Methods in molecular biology (Clifton, N.J.)2027eng
Chlamydia trachomatis, an obligate intracellular pathogen responsible for trachoma and widespread urogenital infections, replicates exclusively within the confines of a membrane-bound vacuole known as the inclusion. This compartment shields the bacterium from host cellular immune detection; thus, clarifying how inclusion integrity is maintained and protected from vacuole-destabilizing host attacks may identify new therapeutic strategies. To enable such insights, we recently developed split-GFP-based microscopic reporters that can detect early forms of membrane damage and track individual bacte

The recent application of experimental and computational drug discovery workflows has identified hundreds of new molecules that display potent, and in some cases highly selective, activity against bacteria of the genus Chlamydia. The full value of these novel antichlamydial molecules, however, depends critically on our ability to determine their molecular targets and modes of action. A particularly powerful and widely applied strategy for identifying candidate targets involves selecting for bacterial mutants that have acquired resistance to a compound's inhibitory activity, followed by determi

BACKGROUND: In low-income and middle-income countries, where typhoid fever remains a major public health problem, WHO recommends the use of typhoid conjugate vaccine (TCV). Here, we report vaccine effectiveness up to 8 years following a single dose of TCV in Nepal.
METHODS: TyVOID was a prospective cohort study that extended the follow-up of children enrolled in a phase 3, double-blind, randomised controlled trial in Lalitpur, Nepal (TyVAC; Nov 20, 2017, to Oct 20, 2021). Children aged 9 months to 15 years were randomly assigned (1:1) to receive Vi-tetanus toxoid conjugate vaccine (Vi-TT) or

Understanding Chlamydia trachomatis infection requires physiologically relevant models tailored to various research questions. One of the main sites of C. trachomatis infection is the columnar epithelium of the endocervix; therefore, a model that accurately mimics the environment of endocervical epithelium is essential. Polarized cell models provide a valuable solution, offering a cost-effective yet biologically relevant alternative to conventional 2D culture systems and more complex and costly 3D or animal models. Immortalized human endocervical epithelial cells (A2EN cells) form a polarized

Chlamydia trachomatis is an obligate intracellular bacterium that infects the columnar epithelium of the human endocervix. While conventional two-dimensional cell cultures and animal models have been instrumental in advancing our understanding of C. trachomatis biology, they are limited in capturing the multicellularity, architecture, and physiological microenvironment of the human cervix. This chapter describes the use of a three-dimensional (3D) microphysiologic model to study Chlamydia trachomatis infection. The model is inexpensively made without specialized equipment and is designed to re

Identification and characterization of essential Chlamydia-specific factors are vital for the generation of effective therapeutics and vaccines. Despite delays in the development of genetic tools for Chlamydia, significant advancements in Chlamydia genetic manipulation have been made over the last decade and a half, now allowing for the targeted interruption or deletion of chromosomal genes. However, because of the obligate intracellular nature of the bacteria and its unique biphasic developmental cycle, it remains challenging to inactivate essential genes and study the function of their encod

Neisseria meningitidis is a notifiable pathogen in Japan when isolated from any sterile site. We describe a rare case of primary meningococcal arthritis without meningitis or bacteremia. A 48-year-old man presented with acute septic arthritis of his knee. A synovial fluid analysis using matrix-assisted laser desorption/ionization time-of-flight mass spectrometry revealed gram-negative diplococci, identified as N. meningitidis. Further antimicrobial susceptibility testing showed the isolate was pan-susceptible, allowing de-escalation from ceftriaxone to ampicillin therapy. The patient recovered

BACKGROUND: The escalating global incidence of multidrug-resistant Neisseria gonorrhoeae necessitates an effective vaccine. While the meningococcal 4CMenB vaccine offers some cross-protection against gonorrhoea, this protection is limited highlighting the need for a targeted gonococcal vaccine. In this study, we evaluate the durability of immune response and characterise the immunogenicity induced by GonoVac, a native Outer Membrane Vesicle (nOMV) vaccine produced from N. gonorrhoeae GC_0817560lpxL1- , across genetically distinct strains of mouse.
METHODS: We evaluated the longevity of the im

The Australian Gonococcal Surveillance Programme (AGSP) has continuously monitored antimicrobial resistance in Neisseria gonorrhoeae (NG) for more than 45 years through the system of jurisdictional Neisseria reference laboratories, the National Neisseria Network (NNN). In 2025, a total of 10,204 isolates across Australia, from public and private sectors, were tested for in vitro antimicrobial susceptibility by standardised methods. In 2025, the AGSP captured antimicrobial susceptibility data for 23.9% of all gonococcal infection notifications nationally. The current treatment recommendation fo

Nocardiosis is a rare but potentially life-threatening infection caused by aerobic, filamentous, gram-positive bacteria of the genus Nocardia found in soil and water. Although traditionally found in immunocompromised patients, up to one-third of cases occur in immunocompetent individuals, contributing to frequent diagnostic delays. Historically fatal before the antibiotic era, outcomes have substantially improved with modern antimicrobial therapy and diagnostic advances. However, nocardiosis remains underrecognized due to its nonspecific presentation and ability to mimic more common diseases.

BACKGROUND: Clostridioides difficile infection (CDI) is one of the leading infectious diarrhea worldwide. C. difficile pathogenicity is mediated by various virulence factors, including toxins and biofilm formation. This study included 313 C. difficile isolates that were previously collected from four medical centers. We aimed to investigate the prevalence, distribution, and associations of virulence factors in C. difficile strains from both healthcare-associated (HA)- and community-acquired (CA)-CDI patients across Israel from 2020 to 2022.
METHODS: Multi-locus sequence typing (MLST) and whol

This review combines historical reports and surveillance data to contextualize Clostridioides difficile infection (CDI) in the United States as a phased epidemic, currently in the deceleration phase. C. difficile was first isolated in 1935. During a prolonged emergence phase, the organism attracted limited clinical attention. In 1978, C. difficile was established as the cause of antibiotic-associated pseudomembranous colitis and from 1978 to a peak in 2010, CDI was in an acceleration phase characterized by fluoroquinolone-resistant, toxin-overproducing strains, large hospital outbreaks, and in

Gram-negative bacteria produce nonulosonic acids, a family of nine-carbon sugars that mimic host sialic acids and may contribute to bacterial virulence. The nonulosonic acid C8-epimeric 5,7-di-N-acetyllegionaminic acid (8-epi-Leg5,7Ac2) is a component of the K-locus 49 (KL49) capsule of hypervirulent Acinetobacter baumannii, and its epimerization is proposed to be mediated by ElaA, ElaB, and ElaC. We identified a KL49 strain that induces lethal sepsis in mice and found deletion of elaA, elaB, or elaC abolished lethality and markedly reduced capsule production. Crystal structures of ElaB, ElaA,

Chronic infectious keratitis represents a distinct clinical entity characterized by indolent progression, diagnostic difficulty, and prolonged therapeutic courses. Unlike acute bacterial keratitis, chronic infections often present with subtle inflammatory signs, recurrent episodes, or poor response to empirical antimicrobial therapy. Among the most important causes are herpetic keratitis, acanthamoeba keratitis, Microsporidial stromal keratitis, and atypical (non-tuberculous) mycobacterial keratitis. Herpes simplex virus keratitis may manifest as recurrent epithelial disease or immune-mediated

Tuberculosis (TB) continues to be a major global health challenge, largely due to the complex nature of Mycobacterium tuberculosis. Its early detection and effective management are heavily reliant on advanced diagnostic methods. New drug delivery systems and repurposing existing drugs show great promise in improving TB treatment. This study explores the progress and hurdles in developing anti-TB drugs, focusing on those currently in clinical trials. Additionally, innovative approaches like immunotherapy, combination therapy, and adjunct therapy, which include the use of phytochemicals, are exa

BACKGROUND: Klebsiella pneumoniae is a 'superbug' characterized by high virulence and carbapenem resistance. CRISPR-Cas3 is capable of performing large-scale deletions in bacterial genomes. The purpose of this study is to sensitize Klebsiella pneumoniae A324 to carbapenem antibiotics by eliminating the multidrug resistance plasmid pA324-IMP using a compact Cascade-Cas3 system.
METHODS: Recombinant plasmids were constructed using molecular cloning techniques. Three sgRNAs targeting different positions within the blaIMP-38 gene were designed. Recombinant plasmids were separately transformed int

We evaluated a semi-automated method for DNA extraction, expanded to herbivore feces a gyrA PCR method for the detection of Campylobacter jejuni in canine feces, added an internal positive control (IPC) DNA to specimens before DNA extraction, and multiplexed the gyrA PCR assay with the IPC assay. The addition of IPC DNA and multiplexing of the IPC assay did not affect the performance of the gyrA PCR assay. The method was repeatable with no significant difference in results across 6 d (F = 0.715; p = 0.614 with R2 = 0.818). In a single-laboratory randomized method test evaluating detection in b
