Molecular biomarkers associated with environmental naphthalene exposure in the respiratory system.

Naphthalene (NPT) is a polycyclic aromatic hydrocarbon (PAH) contaminant found in tobacco smoke and various consumer products, causing widespread toxic exposure to human. NPT-induced cell toxicity depends on a metabolic activation step mediated by cytochrome P450 (CYP) and primarily affects non-ciliated club cells of the conducting airways. Here, we characterize the histological features and gene expression profiles of airway epithelial cells following NPT-induced injury and identify early response genes (ERGs) that are sensitive to NPT in respiratory organs. NPT results in the detachment and

Exosomes derived from avian influenza virus-infected chickens modulate host immune responses.

Exosomes are emerging as key mediators of host-pathogen interactions, particularly as carriers of viral components during infection. This study aims to examine the immunomodulatory effects of serum-derived exosomes from Brown Leghorn chickens infected with low pathogenic avian influenza virus (LPAIV) or highly pathogenic avian influenza virus (HPAIV). These exosomes (CTRL-EXO [noninfected], LPAIV-EXO, and HPAIV-EXO) were intramuscularly injected into naïve chickens, after which tissues and serum were collected. Cytokine gene expression in immune-related tissues (lung, spleen, and trachea) was

Technical Approach to Repair of Tracheogastric Conduit Fistula Following Minimally Invasive Esophagectomy: A Case Report.

BACKGROUND Tracheogastric conduit fistula (TGCF) is a rare but life-threatening complication of esophagectomy, particularly in high-risk patients with comorbidities or prior chemoradiotherapy. It typically develops in the setting of anastomotic leakage, ischemia, or infection. There is no standardized treatment, and outcomes vary depending on timing and surgical approach. We report a delayed TGCF after thoracolaparoscopic esophagectomy and describe the operative technique and key perioperative considerations. CASE REPORT A 78-year-old man with diabetes, hypertension, and a heavy smoking histor

Enhanced MSC spheroid adhesion on 3D-printed leaf-stacked scaffolds for functional tracheal regeneration.

Reconstruction of segmental tracheal defects using three-dimensional (3D)-printed scaffolds remains a formidable challenge. While polycaprolactone (PCL) is widely utilized for its mechanical integrity, its inherent hydrophobicity limits cellular adhesion and tissue integration. In this study, we developed a 3D-printed PCL tracheal scaffold featuring a Leaf-Stacked Structure (LSS) and evaluated a spatially organized Mesenchymal Stem Cell (MSC) delivery strategy for functional regeneration. MSC spheroids were employed to overcome the limitations of monolayer cells, as their 3D configuration crea

IbinA and IbinB regulate the Toll pathway-mediated immune response in Drosophila melanogaster.

BACKGROUND: To combat infection, an immune system needs to be promptly activated but tightly controlled to avoid destruction of host tissues. IbinA and IbinB are related short peptides with robust expression upon microbial challenge in Drosophila melanogaster. RESULTS: Ibin genes are ubiquitously present in flies of the Drosophila subgenus Sophophora, replacing the likely evolutionarily older, related gene, Mibin, which is found across a much wider range of cyclorrhaphan flies and is also upregulated following infection. We observed no direct bactericidal or bacteriostatic activity for IbinA

Quantitative Tissue Proteomics Reveals Protein Signatures Associated with SARS-CoV-2 Variant Infection in Hamsters.

Since its emergence in 2019, circulating SARS-CoV-2 has been dominated by waves of genetically distinct variants with varying pathogenicity. Understanding the multidimensional responses to SARS-CoV-2 infection and their associations with pathogenesis is critical for developing therapies to prevent severe illness and death. Here, we applied quantitative proteome and phosphoproteome analyses to compare host responses to infections with an ancestral variant (WA-1/2020), a Delta variant (B.1.617.2), and an Omicron variant (BA.1) of SARS-CoV-2 in Syrian golden hamster tissues at 5 days postinfectio

TAPI-1 Combined With Silicone Stents Alleviated Severe Traumatic Tracheal Stenosis via the ADAM17/TGF-β1 Pathway.

BACKGROUND: Traumatic tracheal stenosis is characterized by airway granular proliferation after trauma, which may lead to asphyxy. Abnormal activation of the ADAM17/TGF-β1 pathway may trigger excessive repair and airway fibrosis. TAPI-1 is an effective inhibitor of ADAM17, which blocks the "extracellular cleavage release" of inflammatory factors (such as TNF-α) and proliferation signals (such as EGFR ligand TGF-α) by specifically inhibiting ADAM17 (tumor necrosis factor converting enzyme). Silicone stents are considered an important method for the treatment of airway stenosis. This study aimed

Phage-Mediated Presentation of a Conserved HA2 Epitope From Influenza A Virus Elicits Significant IgY Antibody Responses in Broiler Chickens.

Peptides alone often exhibit limited immunogenicity, necessitating the development of effective antigen delivery systems to facilitate recognition and presentation, ultimately eliciting robust immune responses and activating T and B lymphocytes. Filamentous bacteriophages, such as M13, are recognized as efficient platforms for peptide expression and presentation via their capsid surfaces. The conserved amino acid sequence HA2 1-9 (GLFGAIAGF) from the haemagglutinin (HA) transmembrane protein of Influenza A virus (IAV) has been identified as a promising immunogen for eliciting broad-spectrum im

A Nanovaccine Based on Cationic Lentinan Adjuvant Enhances Protective Immunity against H1N1 and H9N2 Influenza Virus Infection.

Influenza viruses are highly prevalent and continuously evolving respiratory pathogens that pose a significant threat to global public health and the poultry industry. Immunization remains the primary approach to preventing infection and reducing the risk of an influenza pandemic. Nanoparticles not only serve as delivery vehicles for antigens but also enhance antigen-specific immune responses. Polysaccharide nanoparticles not only amplify the immunostimulatory capacity of polysaccharides but also exhibit superior adjuvant effects. In this study, we investigated the effect of cationic Lentinan

Delta large-channel endoscopy versus unilateral biportal endoscopy for cervicothoracic junction disc herniation: a prospective randomized controlled trial.

BACKGROUND: Due to the obscuration by the sternum, trachea, and major blood vessels, the anterior surgical approach for cervicothoracic junction disc herniation (CTJDH) is extremely challenging. Delta large-channel endoscopy and unilateral biportal endoscopy (UBE) provide posterior decompression and have demonstrated promising clinical potential. This prospective randomized controlled study aims to compare the clinical outcomes of Delta and UBE endoscopic surgeries. METHODS: A total of 63 patients were randomly assigned to either the Delta group (n = 32) or the UBE group (n = 31). The primary

Primary pulmonary hypoplasia masquerading as pneumonia in a preterm neonate and creating a diagnostic challenge: a case report.

BACKGROUND: Pulmonary hypoplasia is a rare congenital lung disorder characterized by the incomplete development of lung tissue. This condition can be classified as primary or secondary to other congenital anomalies, with primary pulmonary hypoplasia being considerably rarer than the secondary form. Severe cases often manifest as respiratory failure in the immediate newborn period. While secondary pulmonary hypoplasia is often antenatally detected, primary cases-such as ours-may evade prenatal imaging, presenting as refractory neonatal respiratory distress. Secondary causes, such as oligohydram

Girdin controls the pace of 3D tracheal cell intercalation by coupling adherens junctions to the actin cytoskeleton in Drosophila.

Morphogenesis is orchestrated through coordinated cell movements, including cell intercalation, which drives extensive changes in cell shape and position. This process requires precise regulation of interactions between adherens junctions (AJs) and the cortical actin network to generate the necessary mechanical forces. Although the Myosin molecular motor II plays a central role in generating morphogenetic forces, it is dispensable for processes such as dorsal branch morphogenesis in the Drosophila tracheal system, where coordinated cell migration and three-dimensional intercalation rely on alt

Architecture of the insect tracheal system driven by spatially varying limitation of oxygen and carbon dioxide transport.

Insects transport respiratory gases through a system of air-filled tubes (tracheae) that branch extensively to reach individual cells. A century of research has focused primarily on how tracheal systems deliver oxygen, often overlooking the complementary challenge of removing carbon dioxide. Here, we develop and analyse a model of simultaneous O₂ and CO₂ transport, which we parametrize with morphological and metabolic data. The model reveals a fundamental asymmetry: oxygen transport is most limited by the tissue gap between tracheoles and mitochondria; carbon dioxide transport, by contrast, is

Anesthetic Management of Penetrating Tracheal Injury in a 3-Year-Old Child: A Case Report.

Penetrating tracheal injuries in children are uncommon and present significant diagnostic and airway challenges. We report the case of a 3-year-old boy who sustained an accidental penetrating neck trauma and was taken up for tracheal repair. Initial radiological evaluation did not reveal any tracheal/bronchial injury, but there was extensive pneumomediastinum. The child exhibited signs of airway compromise along with a deep penetrating injury on the anterior neck, extensive subcutaneous and musculofascial emphysema in the neck and anterior chest wall. Anesthetic management required careful ind

Tendon decellularized bio-adhesives interpenetrated by genipin crosslinked chitosan with ROS scavenging capability and hemocompatibility.

Decellularized extracellular matrix (dECM) has been widely used in tissue engineering and regenerative medicine due to its excellent biocompatibility and biological activities. In this work, the dECM was obtained by treating porcine Achilles tendon tissue via a combination of chemical, physical, and biological approaches. The obtained dECM was interpenetrated with genipin crosslinked quaternary ammonium chitosan (QCS) network to fabricate the composite genipin/QCS/dECM (QGD) hydrogels. The prepared QGD hydrogels exhibited an interconnected microstructure and a suitable water-absorption capacit

Adult-onset intratracheal xanthogranuloma presented with uncontrolled asthma.

Adult-onset xanthogranuloma (AXG) is a rare form of non-Langerhans cell histiocytosis, with extracutaneous involvement, particularly within the trachea, being exceptionally uncommon. A review of the literature reveals only cases of tracheal juvenile xanthogranuloma and a limited number of laryngeal AXG presentations. We report a unique case of intratracheal AXG in a young adult woman who presented with uncontrolled asthma, without evidence of systemic or cutaneous involvement.

Robotic-assisted thoracic surgery for resectable lung cancer: efficacy, safety and surgical approaches - an umbrella review of meta-analyses.

Lung cancer is still a tumor type with high incidence in the world. Although there are many treatment options for lung cancer, surgical resection is widely used for I-IIIA lung cancer. Video assisted thoracic surgery (VATS) has been proved to have significant intraoperative and postoperative and efficacy and safety advantages compared with traditional thoracotomy. Furthermore, current robotic assisted thoracic surgery (RATS) has created a new development chapter for minimally invasive lung surgery. RATS provides a series of advantages due to its advanced technology, such as three-dimensional v

Inhibition of Rho-kinase restores impaired relaxation of airway smooth muscle in rat pups exposed to neonatal hyperoxia.

Neonatal hyperoxia is a key contributor to bronchopulmonary dysplasia (BPD) which is characterized by airway hyperreactivity due to increased contraction and impaired relaxation of airway smooth muscle (ASM). This study investigated whether inhibition of the Rho/Rho-kinase signaling pathway restored tracheal smooth muscle (TSM) relaxation and reactivated the nitric oxide-guanosine 3',5'-cyclic monophosphate (NO-cGMP) pathway in neonatal rats exposed to hyperoxia. Newborn rats (P4) were exposed to either ambient air (AA; n = 61) or hyperoxia (FiO2 >95%; n = 58) for 7 days. The effects of Rho-ki

Case Report: Emergency management of difficult airway in a thyroid cancer patient with undiagnosed tracheal diverticulum preoperatively and literature review.

Tracheal diverticulum, a rare chronic airway pathology characterized by cystic outpouchings from the trachea or main bronchi, has scant epidemiological documentation. We present a thyroid cancer case with known airway stenosis but an undiagnosed tracheal diverticulum. Perioperatively, repeated displacement of the endotracheal tube into the undiagnosed diverticulum caused critical intubation failure and complicated tracheostomy. Under fiberoptic bronchoscopic guidance, the tracheal tube was successfully advanced past a right subglottic diverticular orifice into the left tracheal lumen, achievin
