Pancreas meets brain: β-hydroxybutyrate as a novel "β-cellular" metabolism therapy.

β-hydroxybutyrate (BHB), the predominant ketone body in human circulation, is synthesized in liver mitochondria and rises markedly during fasting, caloric restriction, ketogenic diets, and high-intensity exercise. Once considered a mere metabolic intermediate, BHB is now recognized as a potent signaling molecule that links nutrient status to gene regulation, inflammation, and cellular stress responses. In fact, beyond serving as an energy substrate, BHB functions as a versatile signaling metabolite that integrates environmental cues to epigenetic regulation, gene expression, and cellular physi

Sepsis prevents the development of experimental type 1 diabetes.

INTRODUCTION: While both sepsis and autoimmunity are characterized by dysregulated immune responses, their mutual influence remains only partially understood. METHODS: In this study, we investigated how sepsis affects the development of type 1 diabetes (T1D), an autoimmune disease characterized by the destruction of pancreatic β-cells. Specifically, we examined the impact of polymicrobial sepsis on the progression of T1D induced by multiple low doses of streptozotocin (MLDS). C57BL/6 mice were subjected to cecal ligation and puncture (CLP) to induce sepsis, and T1D was subsequently induced us

Birth of a Microdomain: Can Glucose Act as a Metabolic Switch?

Glucose homeostasis is an essential physiological process to ensure a stable energy supply to all tissues while preventing the harmful consequences of hypo- or hyperglycemia. The endocrine pancreas plays a central role in maintaining this process, where β-cells and α-cells secrete insulin and glucagon to coordinate systemic glucose metabolism, storage, and production. In this context, β-cells act as the main glucose sensors, closely linking extracellular glucose fluctuations to insulin release. This sensing is dependent on glucose transporters (GLUTs), which regulate cellular glucose uptake th

Molecular mechanisms linking adipose tissue-derived small extracellular vesicles/exosomes to the development or amelioration of obesity, insulin resistance, and diabetes-related complications.

Diabetes mellitus (DM) is a chronic metabolic disorder characterized by impaired glucose storage and utilization in adipose tissue and skeletal muscle. Type 2 diabetes mellitus (T2DM) is particularly prevalent among individuals with obesity, as excess weight fosters significant health complications, including inflammation and insulin resistance, ultimately contributing to the development of T2DM. It is believed that the crosstalk between adipose tissue, skeletal muscle, and the endocrine pancreas underlies the glucose dysmetabolism associated with excess weight. Extracellular vesicles (EVs) ca

Early experience on omaveloxolone in adult patients with Friedreich's ataxia: a real-world observational study.

INTRODUCTION: Friedreich's ataxia (FRDA) is an autosomal recessive neurodegenerative spinocerebellar ataxia caused by a homozygous GAA triplet repeat expansion in the frataxin (FXN) gene. FRDA is a multisystem disorder involving the central and peripheral nervous systems, the musculoskeletal system, the heart, and the endocrine pancreas. In recent years, Omaveloxolone, a potent activator of nuclear factor erythroid 2-related factor 2 signaling, showed a significant neurological improvement compared to placebo, with a good safety profile. With this study, we report an early real-life experience

Adropin modulates pancreatic cell proliferation and glutathione levels in an animal model of type 1 diabetes mellitus.

BACKGROUND: Diabetes mellitus (DM), a metabolic disorder that leads to chronic hyperglycemia, is one of the topmost global public health concerns according to the International Diabetes Federation. Adropin is a peptide hormone that is primarily involved in energy homeostasis, but its involvement in other biological activities such as lowering hyperlipidemia, and diminishing insulin resistance has also been reported. In this study, we aimed to explore additional effects of adropin on oxidative stress, inflammation, and cell proliferation in an animal model of type 1 DM. METHODS: To achieve our

Deletion of CHOP in beta cell protects mice from cardiovascular complications in type 2 diabetes: evidence from a pre-clinical mouse model.

INTRODUCTION: Cardiovascular complications are the leading cause of morbidity and mortality in patients with type 2 diabetes (T2D), which is recognized as a major independent risk factor for coronary artery disease, stroke, peripheral vascular disease, and heart failure. The interrelationship between metabolic dysfunction and cardiovascular disease is complex and multifactorial, involving hyperglycemia, insulin resistance, inflammation, and oxidative stress. Evidence indicates that endoplasmic reticulum (ER) stress and induction of the unfolded protein response (UPR) contribute to metabolic dy

An islet-resident macrophage antioxidant program preserves β cell physiology.

Pancreatic islet-resident macrophages (IRMs) display an activated phenotype and contribute to islet development and remodeling, yet their origin, heterogeneity, and functional roles remain poorly understood. Using complementary fate-mapping systems, we show that, in adult mice, around half of IRMs originate from circulating monocytes and undergo minimal turnover. Integrated multiple single-cell RNA sequencing analyses of mouse and human islets identified four major IRM cell states that collectively reveal their inflammatory and metabolic activation. Among these, a transcriptional program drive

Islet Inflammation and Endocrine Function in Aging - Evaluating the Role of Toll-like receptor 4.

The aging process is accompanied by a gradual decline in tissue function, in part due to chronic low-grade inflammation that contributes to cardiovascular, neurodegenerative, and metabolic disease development. In the endocrine pancreas, Langerhans islets exhibit age-related structural and functional changes, including immune cell infiltration and fibrotic remodeling, as we recently demonstrated. Macrophages, as key immune mediators in both diabetic and aged islets, play a central role in Toll-like receptor 4 (TLR4) signaling, a pathway activated by bacterial lipopolysaccharides and known to ex

Sodium-glucose transporters SGLT1 and SGLT2 in equine renal, hepatic and pancreatic tissue.

BACKGROUND: Insulin dysregulation is a hallmark of equine metabolic syndrome (EMS), and in recent years, pharmacological treatment with sodium-dependent glucose transporter 2 inhibitors (SGLT2i) have shown promise in reducing the risk of hyperinsulinemia-associated laminitis in horses diagnosed with EMS. In humans and laboratory animals, this transporter protein is responsible for the majority of renal tubular glucose reabsorption, however, the presence of this and the related sodium-dependent glucose transporter 1 (SGLT1) have not yet been studied in equine kidneys. Additionally, studies in h

Evaluation of Pancreatic Endocrine Reprogramming in Diabetic Cats.

Transcription factors control the development of the endocrine pancreas in various mammals. In humans, paired box-4 (PAX4) and aristaless-related homeobox (ARX) allocate endocrine progenitor cells toward β-cell and α-cell specification, respectively. In adulthood, PAX4 contributes to reprogramming α-cells into β-cells and exocrine into endocrine cells; induction of ARX in β-cells drives them to reprogram into α-cells. Feline diabetes mellitus has a similar pathophysiology to human type 2 diabetes, but information about the role of these transcription factors is unavailable in diabetic cats. Th

The Hippo terminal effector YAP boosts enterovirus replication in type 1 diabetes.

Type 1 diabetes (T1D) risk has been associated with enteroviral infections, particularly coxsackieviruses B (CVB). Cellular host factors contributing to virus-induced islet autoimmunity remain unclear. We show that the Hippo pathway effector Yes-associated Protein (YAP) is markedly upregulated in the exocrine and endocrine pancreas of T1D and at-risk autoantibody-positive (AAb+) donors, along with its target CTGF. YAP expression correlates with CVB RNA presence, often in or near infected cells. YAP overexpression enhances CVB replication, islet inflammation, and β-cell apoptosis, whereas its i

PLAIN ENGLISH SUMMARY: Parents of children with physical disabilities do a lot to support their child in daily life. In doing this they are faced with many challenges. These parents have a wide range of unmet needs, especially for information, on different topics. It is sometimes hard for them to get the right information at the right moment, and to ask the right questions to physicians and other healthcare professionals. In order to develop a digital tool to help parents formulate questions and find information, we thought it would be crucial to work together in a process of co-creation with

Optimizing equine sperm quality: an alternative to single layer centrifugation for sperm isolation.

ABSTRACT: In vitro semen purification techniques have been developed that seek to mimic the in vivo selection process in order to generate the highest possible chance of oocyte fertilization following artificial insemination. Numerous methods have been developed to isolate functional spermatozoa for artificial insemination, yet only one method, single-layer centrifugation using commercial preparations like EquiPure, has been widely employed. In this study, we have introduced a novel approach for isolating spermatozoa and compared their quality to those isolated using EquiPure. The AI port syst

Specific targeting of the NRF2/β-TrCP axis promotes beneficial effects in NASH.

Non-alcoholic steatohepatitis (NASH) is a common chronic liver disease that compromises liver function, for which there is not a specifically approved medicine. Recent research has identified transcription factor NRF2 as a potential therapeutic target. However, current NRF2 activators, designed to inhibit its repressor KEAP1, exhibit unwanted side effects. Alternatively, we previously introduced PHAR, a protein-protein interaction inhibitor of NRF2/β-TrCP, which induces a mild NRF2 activation and selectively activates NRF2 in the liver, close to normal physiological levels. Herein, we assessed

In vivo models of gestational and type 2 diabetes mellitus characterized by endocrine pancreas cell impairments.

Insulin resistance contributes to the development of various diseases, including type 2 diabetes and gestational diabetes. Even though gestational diabetes is specific to pregnancy, it can result in long-term glucose intolerance and type 2 diabetes after delivery. Given the substantial health and economic burdens associated with diabetes, it is imperative to better understand the mechanisms leading to insulin resistance and type 2 diabetes so that treatments targeted at reversing symptoms can be developed. Considering that the endocrine cells of the pancreas (islets of Langerhans) largely cont

Heterogeneity in the preferential diet of neotropical bats impacts the pancreatic islet mass and α and β cell distribution.

Whether there is a relationship between bats' dietary patterns and evolutionary endocrine pancreas adaptation is not clearly understood. Aiming to contribute to this topic, we evaluated some metabolic and structural parameters in the following adult bats: the frugivorous Artibeus lituratus, the nectarivorous Anoura caudifer, the hematophagous Desmodus rotundus, and the insectivorous Molossus molossus. A. lituratus and A. caudifer diets consist of high amounts of simple carbohydrates, while D. rotundus and M. molossus diets consist of high amounts of proteins or protein and fat, respectively. I

Resistin in endocrine pancreas of sheep: Presence and expression related to different diets.

Resistin (RETN), a recently discovered adipokine, is a cysteine-rich and secretory protein produced by adipocytes. RETN has been detected in several tissues, including human and laboratory animals' pancreas, wherein impairs glucose tolerance and insulin (INS) action and causes INS resistance. This study aims to evaluate the presence and expression of RETN in the pancreas of 15 adult female sheep reared on Apennine pastures, which show a decrease in their nutritional value due to the drought stress linked to the increasing summer aridity. The sheep were divided into 3 groups according to the di
