Autosomal recessive HOXA3 deficiency causes congenital athymia and laryngeal malformation.

BACKGROUND: Approximately 10% of patients with Severe Combined Immunodeficiency (SCID) phenotype lack a known genetic cause. In particular, the molecular basis of thymic defects is poorly understood. Homeobox (HOX) genes encode conserved transcription factors that control spatial body development. The function of human HOXA3 is undefined. OBJECTIVE: We characterize the first human with HOXA3 deficiency and investigate the role of HOXA3 in immune system development. METHODS: We performed clinical and immunological phenotyping, as well as genetic sequencing. A homozygous nonsense variant in HO

Social interdependencies: the deep evolutionary roots of morality and normativity.

The conducting airways of the respiratory system play a crucial role in filtering, humidifying, and directing air into the lungs. Among the specialized cell types within these airways, airway serous cells are notable for their secretion of watery, protein-rich fluids and enzymes, which contribute to maintaining airway surface liquid homeostasis and defending against pathogens. However, the distribution and abundance of serous cells across different species in the conducting airways remain poorly understood. In this study, we addressed this gap by investigating the spatial distribution of the a

'Teleonomes in Tatters': The Welfare Costs of Selecting Dogs for Extreme Phenotypes, and the Case for Change.

The domestic dog is the most morphologically diverse domesticated mammalian species. That diversity has arisen from deliberate, sustained artificial selection over the most recent 1% of the domestic dog's evolutionary history. However, artificial selection has pushed many individual breeds so far from the ancestral canine form that many dogs within these breeds can no longer perform the critical biological functions their bodies evolved to serve. Indeed, much of the popularity of many dog breeds is predicated on them no longer being functional dogs; rather, they are representations or caricatu

Management of Quadriceps Tendon Tear After Total Knee Arthroplasty: A Systematic Review.

OBJECTIVE: Although rare quadriceps tendon (QT) rupture after total knee arthroplasty (TKA) has debilitating complications, often resulting in significant extension deficits, limited mobility, and chronic pain. To address these complications, various surgical techniques have been developed, but a consensus on optimal management is lacking. This study aims to comprehensively review the different management techniques, outcomes, and complications in patients of TKA with QT tear. METHODS: As per PRISMA guidelines, systematic electronic search was conducted of PubMed, Embase, and Scopus databases

Glutathione peroxidase 4 (GPX4) as an oncogenic factor in human tumors: a pan-cancer analysis.

BACKGROUND: Tracheostomy is one of the most common procedures done in intensive care unit (ICU) patients. Decannulation is the weaning off from tracheostomy to maintain spontaneous respiration and/or airway protection. However, this step needs a near perfect coordination of brain, swallowing, coughing, phonation, and respiratory muscles. However, despite its perceived importance, there is no universally accepted protocol for this vital transition. In this systematic review of decannulation we focus attention to this important aspect of tracheostomy care. AIM: To compare the two methods of dec

Scope and Scan Led Airway Care: Therapist Laryngeal Visualization.

Dysphagia and impaired vocal fold motion are common complications following endotracheal intubation in critically ill patients and are associated with adverse clinical outcomes. In the ICU setting, laryngeal dysfunction complicates core physiotherapy and respiratory therapy tasks, including noninvasive therapies, cough evaluation, and airway clearance. This clinically oriented narrative synthesis outlines how bedside visualization with transnasal fiberoptic laryngoscopy (TFL) and translaryngeal ultrasound (TLU) can improve individualized airway clearance strategies and optimization of noninvas

Toward a Functional Terminology: Reconsidering "Minor Laryngeal Clefts" as Interarytenoid Dysfunction.

Much confusion surrounds the terminology used to describe minor laryngeal clefts in pediatric otolaryngology. The widely accepted Benjamin-Inglis classification defined type 1 clefts as those where the interarytenoid mucosal is at the level of the vocal folds. However, many children with aspiration and intact supraglottic mucosa respond favorably to interarytenoid augmentation. This finding has prompted the use of terms to suggest a deficient, and clinically significant, anatomic anomaly including "deep interarytenoid notch," "deep interarytenoid groove," "low interarytenoid height," and "type

Laryngeal Injuries Following General Endotracheal Anesthesia: A Case Series.

OBJECTIVE: Endotracheal intubation can cause laryngeal injury, with clinical presentation ranging from transient hoarseness to persistent vocal cord immobility. This study examines the incidence, clinical features, and outcomes of post-intubation laryngeal injuries following procedures traditionally considered low risk for such complications. METHODS: We conducted a retrospective review of adult patients who underwent tracheal intubation for surgical procedures between May 1, 2018, and December 31, 2024, and subsequently presented with postoperative laryngeal injury-related symptoms. Patients

An Unexpected Finding in an Infant's Feeding Difficulty: A Diagnostic Challenge.

Feeding difficulties represent a common yet clinically nonspecific presentation in infants, characterized by diverse etiologies. Accurate diagnosis is often challenged by limited verbal expression in infants and potentially imprecise medical histories provided by caregivers, contributing to persistent management challenges in certain cases. We report a case of feeding difficulty in an infant by an uncommon etiology. Comprehensive evaluation ultimately revealed a soft foreign body lodged in the pharyngolaryngeal region. Although infrequent, soft foreign bodies should be considered a plausible,

Selective KIT-inhibition Supports Mast Cells as Key Drivers of Allergic Asthma Pathogenesis.

BACKGROUND: Asthma is a chronic respiratory disease characterized by airway inflammation, airway hyperresponsiveness (AHR) and airway remodeling. Mast cell numbers are increased in asthmatic airways and contribute to bronchoconstriction and inflammation; yet their role in asthma development remains elusive. OBJECTIVE: To investigate mast cell contributions to allergic asthma by pharmacologically targeting KIT, a central regulator of mast cell survival and activation, using the selective inhibitor THB001. METHODS: THB001 effects were assessed in human lung mast cells (HLMCs), isolated human b

Clinical and Functional Outcomes of Bone Defect Reconstruction Using the Masquelet Technique: Experience with Rare Talus and Metatarsal Cases.

BACKGROUND: Segmental bone defects remain a major challenge in orthopedic surgery. The Masquelet's induced membrane technique (MIMT) is a two-stage reconstructive approach used for limb salvage in complex defects. This study evaluated radiographic healing and patient-reported outcomes after MIMT in a single tertiary center, including rare talus and metatarsal applications. METHODS: We retrospectively reviewed 14 patients (10 males, 4 females) treated with MIMT for post-traumatic or osteomyelitis-related lower-extremity bone defects (tibia, femur, talus, metatarsal) between 2010 and 2023. Shifting growth-climate limitations in Canadian forests under recent climate change.

INTRODUCTION: Although the etiology of osteoporosis is different between men and women, the underlying pathophysiological mechanism is similar, namely an absolute or relative increase in bone resorption, leading to progressive bone loss. Transforming growth factor (TGF)-beta1 is a growth factor in human bone, which is produced by osteoblasts, and which has various effects on osteoclasts and osteoblasts. The aim of our study was to determine serum TGF-beta1 levels in male patients with idiopathic osteoporosis. METHODS: Twenty five males with idiopathic osteoporosis and 25 age-matched controls

The bovine respiratory syncytial virus (BRSV) known as Bovine orthopneumovirus according to the international classification is one of the most important etiological agents of respiratory diseases in calves. At present, rapid and reliable methods to detect and measure the concentrations of this pathogen are needed. The objectives of the survey are developing the real-time polymerase chain reaction (PCR) to identify and quantify the BRSV RNA and, based on it, determining the number of the virus genomes in the respiratory tract of sick animals during the disease outbreaks.
