UNLABELLED: Bdellovibrio are obligate predators and have been described as living antibiotics since their predatory lifestyle enables them to kill pathogenic bacteria, making Bdellovibrio a promising biological control agent. Bdellovibrio strains were isolated from sampling sites in an urban watershed and tested for their killing activity on Salmonella enterica strains associated with human infections. An enrichment technique was used to isolate Bdellovibrio from surface water samples from local tributaries to the Oconee River, employing Salmonella Infantis as the prey bait. The isolated Bdell

An ultrasmall bimetallic Au2Pd3 nanozyme with self-cascade enzyme-like activities and robust photothermal heating properties is synthesized and encapsulated into a poly(vinyl alcohol)/hyaluronic acid (PVA/HA) matrix via a repeated freezing and thawing process to yield a hydrogel microneedle patch (Au2Pd3@PH). This microneedle displays a good tissue-piercing capability and thermal-responsive melting behavior. Upon exposure to an 808 nm near-infrared laser, Au2Pd3@PH dissolves and releases Au2Pd3, which shows glucose oxidase (GOx)-like catalytic activity for the conversion of endogenous glucose

UNLABELLED: Carbapenem-resistant Proteus mirabilis (CRPM), largely driven by dissemination of blaNDM-1, poses a growing global threat, yet its phylogeographic and genetic patterns remain understudied. We analyzed 16 blaNDM-1-harboring CRPM clinical isolates from a tertiary hospital in China (2017-2024) using antimicrobial susceptibility testing, conjugation assays, whole-genome sequencing, and growth experiments. We found that blaNDM-1 dissemination occurred primarily via plasmids and Salmonella Genomic Island 1 (SGI1), including a novel SGI1-PM16 variant. Early transmission events were associ

Gamma-irradiated inactivated bacterial vaccines have emerged as a safer alternative, overcoming the safety and immunogenicity limitations of conventional live attenuated and inactivated vaccines. This study aimed to develop gamma-irradiated Salmonella Gallinarum (γ-SG) vaccine from a local field strain and evaluate its cell-mediated immune response in a chicken model, focusing on CD4+ and CD8+ T cell activation and IFN-γ production. Radiation doses ranging from 1.5 to 10 kGy were evaluated to determine the optimal level for inactivating bacterial replication while retaining metabolic activity.

Typhoid intestinal perforation (TIP) is a life-threatening, late complication of typhoid fever that disproportionately impacts children in low resource settings and continues to have devastating consequences worldwide. Despite elimination of typhoid fever in most high income countries, typhoid fever and TIP remain endemic in many countries around the world as a result of inadequate investments in water, sanitation, and hygiene (WASH) and lack of access to vaccines. A first National Typhoid Conference was held in Niamey, Niger on July 22, 2023, where surgeons and other medical and health profes

Salmonella enterica (SE) is one of the most prevalent enteric pathogens globally and infects humans through contaminated food and water sources. The rising trend of antibiotic-resistant SE strains poses a critical threat to public health. Bacteriophage-encoded endolysins evolve a promising alternative as antimicrobial agents for combating SE infections. These enzymes target the peptidoglycan layer of bacterial cells, causing cell lysis and death. However, the use of endolysins against Gram-negative bacteria is challenging due to the composition of the outer membrane, which acts as a barrier pr

BACKGROUND: Salmonella enterica Serovar Typhimurium (S. Typhimurium) is a major global foodborne pathogen. In our previous study, the S. Typhimurium ATCC 14028 strain harboring plasmid pHXY0908 survived at fourfold its ciprofloxacin MIC (MIC0908 = 0.125 µg/mL), but the mechanism remained unclear. Heteroresistance, where minority subpopulations show elevated resistance, is among the key contributors to chronic, recurrent, and high-fatality infections. This study aimed to elucidate the molecular mechanisms by which plasmid pHXY0908 promotes ciprofloxacin heteroresistance in S. Typhimurium ATCC 1

Salmonella enterica is a diverse bacterial pathogen consisting of both typhoidal and nontyphoidal clinically distinct serovars. While typhoidal serovars cause in humans a systemic life-threatening enteric fever, nontyphoidal Salmonella (NTS) usually provoke a localized self-limiting gastroenteritis. Factors responsible for the different diseases caused by distinct Salmonella serovars are still poorly understood. Here, we show that at elevated physiological temperature, manifested during enteric fever (39-40°C), the transcription of the flagellar regulon, its protein translation, and flagella-m

UNLABELLED: The obligate intracellular bacterium Chlamydia trachomatis (C.t.) is the leading cause of bacterial sexually transmitted infections and infectious blindness worldwide. C.t. replicates within a specialized membrane-bound compartment known as the inclusion and employs a type III secretion system (T3SS) to deliver effector proteins into the host cell. These effectors reprogram host cellular processes to create an environment conducive to bacterial replication. Until recently, C.t. was genetically intractable, limiting efforts to identify effector proteins secreted during infection. As

Background. Nontyphoidal Salmonella (NTS) is a common cause of enterocolitis and a major cause of death in children in low- and middle-income countries (LMICs). High antimicrobial resistance (AMR) prevalence in LMICs reduces treatment options for individuals at risk of severe infections.Methods. We investigated the use of metagenomics to identify NTS and associated AMR genes in 28 faecal metagenomes from children with culture-confirmed salmonellosis in Vietnam, using accompanying NTS genomes from isolated serovars (one per metagenome). Read-based and assembly-based methods were utilised for NT

Outbreaks caused by Salmonella Enteritidis are commonly linked to eggs and poultry meat internationally, but this serovar had never been detected in Aotearoa New Zealand (NZ) poultry prior to 2021. Locally designated genomic cluster Salmonella Enteritidis_2019_C_01, was implicated in a 2019 outbreak associated with a restaurant in Auckland. Four Enteritidis_2019_C_01 sub-clusters have since been identified, two retrospectively, in the Auckland region. Authorities initiated a formal outbreak investigation after genomically indistinguishable S. Enteritidis was isolated from the NZ poultry produc

Salmonella enterica is a highly diverse group of organisms with over 2,500 serovars described to date. However, not all serovars are pathogenic to humans and animals. Many serovars typically cause self-limiting gastrointestinal infections, but certain host-adapted serovars, as well as those expressing specialized virulence factors (e.g., Vi capsule) have an increased potential to cause bacteremia and trigger systemic responses, necessitating immediate medical attention. Timely identification of Salmonella at the serovar level is, therefore, critical for informing risk assessments, guiding clin

Bacterial infections caused by pathogens like Salmonella typhimurium (S. typhimurium) demand rapid, accurate detection and destruction to mitigate public health risks. Conventional culture-based methods are time-consuming, while existing nanomaterial biosensors often lack clinical validation. Here, we present a novel aptamer-stabilized gold nanoparticles (AuNPs)-based photothermal approach for integrated bacterial identification, detection, and destruction. The system exploits the localized surface plasmon resonance of AuNPs, whose aggregation-triggered by aptamer detachment in the presence of

The effective treatment of bloodstream infections caused by gram-negative bacteria (GNB) poses significant challenges as their distribution and resistance patterns vary across geographic locations and healthcare settings. Data on bloodstream infections (BSIs) and antimicrobial resistance patterns (AMR) in Bangladesh are limited. The objective of this study was to address this challenge by analyzing the prevalence, distribution, and resistance patterns-including multidrug-resistant (MDR) and extensively drug-resistant (XDR) status-of Gram-negative bacteria isolated from blood cultures at Dhaka

Antimicrobial resistance (AMR) in Salmonella enterica serotype Kentucky (S. Kentucky) is a global challenge, with increasing resistance to cephalosporins, ciprofloxacin, and carbapenems significantly limiting treatment strategies, yet its worldwide dissemination patterns and AMR profiles remain unclear. Therefore, this study monitored the current trends in global spatiotemporal distribution and AMR of 9,557 S. Kentucky from 47 countries through large-scale genome-based molecular epidemiology. We revealed that S. Kentucky demonstrated a broad geographic distribution, with ST314 emerging as a no

BACKGROUND: Livestock are recognized as major reservoirs for Campylobacter species and other enteric pathogens, posing infection risks to humans. High prevalence of Campylobacter during early childhood has been linked to environmental enteric dysfunction and stunting, particularly in low-resource settings.
METHODS: A total of 280 samples from Campylobacter positive households with complete metadata were analyzed by shotgun metagenomic sequencing followed by bioinformatic analysis via the CZ-ID metagenomic pipeline (Illumina mNGS Pipeline v7.1). Further statistical analyses in JMP PRO 16 explo

Intracellular vacuolar pathogens replicate within membrane-bound compartments known as pathogen-containing vacuoles (PCVs). Maintaining the integrity of these vacuoles is essential for creating a permissive niche that supports pathogen survival and proliferation. In this study, we show that Salmonella enterica serovar Typhimurium co-opts the host mitochondrial citrate carrier (CIC) to promote its intracellular replication by detoxifying the Salmonella-containing vacuole (SCV). Loss of CIC significantly impairs Salmonella growth within host cells, as CIC recruitment to SCVs regulates local citr

Bacterial enteritis necessitates innovative therapeutic strategies to overcome the significant limitations posed by conventional antibiotics. Here, we introduce a pioneering antibacterial approach, employing a multifaceted "catch-and-kill" mechanism that synergistically integrates targeted pathogen capture, sonodynamic eradication, and toxin neutralization. We present SonoMMT, a microfluidically engineered sonosensitizer-montmorillonite complex. SonoMMT selectively adsorbs pathogenic bacteria and bacterial toxins, shields encapsulated sonosensitizers from gastric degradation, and enables sonod
