Chemotactic factors regulate the recruitment of monocytes-macrophages to inflammatory sites and neoplastic tissues. The purpose of this study was to determine whether MCAF also influences the activation of C3H/HeN macrophages to become tumoricidal. From a large series of experiments involving transfer of high molecular weight total genomic DNA from highly metastatic human and mouse tumour cell lines to other mouse tumour cell lines we have derived a few cell lines with greatly augmented metastatic properties. Patterns of cancer incidence within five population density quintiles in New York State, exclusive of New York City, were investigated between 1978 and 1982. Human skin mast cells (HSMC) in situ were examined by electron microscopy of surgical biopsy specimens obtained from a wide variety of circumstances. From these studies, it is apparent that the general ultrastructural morphology of normal HSMC is similar to that of human mast cells from other sites, except that the most prevalent granule pattern is that of crystal granules. The effect of bacterial lipopolysaccharide (LPS) on macrophage receptors for tumor necrosis factor/cachectin (TNF-R) was studied. Using data from the population-based Danish Cancer Registry, the relative risk (RR) of second primary cancer was assessed among 24,970 women with invasive cervical cancer (1943-1982) and 19,470 women with carcinoma in situ of the cervix. The ME491 gene was molecularly cloned by means of DNA-mediated gene transfer followed by screening a lambda genomic library with human repetitive Alu sequences as a probe. Ninety-seven fresh explants of human tumours have been cultured in vitro in an attempt to predict their sensitivity to subsequent cytotoxic chemotherapy. Only 3/26 solid ovarian tumours were cultured successfully although 12 of the 23 which failed to grow proved later to have benign histology.
