Chemical Profiling and Antimicrobial Prospective of Commercial Essential Oils from Selected Lamiaceae Genera.

Essential oils (EOs) of Lamiaceae family have been reported to possess antimicrobial properties. However, commercial preparations of EOs are often of variable quality and inconsistent efficacy due to adulteration and chemotypic variation. This study comprehensively profiled six retail Lamiaceae EOs (Origanum vulgare, O. vulgare subsp. hirtum, Thymus vulgaris, Lavandula angustifolia, Salvia officinalis, and Melissa officinalis) available in Türkiye using GC-MS, complemented by principal component analysis (PCA) and hierarchical cluster analysis (HCA) to discern chemotypic relationships. Antimic

HOXA3 Knockdown Impairs hESC-Derived Thymic Epithelial-like Cell Differentiation and CXCL12-Associated Early T-Lineage Support.

Thymic epithelial cells (TECs) provide essential signals for T-cell development, yet the mechanisms governing human TEC differentiation and the acquisition of T-cell-supporting function remain incompletely understood. Using a three-stage system that directs human embryonic stem cells (hESCs) through definitive endoderm and third pharyngeal pouch endoderm (3PPE) toward a thymic epithelial-like lineage, we investigated the role of HOXA3 during thymic epithelial-like cell (TEC-like cell) differentiation. HOXA3 knockdown during 3PPE-to-TEC-like cell transition impaired thymic epithelial differenti

Possible CPX-351-associated drug reaction with eosinophilia and systemic symptoms/drug-induced hypersensitivity syndrome-like reaction during severe lymphopenia.

Drug reaction with eosinophilia and systemic symptoms (DRESS), also known as drug-induced hypersensitivity syndrome (DIHS), is a severe delayed drug reaction characterized by skin eruption, systemic inflammation, organ involvement, and human herpesvirus 6 (HHV-6) reactivation. CPX-351 is increasingly used for high-risk acute myeloid leukemia, and skin eruptions are common adverse events; however, CPX-351-associated DRESS/DIHS-like reactions have not been well described. We report a 62-year-old man with acute erythroid leukemia who developed a progressive eruption after CPX-351 therapy. An earl

Autoimmunity induced by epigenome alteration affecting thymic Treg cell development.

Methyl-CpG-binding domain protein 3 (MBD3), an essential component of the nucleosome remodeling and deacetylase (NuRD) complex, controls chromatin accessibility at lineage-specific transcriptional enhancers and promoters during hematopoietic cell differentiation. Here, we have shown that MBD3 deficiency in the T cell compartment resulted in spontaneous development of T cell-mediated multiorgan autoimmune diseases including fatal autoimmune myocarditis. The MBD3-deficient mice notably showed a significant reduction of CD25+Foxp3+ regulatory T (Treg) cells in both the thymus and periphery withou

Thymic epithelial tumors: what's next?

Thymic epithelial tumors (TETs) are rare malignancies arising in an organ central to T cell development and immune tolerance, creating a distinctive interface between tumor biology and immunotherapy. Complete surgical resection remains the mainstay for resectable disease, whereas systemic treatment options for unresectable, recurrent, and metastatic TETs remain limited. The unique immunobiology of thymoma, particularly impaired central immune tolerance, may account for the unusual balance between antitumor activity and severe immune-related adverse events observed with immune checkpoint blocka

Thymic Composition Predicts Radiation Pneumonitis in Locally Advanced NSCLC.

INTRODUCTION: Durvalumab consolidation after concurrent chemoradiotherapy (cCRT) is standard for unresectable stage III NSCLC; however, radiation pneumonitis (RP) remains a major treatment-limiting toxicity. Lung dosimetry incompletely explains the risk of toxicity. Recent studies have established radiographic thymic health as clinically relevant in cancer, but its relationship to treatment-related toxicity remains unclear. We evaluated whether percent thymic tissue (pTT), an interpretable CT-derived measure of residual thymic tissue, is associated with severe pneumonitis after cCRT in NSCLC.

PRRC2A-mediated m6A modification on NCOA4 promotes thymic epithelial tumor progression through PKM2-induced glycolysis.

Thymic epithelial tumors (TETs) are heterogeneous neoplasms. While most early-stage cases are curable after resection, advanced or aggressive subtypes have limited treatment options and poor outcomes. The molecular mechanisms underlying their metabolic reprogramming remain poorly understood. Proline-rich Coiled-coil 2a (PRRC2A), a N6-methyladenosine (m6A) reader protein, is significantly upregulated in TET tissues and is associated with poor clinical outcomes. Multi-omics analyses and molecular experiments revealed that PRRC2A binds to m6A-modified sites within the NCOA4 mRNA, thereby stabiliz

Essential oil nanoemulsion coatings as controlled-release antimicrobial mechanism for postharvest mango preservation.

BACKGROUND: Mango (Mangifera indica L.) is a tropical fruit of high commercial value; however, its postharvest quality is severely limited by anthracnose caused by Colletotrichum gloeosporioides. The pathogen establishes latent infections during fruit development that rapidly activate during ripening, resulting in economic losses. Although synthetic fungicides are used for disease control, their use raises concerns about resistance development, environmental impacts, and food safety. Essential oils (EOs) represent a promising alternative, but their practical application is constrained by limit

Aire and Fezf2 Shape the Medullary Thymic Epithelial Cell Immunopeptidome for Central Tolerance.

Medullary thymic epithelial cells (mTECs) are central to immune self-tolerance owing to their capacity to express and present a diverse repertoire of self-derived peptides to developing thymocytes. This diversity arises from both promiscuous gene expression and specialised differentiation programmes, including thymic mimetic-cell populations, and is largely shaped by AIRE/Aire and FEZF2/Fezf2. Although the roles of Aire and Fezf2 in regulating peripheral tissue antigen transcription are well established, a critical conceptual gap remains regarding how transcriptional promiscuity is translated

Carvacrol exhibits potent anti-Klebsiella activity, whereas the multicomponent composition of essential oils may contribute to their enhanced antibacterial activity.

Multidrug-resistant (MDR) Klebsiella infections represent an increasing therapeutic challenge, underscoring the need for complementary antimicrobial strategies. Essential oils (EOs) are complex mixtures of bioactive compounds, and their antibacterial effects may not be fully explained by their predominant constituents. We compared the antibacterial activity of isolated carvacrol with that of oregano (Origanum vulgare), summer savory (Satureja hortensis), and thyme (Thymus vulgaris) essential oils against MDR Klebsiella isolates and evaluated their ability to enhance conventional antibiotics. Effects of polysaccharides from the rhizome of Menispermum dauricum DC on alleviating chemotherapy-induced immune damage and the immunosuppressive tumor microenvironment associated with PD-1/PD-L1 regulation.

In this study, we established a metastatic tumor model by tail vein injection of fluorescent-labeled Lewis lung cancer cells. We systematically evaluated the in vivo antitumor effects of the rhizome of Menispermum dauricum polysaccharides (MDP), a homogeneous polysaccharide obtained through sequential chromatographic purification, its efficacy in alleviating cyclophosphamide (CTX)-induced chemotherapy damage, and its ability to improve the immunosuppressive tumor microenvironment (TME). We also investigated whether its mechanism is associated with the PD-1/PD-L1 pathway. Concurrently, an in vi

Ligustilide enhances TNTs-mediated mitochondrial transfer to accelerate the reactivation of TECs in sleep deprivation induced postpartum mouse.

ETHNOPHARMACOLOGICAL RELEVANCE: Angelica sinensis (Oliv.) Diels (Danggui) has been widely used by puerperae in China for postpartum recovery for more than a thousand years. Its key active ingredient, ligustilide (LIG), has been documented to exert thymus-repairing effects; however, its role and underlying mechanisms in postpartum thymic regeneration impairment remain unclear. AIM OF THE STUDY: During the puerperium, the acutely atrophied maternal thymus gradually returns to its non-pregnant state. However, frequent sleep deprivation (SD) resulting from neonatal breastfeeding often slows down

Association of structural characteristics with immunostimulatory activity of pectic polysaccharides prepared from Tartary buckwheat sprouts by enzymatic degradation and partial acid hydrolysis.

Although Tartary buckwheat sprout pectic polysaccharides (YM) have been reported to exhibit immunostimulatory activity, their structure-activity relationship remains poorly understood. We therefore compared the structural characteristics and immunostimulatory activities of three YM fractions to explore structure-activity associations. YM was enzymatically digested and partially acid-hydrolyzed to yield two derivatives, YME and YMA, with distinct structures. YME showed an increased proportion of the galactose-rich rhamnogalacturonan-I (RG-I) domain (from 68.66 to 72.65 mol%) and a moderate mole

Thyme essential oil (Thymus vulgaris) modulates methane mitigation and rumen fermentation in vitro: a dose-response study.

This study evaluated the effects of thyme (Thymus vulgaris) essential oil (EO), applied as a conventional bulk extract or a nanosized nanoemulsion at inclusion levels of 0, 15, 30, and 45 µL/g dry matter (DM), on in vitro ruminal gas production kinetics, methane and carbon dioxide emissions, nutrient degradability, and fermentation end-products. Gas chromatography-mass spectrometry identified thymol (33.15%), 1,3,8-p-menthatriene (22.65%), and γ-terpinene (12.8%) as the dominant volatile constituents, classifying the extract within the thymol chemotype. The nanoemulsion showed a unimodal dropl
