First serological investigation of Toxoplasma gondii, Neospora caninum, Leishmania infantum and Leptospira spp. in dogs from a Fulni-ô Indigenous community in Pernambuco, Brazil: a One Health perspective.

Zoonotic diseases pose a major public health threat in indigenous communities due to geographic isolation, inadequate sanitation, and close contact between humans, domestic animals, and wildlife. This study aimed to investigate the presence of antibodies against Toxoplasma gondii, Neospora caninum, Leishmania infantum, and Leptospira spp. in dogs from the Fulni-ô indigenous community, located in Águas Belas, Pernambuco, Brazil. Blood samples were collected from 66 free-roaming or semi-domiciled dogs for serological analysis. Indirect immunofluorescence tests (IFAT) were used to detect antibodi

Phylogenetic evidence of possible zoonotic circulation of Leptospira species between human febrile patients and bats within a same interface.

Leptospirosis is a widespread zoonotic disease caused by pathogenic Leptospira spp., maintained by a wide range of animal reservoirs. In Colombia, it remains a major cause of acute undifferentiated febrile illness, yet the role of wildlife, particularly bats, in its transmission cycle is poorly understood. To investigate the relationship between Leptospira infecting humans and bats in Villeta, Colombia, thirty partial available leptospiral 16S rRNA sequences from febrile patients (n = 13) and bats (n = 17) were analyzed. Hierarchical clustering identified twelve groups (A-L), two (C and G) con

[Leptospirosis].

Two patients with severe leptospirosis, infected in Denmark, developed multi-organ failure. A previously healthy woman presented with poor general condition and was diagnosed with cerebrospinal fluid PCR. A man with hypertension developed cardiopulmonary failure and tested positive for Leptospira in urine. In this case report, both received intensive care with vasopressors and antibiotics, improved clinically, and were discharged for follow-up.

The Leptospira immunoglobulin-like protein LigB from Leptospira borgpetersenii serovar Arborea is not required for either acute or chronic infection.

Leptospirosis is a globally important zoonotic disease caused by more than 40 pathogenic Leptospira spp. that are responsible for more than 1 million human cases and almost 60,000 deaths annually. The disease also affects many companion, domestic, and wild animal species. Leptospiral immunoglobulin-like proteins (Lig), particularly LigA and LigB, are well-established surface membrane proteins that have been extensively studied due to their interactions with the host immune system. Silencing expression of both ligA and ligB in L. interrogans serovar Copenhageni results in attenuation of virulen

Subclinical Leptospiral Infections and Antibody Waning During Pregnancy: A Population-Based Cohort Study from Rural Sri Lanka.

Leptospirosis is a zoonotic infection with significant mortality, yet its burden is underestimated due to diagnostic limitations. The dynamics of naturally acquired anti-Leptospira antibodies, particularly during pregnancy, remain poorly understood. This study examined the seroincidence of leptospiral infection among a cohort of pregnant women and characterized longitudinal antibody kinetics in an endemic setting. Paired sera were collected from a population-based pregnancy cohort in Anuradhapura, Sri Lanka. Women were enrolled before 12 weeks of gestation and followed up between 25-28 weeks.

Epidemiology and associated factors of human and canine leptospirosis in China: a systematic review and meta-analysis.

Leptospirosis, a neglected zoonotic disease caused by pathogenic Leptospira spp., poses ongoing challenges in China due to shared environmental exposure of humans and dogs. To summarize available epidemiological evidence, we conducted a PRISMA-compliant systematic review and meta-analysis of human and canine leptospirosis in China. Six databases (PubMed, Web of Science, ScienceDirect, CNKI, Wanfang, and VIP) were searched for eligible studies published up to 11 November 2025. Cross-sectional data were synthesized using random-effects models, with subgroup analyses applied to explore heterogene

Hybrid neural-mechanistic modeling of leptospirosis transmission with environmental drivers: Evidence from Thailand.

Accurate infectious-disease forecasts are essential for timely public health decision-making. In this study, we develop a hybrid modeling framework that combines compartmental models with Long Short-Term Memory (LSTM) networks to estimate a key time-varying epidemiological parameter as a case study for leptospirosis in Thailand. Our framework uses an LSTM-ODE model trained on environmental covariates (rainfall, flooding, and temperature) and infected human cases to infer the transmission rate, which shows strong seasonal and environmental dependencies. The results demonstrate that including fl

Unique aspects of acute kidney injury associated with infections in the tropics: presentation, challenges, and opportunities.

"The tropics" currently account for around 40% of the world's population and face unique environmental challenges, with limited resources, a deficient health system, and a rudimentary sanitary infrastructure, in addition to a lack of adequate preventive measures and poor surveillance programs. Cases of acute kidney injury in the tropics are to a very large extent driven by infections such as leptospirosis, scrub typhus, dengue, malaria, Ebola virus, yellow fever, and HIV, with a pathogenesis driven by volume depletion, hypotension, and immune-mediated mechanisms. Many of these infections also

Acute pancreatitis due to Leptospira spp.: An uncommon case report of a common zoonosis.

Leptospirosis, a widespread zoonotic disease prevalent in tropical and subtropical regions, causes over one million cases and about 59,000 deaths annually. It is transmitted through contact with infected animal urine, primarily from rodents, and worsened by poor sanitation and favorable climate. Although in Colombia there was a significant decline in cases from 2018 to 2022, severe forms of the disease, including rare complications like acute pancreatitis, still pose significant health risks. A 49-year-old male presented with symptoms including abdominal pain, jaundice, and severe thrombocytop

Leptospirosis on the edge: a case report of critical multi-organ involvement.

Leptospirosis is a bacterial zoonotic disease caused by Leptospira species, commonly transmitted through direct or indirect contact with contaminated water, soil, or animal urine. Although typically a self-limiting illness, severe forms of leptospirosis (Weil’s disease) can lead to multi-organ dysfunction, including hepatic and renal failure, hemorrhagic manifestations, and altered mental status. This case report describes a 52-year-old male who presented with severe jaundice, nausea, vomiting, diarrhea, and multi-organ dysfunction, including acute kidney injury, liver failure, and thrombocyto

Retrospective Molecular Detection and Characterization of Pathogenic Leptospira in the Philippines.

Leptospirosis remains a public health concern in the Philippines. Conventional diagnostic methods, including the microscopic agglutination test (MAT) and qPCR, are routinely used for outbreak response and surveillance. However, these methods often yield discordant results due to cross-reactivity, limited sensitivity, or lack of species-level resolution. To address these diagnostic gaps, this study optimized the Boonsilp 16S rRNA PCR assay and applied Sanger sequencing for accurate species identification of Leptospira in 92 archived DNA samples collected between 2018 and 2020. The sensitivity a

Genomic analysis of human-infecting Leptospira borgpetersenii isolates in Sri Lanka: Expanded PF07598 gene family repertoire and less genome reduction than bovine isolates.

Leptospira borgpetersenii is a causative agent of human leptospirosis, with the potential to lead to severe disease manifestations. The first published analysis of L. borgpetersenii, performed on two strains of serovar Hardjo (L550 and JB197), suggested that the L. borgpetersenii genome is in the process of genome decay with functional consequences leading to a more obligately host-dependent life cycle. Whole genome analysis has only been carried out on few strains of L. borgpetersenii, with limited closed genomes and comprehensive analyses. Herein we report the complete, circularized genomes

Infectious diseases in pregnancy in Sri Lanka: gaps in surveillance, diagnosis, and policy: a scoping review.

BACKGROUND: Infectious diseases during pregnancy are often overlooked in the Global South. Country-level mapping of infectious diseases and their effects on pregnancy would provide possible intervention strategies for diagnosis, prevention, and management. This scoping review aims to map the evidence from 2000–2024 on pathogens, mortality, morbidity, and adverse outcomes caused by infectious diseases in pregnancy among pregnant women in Sri Lanka. METHODS: We conducted a PRISMA-ScR-based systematic search on PubMed, Scopus, EBSCO, and the Web of Science using key terms related to infectious di

Severe Leptospirosis With Multi-Organ Dysfunction Syndrome in a Non-Endemic Region: A Case Report.

Leptospirosis is a common zoonotic infection worldwide but remains rare in the Middle East, including the United Arab Emirates. We report a case of a 36-year-old male who presented with high-grade fever, severe myalgia, headache, vomiting, dark urine, and altered mental status following recent travel from an endemic area where he worked as a paddy field farmer. Clinical examination and laboratory findings revealed septic shock, acute kidney injury, marked hyperbilirubinemia, and severe thrombocytopenia, consistent with multi-organ dysfunction syndrome. Based on the clinical presentation and ex

A multi-epitope approach for development of a universal vaccine against leptospirosis.

INTRODUCTION: Leptospira, a zoonotic pathogen, poses a significant public health threat, causing morbidity and mortality in both humans and animals. Although several bacterin and recombinant vaccines targeting specific serovars have been developed, most provide short-term, serovar-restricted protection and do not consistently induce sterilizing immunity or broad cross-serovar coverage. MATERIALS: To address this challenge, we employed comparative pan-genomic analysis and reverse vaccinology to identify conserved potential vaccine candidates (PVCs) and designed a multi-epitope vaccine (MEV) in

Collaborative mapping: Perceived environmental risk of leptospirosis in Urban communities in Salvador, Brazil.

Environmental risks disproportionately affect socially vulnerable populations, yet most studies on urban leptospirosis have not captured residents' perceptions of these risks. We conducted a cross-sectional study between August 2018 and March 2019 across four communities in Salvador, Brazil (Marechal Rondon, Alto do Cabrito, Nova Constituinte, and Rio Sena) using a collaborative mapping approach to explore residents' perceptions of environmental health risks. The majority of participants identified sewage as the primary health risk (26-39% across communities), followed by garbage (22-28%), whi
