Characterization of the phage ΦK64 depolymerase S2-4 and its therapeutic effect against K1 serotype Klebsiella pneumoniae.

The infection rate and drug resistance of Klebsiella pneumoniae containing capsular polysaccharides (CPSs) have been increasing annually, resulting in severe human and animal infections. Depolymerases derived from bacteriophages can degrade CPSs and thus hold potential for treating bacterial infections. However, little is known about the mechanism by which K. pneumoniae phage depolymerases hydrolyze CPSs. In this study, the S2-4 encoded by the phage ΦK64 was identified as a potent depolymerase against K1 serotype Klebsiella CPSs. Cryo-electron microscopy structural analysis revealed that S2-4

Emerging zoonotic risks: whole-genome sequencing reveals antimicrobial resistance and genomic diversity in Providencia stuartii isolated from broiler chickens in Noakhali, Bangladesh.

Providencia stuartii is emerging as an Extensively Drug-Resistant (XDR) pathogen commonly found in animals, insects, and in burned and immunocompromised conditions. The misuse of antibiotics in poultry feed causes the emergence of XDR bacteria in the poultry industry. The knowledge of zoonotic transmissibility of poultry-derived P. stuartii remains elusive in Noakhali, Bangladesh. Poultry fecal and rectal swab samples were collected from selected farms in Noakhali, Bangladesh. Bacterial isolation and identification were performed using MacConkey agar, biochemical tests, and 16S rRNA Sanger seq

Molecular identification of Bartonella henselae in dogs with clinical suspicion of visceral leishmaniasis.

The genus Bartonella includes species responsible for severe infections. Bartonella henselae, commonly linked to human disease, also infects dogs with or without symptoms. In Brazil, visceral leishmaniasis (VL) caused by Leishmania infantum is prevalent in Mato Grosso State, Brazil, particularly in Rondonopolis city, affecting both humans and dogs. Although reports of co-infection with Bartonella spp. and L. infantum exist in other countries, Brazilian studies are scarce. This study investigates the presence of B. henselae using culture and molecular methods in dogs suspected of having VL in R

Epidemiological investigation of Coxiella burnetii in farms after an outbreak of Q fever in slaughterhouse workers.

Q fever is a worldwide zoonosis caused by the bacterium Coxiella burnetii. In January/2015, a Q fever outbreak occurred among workers at a cattle slaughterhouse in southeastern Brazil. This study investigated if C. burnetii was circulating on farms that sent cattle to the slaughterhouse at the time of the 2015-Q fever outbreak. Blood, milk, vaginal swab and tick samples were collected from livestock on 35 farms surrounding the slaughterhouse. Serological analyses with C. burnetii whole antigens indicated that 68.6 % farms (24/35) had at least one seropositive animal, 76.7 % (23/30) farms had s

Phylogenomic and population genomic insights into the dissemination of ESBL-producing Escherichia coli causing bloodstream infections in the United Arab Emirates.

Extended-spectrum β-lactamase-producing Escherichia coli are globally disseminated pathogens whose success is driven by clonal expansion and horizontal gene transfer. However, the population structure and evolutionary relationships of these organisms in the United Arab Emirates remain insufficiently characterized. In this study, we applied a population genomic and phylogenomic approach to investigate ESBL-producing E. coli causing bloodstream infections and their genetic relatedness to strains from non-human reservoirs within a One Health framework. Forty-five ESBL-producing E. coli isolates r

Zoonotic Bordetella bronchiseptica infection at the swine-human interface: unveiling the evolutionary path from an animal to a human pathogen.

Bordetella bronchiseptica, long regarded as a veterinary pathogen, is now emerging as a zoonotic threat to humans, particularly in immunocompromised individuals. We report a sentinel event involving a synchronized B. bronchiseptica outbreak in swine and their human caretaker, providing a unique opportunity to examine cross-species transmission and adaptation at the genomic level. Comparative genomics revealed that the human-adapted isolate (RL57) and its swine progenitor (XX35) share an identical chromosome, with XX35 harbouring an extra conjugative plasmid. Remarkably, RL57 did not simply los

Discovery of H2 Receptor Antagonists as Colistin Enhancers by Targeting Acid Stress Response.

Plasmid-mediated colistin resistance gene mcr has markedly diminished the effectiveness of colistin in managing multidrug-resistant (MDR) Gram-negative bacterial infections. Employing antibiotic adjuvants to revive colistin susceptibility offers a valuable solution to address this issue. Intestinal pathogens have evolved sophisticated acid resistance systems to withstand the acidic environment, a key adaptation that facilitates subsequent infection. Utilizing transposon sequencing, here we identify yqgB gene as a crucial target for restoring colistin susceptibility in mcr-positive bacteria, pa

UspF Regulates Type III Pili-Mediated Adhesion, Oxidative Stress Resistance, and Virulence in Klebsiella pneumoniae.

Klebsiella pneumoniae (K. pneumoniae, KP) is a significant opportunistic pathogen responsible for both nosocomial and community-acquired infections. Bacterial adhesion is the critical initial step for host colonization and the establishment of disease. In this study, we utilized a mariner transposon mutagenesis system to construct a mutant library from the clinical KP strain KP20, identifying a mutant with significantly impaired epithelial cell adhesion due to an insertion in the uspF gene. Genetic knockout experiments confirmed that uspF deletion markedly reduced the adhesion to human airway

Ecology and antimicrobial resistance of Campylobacter in wildlife: insights into specialist and generalist lineages and zoonotic potential.

Wildlife is a critical reservoir of Campylobacter species, particularly C. jejuni and C. coli, carrying diverse genetic lineages, virulence factors, and antimicrobial resistance (AMR) genes. Birds, especially migratory and synanthropic species, are the primary carriers, though mammals, reptiles, and other vertebrates also contribute to maintenance and dissemination. Wildlife-associated strains include both host-specific lineages and generalist clonal complexes (e.g. ST21, ST45, and ST828) capable of crossing wildlife, livestock, humans, and environmental interfaces, reflecting high zoonotic po
