Phenotypic and Genomic Characterization of Novel Straboviridae Bacteriophages Targeting Multidrug-Resistant Salmonella enterica subspecies enterica Serovar Enteritidis.

Salmonella enterica serovar Enteritidis is a leading cause of foodborne zoonoses worldwide. The rapid emergence of multidrug-resistant (MDR) strains has compromised traditional antimicrobial therapies, necessitating the development of biosafe alternatives such as bacteriophages. This study aimed to isolate and comprehensively characterize novel lytic bacteriophages targeting multidrug-resistant Salmonella enterica subspecies enterica serovar Enteritidis isolates from Lebanon. In this study, four novel Salmonella phages, EDA02, EDA03, EDA05, and EDA06, were isolated from wastewater and poultry

Current perspectives on avian tuberculosis in domestic and wild birds: Molecular diagnostics, genomic surveillance, and vaccination challenges.

Avian tuberculosis (ATB) is a chronic infectious disease of poultry caused by Mycobacterium avium subsp. avium and is widespread in various regions of the world. This disease has a serious impact on poultry health and causes significant economic losses, primarily due to decreased body condition, reduced egg production, and increased mortality, particularly in young birds or individuals with compromised immune systems. Furthermore, ATB poses an opportunistic zoonotic risk, particularly for immunocompromised humans, highlighting its relevance within the One Health framework. This review aims to

Genomic outbreak investigation of biosafety-level-3 pathogens using nanopore sequencing.

Background. Accurate outbreak analysis is essential for effective infectious disease control. While short-read Illumina sequencing is the current gold standard for genotyping pathogens, Oxford Nanopore Technologies (ONT) offers advantages such as portability and real-time sequencing. However, the accuracy of ONT for single nucleotide polymorphisms (SNP) detection and core genome multilocus sequence typing (cgMLST) remains poorly characterized, which is a critical issue when investigating outbreaks of highly dangerous biosafety-level-3 (BSL-3) agents.Aim. This study evaluates the potential of O

Dairy farm waste as a source of novel and globally disseminated multidrug-resistant Escherichia coli clones: A genomic and phylogeographic study.

Dairy farm waste may serve as a reservoir for multidrug-resistant (MDR) Escherichia coli clones, but the genomic characteristics and dissemination potential of such clones remain incompletely understood. Here, we performed whole-genome sequencing and comprehensive genomic analysis of 64 MDR E. coli strains isolated from feces and sewage samples collected from two large dairy farms in Gansu Province, China. Genomic analysis revealed that strains carried 16-32 antibiotic resistance genes (ARGs), 1-6 plasmid replicon types, and 26-96 virulence genes (VGs), with numerically higher (though not stat

In silico metabolic profiling of non-baumannii Acinetobacter species uncovers conserved functions and provides first evidence of siderophore biosynthetic genes in Acinetobacter junii.

The genus Acinetobacter is recognized for its metabolic versatility, which contributes to environmental persistence, virulence, and antimicrobial resistance. This study aimed to elucidate the metabolic pathways, particularly those associated with virulence and antimicrobial resistance (AMR), in non-baumannii Acinetobacter species. Genome-scale metabolic prediction of 19 Acinetobacter isolates identified 104 distinct pathways spanning 32 metabolic categories. Ninety-six pathways were conserved across all genomes, whereas a small subset exhibited species- or strain-specific distributions: four p

Global, regional, and national burden of tuberculosis and multidrug-resistant tuberculosis by HIV status, 1990-2023: a systematic analysis for the Global Burden of Disease Study 2023.

BACKGROUND: Tuberculosis (TB) is the leading global cause of death from a single infectious agent. Recent reductions in global health funding have threatened TB control, making comprehensive assessment of TB, HIV-related TB, and drug-resistant TB burdens before these disruptions essential for shaping effective responses. The WHO End TB Strategy sets targets of a 95% reduction in TB deaths and a 90% reduction in TB incidence between 2015 and 2035. Using results from the Global Burden of Diseases, Injuries, and Risk Factors Study (GBD) 2023, this study aims to assess the burden of TB and multidr

The cholesterol-dependent cytolysin promotes Streptococcus systemic spread and induces arachidonic acid accumulation-mediated lethality in a murine intraperitoneal infection model.

Streptococcal peritonitis is associated with a high mortality risk and sometimes accompanied by streptococcal toxic shock-like syndrome (STSLS). Here, we used a murine intraperitoneal model of Streptococcus suis serotype 2 (SS2) infection and applied bacterial lineage tracing to investigate the pathogen's population dynamics during streptococcal intraperitoneal infection. We found that the cholesterol-dependent cytolysin suilysin (SLY) was essential for acute lethality and facilitated bacterial systemic spread by mitigating infection bottlenecks and enabling vascular leakage in the peritoneal

iNOS/NO-Induced autophagy primes PANoptosis as the central driver of streptococcal toxic shock-like syndrome in zoonotic Streptococcus suis infection.

Streptococcus suis is a major zoonotic pathogen that causes human fulminant meningitis and streptococcal toxic shock-like syndrome (STSLS), the latter being driven by dysregulated inflammatory responses and explosive cytokine storms. It remains unclear whether PANoptosis acts as a coordinated host-protective response or instead serves as a detrimental driver of cytokine storm during bacterial infection. Here, we identify autophagy as an upstream orchestrator of PANoptosis during zoonotic Streptococcus suis infection, redefining STSLS as a disease driven by an integrated PANoptosis cell-death p

Research progress on Avibacterium paragallinarum and related bacterial and viral diseases in poultry and their mixed infections.

Avibacterium paragallinarum (A. paragallinarum) is one of the primary pathogens causing respiratory diseases in poultry flocks, posing a serious threat to the health and production efficiency of the global poultry industry. With the expansion of modern farming scales and changes in environmental factors, mixed infections involving A. paragallinarum alongside other bacteria and viruses have become increasingly prevalent. This has led to more complex and diverse disease manifestations, significantly increasing the difficulty of prevsention and control. Current research on A. paragallinarum encom

FlhF interacts with TonB2 to defend against oxidative stress in Campylobacter jejuni.

Campylobacter jejuni is the leading cause of bacterial foodborne diarrheal disease worldwide. Despite its microaerophilic nature, C. jejuni is ubiquitous in aerobic environments and must possess specific adaptation mechanisms against oxidative stress. Here, we identified a novel role for FlhF, a GTPase essential for proper flagellar assembly, in promoting resistance to hydrogen peroxide (H2O2). Comparative transcriptomic analysis under H2O2 stress revealed that deletion of flhF leads to significant downregulation of oxidative stress-related genes. FlhF directly interacts with TonB2, an iron tr

Human Q fever outbreak investigation and management in six European countries: a One Health compendium of practices and recommendations.

BACKGROUND: Q fever is a zoonotic disease caused by Coxiella burnetii, with domestic ruminants serving as the main reservoir. Infected animals can shed large quantities of the pathogen into the environment after normal parturition or abortion, enabling airborne transmission to humans and potentially triggering outbreaks of variable scale. Effective outbreak management requires the rapid identification of all related cases, tracing of infection source(s), and implementing proportionate control measures that protect human health while considering animal health and ecosystem integrity. OBJECTIVE

Ancient pathogen genomics in Africa: Current evidence and future directions.

Ancient pathogen genomics has redefined how infectious disease histories are reconstructed, revealing unexpected origins, transmission routes, and lineage turnovers that are invisible from modern genomes alone. Yet, this perspective remains heavily biased toward Eurasia, leaving Africa-central to human evolution and home to diverse ecological settings relevant to zoonotic and vector-borne disease emergence-largely unexplored. In this review, we assess the current state of ancient pathogen research in Africa and synthesize insights from bacterial, parasitic, and viral perspectives. We identify

Evaluation of the PA-100 AST system (Sysmex) for point-of-care analysis of bacteriuria and antibiotic susceptibility testing in dogs and cats.

Bacterial presence in urine (bacteriuria) is frequently observed in dogs and cats and may be associated with urinary tract infection (UTI), depending on the clinical context. However, no in-house test for the rapid detection of bacteriuria and subsequent antibiotic susceptibility tests (AST) is yet available. The Sysmex PA-100 AST System (PA-100) is a point-of-care (POC) analyzer using nanofluidic cartridges designed for the detection of bacteria and performance of AST directly from voided urine in female patients with cystitis symptoms. In this study, we tested the suitability of the PA-100 t
