BACKGROUND: Acute undifferentiated febrile illness (AUFI) represents a major health challenge in tropical regions due to its wide range of etiologies. In Villeta, Colombia, previous studies investigated common causes such as malaria, arboviral diseases, leptospirosis and rickettsiosis, as well as several neglected bacterial agents. However, some patients remained without an identified etiology, underscoring the need for broader approaches to uncover other potential causes. Therefore, the aim of the present study was to investigate into other potential bacterial causes of AUFI through advanced

Spirochete pathogens are among the most invasive bacteria known, causing syphilis, Lyme disease, and leptospirosis. Their tissue penetration depends on periplasmic flagellar filaments that, unlike other bacterial flagella, are encased in a spirochete-specific multi-protein sheath and deform the cell body into motile waves. How these filaments achieve the mechanical properties needed for invasive motility has remained unclear. Here we determine complete atomic structures of the Leptospira endoflagellar filament, revealing an elaborate sheath of 9 to 12 distinct asymmetrically arranged proteins.

BACKGROUND: Leptospirosis is a globally important zoonosis maintained by chronically infected animal reservoirs, yet its long-term eco-epidemiological dynamics remain poorly understood. Southeast China has historically been an endemic region, but changing climate and land use may be reshaping Leptospira transmission.
METHODS: We conducted 15-year longitudinal surveillance (2009-2024) in Pan'an County, Zhejiang Province, integrating field sampling of rodents and other hosts, serogroup identification, multilocus sequence typing, and experimental mouse infection. Annual rainfall, host data, and

Leptospirosis is a zoonotic tropical disease with worldwide distribution caused by bacteria from the genus Leptospira. The clinical picture is assorted, from asymptomatic subclinical disease to more severe forms affecting several organs, including lungs, liver, and kidneys. Neurologic manifestations account for 10%-20% of the cases. The most common neurological presentation is aseptic meningitis, but other syndromes, such as cranial nerve palsies, and encephalitis, are also seen. To our knowledge, this is the first reported case of this type of neurological presentation of leptospirosis in Pan

Leptospirosis is a globally distributed zoonotic disease caused by pathogenic bacteria of the Leptospira genus. Currently, 77 genomic species have been described. Leptospira interrogans is the most extensively studied species due to its high prevalence worldwide and the severity of the disease it causes in humans and animals. However, Leptospira santarosai is an important pathogenic species in the Americas, the Caribbean islands, and Taiwan. This species has a high serological diversity: it can infect domestic, wild, and agricultural production animals, causing reproductive problems and substa

BACKGROUND: Leptospirosis has been infrequently associated with persistent kidney dysfunction fulfilling the criteria for chronic kidney disease (CKD). However, the entity remains poorly characterised. This study aimed to describe the clinical profile, laboratory findings, treatment requirements, and kidney outcomes of patients without recognised CKD risk factors who had persistent kidney dysfunction fulfilling CKD criteria at three-month follow-up after leptospirosis-associated Acute Kidney Injury (AKI).
METHODS: This descriptive case series was nested within a prospective cohort of hospital

Studies on the global burden of sepsis confirm its major mortality, morbidity, and societal impact, with 85% of cases and deaths occurring in low-income and middle-income countries. Importantly, these analyses have blurred the boundaries between sepsis, defined as a dysregulated host response to infection causing life-threatening organ dysfunction, and endemic and emerging infectious diseases, such as malaria, leptospirosis, tuberculosis, and gastroenteric illnesses, which often follow clinical trajectories distinct from those of so-called standard sepsis. Framing these disease-specific deaths

Leptospira is a bacterial genus which includes several pathogenic species. Wild mammals can act as reservoir hosts, shedding bacteria in their urine. Leptospirosis is an important health problem in Villeta, but data regarding potential reservoirs hosts and natural sources of infection are still scarce. We aimed to detect and characterize the presence of Leptospira among small wild mammals from Villeta municipality, Colombia. Small wild mammals from three orders: Didelphimorphia, Chiroptera and Rodentia, were sampled in the region. DNA was extracted from kidney samples and screened for Leptospi

Leptospirosis, a global health concern caused by Leptospira sp. is largely unknown due to its elusive phylogeographic distribution and flow. Therefore, this study aimed to understand the geographical distribution and evolutionary changes of pathogenic, non-pathogenic, and opportunistic strains of Leptospira. This study used genome-scale data, 16S rDNA gene sequences, and virulence proteins to construct precise phylogenetic trees using robust bootstrapping techniques. Phylogenomic studies indicated that pathogenic Leptospira strains constituted a distinct cluster from opportunistic and non-path

BACKGROUND: Nepal relies on an agrarian-based economy, with the livestock sector contributing significantly to the national GDP. However, diseases like leptospirosis negatively impact cattle production and pose significant zoonotic risks. This study represents the first attempt to evaluate the risk factors of leptospirosis in cattle in Nepal. A cross-sectional study was conducted from March 2019 to April 2020 in 14 administrative units of the Rupandehi district. A total of 367 blood samples were collected from 206 cattle farms using a proportionate sampling procedure. An indirect ELISA was use

INTRODUCTION: Leptospirosis, caused by Leptospira spp., is one of the most widespread zoonoses worldwide. It affects both domestic and wild animals, with ruminants serving as a primary reservoir for serovar Hardjo. This serovar causes long-term colonisation of the kidney and genital tract. Hardjo strains are taxonomically assigned to two Leptospira species: Leptospira interrogans and Leptospira borgpetersenii. However, the molecular basis of L. interrogans serovar Hardjo adaptation remains poorly understood. Comparative genomic analysis of L. interrogans strains classified as the Hardjo serova

Leptospirosis is a zoonotic disease that affects multiple hosts, with dogs being highly susceptible and frequently developing renal impairment. Leptospira biofilms have been documented in the environment, chronic hosts, and in vitro; however, their role in acute infections remains unclear. Given its association with antimicrobial tolerance and bacterial persistence, biofilm formation poses a One Health concern at the human-animal-environment interface. This study aimed to investigate Leptospira biofilms in the renal parenchyma of naturally infected dogs with acute leptospirosis and to evaluate

Here, we present an integrated suite of protocols for the cultivation, quantitative monitoring, structural and functional characterization of Leptospira spp. biofilms over time. This workflow combines crystal violet microtiter assays to measure biofilm biomass at multiple time points, with a time-resolved fractionation approach that distinguishes attached (biofilm) and non-attached (liquid-phase) bacterial populations, time-lapse phase-contrast imaging for non-destructive kinetic observation, confocal laser-scanning microscopy to generate full 3D reconstructions with matrix-probe readouts, and

BACKGROUND: Leptospirosis is an anthropozoonosis that occurs worldwide but is more common in tropical regions. Severe forms may require intensive care unit (ICU) admission. Whether the clinical patterns and outcomes differ between tropical and non-tropical regions with similar healthcare systems is unclear. Our objective here was to address this issue by comparing two cohorts of ICU patients with leptospirosis managed in mainland France and in the overseas French department of Réunion, respectively.
METHODOLOGY/PRINCIPAL FINDINGS: We compared two retrospective cohorts of patients admitted to

Leptospira is a bacterial genus that includes several pathogenic species related to leptospirosis. In Colombia, leptospirosis is a mandatorily reported disease, widely distributed across the country. In the Villeta municipality, leptospirosis has been identified as an important cause of febrile illness; however, to date, no studies have been performed to identify the circulating species. A genus-specific qualitative qPCR was performed on DNA extracted from febrile patients' acute-phase whole-blood samples targeting a fragment of the rrs gene. Positive qPCR samples were further amplified for th

BACKGROUND: Serology (the detection of antibodies formed by the host against an infecting pathogen) is frequently used to assess current infections and past exposure to specific pathogens. However, the presence of cross-reactivity among host antibodies in serological data makes it challenging to interpret the patterns and draw reliable conclusions about the infecting pathogen or strain.
METHODOLOGY/PRINCIPAL FINDINGS: In our study, we use microscopic agglutination test (MAT) serological data from three host species [California sea lion (Zalophus californianus), island fox (Urocyon littoralis)

INTRODUCTION: Leptospirosis is considered one of the most common zoonotic infections in the world and is characterized by distribution in countries of subtropical and tropical zones. In Georgia A sharp increase in the frequency of leptospirosis began in 2011, and the morbidity rate increased to 16.3. Also, the landscape of detected serovars changed during this period: L.Pomona, L.icterohemorragiae, L.grippotyphosa, L.hebdomadis, among them the most frequently registered - L.pomona -45%.

UNLABELLED: Spirochete bacteria cause Lyme disease, leptospirosis, syphilis and several other human illnesses. Unlike other bacteria, spirochete flagella are enclosed within the periplasmic space where the filaments distort and push the cell body by action of the flagellar motors. We previously demonstrated that the oral pathogen Treponema denticola (Td) catalyzes the formation of covalent lysinoalanine (Lal) crosslinks between conserved cysteine and lysine residues of the FlgE protein that composes the flagellar hook. Although not necessary for hook assembly, Lal is required for motility of T
