ClavF derived from Clavanins as a promising candidate for fighting infections from multiple-drug resistance Aeromonas hydrophila.

The antimicrobial peptide ClavF was designed based on the natural antimicrobial peptide Clavanins, derived from the hemocytes of the stalked ascidian, using the amino acid substitution method. The antimicrobial activity of ClavF was tested against various multiple-drug resistance (MDR) pathogenic bacteria (Staphylococcus aureus, Aeromonas hydrophila, Escherichia coli and Salmonella pullorum), with minimal inhibitory concentrations (MICs) as low as 2 μM and the half-maximal inhibitory concentration (IC50) for Ctenopharyngodon idella kidney (CIK) cells is 50 μM. Its bactericidal mechanism agains

Macrophages downregulate NEDD9 to counteract S. Typhimurium- mediated FAK-AKT activation and lysosome inhibition.

The scaffolding protein NEDD9 coordinates signaling downstream of integrins by interacting with focal adhesion kinase (FAK) and thereby promotes cell migration. NEDD9 expression is altered in a number of clinical conditions such as cancer, but its role in innate immunity against infections remains elusive. Transcriptome analysis of Salmonella Typhimurium (ST)-infected murine macrophages showed downregulation of NEDD9 and genes belonging to its signaling network. Bacterial infections induced host-mediated lysosomal degradation of NEDD9 in macrophages and PBMCs isolated from patients suffering f

Assessment of process hygiene criteria in poultry slaughterhouses: A comparative analysis of own-checks and official controls in Northeast Italy (2021-2023).

Process hygiene criteria (PHC) for Campylobacter and Salmonella provide essential guidance on hygiene standards for poultry processing at slaughterhouses. Data collected by Member States (MSs) in Europe revealed inconsistencies between official verification and internal monitoring results, highlighting the need for a comprehensive investigation to uncover the reasons behind these discrepancies. This study investigated PHC in relation to operational factors at nine poultry slaughterhouses, integrating production characteristics and epidemiological surveillance data from a representative sample

Fibroblast-derived CSF1 maintains colonization of gut mucosal macrophage to resist bacterial infection.

Macrophages play essential roles in immune defense and tissue homeostasis, but the mechanisms underlying their colonization in the gut mucosa remain incompletely understood. Here, we identify CSF1, primarily derived from fibroblasts, as the dominant factor maintaining mucosal macrophage colonization, whereas IL-34 deficiency alone has a minimal impact. We reveal that CSF1R ligands originate from distinct cellular sources: macrophages at the upper villus region depend on fibroblast-derived CSF1 and IL-34, while macrophages in the lower villus and the submucosal (lower villus + SM) region are re

Acute Respiratory Distress Syndrome and Spondylitis Secondary to Salmonella Bacteremia.

Salmonella enterica subsp. enterica includes both typhoidal and non-typhoidal serovars. The former are more likely to cause systemic infections such as typhoid fever, while the latter typically result in localized gastroenteritis. The main symptoms of non-typhoidal Salmonella (NTS)infection are transient diarrhea, which typically resolves within approximately five days. However, in a few cases, it may worsen and cause multiorgan dysfunction. A 39-year-old healthy male developed an NTS infection, which likely manifested as enteritis, followed by Salmonella bacteremia, spondylitis, and acute res

Antimicrobial resistance and biofilm formation in rarely reported Salmonella enterica serovars from patients presenting with gastroenteritis in Nairobi, Kenya.

Non-typhoidal Salmonella infections are a significant global public health concern, causing approximately 150 million illnesses and 60,000 deaths annually, with majority of the cases occurring in low- and middle-income countries. In this study, we used whole genome sequencing to identify and characterize uncommon non-typhoidal Salmonella serovars isolated from patients presenting with gastrointestinal symptoms in the Mukuru area of Nairobi, Kenya. Sixteen less common NTS serovars (excluding Salmonella Typhimurium and S. Enteritidis) were identified from 25 patients, with 1 isolate from blood a

Signal peptides enhance immunogenicity and protection of recombinant Salmonella enterica serovar Choleraesuis vectors against Streptococcus suis.

Streptococcus suis (S. suis) is prevalent worldwide, threatening the pig farming industry and public health. At present, there is no effective vaccine. Salmonella vectors are highly attractive vaccine platforms that can deliver foreign antigens to induce protective responses. However, the limited secretion and release of foreign antigens limit the immunogenicity. Here, we employed 6-phosphogluconate dehydrogenase (6PGD), a protective antigen of Streptococcus suis serotype 2 (SS2), as a model antigen to evaluate how signal peptide (SP) selection influences antigen expression dynamics and immuno

Macrophages form dendrite-like pseudopods to enhance bacterial ingestion.

Macrophages are critical innate immune cells that exhibit remarkable adaptability during pathogen infections. However, the relationship between their morphological plasticity and physiological functions remains largely elusive. Here, we discovered an unprecedented paradigm of macrophage adaptation within a few hours upon severe Gram-negative bacterial infections, characterized by the formation of dendrite-like pseudopods (DLPs). Using in vitro, microfluidic, and in vivo infection models, we demonstrate that these pseudopods enhance bacterial uptake by expanding the macrophage searching radius,

Typhi Mykrobe: fast and accurate lineage identification and antimicrobial resistance genotyping directly from sequence reads for the typhoid fever agent Salmonella Typhi.

BACKGROUND: Typhoid fever results from systemic infection with Salmonella enterica serovar Typhi (Typhi) and causes 10 million illnesses annually. Disease control relies on prevention (water, sanitation, and hygiene interventions or vaccination) and effective antimicrobial treatment. Antimicrobial resistant (AMR) Typhi lineages have emerged and become established in many parts of the world. Knowledge of local pathogen populations informed by genomic surveillance, including of lineages (defined by the GenoTyphi scheme) and AMR determinants, is increasingly used to inform local treatment guideli

Increase of Clostridioides difficile PCR ribotype 002 infection with decreased disease severity in hospitalized patients in the Netherlands.

Clostridioides difficile PCR ribotype (RT) 002 is the second most prevalent RT in the Netherlands. In 2019, an increase in Clostridioides difficile infections (CDIs) due to clonal RT002 strains was reported in Ireland. In retrospect, a significant increase in the proportion of CDI due to RT002 was detected in the Netherlands between May 2017 and May 2018 (11.9% of 683 cases vs 6.5% of 865 in the previous year). Our aim was to analyze and characterize CDI due to RT002 over time and to assess whether RT002 isolates in the Netherlands were related to the Irish RT002 isolates.During the period fro
