Preparation of controlled-released starch-alginate microspheres loaded with thymol nanoemulsions and their therapeutic effects on salmonellosis.

The rise of antimicrobial resistance has spurred interest in natural alternatives such as thymol, a potent broad-spectrum antibacterial agent. However, its clinical potential for treating enteric infections is severely limited by poor stability, low bioavailability, inherent cytotoxicity, and non-targeted release. To overcome these challenges, thymol nanoemulsion was firstly prepared and then incorporated into starch‑sodium alginate microspheres (TMS). This system was comprehensively characterized and evaluated for its antibacterial efficacy, safety, and therapeutic performance both in vitro a

Decontamination Efficacy of Acetic Acid on Microbial Load in Fresh Produce from Open Markets in Khulna City, Bangladesh.

Vegetables are exceptionally nutrient-dense food sources essential to our diet and are mostly preferred to be consumed uncooked for maximum benefits. In Bangladesh, they are predominantly sold in outdoor markets. However, poor handling practices can lead to bacterial contamination with pathogenic microorganisms, increasing the risk of severe foodborne infections. This study aims to quantify the bacterial load of fresh vegetables and evaluate the impact of the antimicrobial agent acetic acid on the microbial quality of fresh produce available in the local markets. Following a completely randomi

Pathogen hijacks focal adhesion signaling by a T3SS effector CteX.

Infections by Gram-negative pathogens like Salmonella and Shigella rely on type III secretion system (T3SS) effectors. While the opportunistic pathogen Chromobacterium violaceum encodes a crucial T3SS (Cpi-1/-1a), its full effector repertoire remains undefined. Here, we performed a comprehensive proteomic analysis of the C.v. Cpi-1/-1a T3SS secretome. Our analysis not only confirmed known effectors but also unveiled CteX, an effector with no prior functional annotation. Structural determination revealed that CteX adopts a papain-like fold, and functional studies demonstrated that it acts as a

The typhoid Mary legacy: Genomic epidemiology uncovers contemporary carriage dynamics across two decades of enteric fever surveillance in England and Wales.

Modern knowledge regarding 'healthy bacterial carriers' traces back to the legacy of typhoid Mary, who unknowingly infected >50 people with enteric fever. Yet, the intricacies of typhoidal Salmonella carriage, inclusive of Salmonella Typhi and Paratyphi A/B, remain largely unknown. Using data collected by the United Kingdom Health Security Agency's Salmonella reference laboratory and enhanced surveillance, we examined cases of enteric fever in England and Wales between 2004-2023, to track disease trends and identify carriers. In total, 8,297 cases of enteric fever were reported during the stud

SeaB is a conserved Salmonella enterica extracellular matrix binding protein involved in biofilm formation and infection.

Salmonella enterica is a leading cause of gastroenteritis worldwide. Exacerbating this issue is the emergence of multi-drug-resistant strains, posing a major threat to human health. Type 5 secretion system proteins play a major role in virulence and are viable vaccine targets. However, only a limited number of these proteins have been functionally characterized to date. In this study, we characterized SeaB, which belongs to the Type 5a secretion system. We demonstrated that SeaB is localized to the cell surface and involved in binding to the extracellular matrix. Our results indicate that SeaB

Impact of extraction method on the chemical composition and antibacterial potential of Salvia fruticosa essential oil.

BACKGROUND: Salvia fruticosa (Lamiaceae) is traditionally used to treat gastrointestinal (GIT) issues, stomach and abdominal pain, and ulcers. In this work, we aimed to extract essential oils (EOs) from the aerial parts of S. fruticosa, a plant commonly found in Egypt, and to evaluate their antibacterial activity against GIT pathogens. METHODS: Fresh samples' essential oils (EOs) were extracted using hydrodistillation (HD) and headspace (HS) techniques, while dried samples' EOs were obtained via supercritical fluid (SF) extraction. GC/MS analysis was used to analyze the EOs. Their antibacteri

Interregional outbreak of Salmonella Typhimurium linked to fresh cheese: A case-case study guided by whole-genome sequencing (WGS), Portugal, March-June 2024.

In 2024, an outbreak of Salmonella typhimurium affected two regions in Portugal. To identify the vehicle, we conducted a case-case study using a 'same disease, different time period' design. We compared S. typhimurium cases linked by whole-genome sequencing (WGS) (cluster cases) with salmonellosis cases notified in 2023 (historical cases) and calculated odds ratios (OR) for food exposures in surveillance data using logistic regression. We performed WGS on 58 isolates from the outbreak period (11/03/2024-2118/06/2024), and all belonged to a single cgMLST cluster (HierCC HC5_410,410). Compared w

Effect of Salmonella Cluster Detection and Investigation Methods on Outbreak Reporting in the United States, 2013-2017.

Cluster detection and outbreak investigation practices and resources vary across states. There is a lack of evidence to evaluate the effectiveness of Salmonella cluster detection and investigation practices on outbreak detection and reporting by states. We conducted an online survey of all 50 U.S. state health departments (HDs) in 2015 to assess how pulsed-field gel electrophoresis (PFGE) cluster detection and outbreak investigation practices and resources at the state affected reporting single-state Salmonella outbreaks to CDC's Foodborne Disease Outbreak Surveillance System (FDOSS) from 2013

Predicting Salmonella Typhi incidence using prevalence metrics from sentinel studies of community-onset bloodstream infections: a secondary analysis of observational data.

BACKGROUND: Typhoid fever incidence estimates are central to policy decisions on vaccine introduction and investments in non-vaccine prevention and control but are often unavailable. We explored whether prevalence metrics from sentinel studies of community-onset bloodstream infections could accurately predict local Salmonella enterica serovar Typhi (Salmonella Typhi) incidence. METHODS: Using a previous systematic review (January 2018-December 2024), we identified studies reporting both typhoid incidence and prevalence of community-onset bloodstream infections from sentinel sites. Vitamin D3 contribution to systemic and local control of Salmonella Typhimurium infection in an in vivo mouse model.

INTRODUCTION: Vitamin D serves as a critical modulator of innate and adaptive immune responses. Its active metabolite, 1,25-dihydroxyvitamin D3 (1,25(OH)2D3), exhibits potent antimicrobial activity in both humans and mice, positioning it as a promising adjunctive or alternative therapeutic for infectious diseases through immune modulation and clearance of pathogens. Among these, Salmonella enterica serovar Typhimurium, a leading cause of foodborne zoonotic infections, presents significant global public health burdens. METHODS: Employing an in vivo murine model, we recapitulated the systemic a
