Adverse effect of age on pancreatic islet morphology, islet cell turnover and lineage-labelled hormone expression in mice.

AIMS: Ageing leads to a gradual decline in the structure and function of many bodily organs, including the endocrine pancreas. The current study examines the impact of ageing on pancreatic islet morphology, and especially the role of islet cell plasticity in this process. METHODS: Transgenic Ins1Cre/+;Rosa26-eYFP mice, with islet beta-cell tracing capabilities, were employed at 12 and 52 weeks of age, and islet morphology, islet cell turnover as well as changes in beta-cell identity and plasticity assessed. RESULTS: Ageing was associated with elevated body weight and circulating glucose leve

Distinct senescent β-cell senotypes differentially drive islet aging and dysfunction.

Biological aging greatly impacts the body's ability to handle glucose, and represents a major risk factor the development and progression of type 2 diabetes (T2D). Nonetheless, despite advances in cellular senescence research and the development of new senolytic therapies, the heterogeneity of cellular senescence in the human endocrine pancreas, as well as its roles in normal aging, remains to be elucidated at the single-cell level. Here, we performed single-cell-resolved spatial proteomics and transcriptomics on intact pancreas from 26 donors (ages 20-80) and multiplexed single-cell RNA seque

Pancreatic mu opioid receptors regulate metabolism and ingestive behaviors.

The mu opioid receptor system has long been recognized for its role in regulating ingestive behaviors and food palatability. However, beyond their robust expression in the brain, opioid receptors are also found throughout the body, including the stomach, intestines, and pancreas. Previous studies suggest that opioids may directly regulate glycemia through actions in the endocrine pancreas, but whether these metabolic effects are linked to their influence on ingestive behavior remains unclear. Here, we used metabolic, ingestive, and genetic approaches to determine how the mu opioid receptor (MO

AI-guided analysis of human pancreatic islet sociology reveals distinct cell compositional changes in type 1 diabetes.

Human pancreatic islets exhibit greater anatomic and cellular heterogeneity than previously appreciated, raising fundamental questions about how their composition varies with age, sex, region, and islet size and how type 1 diabetes (T1D) alters these relationships. Yet these questions remained largely unresolved due to the bottleneck of manual tissue inspection. Here, we developed an integrated artificial intelligence (AI)-guided imaging, processing, and statistical pipeline enabling unbiased, high-throughput analysis of more than 2 million candidate islets from 106 non-diabetic (ND) and T1D d

Mapping histologic and functional maturation of human endocrine pancreas across early postnatal periods.

Human endocrine cell differentiation and islet morphogenesis play critical roles in determining islet cell mass and function, but the events and timeline of these processes are incompletely defined. To better understand early human islet cell development and maturation, we collected 123 pediatric pancreata and mapped morphological and spatiotemporal changes from birth through the first ten years of life. Using quantitative analyses and a combination of complementary tissue imaging approaches, including confocal microscopy and whole-slide multiplex imaging, we developed an integrated model for

Type 2 diabetes genetics in 125,000 admixed adults from Mexico City.

Type 2 diabetes (T2D) is a highly heritable, polygenic disease with over 600 loci identified through genome-wide association studies (GWAS). However, despite possessing unique genetic variation shaped by demographic history and admixture, Latin American populations remain markedly underrepresented in global genomic research. To address this gap, we conducted genome- and exome-wide analyses of 19,431 T2D cases and 105,611 controls from the Mexico City Prospective Study (MCPS). We identified 86 independent GWAS associations, including 21 novel signals, 15 of which replicated in external cohorts.

Functional Inactivation of PAX4 Results in Disrupted Endocrine Pancreas Development and Neonatal Diabetes in Pigs.

Variants in the human PAX4 gene are associated with both monogenic and complex forms of diabetes, yet their pathogenic effects remain difficult to define in models that accurately mimic human islet architecture and neonatal metabolic transitions. Here, we created a porcine PAX4 loss-of-function model using CRISPR/Cas9 cytidine deaminase base editing to introduce a premature stop codon in the PAX4 coding sequence. PAX4 knockout piglets developed severe hyperglycemia within 24 h of birth, followed by rapid postnatal clinical deterioration and uniform death by day 3. Biochemical analysis showed s

Maternal high-fat diet impairs pancreatic β-cell development and reduces VAMP-2 levels in islets of mouse offspring.

Maternal nutrition can influence offspring metabolism through metabolic programming, potentially leading to long-term disorders such as obesity and type 2 diabetes. However, the cellular- and tissue-level mechanisms involved in the metabolic programming, particularly regarding the endocrine pancreas, remain unclear. This study investigated the effects of maternal high-fat (HF) diet exposure on the biometric/metabolic profiles, β-cell morphometry, insulin secretion, and islet levels of the SNARE protein VAMP-2 and cytoskeletal F-actin, both critical to insulin granule exocytosis, in mouse offsp

A multiplex tissue resource for high-resolution spatial protein profiling in the Human Protein Atlas.

Spatially resolved protein expression is essential for understanding tissue organization, cellular specialization, and protein function. The open-access Human Protein Atlas database (www.proteinatlas.org) has generated an extensive antibody-based tissue resource for a majority of the human protein-coding genes using conventional immunohistochemistry, enabling body-wide annotation of protein expression across normal human tissues and major cell types. However, single-marker staining often lacks the cellular and subcellular context required to resolve rare cell populations, closely related cell

Effects of Diabetes on the Transcriptomic Profile of the Endocrine Pancreas: A Systematic Review.

INTRODUCTION: Type 2 Diabetes Mellitus (T2DM) is characterized by progressive pancreatic beta-cell dysfunction and insulin resistance. Although transcriptomic approaches have improved the understanding of molecular mechanisms underlying T2DM, evidence specifically addressing gene expression changes in the endocrine pancreas remains limited and fragmented across experimental and clinical studies. AIM: To systematically review the experimental and clinical evidence on the effects of diabetes on the transcriptomic profile of the endocrine pancreas. METHODS: A comprehensive literature search was

Direct ultrastructural validation of endocrine cell identity in zebrafish pancreatic islets.

The zebrafish is widely used to study glucose homeostasis, diabetes, and endocrine cell plasticity, yet a definitive ultrastructural characterization of adult pancreatic endocrine cell types has remained incomplete. Here, we combined transmission electron microscopy with hormone-specific immunogold labeling to define the ultrastructural characteristics of the major endocrine cell populations in adult zebrafish pancreatic islets. Three principal endocrine cell types were distinguished based on secretory granule morphology, electron density, and intracellular organization. Glucagon-positive α-ce

Spatiotemporal Dynamics of Bihormonal Cells in the Developing Human Pancreas.

Studies of bihormonal cell distribution in the developing human pancreas are limited and focused on the first half of pregnancy. Here, we quantified the distribution of bihormonal insulin+/glucagon+ cells in different forms of the structural organization of the endocrine pancreas across gestational weeks 10-40. It was demonstrated that insulin+/glucagon+ cells are most abundant during early gestation (weeks 10-12) and prevail among single cells and small clusters throughout prenatal development, indicating that these cells belong to a population of immature endocrine cells. Successful management of nesidioblastosis in a dog with persistent hyperinsulinemic hypoglycemia following staged bilateral partial pancreatectomies.

Nesidioblastosis is a rare but important differential diagnosis for hyperinsulinemic hypoglycemia in dogs, a condition most commonly associated with insulinoma. A nine-year-old spayed female dog was presented with recurrent episodes of collapse and documented hyperinsulinemic hypoglycemia. Diagnostic imaging failed to identify a pancreatic mass, prompting exploratory laparotomy with left partial pancreatectomy and regional lymphadenectomy. Histopathological examination excluded insulinoma and established a diagnosis of nesidioblastosis. Although clinical signs initially resolved following surg

Sex-Dependent Effects of Exogenous Irisin on Pancreatic Endocrine-Cell Immunophenotype in Post-Weaning Wistar Rats.

BACKGROUND/OBJECTIVES: Irisin is an exercise-associated myokine with reported effects on β-cell survival, insulin secretion, and pancreatic tissue protection in experimental models of metabolic stress. However, less is known about its influence on the developing endocrine pancreas under physiological conditions. Accordingly, the aim of the present study was to determine whether exogenous recombinant FNDC5/irisin is associated with changes in pancreatic endocrine-cell immunophenotype and islet-level markers of cell turnover in male and female Wistar rats. METHODS: Thirty-two 3-week-old Wistar

Interplay Between Infectious Diseases and the Endocrine System: An Overview and Clinical Insights.

INTRODUCTION: Infectious diseases can affect the endocrine system through multiple mechanisms, including direct cytopathic effects, immune dysregulation, inflammatory pathways, and in some cases the secretion of hormone-like substances by the pathogens themselves. While viral and bacterial infections represent the most frequent triggers, fungal and parasitic agents can also contribute, though less commonly. METHODS: This narrative review, based on a search in PubMed and Google Scholar, examined both paediatric and adult studies, given the limited evidence available in children. The analysis f

Hexavalent Chromium Toxicity in the Pancreas: A Study on the Protective Effects of Hypericum perforatum Extract.

Hexavalent chromium, a widespread heavy metal, induces apoptosis via the mitochondrial pathway through Bax (pro-apoptotic) and Bcl2 (anti-apoptotic) proteins. Hypericum perforatum, rich in antioxidants, can neutralise free radicals. This study investigated the effects of CrVI on the pancreas and the protective role of Hypericum perforatum. Five groups of animals were used: control, Cr (CrVI for 3 months), CrH (CrVI + 2.5% Hypericum perforatum extract made from flowers, for 3 months), Cr2 (CrVI for 3 months + distilled water for 1 month), and CrH2 (CrVI for 3 months + Hypericum perforatum extra
