Neonatal lipopolysaccharide administration causes long-term changes in mouse immunosteroids.

Early-life infections have enduring effects on the immune and endocrine systems. Glucocorticoids (GCs) are produced by the adrenal glands and also produced by lymphoid organs (immunosteroids). We investigated the impacts of early-life lipopolysaccharide (LPS) challenge on GC and mineralocorticoid regulation in blood and lymphoid organs. We administered saline vehicle (nVEH) or LPS (50 µg/kg body wt, ip) (nLPS) to neonatal mice on postnatal day (PND) 4 and 6 ("first hit"). We then administered saline vehicle (aVEH) or LPS (50 µg/kg body wt, ip) (aLPS) to adults on PND90 ("second hit"), in a 2 ×

Molecular and pathological characterization of very virulent infectious bursal disease virus (vvIBDV) strains in Kashmir, India.

Infectious Bursal Disease (IBD), or Gumboro disease, is an acute, highly contagious viral infection that primarily affects young chickens, causing immunosuppression and mortality. This study aimed to characterize the genetic and pathological features of recent IBD outbreaks in Kashmir, India. A total of 32 suspected outbreaks were investigated from 2021 to 2023 across three districts: Srinagar, Ganderbal, and Budgam. Bursal tissues were collected for both histopathology and molecular detection. RT-PCR targeting the hypervariable region (HVR) of the VP2 gene confirmed IBDV infection in 29 sampl

Protocol for studying in vitro mouse primary thymocyte differentiation via retroviral-mediated gene expression and stromal-free stimulation.

T cell differentiation and selection in the thymus are pivotal for establishing antigen specificity and shaping the functional repertoire of peripheral T cells. Here, we present an in vitro protocol to investigate the roles of specific target genes during early thymic development in mice utilizing retroviral vectors. We describe steps for retroviral packaging, the isolation of primary murine thymic cells, retroviral transduction, and the subsequent in vitro differentiation of thymocytes. For complete details on the use and execution of this protocol, please refer to Fan Zhao et al.1.

The Power of Flow Cytometry in the Cytopathologic Diagnosis of Lymphoid Lesions.

BACKGROUND: Cytological examination of a fine-needle aspirate (FNA) is a minimally invasive modality that is increasingly used in the diagnosis of lymphoma. Flow cytometry and other ancillary tests make an important contribution to the diagnostic value of FNAs of lymphoid lesions. This review follows the recent publication of the first edition of the WHO Reporting System for Lymph Node, Spleen, and Thymus Cytopathology. This WHO volume provides a framework for assessment of cytopathological samples and presents in detail the contribution of ancillary studies, including flow cytometry. SUMMARY

The Critical Role of Ancillary Testing in Fine-Needle Aspiration of Lymph Nodes, Thymus, and Spleen.

BACKGROUND: Fine-needle aspiration cytology (FNAC) is a minimally invasive and reliable technique for sampling lymph nodes, thymus, and spleen. However, morphological interpretation alone is often insufficient due to overlapping features among reactive, infectious, and neoplastic processes. SUMMARY: The aim of this study was to review the essential role of ancillary testing in enhancing the diagnostic accuracy of FNAC in lymphoid and mediastinal organs. This is a narrative review of the literature focusing on the applications, strengths, and limitations of flow cytometry, immunocytochemistry,

The differentiation and function of heterogeneous thymic dendritic cell subsets require signals provided by distinct thymocyte cell types.

Dendritic cells are essential for establishing thymic central tolerance; however, mechanisms supporting their homeostasis and activation remain unresolved. Through single-cell transcriptomics and functional assays, we identify seven thymic conventional dendritic cell (cDC) subsets and discriminate their abilities to present self-antigens and induce regulatory T cells. Mice blocked at different stages of T cell development revealed that CD4+ single-positive (CD4SP) and CD8SP thymocytes differentially support homeostasis and activation of type 1 cDCs (cDC1s) versus cDC2s/plasmacytoid DCs (pDCs),

Fezf2 regulates differentiation of Aire-expressing and post-Aire mimetic epithelial populations maintaining thymic homeostasis.

Fezf2 has been proposed to serve principally as a transcriptional regulator of broad self-antigen expression in medullary thymic epithelial cells (mTECs). Here, we demonstrate an additional function for Fezf2 as a developmental regulator of TEC lineage commitment. Fezf2 deficiency in the thymus led to a relative expansion of lung mimetic and Ccl21+ immature mTECs at the expense of MHCII-hi Aire-expressing and all other mimetic populations. Consistent with this, the disruption of mTEC development had functional consequences, including alterations in tuft-associated iNKT polarization and microfo

Generation of thymus-reconstituting T cell progenitors from human pluripotent stem cells.

Generating a large number of progenitors that can repopulate the immune system of a recipient is one of the key steps toward efficient cancer immunotherapy. Here, we describe the engineering of T cell progenitors capable of direct and long-term reconstitution of the thymus. In the thymus, human pluripotent stem cell (hPSC)-derived progenitor T cells (pro-T cells) developed into single-positive human T cells that entered circulation and settled in the spleen. Single-cell transcriptome analysis of differentiating hPSCs attested to the emergence of cells that displayed the transcription signature

Artificial thymic organoid culture generates functional iPSC-derived CD4+ invariant natural killer T cells.

Invariant NKT (iNKT) cells have a T-cell receptor that is common to all individuals and are activated by recognizing glycolipids on MHC class I-like CD1d molecules. Activated iNKT cells are known to exert anti-tumor effects through the activation of other immune cells and have attracted attention as promising T cells for eliciting anti-tumor immunity. However, securing a sufficient number of iNKT cells is an obstacle to treatment because iNKT cells are a very small cell population, less than 0.1% of the peripheral blood lymphocytes. Although previous studies have demonstrated redifferentiation

Early antioxidant capacity, intestinal barrier integrity and gut microbiota drive DHAV-3 resistance in ducks.

BACKGROUND: Selective breeding for disease resistance is an effective strategy to control duck hepatitis A virus type 3 (DHAV-3) in waterfowl. However, the mechanism underlying resistance remains poorly understood, particularly those associated with antioxidant defense, intestinal development and host-microbiota interactions. METHOD: A total of 100 1-day-old Pekin ducklings were used in this study with 50 DHAV-3 susceptible and resistant ducks, respectively. Samples were collected at 7 days post-hatching (D7), D21 and D42, 10 birds per group. We compared DHAV-3 resistant and susceptible ducks

Solid state fermentation of Ginkgo Biloba leaves and its effects on antioxidant capacity and breast muscle quality of broilers.

Ginkgo biloba leaves contain many kinds of active ingredients, which can affect the metabolism of animals to a certain extent, thus promoting the growth performance and disease prevention and resistance of animals. After microbial fermentation, Ginkgo biloba leaves can better retain and release their active ingredients, and achieve full utilization of biomass, making them suitable as feed additives. In this study, we conducted an orthogonal optimization experiment with 7 factors and 3 levels using Ginkgo biloba leaves as the substrate, employing two probiotic strains Bacillus licheniformis and

Cabozantinib for advanced grade 3 neuroendocrine tumors: subgroup analysis of the phase 3 CABINET trial (Alliance A021602).

Well-differentiated grade 3 neuroendocrine tumors (NETs) have recently been described as a distinct category, and randomized data regarding efficacy of therapy for these patients are scarce. In the phase 3 CABINET trial, cabozantinib improved PFS compared with placebo in patients with advanced, previously treated, progressive extra-pancreatic NETs (epNETs) and pancreatic NETs (pNETs) of all grades. Here, we evaluate if these results remain consistent in a subgroup of patients with well-differentiated G3 NETs. Patients with locally advanced or metastatic epNETs or pNETs were randomized 2:1 in i

The impact of growth hormone (GH) on immunosenescence: exploring the role of B and T cells.

PURPOSE: Immunosenescence is a gradual decline in immune function, leading to increased susceptibility to infections and autoimmune conditions. Growth hormone (GH) has been shown to have an effect on both immune function and aging. In fact, the absence of GH-induced intracellular signaling can slow the aging process, as demonstrated by the longest-lived laboratory mouse (GH receptor gene disrupted or GHR-/- mice). Because GH receptors (GHR) are expressed in B and T cells, and these cells undergo age-related changes that impact immune function, we hypothesized that decreased GH action protects

Nanoformulated PDRN Improves Anti-Inflammatory and Wound Healing Activities.

Polydeoxyribonucleotide (PDRN), a bioactive DNA fragment, has been known to promote anti-inflammatory responses and wound healing primarily via adenosine A2A receptor activation. However, low molecular weight PDRN can undergo rapid degradation, limiting its sustained therapeutic efficacy. In this study, we developed a scalable method to produce high-purity, and low molecular weight PDRN (c.a. 325 bp) from calf thymus DNA via physical fragmentation. To enhance its stability and delivery, PDRN was encapsulated in poly(lactic-co-glycolic acid) (PLGA) to form PDRN/PLGA nanoparticles, yielding unif

Identification of Halophenol-Deoxyguanosine Adducts in SV-HUC-1 Cells Exposed to Halobenzoquinones via the Neutral Loss and Information-Dependent Acquisition Strategy.

Halobenzoquinones (HBQs), emerging disinfection byproducts (DBPs) in drinking water, were prioritized as potential bladder carcinogens. This study aimed to elucidate the interaction between HBQs and DNA, which may represent a critical initial molecular event in the adverse outcome pathway toward carcinogenesis. By combining high- and low-resolution mass spectrometry (MS), we developed a highly selective and sensitive analytical strategy integrating pentose neutral loss and information-dependent acquisition to characterize trace-level HBQ-DNA adducts in complex biological matrices.
