Leptospira interrogans HtrA protease is a potent inducer of multifaceted cellular responses.

Leptospirosis, a bacterial zoonosis caused by select pathogenic spirochetes belonging to the genus Leptospira, can trigger a systemic inflammatory response syndrome due to a cytokine storm that can occur during infection. However, the molecular mechanisms underlying the extensive inflammatory responses during leptospiral infection remain poorly understood. In this study, we characterized a high temperature requirement protein A (HtrA) homologue encoded by the L. interrogans gene locus LIC12812, referred herein as LepHtrA. We used AlphaFold 3 to predict the structure of this protein. And we sho

Molecular Detection and Genotyping of Pathogenic Leptospira in Small Mammals from Southern Vietnam.

Leptospirosis is a major zoonosis, yet genetic data on Leptospira strains in animal reservoirs in Southern Vietnam are limited. This study aimed to detect and genotype pathogenic Leptospira in synanthropic small mammals. From 2016 to 2020, 856 animals were captured in three regions. Kidneys were screened by qPCR targeting pathogenic Leptospira 16S rRNA, and positive samples were genotyped via secY gene sequencing. The overall prevalence was 7.8%. Rattus norvegicus was the primary host (12.4% infected). Leptospira interrogans predominated (77.6%), followed by L. borgpetersenii (22.4%). Infectio

Discovery of Novel c-di-GMP-Related Genes in Leptospira interrogans.

Cyclic di-GMP (bis-(3'→5') cyclic dimeric guanosine monophosphate) is a ubiquitous bacterial second messenger that regulates a wide range of cellular processes, including biofilm formation, motility, virulence, and environmental adaptation. Its intracellular levels are dynamically controlled by diguanylate cyclases (DGCs), which synthesize c-di-GMP from GTP, and phosphodiesterases (PDEs), which degrade it into linear pGpG or GMP. The functional effects of cytoplasmic c-di-GMP are mediated through diverse effector proteins, including PilZ domain-containing receptors, transcription factors, and

Flood preparedness and public health policy, Greece.

Floods are among the most frequent and complex climate-related hazards, affecting health and health systems through infectious disease risks, water and chemical contamination, and disruption of essential services. Examining how international guidance reflects the operational realities of flood response, and how it supports countries in practice, is essential. While each event is unique, sharing experiences can strengthen preparedness and resilience. This paper draws lessons from an assessment of the response measures used during the 2023 floods in Greece. The coordination team rapidly adapted

First report of pathogenic Leptospira in Sri Lankan bats: A potential reservoir risk?

BACKGROUND: The genus Leptospira, classified under the phylum Spirochaetes, includes saprophytic, intermediate, and pathogenic species. Pathogenic Leptospira spp. are the causative agents of leptospirosis, a widespread and often neglected zoonotic disease that causes severe illness in humans, particularly in tropical and subtropical regions. In Sri Lanka, leptospirosis causes annual outbreaks, especially during the monsoon seasons. While rodents are recognized as primary reservoirs, bats have also been identified as potential reservoir hosts. This study aimed to detect and characterize Leptosp

Clinical and Laboratory Profile of Patients with Tropical Coinfections Admitted at a Tertiary Care Center in North India.

BACKGROUND: Tropical coinfections (CI) are the simultaneous occurrence of two or more vector-borne diseases in a single host. The prevalence of such illnesses is not uncommon among tropical and subtropical regions such as India; however, these CIs have not been systematically studied prospectively. Mixed infections can prove potentially detrimental if underdiagnosed or undertreated. We undertook this study to estimate the prevalence and compare the clinical profile, laboratory characteristics, and various outcomes among the patients with tropical CI who presented with acute undifferentiated fe

A tale of dual battle against leptospira and scrub typhus coinfection at a peripheral hospital in Kashmir.

Scrub typhus results from infection by Orientia tsutsugamushi, a bacterium transmitted to humans via larval mite (chigger) bites. On the other hand, Leptospira interrogans causes leptospirosis, a zoonotic bacterial infection typically contracted through exposure to contaminated water or soil. Scrub typhus and leptospirosis are important zoonotic infections increasingly recognized as public health concerns in tropical regions like India. Transmitted by mites and contact with contaminated water or soil respectively, they often cause nonspecific febrile illness, leading to frequent misdiagnosis a

CRISPR-based mutagenesis of lipopolysaccharide biosynthesis genes in Leptospira interrogans reveals gene essentiality and confirms the role of an O-antigen polymerase.

Leptospirosis is a worldwide zoonosis caused by pathogenic bacteria of the genus Leptospira. Lipopolysaccharide (LPS) is an immunodominant and protective antigen for Leptospira, but its biosynthesis remains poorly understood. In this study, we employed CRISPR/Cas9-non-homologous end-joining and CRISPR-Prime Editing to mutate key genes within the rfb locus of L. interrogans, including those involved in core oligosaccharide assembly, and the biogenesis, polymerization, and ligation of O-antigen. Mutants were successfully generated in LIC11312 (waaF, heptosyltransferase II) and LIC12137 (wcaJ, un

Functional Validation of the Proteome-Identified LIC_13056 Putative Lipoprotein of Leptospira interrogans and the Potential Role in Pathogenesis.

Leptospirosis is a widespread zoonosis of human and veterinary concern. The etiological agent of the disease is the pathogenic bacteria of the genus Leptospira. Transmission typically occurs through mucosal contact and/or injured skin with the urine of infected animals or contaminated environmental sources. Understanding the biology and pathogenesis of leptospires is the main focus of our study. In this work, we characterized a novel protein encoded by the LIC_13056 gene from L. interrogans serovar Copenhageni, having an OmpA-like domain. We show that this coding sequence (CDS), previously ass

Clinical presentation and laboratory findings: key indicators for early diagnosis and severity prediction of leptospirosis in Sri Lanka.

BACKGROUND: The diagnosis and treatment of leptospirosis in high-burden, low-resource settings pose a challenge due to the limited availability of confirmatory diagnostics. Therefore, the initiation of treatment relies primarily on the clinical assessment of patients suspected of having leptospirosis in resource-poor settings. This study aimed to both early diagnostic indicators and predictors of disease severity. METHODS: A total of 170 patients suspected of having leptospirosis admitted to selected hospitals in Western Province, Sri Lanka were enrolled. MAT, qPCR, and culture were used for c

DMEM and EMEM as alternate growth media for pathogenic Leptospira.

Pathogenic Leptospira species can survive and thrive in a wide range of environments. Distinct environments expose the bacteria to different temperatures, osmolarities, and amounts and sources of nutrition. However, leptospires are mostly cultured in a laboratory setting under in vitro conditions that do not reflect natural environments. This constraint on laboratory cultures limits the applicability of in vitro studies to the understanding of even simple pathogenic processes. Here we report, investigate, and identify a medium and conditions that mimic the host environment during leptospirosis
