Identification and validation of novel reference genes for bovine respiratory and lymphoid tissues using public transcriptomes and BRSV challenge model.

RT-qPCR can be employed to quantify target gene expression normalized to reference genes that are expected to remain stable across experimental conditions. However, previously reported reference genes may not be stably expressed across all experimental conditions and individuals. This research identified novel candidate reference genes from publicly available healthy bovine tissue transcriptomes and subsequently developed and validated primers for five candidate reference genes. Forty-one transcriptomes were obtained from the NCBI Gene Expression Omnibus (GEO) from apparently healthy Bos tauru

dbGVOSCC: a comprehensive database of genetic variation for systems genetics research on oral squamous cell carcinoma.

INTRODUCTION: Oral squamous cell carcinoma (OSCC) is a highly aggressive malignancy of the oral epithelium, marked by a high rate of lymph node metastasis and a profound negative impact on patients' quality of life. Despite its severity, no routine screening program currently exists for OSCC. To address the genetic heterogeneity underlying OSCC, we have developed a database of genetic variation in oral squamous cell carcinoma (dbGVOSCC; http://www.sysbio.org.cn/dbGVOSCC/). METHODS: OSCC literature (1991-2024) was queried from PubMed and screened manually and via PubTator, following predefined

Prediction of heart failure risk factors from retinal optical imaging via explainable machine learning.

Over 64 million people worldwide are affected by heart failure (HF), a condition that significantly raises mortality and medical expenses. In this study, we explore the potential of retinal optical coherence tomography (OCT) features as non-invasive biomarkers for the classification of heart failure subtypes: left ventricular heart failure (LVHF), congestive heart failure (CHF), and unspecified heart failure (UHF). By analyzing retinal measurements from the left eye, right eye, and both eyes, we aim to investigate the relationship between ocular indicators and heart failure using machine learn

3D Flipwell Engineering for Developing Asynchronous Systems for Toxicologic and Immunomodulatory Therapies in Bacterial, Gut, and Immune Cells.

To study the roles of bacterial secretome in immune cell polarization and gut mucosal homeostasis, we developed a 3-dimensional co-culture model using commercially available inserts in a back-to-back stacked format. We termed this system 3D Flipwells, and successfully co-cultured bacteria, colon epithelial, and immune cells in stratified layers to recapitulate the gut mucosal microenvironment. This co-culture system enables the seeding of colon epithelial cells on one side of the membrane and undifferentiated non-adherent monocytes on the other, allowing for the synchronous progression of colo

Giant mucinous borderline tumor of the ovary in a postmenopausal woman: A rare case report and review of the literature.

INTRODUCTION AND IMPORTANCE: Mucinous cystadenoma is a benign cystic ovarian tumor arising from the surface epithelium of the ovary that usually presents with vague, unspecified abdominal symptoms. If not detected early, this tumor has the potential to grow to a substantial size and can present with huge abdominal distention leading to various compression symptoms. The reported incidence of giant ovarian cystadenoma in postmenopausal women is relatively unknown due to the widespread use of ultrasound and other radiological imaging modalities. CASE PRESENTATION: Here, we present a rare case of

Dacryolithogenesis or Dacryolithiasis-The Story So Far.

The term dacryoliths refers to the concretions found within the lacrimal system. When the term dacryoliths is unspecified, it usually refers to the noninfectious dacryoliths commonly isolated from the lacrimal sac and the nasolacrimal duct. More often, they are diagnosed incidentally during a dacryocystorhinostomy, and the reported incidence among all dacryocystorhinostomy surgeries is 5.7% to as high as 18%. Dacryolithiasis is a complex process occurring within the lacrimal system, and current evidence suggests a multifactorial etiology. The sequence of events can be summarized broadly into 4

A novel quantification method for retinal pigment epithelium phagocytosis using a very-long-chain polyunsaturated fatty acids-based strategy.

INTRODUCTION: The vertebrate retinal pigment epithelium (RPE) lies adjacent to the photoreceptors and is responsible for the engulfment and degradation of shed photoreceptor outer segment fragments (POS) through receptor-mediated phagocytosis. Phagocytosis of POS is critical for maintaining photoreceptor function and is a key indicator of RPE functionality. Popular established methods to assess RPE phagocytosis rely mainly on quantifying POS proteins, especially their most abundant protein rhodopsin, or on fluorescent dye conjugation of bulk, unspecified POS components. While these approaches

Diagnosis of Metastatic Carcinoma Using Body Cavity Fluid Specimens: A Comparison of Diagnostic Panels.

INTRODUCTION: Body cavity effusions are routinely used as cytologic specimens. The distinction between metastatic carcinoma, mesothelioma, and reactive mesothelial cells remains a major challenge. Immunohistochemistry (IHC) is a supplemental method that can aid in diagnosis and often involves many markers as part of an IHC panel. Several immunohistochemical markers are now widely used. This study aims to determine the optimal immunomarkers and IHC panels to differentiate reactive mesothelial cells from metastatic cancer in body cavity fluid samples. METHODS: IHC was performed for claudin-4, M

Clinical and Histologic Features Associated With Malignant Transformation of Oral Cavity Dysplasia Managed at a Single North American Institution.

IMPORTANCE: Reports characterizing clinical and histologic features associated with a higher risk for development of malignant lesions in the background of an oral potentially malignant disorder have largely reflected East Asian populations. Long-term studies among the North American population are rare. OBJECTIVE: To evaluate risk of malignant transformation (MT) of oral dysplastic lesions by investigating the demographic, social, clinical, and histologic factors that may be associated with an increased rate as well as a decreased time to MT. DESIGN, SETTING, AND PARTICIPANTS: This was a re

Characterization of a flexible AAV-DTR/DT mouse model of acute epithelial lung injury.

Animal models are important to mimic certain pathways or biological aspects of human pathologies including acute and chronic pulmonary diseases. We developed a novel and flexible mouse model of acute epithelial lung injury based on adeno-associated virus (AAV) variant 6.2-mediated expression of the human diphtheria toxin receptor (DTR). Following intratracheal administration of diphtheria toxin (DT), a cell-specific death of bronchial and alveolar epithelial cells can be observed. In contrast to other lung injury models, the here described mouse model provides the possibility of targeted injur

Epithelial expression of Gata4 and Sox2 regulates specification of the squamous-columnar junction via MAPK/ERK signaling in mice.

The squamous-columnar junction (SCJ) is a boundary consisting of precisely positioned transitional epithelium between the squamous and columnar epithelium. Transitional epithelium is a hotspot for precancerous lesions, and is therefore clinically important; however, the origins and physiological properties of transitional epithelium have not been fully elucidated. Here, by using mouse genetics, lineage tracing, and organoid culture, we examine the development of the SCJ in the mouse stomach, and thus define the unique features of transitional epithelium. We find that two transcription factors,

Multinucleate parietal cells and cytoplasmic inclusion bodies in the gastric epithelium of callitrichids.

Inclusion bodies (IBs) and multinucleate cells can be associated with viral infections; however, IBs and multinucleate cells have been described in normal tissue and with non-viral disease processes in multiple species. We examined fundic stomach from 50 callitrichids histologically for bi- and multinucleate parietal cells and cytoplasmic IBs in gastric epithelial cells. Callitrichids represented included 6 genera: Saguinus (4 spp.), Leontopithecus (1 sp.), Mico (3 spp.), Cebuella (1 sp.), Callithrix (1 sp.), Callimico (1 sp.), and 13 unspecified marmosets. Gastric epithelial IBs were present

The development and stem cells of the esophagus.

The esophagus is derived from the anterior portion of the foregut endoderm, which also gives rise to the respiratory system. As it develops, the esophageal lining is transformed from a simple columnar epithelium into a stratified squamous cell layer, accompanied by the replacement of unspecified mesenchyme with layers of muscle cells. Studies in animal models have provided significant insights into the roles of various signaling pathways in esophageal development. More recent studies using human pluripotent stem cells (hPSCs) further demonstrate that some of these signaling pathways are conser

A survey of proteins in midgut contents of the tick, Haemaphysalis flava, by proteome and transcriptome analysis.

Tick blood meals are stored and digested in their midguts. Blood digestion is complex, and many proteins are involved. Study of the tick-derived proteins in the midgut content may aid in the discovery of active molecules that would be useful for anti-tick vaccines. We analyzed the midgut content proteomes of partially engorged female Haemaphysalis flava, fully engorged female H. flava, and hedgehog serum using liquid chromatography tandem-mass spectrometry and label-free quantitation. In this study, high-confidence protein profiling of tick midgut content was determined. Based on the search ag

Modified Fusarium Mycotoxins in Cereals and Their Products-Metabolism, Occurrence, and Toxicity: An Updated Review.

Mycotoxins are secondary fungal metabolites, toxic to humans, animals and plants. Under the influence of various factors, mycotoxins may undergo modifications of their chemical structure. One of the methods of mycotoxin modification is a transformation occurring in plant cells or under the influence of fungal enzymes. This paper reviews the current knowledge on the natural occurrence of the most important trichothecenes and zearalenone in cereals/cereal products, their metabolism, and the potential toxicity of the metabolites. Only very limited data are available for the majority of the identi
