Pseudomonas aeruginosa 3-oxo-C12HSL-induced apoptosis involves bitter taste receptor T2R14 and the mitochondrial calcium uniporter.

Pseudomonas aeruginosa is an opportunistic pathogen that infects immunocompromised patients. P. aeruginosa infects airways in cystic fibrosis (CF) and chronic rhinosinusitis (CRS) and also infects oral and nasal epithelia in immunocompromised head and neck squamous cell carcinoma patients. P. aeruginosa produces several apoptosis-inducing factors, including N-3-oxo-dodecanoyl-L-homoserine lactone (3-oxo-C12HSL). Additionally, 3-oxo-C12HSL activates several human bitter taste G protein-coupled receptors (taste family 2 receptors, or T2Rs), including T2R14 expressed throughout the body. We previ

DICER1 -Related Primitive Polyphenotypic Neoplasm : A Report of 15 Cases of an Underrecognized Tumor of the Gynecologic Tract and Peritoneum.

Somatic or germline pathogenic/likely pathogenic variants in DICER1 have known associations with certain neoplasms in the gynecologic tract, including Sertoli-Leydig cell tumors, embryonal rhabdomyosarcoma, and adenosarcoma. However, recent studies have highlighted DICER1- related malignant neoplasms with complex admixtures of sarcomatous, primitive glandular, and/or neuroectodermal elements, which are underrecognized and lack consistent nomenclature. We report the largest series of these primitive polyphenotypic DICER1 -related neoplasms arising in the gynecologic tract or peritoneum. The 15

Record of occurrence and histopathological changes induced by nematodes of the family Capillariidae in the tongue of fish, reptiles, birds, and mammals.

The family Capillaridae comprises of nematodes within the order Trichinellida that parasitize a wide range of vertebrate hosts, targeting the gastrointestinal, respiratory, and urinary tracts. While these helminths are frequently recorded in scientific literature, few studies report their presence in other anatomical sites such as the tongue. With that in mind, this study provides a systematic review of articles from 1975 to January 2026 focusing on the description of the presence and pathological alterations caused by helminths from the family Capillariidae whose habitat is the tongue of anim

Peritoneal Inclusion Cyst in an Unusual Location in a "Virgin" Abdomen: A Diagnostic Dilemma Expanding the Clinical Spectrum.

Peritoneal inclusion cysts (PICs) are benign, multilocular fluid-filled lesions that predominantly affect women of reproductive age, often arising after abdominal surgery or chronic inflammation. We report a case of a 28-year-old female presenting with chronic left upper-quadrant abdominal pain for 1 year. Radiological evaluation revealed a large cystic lesion closely abutting the left adrenal gland and adjacent organs, creating a diagnostic dilemma regarding its tissue of origin. The patient was planned for diagnostic laparoscopy, which revealed a large, thin-walled cyst containing clear sero

Antisenescent and antiinflammatory effect of N-acetylcysteine in peritoneal mesothelial cells.

BackgroundPeritoneal dialysis induces an intraperitoneal inflammatory reaction, which causes damage to the peritoneum. Inflammation also accelerates cellular senescence. We studied in vitro effect of the dialysates from peritoneal dialysis patients on the senescence of the peritoneal mesothelial cells (MCs). The effect of N-acetylcysteine (NAC) on that process was studied.MethodsReplicative senescence was induced in MC cells exposed to culture medium, medium mixed with the dialysate ± NAC 0.025 mmol/L. After 10 passages, markers of the cellular senescence and secretory activity of the cells we

Generation of functional mesothelial cells from human iPSCs that restore peritoneal integrity in experimental peritoneal injury.

BACKGROUND: Long-term peritoneal injury results in loss of mesothelial integrity and impaired ultrafiltration, limiting the durability of peritoneal dialysis. Because primary human peritoneal mesothelial cells (PMCs) are difficult to obtain and expand, regenerative strategies for restoring peritoneal function have remained limited. METHODS: We developed a protocol to differentiate human induced pluripotent stem cells (iPSCs) into functional peritoneal mesothelial-like cells (iPMCs). The molecular and functional characteristics of iPMCs were evaluated using gene expression analyses and functio

[Mesothelial impairment induced by peritoneal dialysis and its potential protective strategies].

Peritoneal dialysis serves as a critical treatment modality for end-stage renal disease. Extensive clinical practice has demonstrated that long-term exposure to high-glucose non-physiological peritoneal dialysis solutions can induce peritoneal injury, which is directly associated with a decline in ultrafiltration function and solute clearance capacity. This represents a significant cause of technical failure in dialysis and adverse clinical outcomes, such as peritonitis. Mesothelial cells and the glycocalyx on their surface, acting as the direct barrier between the peritoneum and the dialysis

Localization of TCR-expressing natural intraepithelial lymphocytes in the colon epithelium depends on GPR15 and epithelium-derived C10ORF99.

Intraepithelial lymphocytes (IELs) are among the largest lymphocyte populations in the body and play a crucial role in maintaining the integrity of epithelial barriers at mucosal sites, which are highly immunostimulatory. Therefore, understanding how these cells are generated, localized within the epithelium, and contribute to barrier immunity is essential. Although most research on IEL biology has focused on the small intestine, IELs are also present in the large intestinal epithelium. Additionally, the homing receptor-ligand pairs that regulate T cell localization in the lamina propria benea

Peritoneal dialysis-induced senescence contributes to treatment failure in long-term PD.

Peritoneal dialysis (PD) is an effective method of kidney replacement therapy. It offers patients a high degree of autonomy. However, long-term PD remains problematic. Continuous exposure of the peritoneal membrane to high-glucose solutions causes structural changes over time. This can ultimately result in a failure of ultrafiltration. It is known that PD patients exhibit elevated levels of oxidative stress, which can trigger senescence. However, it remains unclear, whether senescence occurs in the peritoneal membrane of PD patients. In this study, we demonstrate that senescent cells are mainl

Ultrafine particulate matter emitted from ships drives inflammation and susceptibility to viral infection.

Exposure to ambient airborne particulate matter (PM) has been associated with almost 9 million premature deaths per year worldwide. Ports are essential infrastructure for trade and transport, with over 80% of the world's international trade volume travelling by sea, yet there is little understanding of the potential health effects of PM from ships. Here, we studied the elemental and toxicological characteristics of size-fractionated PM collected at different areas of a busy European mixed-use port. We found that unregulated ultrafine PM, a PM size fraction <100 nm in aerodynamic diameter, orig

Asiaticoside alleviates mesothelial cell senescence and peritoneal fibrosis by targeting macrophage-mesothelial communication in single-cell peritoneal landscape.

BACKGROUND: Peritoneal fibrosis (PF) is a critical factor limiting long-term peritoneal dialysis (PD). The specific composition of peritoneal cells and intercellular communication remain unelucidated in PF context. Asiaticoside (ASI) possesses anti-fibrotic properties, but its therapeutic effects on PD-related PF and underlying mechanisms stay unclear. PURPOSE: To investigate changes in peritoneal cell components and their interactions during PF progression. To explore the pharmacological effects and potential mechanism of ASI against PF. METHODS: A mouse PF model was induced with high-gluco

From Primary Tumor to Peritoneal Niche: Microenvironmental Divergence in Gastric Cancer Peritoneal Metastasis.

Gastric cancer peritoneal metastasis is not simply an extension of the primary tumor into the abdominal cavity. It represents a biologically distinct disease context shaped by interactions between disseminated tumor cells, peritoneal fluid, mesothelial surfaces, submesothelial stroma, extracellular matrix, immune populations, and malignant ascites. In this narrative review, we examine peritoneal metastasis as a transition between three related but physiologically different states: the primary gastric tumor, free-floating tumor cells or spheroids in the peritoneal fluid, and established mesothe

Integrated Pathologic, Genomic, and Transcriptomic Analysis of Renal Neuroendocrine Tumors Reveals Neuroendocrine Transcriptional Programs and Associated Gastrointestinal-Type Epithelium in a Subset of Cases.

Renal well-differentiated neuroendocrine tumors (RenNETs) are rare primary renal neoplasms with enigmatic histogenesis. To define the molecular features of RenNETs and clarify their relationship to neuroendocrine tumors (NETs) from other sites, we performed whole-exome DNA sequencing and transcriptomic profiling on six RenNETs. In one case, adjacent non-neuroendocrine cystic epithelium and a regional lymph node metastasis were independently analyzed. RenNETs demonstrated a low-to-intermediate tumor mutational burden and recurrent copy number alterations, including losses involving chromosomes

MFAP2 secreted by TGF-β1-induced cancer-associated fibroblast cells promotes gastric cancer peritoneal metastasis through Src-STAT3-PTK7 axis.

Peritoneal metastasis is the leading risk factor for gastric cancer (GC). However, the mechanism of gastric cancer peritoneal metastasis (GCPM) is still unclear. GCPM depends not only on the "seeds" of tumor cells but also on the "soil" of the peritoneal microenvironment. This study aimed to analyze the changes in the peritoneal tissue microenvironment of nine patients with early-stage GC, advanced-stage GC, and GCPM using single-cell transcriptome sequencing. It found that the number of microfiber-associated protein 2 (MFAP2)-positive cancer-associated fibroblasts (CAFs) gradually increased w

Mesothelial cells derived extracellular vesicles promote angiogenesis through the transfer of angiopoietin-2.

BACKGROUND: Peritoneal mesothelial cells play a critical role in shaping the peritoneal tumor microenvironment and are increasingly recognized as active regulators of angiogenesis in cancers that metastasize to the peritoneum, including gastrointestinal (GI) and ovarian malignancies. Through complex interactions with tumor and stromal cells, peritoneal mesothelial cells contribute to the establishment of a pre-metastatic niche in the peritoneal cavity. Extracellular vesicles (EVs), nanosized vesicles secreted by most cell types, mediate intercellular communication by transferring bioactive mol

Astragalus polysaccharide as a protective agent against peritoneal dialysis-associated peritoneal injury: Role of the Nrf2/SLC7A11 axis in ferroptosis regulation.

Astragalus polysaccharide (APS) is an active component in Astragalus membranaceus. It has immunoregulatory, antioxidant-stress, anti-inflammatory, and anti-fibrotic actions. APS has favorable therapeutic effects upon tissue injury, but its effects on peritoneal mesothelial cells (PMCs) ferroptosis-associated peritoneal injury have not been reported. In this study, high-glucose peritoneal dialysis solutions (PDS) were found to induce ferroptosis similarly to erastin, a well-established ferroptosis inducer. Different concentrations of APS were administered in vitro and in vivo, with ferrostatin-

Pure isolation, culture, and postinjury lineage tracing of mouse visceral pleural mesothelial cells.

Pleural mesothelial cells (PMCs) are a major component of the pleura. The involvement of visceral PMCs has been suggested in several pleural or subpleural lung diseases, thus highlighting the importance of animal experiments using these cells. However, in wild-type rodent lungs, no direct and pure isolation method for visceral PMCs has been established so far. Using reanalyzed single-cell RNA sequencing data, we identified that mesothelin (Msln), a specifically expressed gene in visceral PMCs, can be useful for live cell sorting. After collecting cells by scraping the visceral pleura, MSLN+EpC

A case of delayed recurrence of pleural mesothelioma with intradural invasion.

BACKGROUND: Pleural mesothelioma (PM) is a tumor of the pleural epithelium with a poor prognosis. Asbestos exposure is closely related to its occurrence. To date, there have been 13 reports of PM with intradural invasion. CASE DESCRIPTION: A 73-year-old male was admitted with rapid progression of back pain and paraparesis. At age 67, he was diagnosed with PM (epithelial type) and underwent chemotherapy and a left thoracotomy for tumor resection. At present, the magnetic resonance newly showed T7-T9 spinal cord compressed due to an intradural tumor. Originating from the left pleura and invadin

Unraveling the multidimensional pleural ecosystem of lung cancer-associated malignant pleural effusion.

Lung cancer–associated malignant pleural effusion (LC-MPE) is a defining manifestation of advanced disease and a clinically consequential pleural compartment associated with dyspnea, repeated pleural interventions, and poor outcomes. This review synthesizes current evidence across mechanisms, diagnostics, and therapeutics to reframe LC-MPE as an active pleural ecosystem rather than a passive fluid collection. We summarize how partially reversible CD8⁺ T-cell dysfunction, regulatory T-cell and macrophage programs, mesothelial/fibro-inflammatory remodeling, vascular leak with impaired drainage,
