Organ Dosimetry in Radionuclide Therapy: Biokinetics of 131 I and Dose with a Healthy Thyroid and after Thyroidectomy Using MCNP6.

An accurate internal dosimetry is important in 131 I radionuclide therapy. In patients who have undergone a thyroidectomy, organs other than the thyroid absorb higher doses as the radionuclide is transported through the body because most of the thyroid gland is removed, and it cannot retain the 131 I. The primary objective of this study is to compare the absorbed doses in at-risk organs due to intake of 131 I after thyroidectomy vs. a healthy thyroid scenario. The biokinetic model of internal dosimetry for healthy thyroid as proposed by the International Commission on Radiological Protection (

Feasibility study of a newly developed solid gel pad containing tamarind seed gum for diagnostic ultrasonography in human subjects.

Ultrasonography-based diagnosis is increasingly used owing to its reliability. However, the conventional ultrasound gel has limitations of patient discomfort and cost. This study evaluated the clinical utility of a newly developed gel pad for ultrasound diagnosis composed of tamarind seed gum (0.1–5.0 wt%), polyhydric alcohol (25.0–70.0 wt%), and water (30.0–70.0 wt%). Ultrasound imaging was performed in four healthy volunteers using our new solid gel and the conventional liquid gel. Linear, convex, and sector probes were used for imaging of the common carotid artery and thyroid gland, liver,

Androgen production in adrenocortical H295R cells is regulated by thyroid hormone T3 without reciprocal thyroid axis modulation in pediatric CAH.

Thyroid hormones (THs) are critical regulators of human development, cellular differentiation, and metabolism. While their systemic effects are well established, their role in adrenal androgen production remains poorly defined. Moreover, potential feedback regulation of the hypothalamic-pituitary-thyroid (HPT) axis by adrenal androgens has not been thoroughly investigated. This study aimed to clarify the regulatory effects of THs on adrenal androgens and to investigate potential feedback mechanisms in patients with congenital adrenal hyperplasia (CAH). In an in-vitro experiment, treatment with

Differentiated high-grade thyroid carcinoma (DHGTC): clinicopathological analysis of a new entity in a chilean center.

INTRODUCTION: High-grade differentiated thyroid carcinoma (DHGTC) is a rare cancer with an intermediate behavior between well-differentiated thyroid carcinoma (WDTC) and anaplastic thyroid carcinoma. In 2022, the WHO defined DHGTC by the presence of ≥ 5 mitoses per 2 mm² and/or tumor necrosis in tumors that retain the morphological and immunohistochemical features of carcinomas derived from follicular thyroid cells. OBJECTIVES: To report the clinicopathological characteristics and outcomes of patients with DHGTC treated at a national center. DESIGN: Retrospective observational study. METHODS:

Integrative multi-omics analyses identify key genes and elucidate bidirectional regulatory mechanisms in thyroid dysfunction.

OBJECTIVE: Hyperthyroidism and hypothyroidism are globally prevalent endocrine disorders, with their pathogenesis involving multifactorial mechanisms including genetics, immunity, and metabolism. Although genome-wide association studies (GWAS) have identified risk genes such as PDE8B, critical gaps remain in the annotation of causal variants in non-coding regions, characterization of tissue-specific regulatory networks, and understanding of ethnic heterogeneity. This study aimed to systematically identify genes associated with hyperthyroidism and hypothyroidism and unravel their underlying mol

Investigating the impact of estetrol and 17α-ethinylestradiol on thyroid hormonal signaling during zebrafish (Danio rerio) metamorphosis.

The thyroid axis orchestrates key biological functions in fish, including metamorphosis. Disruption of thyroid signaling by endocrine disrupting compounds (EDCs), particularly estrogens, remains understudied during this sensitive period. This study compared the thyroidal effects of two estrogens: 17α-ethinylestradiol (EE2), a synthetic compound widely used in combined oral contraceptives (COCs), and estetrol (E4), a natural estrogen produced only during pregnancy and recently introduced as the estrogenic component of a new COC. Zebrafish (Danio rerio) were exposed to EE2 and E4 at concentratio

Molecular testing and other metrics in thyroid cytology as quality-assurance measures in evaluating variation among pathologists in the diagnosis of atypia of undetermined significance.

BACKGROUND: Atypia of undetermined significance (AUS) is a subjective diagnosis with reported interinstitutional rates ranging from 1% to 20%. The Bethesda System for Reporting Thyroid Cytology (TBS) endorsed ≤10% as an achievable target; however, lowering AUS rates without informed, adjusted diagnostic thresholds can compromise the sensitivity of thyroid cytology. The authors conducted this study to identify quality-assurance measures that can assess individual pathologist's metrics compared with laboratory averages and published benchmarks and can provide feedback to pathologists on the caus

A Randomized Trial Evaluating Intraoperative Ischemic Preconditioning of Parathyroid Glands During Total Thyroidectomy: A Signal for Earlier Parathyroid Function Recovery.

BACKGROUND: Ischemic preconditioning of parathyroid glands (IPCP) is biologically plausible but clinical evidence is limited. In this single-center randomized trial (ChiCTR2000039788), we compared IPCP versus control during total thyroidectomy with central neck dissection. METHODS: A total of 135 patients with differentiated thyroid carcinoma were analyzed (IPCP, n = 67; control, n = 68). IPCP consists of three cycles of 60-second occlusion followed by 60-second reperfusion of the ipsilateral superior and inferior thyroid arteries. The baseline characteristics, postoperative hypoparathyroidis

Associations of Multimarkers of Metabolic Malnutrition and Inflammation With All-Cause Mortality and Their Interplay With Thyroid Function.

INTRODUCTION: The metabolic vulnerability index (MVX)-a composite biomarker reflecting metabolic malnutrition and inflammation-has been linked to increased mortality risk in populations with cardiovascular disease. Thyroid function, a key regulator of metabolism and inflammation, may confound or modify this relationship, but evidence in the general population is limited. OBJECTIVES: To evaluate the interplay between MVX and its subcomponents (inflammation vulnerability index, IVX and metabolic malnutrition index, MMX), thyroid function, and mortality risk in the general population. METHODS:

Hypothalamic-Pituitary-Thyroid and Adrenal Axis Modulation in Response to Fetal Porcine Reproductive and Respiratory Virus Infection.

Porcine reproductive and respiratory virus (PRRSV) has been shown to cause a substantial decrease in circulating thyroid hormone levels, consistent with nonthyroidal illness syndrome (NTIS) observed in response to other nidoviruses. This effect is particularly profound following fetal infection, whereby the ability to decrease circulating triiodothyronine is associated with resilience following late-gestation infection. We have previously shown that the thyroidal response to fetal infection is associated with peripheral changes in deiodinase activity, but the role of the central regulatory axi

Read-across prediction of nongenotoxic thyroid carcinogenicity of pesticides in rats using mechanism-based in vitro assay data.

Developing alternatives to rat carcinogenicity studies remains a major challenge. In our previous study, we found that while structural similarity provided a basis for grouping carcinogens, integrating mechanistically relevant in vitro cytotoxicity data substantially increased the accuracy of read-across predictions for tumors in the nasal cavity, stomach, and bladder/urethra. In this study, using the same dataset of 126 pesticides, we investigated whether a toxicity mechanism-based read-across approach could be applied to predict thyroid carcinogenicity. We evaluated nuclear receptor activati

Micro/nanoplastics induce thyroid follicular cell pyroptosis to trigger thyrotoxicity by activating NF-κB signaling.

BACKGROUND: Micro/nanoplastics (MNP) have emerged as ubiquitous environmental contaminants with demonstrated bioaccumulation potential in organisms through multiple exposure pathways, posing substantial health risks globally. While mounting evidence indicates that MNP exposure adversely affects various organ systems including the nervous, reproductive, and digestive systems, the specific mechanisms underlying MNP-induced thyrotoxicity remain enigmatic. METHODS: 4-week-old male C57BL/6 mice were administered microplastics (MP, 5 μm) or nanoplastics (NP, 50 nm) via intragastric gavage at 30 mg/

Rare RET Variants in a Patient With MEN2A and Multiple Follicular-Derived Thyroid Tumors: A Case Report and Review of the Literature.

Multiple endocrine neoplasia (MEN) syndromes are a group of rare autosomal dominant hereditary disorders that are classified into 5 distinct types. MEN2A, the most common type of MEN2, is associated with mutations and variants of RET, which are continually being updated. However, studies regarding RET mutations/variants related to MEN2A in the Chinese population are scarce, and patients involving MEN2A along with other differentiated thyroid tumors are rarer. We report a 54-year-old Chinese woman diagnosed with MEN2A and multiple follicular-derived thyroid tumors. Ultrasonography and computed

Real-World Outcomes Following the Implementation of ACR TI-RADS for Evaluation of Thyroid Nodules.

OBJECTIVES: Detection of thyroid nodules is increasing. The American College of Radiology Thyroid Image Reporting and Data System (ACR TI-RADS) purports to decrease unnecessary thyroid fine needle aspiration (FNA) in selected retrospective groups. Limited reports of real-world performance suggest otherwise. This large series compares our real-world outcomes pre- and post-implementation of ACR TI-RADS for thyroid ultrasound (US) reporting. METHODS: ACR TI-RADS was adopted at our tertiary institution in 2018. In this retrospective observational study, all patients undergoing thyroid US were div

Single-cell analysis reveals the cellular and transcriptional heterogeneity of the normal pediatric thyroid.

To enhance the understanding of cellular heterogeneity within the pediatric thyroid, single-nuclei RNA sequencing was used to recover 38,069 non-pathogenic cells from thyroid tissue of three female pediatric/young adult patients. The recovered cells were analyzed using the SWANS (Single Entity Workflow ANalysiS pipeline (version 1.0). Analysis revealed seven major cell types: thyrocytes, endothelial cells, fibroblasts, C cells, T cells, B cells, and myeloid cells. Thyrocytes were the most prominent and heterogeneous cell type. Initially, two dominant thyrocyte subsets were identified based on

Age-specific risk of malignancy in pediatric thyroid cytology: Reframing ROM based on pre-test probability.

OBJECTIVE: Pediatric thyroid nodules are uncommon but have a higher malignancy rate than adult nodules. Existing Bethesda risk-of-malignancy (ROM) estimates are not age-stratified and are affected by verification bias. We aimed to generate age-specific ROM, likelihood ratios (LRs), and post-test malignancy probabilities for pediatric and young adult thyroid cytology. METHODS: We analysed 2728 thyroid fine-needle aspirations (FNAs) from patients aged 0-25 years across multiple tertiary centers (2000-2023). All cases were classified or reclassified using the 2023 Bethesda System and grouped int
