Molecular evolution and mechanisms underlying extremely high-level carbapenem resistance in the emerging CRPA clone ST1976.

BACKGROUND: Carbapenem-resistant Pseudomonas aeruginosa (CRPA) represents a critical threat to clinical therapy, yet the genetic basis underlying extremely high-level carbapenem resistance remains poorly understood. An emerging CRPA clone, ST1976, has recently been identified in China with unusually high meropenem MICs. METHODS: ST1976 CRPA isolates collected between 2018 and 2022 were subjected to antimicrobial susceptibility testing, whole-genome and Nanopore sequencing, and bioinformatic analyses. Cloning assays and sodium mercaptoacetic acid inhibition tests were performed to investigate

High prevalence of carbapenem-resistant Pseudomonas spp. in the pork production chain: a potential reservoir for zoonotic transmission.

Multidrug-resistant Pseudomonas aeruginosa is currently one of the primary threats to human health, accounting for a significant proportion of difficult-to-treat infections globally. Although pork meat is frequently contaminated with Pseudomonas spp., well-known spoilage organisms, their potential role as reservoirs of antibiotic-resistant strains and associated risks to human health remains unclear. This study investigated the prevalence of carbapenem-resistant Pseudomonas spp. (CRP) throughout the same pork processing chain, and assessed their pathogenicity and genetic relatedness. We sample

Therapeutic significance of surface-exposed virulence factors in Acinetobacter baumannii pathogenicity.

Acinetobacter baumannii is a rapidly evolving opportunistic pathogen that has developed strategies to resist multiple antimicrobials and evade the host immune system. Its global emergence as a multidrug-resistant (MDR) pathogen has severely limited treatment options and increased mortality in hospital settings. Virulence factors associated with the cell envelope and outer membrane vesicles play central roles in bacterial survival and pathogenicity. In this context, this review aims to provide a concise overview of the virulence apparatus present on the surface of A. baumannii and evaluate its

Frontiers in aptamer-based diagnostics and therapeutics for zoonotic diseases.

Zoonotic diseases have become a significant public health concern due to their capacity to cross species barriers and cause widespread outbreaks. Conventional diagnostic and therapeutic strategies often lack the specificity, sensitivity, and rapid adaptability required for effective disease management. Therefore, aptamers, single-stranded oligonucleotides, have emerged as powerful tools in detection and therapeutics. This review elucidates the role of aptamer-based technologies in diagnosing and treating bacterial, viral, and parasitic zoonoses, including Leptospira, Helicobacter pylori, rabie

Ehrlichia ruminantium infection is associated with tissue-specific microbial community shifts in Amblyomma gemma ticks from cattle in Kenya.

Tick-borne pathogens can reshape vector microbiomes in ways that influence pathogen colonisation and transmission, yet the interplay between Ehrlichia ruminantium and the microbiota of its tick vectors remains uncharacterised. We profiled bacterial communities in haemolymph, midgut, and salivary glands of infected (n = 11) and uninfected (n = 12) Am. gemma ticks, a vector of E. ruminantium in East Africa, collected from cattle in Kajiado County, Kenya, using near-full-length 16S rRNA gene amplicon sequencing on the Oxford Nanopore platform. Community composition, alpha and beta diversity, co-o

Rickettsiosis: existing advances and future opportunities for the development of vaccines.

Rickettsioses is a zoonotic disease caused by an intracellular bacterium transmitted through the bites of hematophagous arthropods. While antibiotics can effectively treat this disease, overuse may lead to the development of antibiotic resistance. To combat this issue, vaccine development has been pursued to address this bacterial infection. The types of vaccines formulated against Rickettsia include inactivated, live-attenuated, subunit, DNA, and nanovaccines, all of which induced immune responses, and some demonstrated protective efficacy against infectious challenges. These outcomes have en

Carbapenem-Resistant Acinetobacter baumannii: Resistance, Virulence, and Possible Dissemination Between Different Hospitals in Western Iran.

The capacity for biofilm formation, along with the acquisition of antimicrobial resistance, could be the primary factors enhancing the persistence and spread of Acinetobacter baumannii (AB). We aimed to identify the trends in molecular epidemiology of AB isolated from western Iran. Ninety-nine pathogenic AB isolates were collected from patients in three major hospitals in Sanandaj and Kermanshah (the capitals of Kurdistan and Kermanshah provinces in western Iran, respectively) in one year. Isolates were tested for the presence of class D and B carbapenemases, integrons, and biofilm-associated

Co-occurrence of resistance and virulence plasmids in a blaNDM-5-positive Escherichia coli ST602 clone across One Health sectors in China.

Carbapenem-resistant bacteria circulating across human, animal, and environmental sectors are a growing public health concern. We investigated blaNDM-5-positive Escherichia coli ST602 from an integrated duck-egg farming ecosystem in China using whole-genome sequencing with plasmid characterization. Among 44 representative carbapenem-resistant E. coli (CREC) isolates selected from a larger collection of 103 CREC isolates, seven belonged to ST602 and formed a genetically related O103:H21 cluster recovered from human, duck, and environmental samples within the same farm ecosystem. To place these

Functional characterization of the oligopeptide transporter operon oppABCDF-1 in Escherichia coli Nissle 1917.

BACKGROUND: The ABC-type oligopeptide transporter (Opp system), composed of five subunits (OppA-F), plays an essential role in microbial fitness and survival by mediating the uptake of short peptides. Paradoxically, although the probiotic strain Escherichia coli Nissle 1917 (EcN) harbors multiple Opp systems, it exhibits a defect in the import of oligopeptides comprising three or more amino acids, a limitation that may compromise its ability to colonize the intestinal tract. This functional paradox underscores the need for further investigation into the substrate specificity and physiological

Infection with multiple strains of the Lyme disease bacterium in female mice can affect the strain-specific maternal antibody response in their offspring.

BACKGROUND: The tick-borne spirochete Borrelia burgdorferi causes Lyme borreliosis in North America. In nature, rodent reservoir hosts are often infected with multiple strains of B. burgdorferi, which induce strain-specific antibody responses that are mediated by outer surface protein C (OspC). During pregnancy and lactation, female rodents infected with B. burgdorferi will transfer a finite amount of strain-specific maternal antibodies to their offspring. The purpose of our study was to investigate whether infection with multiple strains of B. burgdorferi in female mice reduces the per strain

Bartonella transmission and gut microbiome dynamics in Ceratophyllus sciurorum fleas and their edible dormouse hosts (Glis glis).

BACKGROUND: The genus Bartonella comprises facultative intraerythrocytic bacteria capable of causing long-lasting bacteremia in their natural hosts, with zoonotic potential across multiple species. Rodents serve as important reservoirs for a broad diversity of Bartonella spp., with blood-feeding arthropods, particularly fleas, mediating transmission. Despite their frequent association with humans, the role of edible dormice (Glis glis) and their fleas (Ceratophyllus sciurorum) in Bartonella ecology remains poorly understood. METHODS: We combined long-term ecological and epidemiological data w

Zoological gardens as a neglected One Health interface for tigecycline-resistant Escherichia coli.

The tigecycline resistance gene tet(X4) is increasingly reported from food-producing animals and humans, but its circulation at wildlife-environment interfaces, including zoos, remains poorly resolved. Here, we combined systematic sampling of zoo animals and zoo-associated environments with comparative genomics against a global collection of 1102 tet(X)-positive Escherichia coli genomes to characterize the occurrence, genomic context and potential dissemination of tet(X4) in zoological settings. We recovered 60 tet(X4)-positive isolates (58 E. coli and two Enterobacter hormaechei) from 188 sam
