The spa gene of Staphylococcus aureus encodes protein A, a key virulence factor contributing to molecular typing. spa typing provides valuable information encoding diversity and transmission, particularly for methicillin-resistant S. aureus (MRSA), which remains a major public health concern. A one-year prospective surveillance study was conducted involving S. aureus bacteremia (SAB) patients admitted to Haji Adam Malik General Hospital, Medan, Indonesia. SAB isolates were phenotypically analysed for cefoxitin resistance using the VITEK-2 system and genotypically characterised by PCR for the m

INTRODUCTION: Staphylococcus simulans is an animal pathogen belonging to the coagulase-negative staphylococci (CoNS) group that was occasionally identified in the skin of healthy people. Although it was suggested that S. simulans share some virulence factors with S. aureus, little is known about human infections, and clinical relevance is limited to a few reports. Herein, we report a case of bacteremia caused by S. simulans.
CASE PRESENTATION: A 39-year-old man was brought to the hospital due to heart attack. During the course of admission, his procalcitonin, C-reactive protein, and erythrocy

Staphylococcal toxic shock syndrome is a rare toxin-mediated illness. A 9-year-old girl on automated insulin delivery presented with shock, multiorgan dysfunction, rash, and gastrointestinal symptoms. An infected infusion site grew methicillin-susceptible Staphylococcus aureus. She met CDC criteria for TSS, highlighting infusion sites as sepsis foci and supporting continuation of automated insulin delivery during critical illness.

Staphylococcus aureus is an opportunistic pathogen implicated in skin and soft-tissue infections and chronic inflammatory skin diseases, such as atopic dermatitis. These infections can range from mild to potentially life-threatening systemic illnesses progressing to bacteremia, endocarditis, and sepsis. Mast cells, traditionally recognized for their role in allergies, are highly abundant within the skin and are gaining recognition for their contribution to bacterial defense. Here, we discuss the role of mast cells in three models of S. aureus skin infection according to skin depth: epicutaneou

Therapeutic interventions for bacterial infections are increasingly challenging due to the growing threat of antibiotic resistance, particularly from Methicillin-resistant Staphylococcus aureus (MRSA), which poses significant risks to global health. Generally, MRSA has a high mortality rate and can lead to severe illnesses such as sepsis and pneumonia, mainly due to the lack of effective treatments and growing drug resistance. To this end, the present study introduces azobenzene-conjugated photo-responsive polypeptide nanofibers (PNFs) that mimic host-defense peptides to combat antibiotic-resi

Methicillin-resistant Staphylococcus aureus (MRSA) remains one of the most significant multidrug-resistant bacterial pathogens responsible for a broad spectrum of infections ranging from mild skin infections to severe invasive diseases, including bacteremia, pneumonia, endocarditis, osteomyelitis, and sepsis. The rapid global dissemination of MRSA is primarily driven by the acquisition of the mecA gene encoding penicillin-binding protein 2a, which confers resistance to β-lactam antibiotics. In addition to β-lactam resistance, MRSA exhibits resistance to multiple antimicrobial classes through d

Multidrug-resistant pathogens are among the greatest global health threats. Among Gram-negative pathogens, the worldwide rise in carbapenem resistance is of particular clinical significance; among Gram-positive pathogens, methicillin-resistant Staphylococcus aureus and vancomycin-resistant enterococci are of primary concern. Resistance mechanisms include changes in penicillin-binding proteins, enzymatic inactivation by β‑lactamases, porin modifications, and efflux pumps. Targeted therapy for infections caused by multidrug-resistant pathogens requires knowledge of the resistance mechanism and a

Staphylococcus aureus adaptation and persistence within host environments are strongly shaped by lipid availability and host-derived lipid fluxes encountered during infection. The glycerol ester hydrolase Geh, initially characterized as a secreted lipase hydrolyzing triacylglycerols and cholesteryl esters, is now emerging as a multifunctional enzyme operating at the host-lipid interface. By contributing to fatty acid release and re-esterification, Geh appears to participate in lipid flux and may thereby influence membrane composition and bacterial physiology. These Geh-associated processes hav

Background and Clinical Significance: Skin-limited Langerhans cell histiocytosis (LCH) is a clinically heterogeneous disease, ranging from self-healing forms to fulminant multi-organ failure, the latter being more often described in infants, especially preterm neonates. The optimal therapy for cutaneous LCH remains controversial; the possibilities vary from a watchful waiting approach to systemic chemotherapy. Case Presentation: This case report describes an exceptionally rare and clinically challenging course of skin-limited LCH in a prematurely born infant treated at the Department of Oncolo

BACKGROUND: Infective endocarditis (IE) often presents with non-specific symptoms, which may delay diagnosis and treatment. Previous studies exploring symptom duration have been limited by small cohorts or single-centre studies. We aimed to investigate patient characteristics, microbial aetiology, treatment, and all-cause mortality of patients with IE according to symptom duration prior to diagnosis.
METHODS: We included all patients with first-time IE from the NatIonal Danish Endocarditis StUdies (NIDUS) registry (2016-2021). Patients with left-sided IE and available symptom duration were st

BACKGROUND: Sepsis is associated with an extremely high global mortality rate, primarily due to the lack of effective therapeutic strategies for clearing bacteria from infected tissues. However, the underlying mechanisms by which methylsulfonylmethane (MSM) protects mice against lethal doses of MRSA infection remain incompletely understood.
METHODS: RNA sequencing was used to identify transcriptional differences of peritoneal macrophages (PMs). Immunofluorescence, Western blotting, and RT-qPCR were used to assess target protein and RNA levels. Molecular docking, flow cytometry, and pharmacolo

BACKGROUND: Patients receiving dialysis, particularly those on hemodialysis with central venous catheters (CVC), are at higher risk of bloodstream infections, often caused by Staphylococcus aureus. After years of declining CVC prevalence, pandemic-related shifts in dialysis practices led to higher CVC use and increased concern regarding contemporary infection risk. This study used national data spanning pre-, intra-, and post-pandemic periods to determine current rates and risk factors for S. aureus-related sepsis in the US dialysis population.
METHODS: A retrospective cohort study was conduc

BACKGROUND: Sepsis is a life-threatening condition driven by a dysregulated immune response to infection, often leading to multi-organ failure and high mortality. Beyond oxygen transport, red blood cells (RBCs) may contribute to sepsis pathophysiology through eryptosis, a form of programmed RBC death. Characterized by cell shrinkage, membrane changes, and phosphatidylserine exposure, eryptosis can impair microcirculation and tissue oxygenation. This study explored the association between bacterial exposure, eryptosis, and inflammation, assessing its potential as a biomarker for sepsis progress

Gangrenous mastitis is a severe disease caused by bacterial infection. It can lead to sepsis and even death in affected goats, posing a significant threat to the development of the dairy goat industry. The pathogenesis of this disease is not fully understood. In this study, a Staphylococcus aureus isolate obtained from a goat with gangrenous mastitis was used to establish in vivo and in vitro models. Using omics sequencing and detection of iron, MDA, GSH, and ROS levels, along with RT-qPCR and western blot, the occurrence of ferroptosis was assessed. Furthermore, by employing ferrostatin-1 (Fe

The chemokine CCL20/MIP3α plays an important role in the migration of dendritic cells and several lymphocytes. This 70-residue protein is also implicated in various diseases, such as rheumatoid arthritis, psoriasis, sepsis and numerous forms of cancer. In addition, as the most basic human chemokine, CCL20 also displays direct antimicrobial and antibiofilm activities. The binding and oligomerization of chemokines to cell surface glycosaminoglycans (GAG) can play an important role during receptor activation; therefore we have studied the interactions of CCL20 with fondaparinux. This anticoagulan
