NONO is essential in maintaining airway smooth muscle contractility through PDE4/cAMP signaling pathways.

Non-POU domain containing octamer binding protein (NONO) is a multifunctional nuclear protein which plays important roles in regulating nuclear processes such as transcription and splicing. However, the role of NONO played in regulating airway smooth muscle (ASM) contraction remains largely unknown. In this study, we aimed to delineate the effects and the underlying mechanisms of NONO on ASM contraction. By deploying NONO gene knockout (NONO K.O.) mice, we examined tracheal contractility using a mechanical recording system. The expression of PDE4 was quantified by real-time PCR, the contents o

Efficacy of tracheal and gastric tube consecutive insertion under videolaryngoscopy in reducing complications associated with gastric tube insertion: a randomized controlled trial.

Insertion of a gastric tube in anesthetized patients is frequently difficult. The "consecutive insertion method", in which a videolaryngoscope is inserted, a tube is inserted to the trachea and a gastric tube insertion guide (with a gastric tube in it) to the esophagus before removing the videolaryngoscope, may reduce complications associated with insertion of a gastric tube. Sixty patients were randomly allocated to the consecutive insertion method (in group C) and blind method (in group B), to compare the incidence of complications, the success rate, and insertion time. The incidence of comp

Designing of porous scaffolds for tissue engineering and regenerative medicine.

The potential of the porous scaffolds lies in its dynamic architectural features, such as porosity, pore size & shape, interconnectivity, and spatial distribution which are highly essential in tissue engineering applications. The porous structure of the scaffolds influences the biological and physicochemical activities in the tissue regeneration process. The porosity also allows better distribution of nutrients and diffusion of biological fluids thereby dispersing the seeding material uniformly. Generally, damaged tissues can be regenerated on their own but many times due to unavoidable factor

Passive cigarette smoke induces reversible tracheal hyperreactivity via oxidative stress and m₃ receptor desensitization in rats.

Passive (second-hand) exposure to cigarette smoke contributes to airway dysfunction through oxidative stress, inflammation, and smooth muscle remodelling. This study investigated the effects of chronic passive cigarette smoke exposure on tracheal smooth muscle (TSM) contractility and oxidative stress biomarkers in adult male Wistar rats. Fifteen adult male Wistar rats were assigned to control, cigarette-exposed, or recovery groups. Rats underwent passive cigarette smoke exposure for 3 weeks, with the recovery group allowed an additional 3-week smoke-free period. The recovery group underwent th

Biointegration of a partially decellularized tracheal scaffold in a porcine model - preliminary results.

Some pediatric tracheal pathologies remain therapeutic dead ends for which current palliative strategies are fraught with serious complications. With the aim of a tracheal replacement, our team has previously developed and patented a clinical grade partially decellularized trachea (PDT) from porcine tracheas. The aim of this work was to study and compare the biointegration mechanisms of this PDT in vivo, in a pig cervical muscle, with or without immunosuppressant. The secondary objective was to evaluate the optimal maturation time of the PDT in this heterotopic position. In total, 11 female La

Circumferential Extramural Spread of Tracheal Adenoid Cystic Carcinoma Causing Unexpected Positive Margin.

Tracheal adenoid cystic carcinoma (ACC) often shows submucosal extension, leading to positive margins. A male patient presented with dyspnea due to an intraluminal tumour arising from the membranous mid-trachea. Rigid bronchoscopy debulking confirmed ACC. In a second-stage operation, circumferential resection of approximately 3.8 cm of trachea with end-to-end anastomosis achieved macroscopic clearance. Intraoperative frozen sections, however, revealed tumour cells within lymphatic channels of the tracheal sheath at the distal cartilaginous-side margin opposite the membranous lesion, precluding

Improving Recognition and Reporting of Malignant Central Airways Obstruction on CT: The Need for Standardized Terminology and Structured Reporting.

Background Malignant central airways obstruction (CAO) is a clinically significant complication of lung cancer that can lead to severe morbidity if not promptly recognized and is frequently under-reported on CT. Methodology All new annual lung cancer baseline CT scans were retrospectively reviewed as part of an audit process in 2014, 2019, and 2020. CT scans were assessed by the interventional pulmonology team. Patients with radiological CAO, defined as obstruction >50% in the trachea, main bronchi, or bronchus intermedius, were assessed. Narrative reports were separated into the main text sec

Acepromazine Reduces Airway Contraction in a Murine Model of Asthma.

Acepromazine (ACP) is one of the most used sedatives in veterinary medicine, and its cardiovascular effects are well documented. In contrast, its effects on the respiratory system remain less well understood, particularly in the context of assessment of respiratory mechanics in murine models of asthma, where its use has become more common in recent years. This study aims to investigate the effect of ACP on respiratory mechanics in BALB/c mice, specifically in an ovalbumin (OVA)-induced model of allergic asthma. Mice received ACP (2.5 mg/kg) in combination with standard ketamine (100 mg/kg) and

Spatiotemporally engineered microneedle for microenvironment remodeling propels mucosal regeneration after tracheal mucosal injury.

Remodeling the mucosal microenvironment is a pivotal step in severe tracheal mucosal injury (TMI), which may initiate a cascade of pathological events such as infection, inflammation, oxidative stress, and microvascular disruption. Current therapeutic strategies for TMI primarily focus on anti-infective or anti-inflammatory pathways, or relay on surgical interventions for complications such as stenosis. However, precise modulation of the mucosal microenvironment remains insufficient, and the unique anatomy of trachea hampers drug delivery and therapeutic concentrations maintenance. To address

Pathologic wall shear stress in the trachea decreases epithelial differentiation and increases fibrosis.

RATIONALE: Mechanical ventilation is integral to the medical management of the critically ill. However, prolonged exposure to pathologic airflow during mechanical ventilation places patients at risk for airway injury. Here, we developed an innovative microsurgical mouse model to examine how changes in airflow affect cellular regeneration and repair in the tracheobronchial airway. We examined wall shear stress (WSS), a known consequence of airflow constriction, and evaluated clinical significance using Heliox, a helium-rich environment, which reduces pathologic airflow in human patients. PET/CT Features of Granulomatosis With Polyangiitis Mimicking Left Lung and Tracheal Malignancy.

Granulomatosis with polyangiitis is a systemic disorder that can affect nearly any organ system, with frequent involvement of the kidneys, lungs, skin, and eyes. Although some patients relapse months to years after treatment, relapsing more than a decade later, particularly with tracheal involvement, is rare. We report a case of granulomatosis with polyangiitis with PET/CT findings mimicking malignancy in the left lung and trachea.

Impact of captivity on gut microbiota, tracheal microbiota and metabolic pathways in wild chipmunks.

BACKGROUND: Understanding the effects of captivity on wild animals is essential, as it helps to improve the physical health and welfare of captive wild animals. The changes in environment, diet and other factors during the captivity may reshape their internal microbiota and affect the body’s metabolism. Using 16S rRNA gene sequencing, we analyzed gut and tracheal microbiota from wild and captive chipmunks, and examined differences in serology, histopathology, fat metabolism, and muscle quality. RESULTS: The dominant bacterial phyla in the gut and tracheal microbiota of chipmunks are Firmicutes

A Two-Level Ensemble Machine Learning Framework for OSA Classification Whilst Awake from Noisy Tracheal Breathing Sounds.

Obstructive sleep apnea (OSA), defined by repetitive airway obstruction during sleep, is significantly underdiagnosed, mainly due to the resource-intensive and time-consuming nature of sleep assessment technologies. Machine learning analysis of the tracheal breathing sounds (TBS) whilst awake offers an alternative approach for OSA quick screening. This study aimed to address the challenge of wakefulness OSA detection using TBS recorded with an inexpensive microphone in a noisy environment. Data of 247 individuals with various degrees of OSA severity were analyzed. Recorded data were segmented

3D-printed bionic tracheal stents based on tunable triblock thermoplastic copolyester elastomers for tracheal stenosis.

Poly(ε-caprolactone) (PCL) has attracted significant attention in biomedical applications, including drug delivery, tissue engineering, and biodegradable implants. However, its inherently poor elasticity restricts its applicability in mechanically dynamic tissues and organs. Here, 4-methyl-ε-caprolactone (MCL), a biobased monomer structurally similar to ε-caprolactone (CL), was incorporated to produce poly(ε-caprolactone)-b-poly(4-methyl-ε-caprolactone)-b-poly(ε-caprolactone) (PCL-PMCL-PCL) triblock polymers via sequential ring-opening polymerization. The resulting triblock polymers exhibited

Estimation of anaerobic threshold and peak oxygen uptake from tracheal sound during cycle ergometer cardiopulmonary exercise test.

Cardiopulmonary exercise test (CPET) is widely used for assessing exercise capacity of individuals through ventilatory anaerobic threshold (AT) and peak oxygen uptake (VO2peak). However, CPET requires specialized equipment, resources, time, and trained personnel. In this study, we investigated the feasibility of estimating ventilatory AT and submaximal VO2peak using tracheal respiratory sounds recording with a wearable sensor (The Patch) during incremental cycle ergometer testing. Twenty-four healthy adults (18 females, age: 31.0 ± 9.1 years, body mass index: 23.5 ± 3.3 kg/m2) underwent CPET w
