Somatic mutation and selection at population scale.

As we age, many tissues become colonized by microscopic clones carrying somatic driver mutations1-7. Some of these clones represent a first step towards cancer whereas others may contribute to ageing and other diseases. However, our understanding of this phenomenon remains limited due to the challenge of detecting mutations in small clones. Here we introduce a new version of nanorate sequencing (NanoSeq)8, a duplex sequencing method with an error rate lower than five errors per billion base pairs, which is compatible with whole-exome and targeted capture. Deep sequencing of polyclonal samples

Ultrasound Findings in Endosalpingiosis: A Closer Look at an Overlooked Condition.

Endosalpingiosis is a rare, benign condition characterized by ectopic cystic glands lined with ciliated cuboidal epithelium, histologically resembling the fallopian tube. Its etiology remains uncertain but is hypothesized to result from either ectopic implantation of fallopian tube tissue or metaplastic transformation of multipotential peritoneal cells. While frequently misdiagnosed as endometriosis, endosalpingiosis coexists with endometriosis in ~35% to 40% of cases. The condition can affect both women of reproductive age and postmenopausal individuals, with a median age of diagnosis between

Donor Diabetes and 1-Year Descemet Membrane Endothelial Keratoplasty Success Rate: A Randomized Clinical Trial.

IMPORTANCE: If the success of Descemet membrane endothelial keratoplasty (DMEK) is not affected by whether the donor has diabetes, then the donor pool should expand. OBJECTIVE: To determine whether the 1-year DMEK success rate is affected by the presence of diabetes in the donor. DESIGN, SETTING, AND PARTICIPANTS: This was a multicenter, double-masked, randomized clinical trial conducted from February 2022 to July 2025 at 28 clinical sites (46 surgeons), with donor corneas provided by 13 eye banks in the US. Included in the study were individuals undergoing low to moderate risk DMEK (95% for

Expanding the Molecular Characterization of Adenoid Ameloblastoma by Assessing a Panel of Oncogenes and Tumor Suppressor Genes.

Adenoid ameloblastoma (AA) is a rare epithelial odontogenic tumor microscopically characterized by ameloblastoma-like epithelium, duct-like structures, epithelial whorls, cribriform architecture, and dentinoid matrix. Wnt/β-catenin activation, usually by CTNNB1 mutations, is thought to characterize AA, which are BRAF and KRAS wild type. The present study aimed to expand the genetic characterization of AA by interrogating variants at several sites known to be linked to solid tumor pathogenesis. Six AA samples were sequenced at an unprecedented sequencing depth using a 22-gene panel including on

Bioresorbable Membrane Composed of Sodium Hyaluronate and Carboxymethylcellulose (Seprafilm) Prevents Cancer Implantation at Peritoneal Defects in 3D-Peritoneum and Murine Models.

BACKGROUND/AIM: The peritoneum is one of the most frequent sites of post-operative recurrence in advanced gastric cancer (GC). Peritoneal defects from laparotomy and GC resection facilitate the implantation of cancer cells shed from the primary tumor into the abdominal cavity. Sodium hyaluronate and carboxymethylcellulose film (Seprafilm) was developed for the purpose of reducing the incidence of postoperative adhesions between the abdominal wall and the underlying visceral organs in patients undergoing abdominal or pelvic surgery. However, it is unknown whether Seprafilm can help prevent canc

Fetal development of chromogranin A-positive gastrointestinal endocrine cells revisited: a histological study using human fetuses.

Initial gastrointestinal endocrine cells (GIECs) likely appear at the proximal and distal sites of abdominal intestines and may take a close topographical relation with neural elements in the gut. We examined immunohistochemically-stained sections from 10 fetuses at approximately 8-18 weeks of gestational age (36-155 mm of crown-rump length). Irrespective of whether physiological herniation was present (early 5 specimens) or absent (the other 5), the duodenum and jejunum had well-developed mucosa with villi containing abundant flask-like chromogranin-positive cells. In the earlier 5 specimens,

Mesenteric Cyst: A Case Report of a Rare Entity.

Mesenteric cysts are rare entities of the abdominal cavity, typically benign, that originate within the mesenteric tissue of the gastrointestinal tract. Their occurrence in adults is extremely uncommon and rarely encountered in clinical practice. These lesions may occur at any site along the mesentery, most often in the small bowel mesentery, and their clinical presentation varies from incidental findings to symptomatic abdominal pain, distension, or acute complications. Preoperative diagnosis can be challenging due to nonspecific symptoms, but advances in imaging modalities such as CT and MRI

Adenosine A2AR agonist blocks mesenteric lymphatic epithelium, adipose tissue and Treg cells links of metabolic dysfunction-associated steatotic liver disease mice.

Recent studies in diet-induced obese mice indicate crosstalk among mesentery, spleen, serum, and liver metabolites. Observations include decreased expression of the adenosine A2A receptor (A2AR), mesenteric lymphatic hyperpermeability, and reduced Treg cell populations in splenic and adipose tissues in metabolic dysfunction-associated steatotic liver disease (MASLD). This study aims to investigate the mechanisms and effects of a four-week treatment with the A2AR agonist CGS21680 in mice on a high-fat, high-sucrose diet-induced MASLD. In MASLD mice, reduced levels of mesenteric A2AR and the lym

Molecular Relationship Between Ovarian Sertoli-Leydig Cell Tumors and Their Heterologous Elements: Emphasis on the Possible Prognostic Significance of TERT Pathogenic Variants.

Since the histogenesis of heterologous elements within Sertoli-Leydig cell tumors (SLCTs) is poorly understood, we aimed to study the molecular relationship between Sertoli cells and the heterologous elements in 16 ovarian SLCTs. We performed a comprehensive molecular study on both SLCT and heterologous components, separately. Eleven tumors (68.7%) had one heterologous element and 5/16 (31.3%) had 2. Heterologous elements were epithelial (7/21 [81%]) (benign mucinous epithelium [9/21, 42.9%], borderline mucinous tumor [1/21, 4.8%], infiltrative mucinous adenocarcinoma [3/21, 14.3%], carcinoid

Subcutaneous Endosalpingiosis of the Buttock Without Prior Surgery or Trauma: A Case Report and Literature Review.

Endosalpingiosis is a rare, benign condition characterized by the presence of ectopic glandular epithelium that resembles the epithelium of the fallopian tubes. It typically affects the pelvic peritoneum and is often discovered incidentally. We present a rare case of subcutaneous endosalpingiosis localized in the left buttock of a 49-year-old woman with no history of surgery or trauma in the area. The patient presented with a palpable, painless subcutaneous mass that had been present for over 15 years and caused discomfort while sitting. Imaging and aspiration suggested a cystic lesion, and su

Aloe-Emodin Targeting FOXC2 Disrupts NETs Formation and EMT-Driven Postoperative Peritoneal Adhesion Through TGF-β1-Smad2/3 Pathway.

Postoperative peritoneal adhesion (PPA) develops through TGF-β1-driven fibrotic remodeling, characterized by neutrophil extracellular trap (NETs)-induced aberrant epithelial-to-mesenchymal transition (EMT) deposition. Although aloe-emodin (AE) exhibits anti-fibrosis potential, its molecular mechanisms remain elusive. Forkhead box protein C2 (FOXC2) is a critical regulator of fibrotic tissue formation, yet its role in PPA is unknown. Here, it is demonstrated that FOXC2 expression is elevated in human ileostomy tissue, PPA rodent model, and TGF-β1-exposed peritoneal mesothelial cells (PMCs), whe

Intraoral Epidermoid Cyst: Case Report and Literature Review.

BACKGROUND/OBJECTIVES: Epidermoid cysts are benign lesions originating from germinative epithelium, characterized by a keratin-filled cavity. They are histopathologically classified as epidermoid, dermoid, or teratoid. Intraoral cases are exceedingly rare, comprising less than 0.01% of all oral cystic lesions, most frequently affecting the floor of the mouth. While usually asymptomatic, they may become painful due to infection or growth. Because of their rarity in sites such as the upper lip, they may be clinically misdiagnosed, making awareness crucial for accurate management. METHODS: Diagn

Interaction between gastric cancer organoids and peritoneal mesothelial cells suppresses the growth of gastric cancer organoids and induces drug resistance.

The prognosis of gastric cancer with peritoneal dissemination is poor because of its resistance to chemotherapy. To investigate the mechanism of drug resistance in peritoneal metastasis, cancer organoids were established from the ascites of a patient with peritoneal metastases of gastric cancer. The histological characteristics of the tumors were preserved in the organoids. A co-culture system was established by overlaying human-derived mesothelial cells on gastric cancer organoids embedded in type IA collagen, mimicking peritoneal dissemination foci. When co-cultured with mesothelial cells, t

Sinonasal DICER1‑mutated embryonal-like (botryoid-like) rhabdomyosarcoma in an adult: report of the first case.

The DICER1 gene, essential for microRNA biogenesis and posttranscriptional gene regulation, has been implicated in a variety of benign and malignant neoplasms, particularly within the context of the DICER1-related tumor predisposition syndrome. While DICER1-associated rhabdomyosarcomas (RMS) are predominantly documented in the genitourinary tract, we present the first case of a DICER1-mutated embryonal-like (botryoid-like) RMS of the nasal fossa. A 63-year-old woman without relevant family history presented with nasal obstruction, headaches, and epistaxis and underwent resection of a polypoid

Single-Cell RNA Sequencing Reveals Peritoneal Environment and New Insights into Fibrosis in a Continuous Ambulatory Peritoneal Dialysis Patient: A Longitudinal Self-Controlled Study from Dialysis Initiation to 3-Year Follow-Up.

INTRODUCTION: Peritoneal fibrosis (PF) from long-term peritoneal dialysis (PD) is one of the main reasons for the ultrafiltration failure and the abandonment of PD in patients, and there is no effective treatment available. The dynamic changes in the PD microenvironment and their relationship to PD-related fibrosis remain unclear. METHODS: We performed single-cell RNA sequencing (scRNA-seq) on overnight PD effluent collected from a continuous ambulatory PD (CAPD) patient at two critical time points: the initiation of dialysis and after 3 years of treatment, so as to reveal the dynamic changes

Hair follicle gene expression profiling in the SubPopulations and InteRmediate Outcome Measures in COPD Study (SPIROMICS).

BACKGROUND: Transcriptomic analysis is common in large cohort studies but is generally restricted to cells in blood, which limits inferences about organs of interest, and direct organ sampling is mostly infeasible in large cohorts. New techniques for RNA-seq from noninvasive biosamples may provide the opportunity to profile transcriptomes of additional tissues for more organ-relevant insights at scale. We investigated the feasibility and utility of hair follicle gene expression profiling in a multi-center study of chronic obstructive pulmonary disease (COPD). METHODS: Bulk RNA-seq was perform

Interplay Between Glutamine Metabolism and Other Cellular Pathways: A Promising Hub in the Treatment of HNSCC.

Head and neck squamous cell carcinoma (HNSCC) is the most common and aggressive histologic subtype of head and neck cancer (HNC), difficult to treat effectively. Here, we discuss several studies on human and mouse HNSCC cell lines arising from the mucosal epithelium of various anatomical sites, as well as recent studies using murine models, focused on targeting key checkpoints in the glutamine (Gln) metabolism pathway, either alone or in synergy with other signaling pathways, as a potential therapeutic strategy for HNSCC. Emerging evidence demonstrates a complex interplay between Gln metabolis

Sodium Alginate Microgel-Wrapped Primary Human Peritoneal Mesothelial Cells for the Treatment of Peritoneal Fibrosis Caused by Dialysis via the POSTN/NF-κB/CXCL8 Pathway.

BACKGROUND: Peritoneal fibrosis is a significant complication arising from long-term peritoneal dialysis (PD), primarily due to the loss of peritoneal mesothelial cells (PMCs). Recent studies have implicated periostin (POSTN) in the progression of various fibrotic diseases; however, its specific role in PD-induced peritoneal fibrosis remains unclear. Sodium alginate (SA) microgels have emerged as promising carriers for cell encapsulation in tissue engineering and regenerative medicine. This study investigated the therapeutic potential of PMCs encapsulated in SA microgels (SA/PMC) for reducing

Pregnancy with vitrified oocytes after fertility sparing surgery in a patient with fallopian tube cancer and a BRCA2 mutation: A case report and literature review.

High-grade carcinomas affecting the ovary or peritoneum may originate from the epithelium of the fallopian tubes. Whilst gynecological cancers are more frequent in post-menopausal patients, 12.1% of ovarian malignancies are diagnosed in patients of childbearing age, aged <45 years. There is not enough data to provide an accurate estimate on the incidence of relapse, the chance of pregnancy and outcomes of assisted reproduction and fertility-sparing surgery techniques in young patients with ovarian cancer. Moreover, there is currently a lack of data regarding fertility preservation techniques f

Ovarian cancer metastasis to the human omentum disrupts organ homeostasis and induces fundamental tissue reprogramming.

The omentum, a visceral adipose tissue with critical metabolic, immunological, and stem cell functions is the preferred site for ovarian cancer metastasis. However, its role in maintaining homeostasis and its responses to metastatic colonization remain incompletely understood. Using single-cell transcriptomics, we profile different anatomical regions of the omentum in patients with benign conditions and metastasis. We catalog the benign omentum and observe a stable cell type composition and a preserved stem and progenitor niche. Upon metastatic colonization, we report on increased immune heter
