Rapid and visual detection of Mycoplasma pneumoniae using a novel ERA-CRISPR/Cas12a-based lateral flow assay.

OBJECTIVE: Mycoplasma pneumoniae (MP) is a leading cause of community-acquired pneumonia in children. Conventional detection methods often lack the speed or accessibility required for point-of-care testing (POCT). This study aims to develop a rapid, highly sensitive, and instrument-free diagnostic platform integrating Enzymatic Rapid Amplification (ERA), CRISPR/Cas12a cleavage, and Lateral Flow Assay (LFA) for visual MP detection. METHODS: Specific primers targeting the conserved P1 adhesin gene of MP were screened for ERA efficiency. A CRISPR/Cas12a system was designed to specifically recogn

First endogenous case of Blastomyces dermatitidis ARDS in an immunocompetent man in Newfoundland and Labrador: Case report and mini-review.

BACKGROUND: Blastomycosis is a fungal infection that can be localized to the lungs or disseminated to the skin, bone, and the central nervous system. Blastomycosis is endemic in parts of North America, specifically the Great Lakes region; the endemicity of the pathogen is a crucial diagnostic clue. CASE PRESENTATION: An adult immunocompetent man with no travel history outside of Newfoundland presented to a community centre with respiratory symptoms and several months of constitutional symptoms. He had several new papulo-ulcerated skin lesions and a single mucosal ulcer on the dorsal posterior

Susceptibility Weighted and Perfusion Imaging for Differentiating Recurrence and Radiation Necrosis in Post-Operative Malignant Brain Tumors: A Retrospective Cohort Study.

BACKGROUND: Brain tumors present a challenge in oncology, necessitating comprehensive treatment strategies. Despite treatment, recurrence rates, especially in malignant tumors, remain high. Novel therapies have emerged, but treatment-related effects such as pseudo-progression (PsP) and radiation necrosis (RN) complicate diagnosis. Imaging techniques such as perfusion imaging and susceptibility-weighted imaging (SWI) show promise in distinguishing tumor progression from treatment effects. This retrospective study evaluated the effectiveness of these modalities in detecting brain tumours, their

ALG-000184 (pevifoscorvir sodium) monotherapy in participants with chronic HBV infection: a phase 1, multicentre, randomised, dose escalation trial.

BACKGROUND: ALG-000184 (pevifoscorvir sodium) is a prodrug of the class-E (empty) capsid assembly modulator (CAM), ALG-001075. CAM-Es prevent hepatitis B virus (HBV) replication by inhibiting capsid assembly and pregenomic RNA encapsidation and preventing covalently closed circular DNA (cccDNA) establishment. This part of the study aimed to assess the safety, tolerability, pharmacokinetics and short-term antiviral activity of ALG-000184 in participants with HBeAg-negative and HBeAg-positive chronic HBV infection to determine the subsequent development pathway for this novel therapeutic. Aerosolized adenoviral IL4/IL10 delivery alleviates LPS-induced acute lung injury.

Acute lung injury (ALI) and acute respiratory distress syndrome (ARDS) are severe inflammatory lung diseases with high morbidity and mortality, lacking specific treatments. Inhalation offers direct access to the damaged lung epithelium, making tracheal gene delivery a promising approach. However, challenges such as low efficiency, off-target effects, and repeated dosing limit its effectiveness. In this study, we developed an inhalable recombinant adenoviral vector (Ad-IL4/10) carrying dual reporter genes to deliver interleukin-10 (IL10) and interleukin-4 (IL4) to the lungs, enabling efficient

Trends in Triple-Site Testing for Chlamydia and Gonorrhea in New York City: A Cross-Sectional Study Using Health Information Exchange Data.

BACKGROUND: Sexually transmitted infections pose a threat to public health and continue to rise in New York City despite reductions in other parts of the United States. Chlamydia and gonorrhea can infect multiple anatomic sites, including the urogenital tract, pharynx, and rectum. Guidelines recommend multisite testing for select populations, yet multisite testing is not routinely performed, limiting our understanding of trends in positivity and missed infections. METHODS: We conducted a retrospective cross-sectional study of patients who underwent triple-site testing for chlamydia and gonorr

Lower airway dysbiosis in nontuberculous mycobacteria-positive bronchiectasis is associated with neutrophil extracellular trap-predominant severe phenotypes.

RATIONALE: The discoveries of neutrophilic inflammation and Pseudomonas-dominant pulmonary dysbiosis have helped pave the way for host-directed therapy in bronchiectasis. Substantial knowledge gaps still remain about the interplay between neutrophilic signatures and microbes in nontuberculous mycobacterial lung disease (NTM-LD), a phenotypically diverse lung infection that is increasingly prevalent in the United States and other parts of the world. OBJECTIVES: To evaluate the lower airway microbiota and neutrophilic traits in NTM-negative (NTM-) and NTM-positive (NTM+) bronchiectasis. METHOD

Adolescent health across Asia Pacific, 2000-23: a systematic analysis for the Global Burden of Disease Study 2023.

BACKGROUND: The Asia Pacific region is home to more than half of the world's 1·93 billion adolescents (aged 10-24 years). Addressing adolescent health in this region is of global importance, but to date a systematic analysis of key contributors to disease in adolescents has not been done, which is a barrier to responsive action. This systematic analysis of the Global Burden of Diseases, Injuries, and Risk Factors Study (GBD) 2023 aims to provide a comprehensive assessment of adolescent health across the Asia Pacific region, at both the subregional and national levels, encompassing burden of di

Prevalence of Erysipelothrix rhusiopathiae in tonsils of domestic pigs and wild boars in Sweden.

Erysipelothrix rhusiopathiae (ER) causes erysipelas in multiple animal species and may persist in the environment or be carried asymptomatically. It is estimated that 30-50% of apparently healthy or convalescent pigs harbour ER in their tonsils and other lymphoid tissues. This study aimed to determine the prevalence of ER in the tonsils of healthy Swedish fattening pigs and wild boars. Tonsils were collected from 200 fattening pigs at slaughter from ten abattoirs across Sweden in 2017, with one pig per herd sampled. Wild boars (n = 180) were sampled during hunting, primarily in Östergötland Co

New Findings in Developmental Studies of Laryngeal Mound With a Reference to Its Attached Structures in Japanese Quail (Coturnix coturnix Japonica).

The laryngeal mound (LM) formed the caudal part of the pharyngeal floor, which varied in position, shape, and length at different ages. This work aimed to study the morphogenesis of the LM in the embryonic and post hatching periods grossly, histologically, and by scanning electron microscopy using forty-eight Japanese quails. The LM primordia appeared on the 8th day of incubation as a raised elevation carried on a deep median symmetrical sulcus (glottis primordium). As a result of rapid differential LM parts growth, LM took different shapes with advanced ages, finally ending in a heart shape.

Automated system for diagnosing pulmonary fibrosis using crackle analysis in recorded lung sounds based on iterative envelope mean fractal dimension filter.

Patients with pulmonary fibrosis (PF) often experience long waits before getting a correct diagnosis, and this delay in reaching specialized care is associated with increased mortality, regardless of the severity of the disease. Early diagnosis and timely treatment of PF can potentially extend life expectancy and maintain a better quality of life. Crackles present in the recorded lung sounds may be crucial for the early diagnosis of PF. This paper describes an automated system for differentiating lung sounds related to PF from other pathological lung conditions using the average number of crac

Redox-Modulating Capacity and Effect of Ethyl Acetate Roots and Aerial Parts Extracts from Geum urbanum L. on the Phenotype Inhibition of the Pseudomonas aeruginosa Las/RhI Quorum Sensing System.

Pseudomonas aeruginosa is an opportunistic pathogen that causes nosocomial infections of the urinary tract, upper respiratory tract, gastrointestinal tract, central nervous system, etc. It is possible to develop bacteremia and sepsis in immunocompromised patients. A major problem in treatment is the development of antibiotic resistance. Therefore, new preparations of natural origin are sought, such as plant extracts, which are phytocomplexes and to which it is practically impossible to develop resistance. Geum urbanum L. (Rosacea) is a perennial herb known for many biological properties. This
