The impact of STING signaling on myeloid cells highlights macrophages as the major player in controlling Brucella abortus.

INTRODUCTION: Brucellosis, caused by Brucella abortus (Ba), is one of the most common zoonoses worldwide. Some components of the bacteria can be recognized by the innate immune system and understanding these mechanisms is necessary for directing future vaccines and therapies against brucellosis. Our group has demonstrated that STING molecule contributes to host protection against Ba in vivo. Considering that Ba has ability to subvert the immune response, the study of the mechanisms of host cell activation becomes fundamental for the development of measures to control bacterial replication. Enzymatic Synthesis of 2-Amino-adenosine Triphosphate as a Noncanonical Nucleotide Precursor of Z-Modified RNA.

2-amino-deoxyadenosine (dZ) occurs naturally in certain bacteriophage genomes, where it replaces deoxyadenosine, forming three hydrogen bonds with thymidine. This noncanonical deoxyribonucleoside underlies the unique biophysical properties of dZ-DNA. Its corresponding ribonucleoside has been introduced to RNA to form Z-modified RNA, with promising applications in vaccine production and biomedicine. Unlike dZ-DNA, Z-modified RNA has only been synthesized in vitro, which required the addition of chemically synthesized 2-amino-adenosine triphosphate (ZTP) as a precursor. Here, we describe enzyme

Seasonal dynamics, antimicrobial resistance, and genetic lineages of thermotolerant Campylobacter isolates from South American camelids.

BACKGROUND: South American camelids (SACs) are increasingly kept as companion animals, but their role in zoonotic transmission is poorly understood. Thermotolerant Campylobacter spp. are a leading cause of bacterial zoonoses globally. This study investigated the occurrence and zoonotic potential of thermotolerant Campylobacter spp. in SACs on German farms. METHODS: Fecal swabs from up to 20 animals on 10 farms were collected across four seasons. Campylobacter spp. were isolated, and genomes were typed to assess multilocus sequence types (STs), virulence, and antimicrobial resistance. RESULTS

Vaccination against H5 HP avian influenza virus leads to persistent immune response in wild king penguins.

Since 2021, the panzootic nature of high pathogenicity avian influenza (HPAI) represents an increasing threat to wild vertebrate populations. In this context, recent vaccines developed for poultry could provide tools for the conservation of wild endangered birds populations. The king penguin (Aptenodytes patagonicus), a long-lived seabird breeding in dense colonies with an extended chick-rearing period, has been identified as a possible surrogate species for a vaccination trial in a sub-Antarctic natural setting. Here we investigate the immune response of king penguin chicks to a self-amplifyi

Exploring antibiotic stewardship interventions within a One Health context: a scoping review.

Antibiotic resistance (ABR) presents a global threat to human health, animal health, and the environment. While ABR is a natural phenomenon, the overuse and misuse of antibiotics in human health, animal health, and food production is a major driver of ABR. As interactions between humans, animals, and the environment are central to the emergence and spread of ABR, adopting a One Health approach is essential to effectively address the issue. In this context, a large range of antibiotic stewardship interventions has been developed to optimize the use of antibiotics. However, we lack a comprehensi

AddaVax-Adjuvanted Inactivated Vaccine Confers Protection Against Listeria monocytogenes Challenge in Mice.

Listeria monocytogenes (Lm) is a facultative intracellular pathogen responsible for life-threatening listeriosis, characterized by high mortality rates. Currently, there is no vaccine available for the prevention or treatment of listeriosis. Here, we developed two adjuvanted inactivated vaccines and evaluated their safety, immunogenicity, and protective efficacy. The hypervirulent Lm strain XYSN was inactivated with β-propiolactone (BPL) and formulated with AddaVax or Alum to produce inactivated vaccines (ADIV and ALIV) respectively. Local reactogenicity and histopathological observations reve

Detection of Zoonotic Photobacterium damselae Subspecies damselae in a Razorbill (Alca torda): The First Report of a Potential Cross-Species Transmission in Birds.

A razorbill (Alca torda) was found dead in Bacoli, Italy, on January 16, 2023, during an exceptional irruptive event. Post-mortem examination revealed coelomitis with severe congestion of the liver, lungs, kidneys, and myocardium. Bacterial isolation, MALDI-TOF MS analysis and PCR confirmed the presence of Photobacterium damselae subsp. damselae (Pdd), supported by 16S rDNA gene sequencing and detection of the ureC gene. PCR screening for virulence factors identified the hlyAch gene in lung samples, suggesting a potentially pathogenic strain in avian species. Histopathological examination show

Autophagy in emerging and highly concerned severe zoonotic infectious diseases.

Autophagy is a conserved cellular process that mediates degradation of damaged organelles, misfolded proteins, and invading pathogens, playing critical roles in intracellular homeostasis and immune regulation. Given that over 70% of infectious diseases and 60% of emerging infectious diseases are zoonotic, posing a major threat to global health, this review aims to summarize the cellular and molecular mechanisms underlying the crosstalk between autophagy and key zoonotic pathogens. We comprehensively retrieved relevant research literature from the PubMed, Web of Science, and Scopus databases (w

The Circular RNA hsa_circ_0004771 Regulates the Intracellular Survival of Mycobacterium tuberculosis in Macrophages by Targeting hsa-miR-3921 to Increase TREM1 Expression.

The interactions among the host's circular RNAs (circRNAs), microRNAs (miRNAs), and target genes are crucial for antibacterial resistance and intracellular pathogen clearance. However, this process remains poorly understood during Mycobacterium tuberculosis (M.tb) infection. Our previous study identified hsa_circ_000477, a novel circRNA formed by the NRIP1 gene on human chromosome 21, which was upregulated in M.tb-infected THP-1 macrophages. The present study systematically investigated the effect of hsa_circ_0004771 on M.tb infection and the underlying molecular mechanism. First, hsa_circ_000

Helicobacter mastomyrinus Cytolethal Distending Toxin Induces Calcium Dyshomeostasis and Liver Injury via Endoplasmic Reticulum Stress.

Helicobacter mastomyrinus (H.m), a Helicobacter species colonizing rodent liver and intestine, produces cytolethal distending toxin (CDT), which induces host cell damage. While the active CdtB subunit is linked to various diseases, the pathogenic mechanisms of H.m in the liver and the precise actions of CdtB remain incompletely defined. This study generated a CdtB-deficient strain (H.m ΔCdtB) and compared it with the wild-type H.m (H.m WT). Male BALB/c mice were orally infected via gavage. The impacts of CdtB on liver injury, inflammation, oxidative stress, endoplasmic reticulum (ER) stress, a

[Bacterial dermato-zoonoses].

The transmission of bacterial, viral, fungal, or parasitic agents from vertebrate animals to humans constitutes a group of diseases called zoonoses. The frequency of these zoonoses is increasing, and new zoonoses are emerging due to travel, factory farming, biodiversity loss, climate change, and deforestation. These diseases have a significant impact on public health, both human and animal, as well as on food, environmental management and the economy. This article reviews bacterial zoonoses with cutaneous manifestations, known as dermato-zoonoses. A second section will address viral, fungal, a

Comprehensive genomic analysis of multidrug-resistant Pseudomonas aeruginosa isolates from companion animals in Germany.

Pseudomonas (P.) aeruginosa is a highly adaptable, opportunistic nosocomial pathogen that poses significant risks to public health and veterinary medicine. This bacterium carries a wide range of antimicrobial resistance (AMR) determinants and produces various virulence factors that enable it to invade hosts and increase disease severity. Recognised as a One Health pathogen, P. aeruginosa can be isolated from multiple sources, including humans, animals, food, and the environment. Despite its importance in clinical settings, there are still limited genomic and epidemiological data on P. aerugino

Characterization of the phage ΦK64 depolymerase S2-4 and its therapeutic effect against K1 serotype Klebsiella pneumoniae.

The infection rate and drug resistance of Klebsiella pneumoniae containing capsular polysaccharides (CPSs) have been increasing annually, resulting in severe human and animal infections. Depolymerases derived from bacteriophages can degrade CPSs and thus hold potential for treating bacterial infections. However, little is known about the mechanism by which K. pneumoniae phage depolymerases hydrolyze CPSs. In this study, the S2-4 encoded by the phage ΦK64 was identified as a potent depolymerase against K1 serotype Klebsiella CPSs. Cryo-electron microscopy structural analysis revealed that S2-4

Emerging zoonotic risks: whole-genome sequencing reveals antimicrobial resistance and genomic diversity in Providencia stuartii isolated from broiler chickens in Noakhali, Bangladesh.

Providencia stuartii is emerging as an Extensively Drug-Resistant (XDR) pathogen commonly found in animals, insects, and in burned and immunocompromised conditions. The misuse of antibiotics in poultry feed causes the emergence of XDR bacteria in the poultry industry. The knowledge of zoonotic transmissibility of poultry-derived P. stuartii remains elusive in Noakhali, Bangladesh. Poultry fecal and rectal swab samples were collected from selected farms in Noakhali, Bangladesh. Bacterial isolation and identification were performed using MacConkey agar, biochemical tests, and 16S rRNA Sanger seq
