Understanding and clinical approach to early T-cell precursor acute lymphoblastic leukemia.

Early T-cell precursor acute lymphoblastic leukemia (ETP-ALL) is a distinct biological subtype of T-cell acute lymphoblastic leukemia (T-ALL) that was first described in 2009 based on a gene expression profile resembling that of early T-cell precursors in the thymus. ETP-ALL is characterized by an immature immunophenotype featuring positivity for CD7 and cytoplasmic CD3, with co-expression of myeloid/stem cell markers, and absence of CD1a, CD8, with weak or absent CD5 expression. ETP-ALL accounts for 5.5%-17.1% of pediatric and 15.8%-47.3% of adult T-ALL cases. Early studies identified ETP-ALL

TET loss of function selects for aneuploidies that confer a proliferative advantage.

Mutations in the Ten-Eleven Translocation (TET) family of DNA dioxygenases are among the most common epigenetic alterations in clonal hematopoiesis and hematologic malignancies, yet how TET loss of function reshapes genome evolution during cellular expansion remains to be fully understood. Here we show that loss of Tet2/3 in invariant natural killer T (iNKT) cells drives aggressive lymphoid expansion that is consistently associated with recurrent chromosome 17 trisomy, a dominant aneuploid state observed across more than ten independently derived tumors following transplantation into immunocom

Distinct Molecular Pathways Driving Thymic and Pulmonary Neuroendocrine Neoplasms: Organ of Origin Defines Molecular Identity and Yields CD44 as a Marker of Pulmonary Neuroendocrine Tumors.

Thymic and pulmonary neuroendocrine neoplasms (NEN) are morphologically similar but arise from different embryonic tissues. Whether this similarity extends to shared molecular programs is unclear. We compared 45 thymic and 37 pulmonary NEN by immunohistochemistry, targeted and exome sequencing, NanoString RNA profiling and mass spectrometry-based proteomics. The RNA and proteomic profiles of pulmonary and thymic neuroendocrine tumors (LuNET and TNET) were clearly different, while the profiles of neuroendocrine carcinomas of both organs (LuNEC and TNEC) were largely overlapping. A subgroup of T

First-line toripalimab in combination with platinum-doublet chemotherapy for advanced B3 thymoma and thymic carcinoma: an open-label prospective phase Ⅱ trial.

BACKGROUND: Several studies have demonstrated the efficacy of programmed cell death protein 1/programmed death-ligand 1 (PD-L1) inhibitors in patients with thymic epithelial tumors (TETs). Previous trials suggest that the efficacy of immunotherapy in TETs is associated with specific features of the tumor immune microenvironment. PATIENTS AND METHODS: Patients received a combination of paclitaxel, carboplatin, and toripalimab every 3 weeks for up to six cycles, followed by toripalimab maintenance every 4 weeks until disease progression or other discontinuation criteria. The primary endpoint wa

Thymic stromal defects and immune dysregulation in hypomorphic Rag deficiency: implications for hematopoietic cell transplantation.

BACKGROUND: Thymic epithelial cells (TEC) are essential for positive and negative selection during T-cell development. Hypomorphic RAG deficiency is characterized by T-cell lymphopenia and an increased risk of infections and autoimmunity. However, whether abnormalities of TEC composition and function also contribute to these clinical features is unknown. OBJECTIVE: Using a Rag1 hypomorphic (Rag1mut) mouse model, we examined composition and function of the TEC compartment, both before and after hematopoietic stem cell transplantation (HSCT). METHODS: Flow cytometry, immunofluorescent imaging,

Lymph node contraction links sex-biased naive CD8+ T cell decline to compromised antigen recognition during middle age.

The abundance of diverse naive CD8+ T cell clones is essential for broad protection against infection and cancer, but how sex and aging jointly shape this compartment remains poorly understood. Here, using mouse models with supporting human data, we uncover a sex-biased mechanism of immune aging in which early male-biased depletion of naive CD8+ T cells driven by accelerated, antigen-agnostic differentiation into virtual memory cells combines with thymic involution, limiting naive CD8+ T cell replenishment. These mechanisms led to more rapid lymph node contraction and reduced local naive T cel

A Rare Case of Large Cell Neuroendocrine Carcinoma of the Thymus Presenting as a Lower Neck Mass in a Young Male Patient.

Large cell neuroendocrine carcinoma (LCNEC) of the thymus is a strikingly unusual and high-grade thymic tumor with a poor prognosis. Studies with large series of patients on the clinical, epidemiological, and biological behavior of the disease are not available due to its low prevalence and short survival. We report here such a rare case of LCNEC in a 33-year-old male patient who primarily presented with a mass in the suprasternal and left supraclavicular regions with multiple bilateral cervical lymph nodes. Fine needle aspiration cytology from the neck mass demonstrated hypercellular smears w

Physiological upregulation of RAG1 expression compromises the survival of CD4+CD8+ thymocytes in vitro.

Thymocytes upregulate expression of recombination activating genes (RAGs) upon developmental transition from the CD4-CD8- double-negative to the CD4+CD8+ double-positive (DP) stage. Although enhanced RAG expression is necessary to generate a complete Tcra repertoire, here we demonstrate that elevated RAG expression simultaneously compromises DP thymocyte survival in vitro. Using a series of mouse models in which the expression and activity of RAG proteins varied, we attribute this apparent toxicity to elevated expression of RAG1 protein, independent of its association with RAG2 and its canonic

Amygdalin Targets PLAU to Modulate Immune-Inflammatory Signaling and Ameliorate Atopic Dermatitis: Evidence From Multi-Omics and an Animal Model.

BACKGROUND: Atopic dermatitis (AD) is a chronic inflammatory skin disorder with limited therapeutic options. OBJECTIVE: This study evaluated the therapeutic potential of amygdalin by identifying urokinase-type plasminogen activator (uPA/PLAU)-associated molecular targets and immune-inflammatory signaling pathways, aiming to explore a potential strategy for natural product-based interventions. METHODS: Transcriptomics, network pharmacology, and molecular docking were integrated to predict potential targets of amygdalin and intersect them with AD-related differentially expressed genes to ident

Heterogeneity and ontogeny of mouse thymic macrophages reveal a requirement for Csf1r-expressing myeloid cells during early T cell development.

Thymic macrophages (TMs) maintain tissue homeostasis by clearing the large numbers of apoptotic cells generated during T cell development, but how TM heterogeneity relates to their developmental origin and role in thymocyte maturation remains incompletely understood. Using complementary flow-cytometric, single-cell transcriptomic, and genetic approaches, we resolved two major TM populations corresponding to TIMD4+ cortical and CX3CR1+ medullary/cortico-medullary macrophages. TIMD4+ VCAM1+ TMs displayed a prominent efferocytosis and apoptotic-cell-clearance program, whereas TIMD4- VCAM1+ TMs we

Long-term immunological impairment after early thymectomy in congenital heart disease.

Early thymectomy during congenital heart disease repair may impair thymic output, but long-term effects across age groups remain incompletely defined. We evaluated T-cell receptor excision circles (TRECs) and a comprehensive panel of lymphocyte subsets in 20 adults (mean age, 29.6 years) and 16 children (mean age, 10.7 years) who underwent thymectomy within the first year of life, and in age- and sex-matched controls. TRECs were lower in patients than controls in adults (34 ± 25 vs. 79 ± 45 copies/μL; FDR-adjusted p = 0.001) and children (9 [7-15] vs. 95 [65-160] copies/μL; FDR-adjusted p < 0.

Single-cell transcriptomic reference map of human peripheral blood γδ T cells from 1 year to 90 years reveals major changes before adulthood.

γδ T cells have an unconventional biology as they develop as functional cells in the thymus, to thereafter adapt to postnatal cues and persist into adulthood in peripheral blood. Single-cell transcriptomics enables investigation of underlying molecular programs of γδ T cell effector functions in health and disease. However, the effect of age is often underestimated, and control transcriptome datasets of young healthy individuals are rare. Here, we generate a cost-effective single-cell transcriptomic reference of peripheral blood γδ T cells (106,711 cells) from 223 individuals from 1 year to 90

Spontaneous organ-specific eye autoimmune disease resulting from two engineered gene mutations compromising immune tolerance.

In order to address the question of how genetic compromises in multiple immune tolerance mechanisms might confer susceptibility to autoimmune disease development, we have bred together a partial loss of function in Aire, which is important for central tolerance of T cells, with a deficiency of the intracellular protein tyrosine kinase Lyn, which is important for function of inhibitory receptors of dendritic cells, B lymphocytes, and myeloid cells. These AireGW/+Lyn-/- mice spontaneously develop severe eye autoimmunity, characterized by retinal destruction and damage to the surrounding uvea, re

Effect of tannic acid and chitosan supplementation on growth performance, antioxidant status, immune function, and caecal microbial diversity of Pekin duck aged 0 to 21 days.

1. Tannic acid (TA) and chitosan (CH) are natural compounds with antioxidant, antimicrobial and immunomodulatory bioactivities. This study evaluated the effects of dietary TA, CH and their combination on White Pekin ducks aged 0 to 21 d.2. A total of 336 one-day-old Pekin ducks were randomly assigned to one of four dietary treatments with six replicate cages per treatment and 14 birds per cage. The treatments included a basal diet (CON), CON +0.2% TA, CON +0.2% CH and CON +0.2% TA + 0.2% CH. The trial lasted 21 d.3. Dietary supplementation with 0.2% TA alone increased average daily gain (p < 0

Electrochemical quantification of thymol in Thymus vulgaris essential oil with a 3D-printed electrode.

Thymol (TM), a monoterpenoid phenol with diverse antioxidant and antimicrobial properties, is of significant interest to the pharmaceutical, food, and agricultural sectors. Due to its importance, this study developed an electrochemical method for its quantification using a 3D-printed electrode made from a conductive carbon black/PLA filament (CB/PLA electrode). Electrochemical measurements were performed using square-wave voltammetry (SWV) in 0.1 mol L⁻1 Na2SO4 (pH 13). The potential was scanned from 0 to +0.80 V, exploiting the oxidation process of TM at approximately +0.30 V (vs. Ag|AgCl|KCl

Time-dependent synergistic pathogenicity of sequential avian reovirus and Mycoplasma synoviae co-infection in SPF chickens.

Avian reovirus (ARV) and Mycoplasma synoviae (MS) are major pathogens associated with tarsal synovitis and growth suppression in broilers. Yet the effect of infection order on their combined virulence remains poorly understood. A survey of 202 swollen joint samples from northern Chinese farms (2024-2025) revealed 35.1% ARV-MS co-detection, identifying this dual infection as a frequent co-infection in broiler arthritis. To assess timing effects, we inoculated 240 SPF chickens with ARV, MS, or both simultaneously or sequentially at 12- or 24 h intervals. All co-infection protocols were associate

Integrative systematics reveal an unusual species of pedunculate ground beetles lacking fossorial legs from Late Cretaceous Kachin amber (Coleoptera: Carabidae: Scaritinae).

CONTEXT: The subfamily Scaritinae (Coleoptera: Carabidae) is a speciose, cosmopolitan group of ground beetles characterised by a pedunculate body form and modified fossorial forelegs, adaptations associated with a burrowing lifestyle in soil or sandy substrates. The fossil record of Scaritinae is scarce and only one Mesozoic species has been described up to the present. AIMS: A new genus and species of scaritine ground beetles is described from Late Cretaceous amber in northern Myanmar. Its phylogenetic relationship with extant lineages of Scaritinae was investigated and divergence times amon

Encapsulation of thyme essential oil: a review of mechanistic insights and emerging applications.

Thyme essential oil (TEO) demonstrates potent antimicrobial and antioxidant activities, making it a promising candidate for use as a natural preservative, antimicrobial agent, feed additive, pest-control agent, and wound-dressing material. However, its practical application is limited by low water solubility, high volatility, and environmental sensitivity. Encapsulation within carrier matrices, which form micro/nanoparticles, can improve TEO stability, bioavailability, controlled release, thereby preserving or improving its bioactivity. This review summarizes the antimicrobial mechanisms of TE
