BACKGROUND/OBJECTIVES: Epidermoid cysts are benign lesions originating from germinative epithelium, characterized by a keratin-filled cavity. They are histopathologically classified as epidermoid, dermoid, or teratoid. Intraoral cases are exceedingly rare, comprising less than 0.01% of all oral cystic lesions, most frequently affecting the floor of the mouth. While usually asymptomatic, they may become painful due to infection or growth. Because of their rarity in sites such as the upper lip, they may be clinically misdiagnosed, making awareness crucial for accurate management.

The DICER1 gene, essential for microRNA biogenesis and posttranscriptional gene regulation, has been implicated in a variety of benign and malignant neoplasms, particularly within the context of the DICER1-related tumor predisposition syndrome. While DICER1-associated rhabdomyosarcomas (RMS) are predominantly documented in the genitourinary tract, we present the first case of a DICER1-mutated embryonal-like (botryoid-like) RMS of the nasal fossa. A 63-year-old woman without relevant family history presented with nasal obstruction, headaches, and epistaxis and underwent resection of a polypoid

BACKGROUND: Transcriptomic analysis is common in large cohort studies but is generally restricted to cells in blood, which limits inferences about organs of interest, and direct organ sampling is mostly infeasible in large cohorts. New techniques for RNA-seq from noninvasive biosamples may provide the opportunity to profile transcriptomes of additional tissues for more organ-relevant insights at scale. We investigated the feasibility and utility of hair follicle gene expression profiling in a multi-center study of chronic obstructive pulmonary disease (COPD).
METHODS: Bulk RNA-seq was perform

Head and neck squamous cell carcinoma (HNSCC) is the most common and aggressive histologic subtype of head and neck cancer (HNC), difficult to treat effectively. Here, we discuss several studies on human and mouse HNSCC cell lines arising from the mucosal epithelium of various anatomical sites, as well as recent studies using murine models, focused on targeting key checkpoints in the glutamine (Gln) metabolism pathway, either alone or in synergy with other signaling pathways, as a potential therapeutic strategy for HNSCC. Emerging evidence demonstrates a complex interplay between Gln metabolis

The proper function of the lower urinary tract depends on its ability to sense and react to mechanical forces as urine is produced, transported, stored, and eliminated; however, our current understanding of the mechanosensors involved in these events is limited. TMEM63 ion channels are reported to function as mechanosensors/osmosensors in other organs, and our studies revealed that the primary site of Tmem63a and Tmem63b gene expression and TMEM63B protein expression in the mouse bladder wall was the urothelium. Despite this localization, voiding behavior in conditional urothelial Tmem63b knoc

Choristomas are rare, benign embryonic tumors characterized by normal tissue appearing in abnormal sites. When lined with respiratory epithelium, they are classified as bronchogenic cysts. While these cysts most commonly occur in the mediastinum or other thoracic regions, their occurrence on the tongue is exceedingly rare. Here, we present the case of a four-year-old boy diagnosed with a bronchogenic cyst of the tongue, highlighting its clinical and histological features, diagnosis, and management, along with a review of the literature.

Introduction: Epidermoid cysts are benign lesions that may occur in many different sites of the body. They are classified into 3 types: epidermoid cysts when the lining presents only epithelium, dermoid cysts when skin adnexa are found, and teratoid cysts when mesodermal elements are present. Case Presentation: A 13-year-old boy presented with a gradually enlarging sublingual mass over a period of 4 years. The mass was painless at start but in the last 3 months, it was associated with pain and difficulties during eating and dysarthria. The general examination was unremarkable and examination o

BACKGROUND: Sporadic measles outbreaks have continued to occur in Korea, mainly in adults in their 20s and 30s, most notably in 2014 and 2019. We here evaluated the possible causes of a low seroprevalance of measles by testing young healthcare workers (HCWs).
MATERIALS AND METHODS: This study was conducted in a 2,743-bed tertiary-care hospital in Seoul between 2020 and 2021. We performed a measles antibody test (chemiluminescence immunoassay), measured the IgM/IgG index ratio, and conducted an avidity test at 1-month after Measles-Mumps-Rubella (MMR) vaccination in HCWs who had been seronegat

CONTEXT: Yolk sac tumor (YST) is a germ cell tumor, arising commonly from the gonads and rarely from other midline sites. Somatic Yolk sac tumors, arising from somatic malignancy, have been rarely reported in the gastrointestinal tract, gynecologic tract, and recently, in the urinary tract.
OBJECTIVE: We share our institutional experience of somatic YST arising from the urothelium.
DESIGN: Electronic medical records (EMR) were searched for cases of somatic YST differentiation in the urinary tract. Clinico-pathologic parameters were collected and reviewed, including clinical presentation, his

Dilated pore of Winer (DPW) is a benign follicular adnexal tumor that typically occurs in middle-aged and elderly individuals, most commonly on the face and neck. Rarely, it can present at unusual sites or in younger patients, leading to diagnostic challenges. We present the case of a 28-year-old female with a solitary, asymptomatic, comedo-like lesion on the right thigh that had been slowly enlarging over two years. Histological evaluation following surgical excision demonstrated a dilated follicular infundibulum filled with keratin and lined by acanthotic epithelium with radiating strands, c

The success and sustainability of U.S. EPA efforts to reduce, refine, and replace in vivo animal testing depends on the ability to translate toxicokinetic and toxicodynamic data from in vitro and in silico new approach methods (NAMs) to human-relevant exposures and health outcomes. Organotypic culture models employing primary human cells enable consideration of human health effects and inter-individual variability but present significant challenges for test method standardization, transferability, and validation. Increasing confidence in the information provided by these in vitro NAMs requires

The mammalian kidney achieves massive parallelization of function by exponentially duplicating nephron-forming niches during development. Each niche caps a tip of the ureteric bud epithelium (the future urinary collecting duct tree) as it undergoes branching morphogenesis, while nephron progenitors within niches balance self-renewal and differentiation to early nephron cells. Nephron formation rate approximately matches branching rate over a large fraction of mouse gestation, yet the nature of this apparent pace-maker is unknown. Here we correlate spatial transcriptomics data with branching 'l

R-loops that accumulate at transcription sites pose a persistent threat to genome integrity. PSIP1 is a chromatin protein associated with transcriptional elongation complex, possesses histone chaperone activity, and is implicated in recruiting RNA processing and DNA repair factors to transcription sites. Here, we show that PSIP1 interacts with R-loops and other proteins involved in R-loop homeostasis, including PARP1. Genome-wide mapping of PSIP1, R-loops and γ-H2AX in PSIP1-depleted human and mouse cell lines revealed an accumulation of R-loops and DNA damage at gene promoters in the absence

Outer retinal degenerations, including age-related macular degeneration (AMD), are characterized by photoreceptor and retinal pigment epithelium (RPE) atrophy. In these blinding diseases, macrophages accumulate at atrophic sites, but their ontogeny and niche specialization remain poorly understood, especially in humans. We uncovered a unique profile of microglia, marked by galectin-3 upregulation, at atrophic sites in mouse models of retinal degeneration and human AMD. In disease models, conditional deletion of galectin-3 in microglia led to phagocytosis defects and consequent augmented photor

The barrier function of the epidermis poses a significant challenge to nanoparticle-mediated topical delivery. A key factor in this barrier function is the thickness of the stratum corneum (SC) layer within the epidermis, which varies across different anatomical sites. The epidermis from the palms and soles, for instance, have thicker SC compared to those from other areas. Previous studies have attempted to bypass the SC layer for nanoparticle penetration by using physical disruption; however, these studies have mostly focused on non-thick skin. In this study, we investigate the role of SC-dis

Physiologically, claudin 18 splice variant 2 (CLDN18.2) expression is restricted to the gastric epithelium, but its expression has been detected in solid cancers. Zolbetuximab, a chimeric IgG1 antibody targeting CLDN18.2, has demonstrated promising effects in patients suffering from CLDN18.2-positive, HER2-negative locally advanced gastric cancer and is currently being studied further. To date, little is known about CLDN18.2 expression in other histological subtypes of tubo-ovarian carcinoma (TOC) and their matching metastases.Using a cohort of all histological TOC subtypes, we investigated th

The dynamic adhesion between cells and their extracellular matrix is essential for the development and function of organs. During insect wing development, two epithelial sheets contact each other at their basal sites through the interaction of βPS integrins with the extracellular matrix. We report that Osiris17 contributes to the maintenance of βPS integrins localization and function in developing wing of Drosophila and locust. In flies with reduced Osiris17 expression the epithelia sheets fail to maintain the integrity of basal cytoplasmic junctional bridges and basal adhesion. In contrast to
