Serological evidence of Rickettsia in horses and survey of tick-borne agents in ticks from horses and wildlife in the state of Pernambuco, Brazil.

Tick-borne diseases are among the most widespread vector-borne infections, including zoonoses caused by bacterial and protozoal microorganisms. This study consisted of a serosurvey for Rickettsia spp. in 270 horses from 11 farms in the coastal region of Pernambuco, Brazil. Through the indirect immunofluorescence assay (IFA) based on antigens of five Rickettsia species, 164 (60.7%) horses were seroreactive to Rickettsia spp., with seroreactivity among farms ranging from 10 to 100%. Through molecular analysis, all 270 horse blood DNA samples tested negative by PCR assays targeting Anaplasmatacea

The potential immunological mechanisms of gut microbiota dysbiosis caused by antibiotics exacerbate the lethality of influenza viruses.

BACKGROUND: Antibiotics are not recommended to treat influenza A virus (IAV). However, antibiotic misuse for IAV persists worldwide. How to scientifically use antibiotics for IAV-infected patients remains a considerable challenge. RESULTS: Here, we investigated the impact of antibiotics on viral pathogenicity, pulmonary-intestinal antiviral immunity, and antiviral drug efficacy. Our findings indicated that antibiotic intervention exacerbated IAV-caused mortality and lung injury in mice, manifested as increased mortality rates, shortened survival time, aggravated pulmonary injury, and excessiv

Antimicrobial effects of Bacillus safensis BS05 cell free supernatant against Campylobacter jejuni and its probiotic evaluation.

Campylobacter jejuni (C. jejuni) is a significant foodborne pathogen. The rise of antibiotic-resistant strains has diminished the effectiveness of conventional treatments, highlighting the urgent need for safe and effective alternatives. In this study, a Bacillus safensis (B. safensis) strain (BS05) was isolated from goats and assessed for its probiotic properties and antagonistic activity against C. jejuni. BS05 formed rough colonies on agar, and scanning electron microscopy revealed predominantly short rod-shaped cells. The cell-free culture supernatant (CFS) of BS05 inhibited multiple C. je

A survey of vector-borne bacteria in bat-associated mites (Mesostigmata: Macronyssidae and Spinturnicidae) from Brazil reveals the presence of Ehrlichia spp.

Macronyssid and spinturnicid mites are globally distributed ectoparasites of medical and veterinary relevance, with various species known to parasitize bats. Beyond their ecological importance, some macronyssid mites, such as Ornithonyssus bursa, are known to occasionally bite humans and cause dermatitis, highlighting the interface between wildlife, ectoparasites, and human habitats. Understanding the microorganisms harbored by bat-associated mites is therefore important from a One Health perspective, as these arthropods may act as bridge hosts for pathogens among bats, domestic animals, and h

Reverse zoonosis in bovine tuberculosis: The neglected threat of Mycobacterium tuberculosis infection in cattle.

Tuberculosis (TB), caused by members of the Mycobacterium tuberculosis complex (MTBC), remains a significant global health concern. While zoonotic transmission of M. bovis from cattle to humans is well documented, reverse zoonotic transmission of M. tuberculosis from humans to cattle has received far less attention. This review provides the first comprehensive synthesis of M. tuberculosis infections in cattle, drawing on evidence from farms, households, and slaughterhouses where human-animal contact is most intense. Available data indicate that such spillover events are uncommon compared with

Molecular detection of Orientia tsutsugamushi infection in bats from the China-Myanmar border.

Orientia tsutsugamushi (Ot), the causative agent of tsutsugamushi disease (TD), has been detected in Muridae, Soricidae, Cricetidae, Canidae, Artiodactyla, and birds, as well as in its trombiculid mite vectors. However, to date, scarce reports have documented Ot infection in bats. TD is an ancient zoonotic disease transmitted through the bite of infected trombiculid mites. The global disease burden of TD, particularly in impoverished regions, warrants renewed attention and a reevaluation of public health strategies. In this study, we analyzed bat samples for Ot using nested PCR (nPCR) and real

Emerging Challenges and Advances in Porcine Circovirus: A Decade in Review.

Over the past decade, porcine circoviruses (PCVs) have continued to pose a significant threat to global swine health, and pivotal discoveries have significantly reshaped our understanding of their biology and control. Extensive genomic surveillance has expanded porcine circovirus 2 (PCV2) genotyping from four to at least eight lineages, with PCV2d now globally dominant under vaccine-driven selection pressure. Since 2016, three novel species, PCV3, PCV4, and PCV5, have been identified, linked to reproductive failure, myocarditis, multisystemic inflammation, and potential neuroinvasion; however,

From pandemic influenza to novel coronaviruses: emerging infectious diseases of the 21st century.

Emerging infectious diseases have risen significantly in the twenty-first century as ecological disruption, climate change, expanding human-animal interfaces, and global mobility intensify opportunities for pathogen transmission. This review synthesizes historical and contemporary evidence across viral, bacterial, fungal, and parasitic threats to characterize how diverse pathogens emerge and spread. Foundational events such as the 1918 influenza pandemic, mid-century influenza pandemics, the emergence of HIV/AIDS, and the eradication of smallpox provide context for understanding modern disease

Deciphering the roles of AcrAB-TolC efflux pump in promoting the transmission of antibiotic resistance.

Plasmid-mediated conjugative transfer drives the global dissemination of antimicrobial resistance, posing a global threat to public health. Besides extruding antibiotics, bacterial multidrug efflux pumps modulate virulence, yet their influence on resistance plasmid spread in antibiotic-free settings remains undefined. Herein, we demonstrate that the AcrAB-TolC efflux pump is critical for the horizontal transfer of model plasmid RP4-7 and diverse clinical resistance plasmids. Single deletions of acrA, acrB or tolC significantly reduce plasmid transfer, and complementation fully restores conjuga

Rapid and simultaneous quantification of viable Lactobacillus and Streptococcus thermophilus cells in yogurt by flow cytometry combined with novel oligonucleotide probes.

Yogurt, one of the most consumed fermented dairy products, is produced with Lactobacillus and Streptococcus thermophilus as classical starter cultures. The enumeration of their viable bacterial cells is extremely important for yogurt production and supervision. This study developed 2 novel oligonucleotide probes (NOP), along with flow cytometry (FC) method for rapid and simultaneous quantification of the 2 probiotics. The NOP exhibited excellent specificity, as validated by 5 Streptococcus thermophilus strains, 11 Lactobacillus strains, and 32 other bacterial strains, and the FC method could a

Rickettsia parkeri subverts nicotinamide adenine dinucleotide (NAD+) metabolic pathway for its survival in endothelial cells.

Rickettsia parkeri rickettsiosis is an emerging tick-borne disease in humans. The bacterium R. parkeri is primarily transmitted to humans by the Gulf Coast tick, Amblyomma maculatum. Upon transmission to the host, R. parkeri infects and multiplies in host endothelial cells. In this study, we provide evidence that R. parkeri modulates the host nicotinamide adenine dinucleotide (NAD+) pathway for its survival in endothelial cells. Analysis performed with arrays containing antibodies against proteins involved in different metabolic pathways revealed increased levels of BST1 (an NAD+-dependent mol

The iucC gene is a major contributor to the virulence of hypervirulent Klebsiella pneumoniae.

BACKGROUND: Hypervirulent Klebsiella pneumoniae (hvKp) is a critical pathogen that causes highly lethal invasive infections with its virulence linked to that of aerobactin, the core siderophore system of hvKp. METHODS: Here, 20 porcine-derived K. pneumoniae strains were subjected to capsular genotyping and the virulence of the strains was evaluated through mouse lethality assays. Whole-genome sequencing and analysis was performed on two K2 serotype strains (including highly and weakly virulent strains), and the differential virulence genes were analyzed. The virulence-related gene iucC, ident

Two Cases of Q Fever in Pregnancy, including Management of the Newborn, Australia.

Optimal management of the birthing parent with Q fever in pregnancy and of the infant has not been established. Coxiella burnetii expresses a tropism for the placenta; resulting infection can potentially lead to spontaneous abortion and fetal demise. Although evidence around preventing transmission and infection in the peripartum and postpartum period is lacking, reports of healthy babies born to mothers with acute or chronic Q fever in pregnancy are increasing. Historically, many clinicians have recommended against breastfeeding in this setting because of a theoretical risk for bacterial tran

Predictive multi-omic biomarkers for urban zoonotic spillover detection: an integrative review.

Urban wildlife is an overlooked yet critical component of zoonotic disease surveillance, especially in biodiversity hotspots where human-animal interfaces accelerate spillover risk. This review synthesizes five complementary omics layers: Host microRNAs, host-pathogen genetic markers, bacterial microbiome profiling, viromics, and host transcriptomics into a single predictive framework for early spillover detection. Across taxa and pathogen classes, we highlight convergent molecular signatures of infection, from receptor polymorphisms and shifts in MHC diversity to pathogen-responsive miRNAs, h
