suspicio

Clinical presentations of the most common histiocytic disorders.

BACKGROUND: Histiocytic diseases are significantly rarer than diseases derived from lymphocytic, plasmacytic, or myeloid lineages, and thus are encountered infrequently in hematology and oncology clinics. The most common form is Langerhans cell histiocytosis, which in adults has an incidence of 1-2 cases per 1 million; the others are considerably rarer, with their occurrence reported only by the number of described cases rather than through incidence or prevalence. Their rarity leads to delays in establishing an accurate diagnosis. OBJECTIVE: The group of histiocytic diseases includes seven c

SHIP1 is required for T cell surveillance of occult malignancies.

SHIP1-deficient mice develop severe, transmural Crohn's-like ileitis and die prematurely. Recent analysis of a human Crohn's disease (CD) cohort revealed a significant subset of patients have severely reduced levels of SHIP1 expression suggesting SHIP1 deficiency leads to CD in both rodents and humans. The mucosal inflammatory disease that causes the premature death of SHIP1-/- mice is a consequence of the requirement of SHIP1 expression for the survival of effector T cells in mucosal tissues. T cell specific ablation of SHIP1 expression in CD4CreSHIP1flox/flox mice also results in Crohn's dis

Extranodal Intra-abdominal Histiocytic Sarcoma Initially Diagnosed as Abdominal Tuberculosis.

Histiocytic sarcoma (HS) is a rare and aggressive hematolymphoid neoplasm that frequently presents as extranodal disease. Intra-abdominal involvement is exceedingly rare and may clinically resemble abdominal tuberculosis. We present the case of a 29-year-old woman with abdominal pain, unintentional weight loss of 15 kg (25% of her baseline body weight) over four months, and altered bowel habits. CT demonstrated a large intra-abdominal mass measuring 14 × 11 × 7 cm with internal calcifications and mass effect on adjacent structures. Initial diagnostic evaluation included image-guided fine-needl

Identification of divergent PTPN11 mutations in canine histiocytic sarcomas reveals evidence of an independent clonal origin.

BACKGROUND: Histiocytic sarcoma (HS) is a rare but highly aggressive neoplasm in humans, with limited knowledge on indicators markers and no standardized treatment. Dogs naturally develop HS, offering a powerful comparative model. Activating mutations in PTPN11, affecting two recurrent hotspots (E76 and G503), are the most frequent drivers of canine HS and are strongly associated with the disseminated form (DHS). While DHS is typically considered metastatic, we hypothesize that multiple independent tumor clones may arise within the same individual. METHODS: We investigated clonal heterogeneit

Cross-species oncolytic virotherapy in vitro: BoHV4EGFPΔTK as a novel candidate against canine osteosarcoma and histiocytic sarcoma.

Canine osteosarcoma (cOSA) and histiocytic sarcoma (cHS) are aggressive tumors characterized by poor prognoses and limited therapeutic options. Oncolytic viruses represent a promising treatment option for these tumors. Based on this hypothesis, this study aimed to evaluate direct and indirect oncolytic effects of Bovine Herpes virus type 1 (BoHV1) and a recombinant type 4 (BoHV4EGFPΔTK). Here we used cOSA, cHS and cutaneous fibroblast (FDOG) cell lines. BoHV1 and BoHV4EGFPΔTK can successfully infect cOSA and cHS cell lines with 1 multiplicity of infection (MOI). Notably, BoHV4EGFPΔTK displayed

Myeloid-lineage blasts with histiocytoid differentiation in TP53 mutated acute myeloid leukemia: a deceptive morphologic overlap with mature malignant histiocytic neoplasms.

Here, we report a case of TP53-mutated acute myeloid leukemia (AML) with monocytic differentiation that showed striking histiocytoid morphologic transformation following induction chemotherapy. Post-treatment blasts demonstrated significant morphologic overlap with mature malignant histiocytic neoplasms, particularly histiocytic sarcoma subtype, creating a potential diagnostic pitfall. Retained myeloperoxidase expression, together with the absence of cyclin D1 expression, supported a diagnosis of persistent AML rather than divergent histiocytic differentiation.

Gastric histiocytic sarcoma: a case report and literature review.

Histiocytic sarcoma is an exceptionally rare and highly aggressive neoplasm that exhibits the morphological and immunophenotypic hallmarks of mature histiocytes. It is characterized typically by extranodal presentation and a dismal clinical course, particularly in patients with disseminated disease. A history of hematolymphoid disorder can be identified in a subset of patients, suggesting transdifferentiation of a preexisting hematolymphoid neoplasm in its pathogenesis. A 38-year-old male patient presented for consultation following the detection of a gastric mass during a routine health exami

Primary Intramedullary Histiocytic Sarcoma with Leptomeningeal Dissemination: Imaging, Pathologic, and Molecular Correlation.

Central nervous system histiocytic sarcoma is exceedingly uncommon, particularly with intramedullary spinal cord involvement. We describe a 27-year-old man presenting with motor weakness and radiating pain. MRI revealed a homogeneously enhancing intramedullary mass at the C2-3 level with diffuse leptomeningeal dissemination, raising suspicion for a disseminated neoplastic process. Histopathology demonstrated pleomorphic large histiocyte-like cells with strong CD163, CD68, and lysozyme expression, confirming histiocytic sarcoma. Targeted next-generation sequencing detected a TP53 hotspot mutati

Computed tomographic features of pulmonary lesions in periarticular versus systemic histiocytic sarcomas in dogs.

INTRODUCTION: The aim of this retrospective study was to characterize and compare the computed tomographic (CT) appearance of lung lesions in periarticular histiocytic sarcomas (PAHS) and systemic histiocytic sarcomas (HS), identifying potential radiological differences. METHODS: The medical records of 45 dogs diagnosed with PAHS or systemic HS, undergoing thoracic CT imaging were reviewed. CT images were evaluated for various parameters. RESULTS: Results revealed that pulmonary lesions associated with systemic HS had larger lesion volumes, were mainly located ventrally 20/26 (77%) and were
