A microbiota-host axis mediates prostaglandin sensitivity: Lactobacillus crispatus as a biomarker and regulator of human labor induction.

Labor induction failure increases the risk of unplanned cesarean delivery and maternal-neonatal complications. However, the determinants of induction sensitivity remain poorly understood. In this prospective cohort of 85 term pregnant women undergoing labor induction with Propess® (prostaglandin E2), we combined full-length 16S rRNA sequencing of the vaginal microbiota, untargeted metabolomics of vaginal secretions, and transcriptomic analysis of cervical stromal cells exposed to Lactobacillus crispatus supernatant to identify predictive factors and underlying mechanisms. We found that women w

Development of Covalent Small-Molecule Fluorescent Probes for DNA Methyltransferase 1 Detection in Cancer Cells and Cervical Exfoliated Cells.

DNA methyltransferase 1 (DNMT1) overexpression is associated with aberrant methylation and tumorigenesis, making its detection vital for tumor diagnosis. In this study, RG108 derivatives bearing cysteine-targeted covalent moieties were constructed as warheads for the DNMT1 detectors. Following affinity assessment by surface plasmon resonance, warheads containing a 2-fluoroacrylamido moiety were selected for preparing fluorescein-labeled probes 20a and 20b. In-gel fluorescence scanning and competitive assays confirmed that the probes can covalently bind to DNMT1 at the S-adenosyl-l-homocysteine

Arachidonic Acid-Induced Contraction of Smooth Muscle Is Mediated by MLC Phosphorylation in Preterm Birth.

Preterm birth (PTB) remains a challenging issue in the reproductive field, and cervical maturation is an essential physiological prerequisite for parturition. The cervix is rich in smooth muscle cells, and their abnormal contractility is a key trigger for premature cervical remodeling, which may further lead to spontaneous PTB. Herein, we found that high serum arachidonic acid (AA) expression in PTB mice may predict potential harms through untargeted metabolomics analysis. After AA intervention, immunofluorescence/qPCR/WB revealed that the expression of cervical smooth muscle contraction index

Trends in 5-year net survival for women diagnosed with breast, cervical or ovarian cancer in Japan, 2000-14 (CONCORD-3).

BACKGROUND: Breast, cervical and ovarian cancers significantly affect young and middle-aged women, both physically and socially. However, relevant comprehensive stratified analyses are limited. Using Japanese data from CONCORD-3, a global cancer survival surveillance program, we analyzed long-term survival trends. METHODS: Data from 16 Japanese population-based cancer registries were analyzed for women diagnosed aged 15-99 years during 2000-2014 with a tumor originating in the breast, cervix uteri or ovaries. Follow-up was extended to five years post-diagnosis or until December 31st, 2014. In

Validation of conformal prediction in cervical atypia classification.

Deep learning based cervical cancer classification can potentially increase access to screening in low-resource regions. However, deep learning models are often overconfident and do not reliably reflect diagnostic uncertainty. Moreover, they are typically optimized to generate maximum-likelihood predictions, which fail to convey uncertainty or ambiguity in their results. Such challenges can be addressed using conformal prediction, a model-agnostic framework for generating prediction sets that contain likely classes for trained deep-learning models. The size of these prediction sets indicates m

Evaluating the Effectiveness of a Cervical Pessary to Improve Neonatal Outcome by Preventing Preterm Birth in Individuals With Twin Pregnancy and Short Cervix: QUAD-P Twins.

BACKGROUND: Both twin pregnancy and short cervix are risk indicators for preterm birth. Cervical pessary and progesterone have been suggested to prevent preterm birth. A randomised trial was conducted in individuals carrying twins with cervix below the 25th centile (less than 38 mm). METHODS: We conducted a single centre open-label randomised clinical trial at Monash Medical Centre in Victoria, Australia (Trial registration number: ACTRN12616000875404). Individuals with a twin pregnancy and mid-trimester (16+0 to 23+6 weeks) cervical length < 25th percentile were invited to participate. After

Total cervical occlusion: A solution after cervico-isthmic cerclage failure.

OBJECTIVE: Spontaneous preterm birth is mainly caused by cervical incompetence. Cervical cerclage is the gold standard for preventing mid-trimester losses and preterm deliveries. If cervical cerclage fails, cervico-isthmic cerclage may be indicated. This procedure can be performed via an abdominal or vaginal approach and allows for a neonatal survival rate of around 90%. If a cervico-isthmic cerclage fails, there is no gold standard. Total cervical occlusion (TCO) could be an option. The present study aimed to analyze the efficacy of TCO using the Saling technique in cases where cervico-isthmi

Generation and Genetic Manipulation of Human Cervical Organoids.

Cervical cancer is the fourth most common cancer in women and most frequently affects the uterine ectocervix. The uterine ectocervix is lined by stratified squamous epithelium, comprising basal cells and differentiating parabasal and suprabasal cells. The regeneration observed after cervical conization suggests the presence of robust stem cell activity. However, our understanding of the identity and regulatory mechanisms of cervical stem cells and their malignant transformation process has been limited due to the restricted access to human cervical tissues and the lack of optimal systems to as
