Adverse events associated with use of immunoglobulin in pediatric patients reported to the US Food and Drug Administration Adverse Event Reporting System, 2001-2023.

BACKGROUND: Immunoglobulin products are widely used for the treatment of immunodeficiency and autoimmune disorders. Although clinical trials have demonstrated their efficacy and tolerability, data describing their postmarketing safety profile in pediatric populations remain limited, particularly regarding rare and serious adverse events. This study aimed to characterize adverse event reports associated with immunoglobulin use in children during the postmarketing period. METHODS: Pediatric adverse event reports (<18 years) submitted to the U.S. Food and Drug Administration Adverse Event Report

Histologically indolent but widely disseminated diffuse midline glioma H3K27-altered revealed by autopsy.

Diffuse midline glioma, H3K27-altered is a highly aggressive central nervous system tumor classified as grade 4 in the 2021 World Health Organization Classification of Tumors of the Central Nervous System. This malignancy typically arises in midline structures such as the thalamus, brainstem, and spinal cord, and is characterized by its infiltrative capacity. Despite the use of magnetic resonance imaging to evaluate tumor extension, its microscopic dissemination can extend far beyond radiological detection. We report the case of a 46-year-old woman with a thalamic diffuse midline glioma harbor

Bridging veins govern discrepancy of CSF efflux into dura mater in mice.

Meningeal lymphatic vessels within the dura provide a route for cerebrospinal fluid (CSF) outflow, but how CSF crosses the arachnoid barrier, particularly toward basal dural compartments, remains unclear. Using in vivo CSF tracer infusion and ultrastructural analysis in mice, we show that bridging vein-associated arachnoid cuff exit (ACE) domains provide specialized routes for CSF movement from the SAS into the dura. These ACE-associated structures are abundant in the basal than dorsal meninges, supporting preferential CSF efflux toward basal meningeal lymphatics. In contrast, spinal meninges

The Aging Meninges: Anatomical and Immunological Remodeling as Emerging Diagnostic Biomarkers of Cerebral Senescence-A Narrative Review.

Background/Objectives: Aging is a universal biological process characterized by progressive structural, cellular, and molecular alterations that contribute to functional decline and increased susceptibility to disease. In recent years, the cerebral meninges have emerged as an active regulatory interface between the central nervous system and peripheral tissues, with growing evidence supporting their involvement in immune surveillance, fluid homeostasis, and age-related neuropathology. Methods: This narrative review provides a comprehensive overview of the current state of knowledge regarding a

Antineutrophil cytoplasmic antibody-associated pachymeningitis: a systematic review of clinical features, diagnosis and treatment outcomes.

Antineutrophil cytoplasmic antibody (ANCA)-associated pachymeningitis is a rare inflammatory disorder of the dura mater that may occur in isolation or as part of systemic vasculitis. The clinical features and outcomes of proteinase 3 (PR3)- and myeloperoxidase (MPO)-ANCA-associated pachymeningitis remain poorly defined. We aimed to synthesize clinical presentation, investigations, and treatment outcomes to improve diagnosis and guide management. A systematic review was conducted in accordance with PRISMA guidelines. PubMed, Embase, and Scopus were searched to June 15, 2023. Eligible studies we

Subnational analysis of pediatric sepsis incidence and mortality from official records in Chile and Mexico: a longitudinal study from 2014 to 2024.

BACKGROUND: Pediatric sepsis is a leading cause of global morbidity and mortality, yet high-resolution, granular subnational assessments remain scarce. Chile and Mexico are the only countries in Latin America that possess robust vital registration systems and open access databases with marginal levels of missing cases. This offers a unique opportunity to quantify the subnational burden of pediatric sepsis, identify healthcare system constrictions, and guide targeted public health interventions. METHODS: This retrospective longitudinal study analyzed official hospital discharge and non-fetal d

Long-term risk of dementia following encephalitis: a large-scale retrospective cohort study of electronic health records.

BACKGROUND: Encephalitis is an acute inflammation of the brain parenchyma, typically caused by microbial infections or autoimmune mechanisms, and represents a potentially life-threatening neurological condition affecting thousands of individuals each year in the United States. Although survivors may experience persistent neurological sequelae, long-term association of encephalitis and subsequent dementia and related diagnoses across different age groups and encephalitis etiologies remains poorly characterized. This study aimed to quantify the association between encephalitis and the risk of de

Decellularized CNS-Derived ECM Hydrogels as Dual-Action Scaffolds for Angiogenesis and Neurogenesis.

Neurodegenerative disorders of the central nervous system (CNS) remain a significant clinical challenge due to the limited regenerative capacity of neural tissues. Herein, bioactive scaffolds were developed from decellularized extracellular matrices (dECMs) derived from four CNS tissues: brain, spinal cord, spinal meninges, and optic nerve. The scaffolds were comprehensively characterized with respect to biochemical composition, mechanical properties, and biological performance. Optimized decellularization protocols efficiently removed cellular components while preserving key matrix constituen

Primary spinal meningeal melanoma with intramedullary and intradural extramedullary components-a case report.

Primary melanomas of the spinal meninges are exceedingly rare. While both intramedullary and extramedullary spinal melanomas have been reported, to the best of our knowledge, this is the first noted case of primary spinal melanoma that has both intramedullary and intradural extramedullary components. We present a case of a 61-year-old female presenting with a 1-year history of lower back pain, bilateral lower extremity pain, and perceived weakness of left foot. Magnetic resonance imaging of the thoracic spine suggested intramedullary and intradural extramedullary mass at levels T8-T12. A T7-T1

An integrated classification to guide the management of spinal meninges pathologies.

PURPOSE: The terminology used for spinal meningeal pathologies is often inconsistent, with multiple terms referring to similar or related anatomical anomalies. This lack of standardization leads to variability in the management of these lesions, with differing diagnostic and therapeutic approaches for similar conditions. Our aim was to revise the classification of spinal meningeal pathologies by integrating pathogenesis, diagnostic criteria, and therapeutic strategies. METHODS: We first conducted a descriptive cohort study over a ten-year period at a single neurosurgical reference center. Cli

Diabetes is associated with a decreased risk of meningiomas: a mendelian randomization study.

BACKGROUND: Meningiomas are the commonest primary intracranial tumors, but the risk factors are complex and not fully understood. Type 1 and 2 diabetes mellitus (T1DM and T2DM) are frequently occurring endocrine diseases, but their relationship with meningiomas is debated. Obesity and elevated body mass index (BMI) are known risk factors for meningiomas but often confound with diabetes. We aim to address this controversy using mendelian randomization a method inherently robust to confounding. METHODS: We used mendelian randomization on genetic instruments for T1DM and T2DM to assess the risks

Global Assessment of Health Utilities Associated with Pneumococcal Disease in Adults: Targeted Literature Reviews.

BACKGROUND: Pneumococcal disease can significantly reduce quality of life (QoL). Health utilities are commonly used to assess QoL burden and calculate quality-adjusted life years (QALYs), which are critical inputs in cost-utility analyses (CUAs) to evaluate the economic value of pneumococcal vaccines. OBJECTIVES: This study aimed to synthesize utility values used in CUAs of pneumococcal vaccines in adult populations in North America and Europe and to summarize global utility studies on pneumococcal disease and post-meningitis sequelae (PMS) in adults. METHODS: Two targeted literature reviews

The Burden of Hospital Illness Associated with Disseminated Versus Isolated Pulmonary Coccidioidomycosis in the United States.

There are scarce data comparing inpatient mortality, length of stay (LOS) and all-cause hospital costs in disseminated coccidioidomycosis (DCM) vs. isolated pulmonary coccidioidomycosis (IPCM). We assessed the burden of hospital illness associated with DCM versus IPCM. This study was performed using National Inpatient Sample data from 2019 to 2021. DCM was defined as having a primary International Classification of Diseases-Tenth Revision (ICD-10) code for coccidioidal meningitis, a non-primary code for coccidioidal meningitis in the presence of a primary code for a meningitis complication or

Neuronal Autoantibodies in Adults After Community-Acquired Bacterial Meningitis.

BACKGROUND AND OBJECTIVES: Long-term cognitive impairment is observed in 14%-32% of patients surviving community-acquired bacterial meningitis. We hypothesized that the impairment might be linked to secondary immune activation due to the development of neuronal autoantibodies, similar to postinfectious autoimmune encephalitis after viral encephalitis. METHODS: In this cross-sectional observational study, we included adult patients from a prospective, nationwide cohort study of community-acquired bacterial meningitis in the Netherlands, the MeninGene study. The presence of neuronal autoantibod
