Synchronous Metabolic Interference and Immune Reprogramming by a Self-Assembled Bismuth-Phenolic Nanoparticle against Pseudomonas aeruginosa-Induced Infections.

Effective eradication of Pseudomonas aeruginosa (P. aeruginosa) infection remains challenging due to its multifaceted antibiotic resistance mechanisms and biofilm-associated protection. Furthermore, inflammatory dysregulation within the infected microenvironment compromises both antibacterial efficacy and tissue repair. Therapeutic strategies capable of simultaneously eliminating pathogens and restoring microenvironmental homeostasis are therefore highly desirable. Herein, we report a bismuth-phenolic nanoparticle (BAQ NP) for synergistic antibacterial and immunomodulatory therapy of bacterial

Sunlight-Triggered Bacterial Membrane-Targeting Aggregation-Induced Emission "Nanoantibiotic" Hydrogel for Ultrafast Multidrug-Resistant Bacteria Eradication and Diabetic Wound Healing.

Drug-resistant bacterial infections and persistent biofilm formation pose critical bottlenecks for diabetic wound management. To overcome these challenges, a sunlight (SL)-triggered aggregation-induced emission (AIE) photosensitizer, designated as MTP, was engineered and incorporated into an injectable thermosensitive hydrogel. Featuring a donor-acceptor architecture, MTP exhibits NIR fluorescence emission, while its visible light absorption enables sunlight-triggered dual-type photodynamic activity. Crucially, MTP achieves ultrafast bacterial cell membrane anchoring within 15 seconds, deliver

Water-Soluble Chlorophyll Proteins as Biocompatible Photosensitizers for Photodynamic Therapy to Combat Antibiotic Resistance and Biofilm Through Disrupting Bacterial Membranes.

The escalating crisis of antibiotic resistance and biofilm-associated infections urgently demands therapeutic strategies with distinct mechanisms of action. Antibacterial photodynamic therapy (abPDT) offers a promising alternative, yet its clinical translation is often limited by the poor solubility, low biocompatibility, or complex delivery requirements of conventional photosensitizers (PSs). Here, we report the application of recombinant water-soluble chlorophyll proteins (WSCPs) from the Brassicaceae family as fully natural, self-contained PSs for abPDT. WSCP forms exceptionally stable tetr

Helicobacter pylori-induced immune priming requires type I interferon signaling.

Helicobacter pylori (H. pylori) infection induces persistent colonization and chronic inflammation, yet the mechanisms by which bacterial exposure establishes sustained inflammatory responses remain incompletely understood. We previously demonstrated that priming human monocytes with viable, but not heat-killed H. pylori, establishes immune priming, characterized by a heightened response to subsequent LPS challenge. Here, we delineate the signaling pathways that establish this inflammatory adaptation. Unbiased whole-cell proteomics identified type I and type II interferon (IFN) signaling as th

Protective-Antigen Scaffold Nanoparticle Vaccine Overcomes Carrier-Induced Suppression and Immunosenescence to Protect against Pseudomonas aeruginosa Pneumonia.

Multidrug-resistant Pseudomonas aeruginosa (PA) causes severe infections, with severe burden especially in older adults. Vaccines remain effective and are urgently needed despite immunosenescence. Self-assembling nanoparticles can enhance vaccine immunogenicity, but their translational use is constrained by the useless anticarrier immunity and carrier-induced epitope suppression (CIES). Herein, we developed a protective-antigen-based nanoparticle platform in which the PA chaperonin GroEL, an intrinsically self-assembling protective antigen, serves as the scaffold for a fusion immunogen reGroEL

Nanoparticles Against Antimicrobial Resistance: Chemical Design, Assembly, and Emerging Applications.

Antimicrobial resistance (AMR) is an escalating global health concern, with increasing rates of morbidity and mortality associated with multidrug-resistant infections worldwide. Despite intensive efforts to discover antimicrobial compounds, few candidates progress to clinical trials, and even fewer achieve market approval. A promising approach to address the current stagnation in antibiotic discovery is the development of advanced antimicrobial delivery platforms, which can enhance the efficacy, stability, and reusability of both existing and engineered antimicrobial agents. In this review, we

Preparation and Characteristics of Selenium Nanoparticles Synthesized by Limosilactobacillus reuteri CICC 6118 and Their Protective Effects on Intestinal Injury in Early-Weaned Piglets.

In this study, biogenic selenium nanoparticles (SeNPs) were synthesized through biotransformation of Na2SeO3 by the porcine-derived strain Limosilactobacillus reuteri CICC 6118 (L. reuteri CICC 6118) and subsequently characterized. Their protective effects were evaluated in early-weaned piglets challenged with enterotoxigenic Escherichia coli K88 (ETEC K88). L. reuteri CICC 6118 synthesized SeNPs with diameters of approximately 70-190 nm and achieved a Na2SeO3 conversion efficiency of 97%. The purified SeNPs exhibited antioxidant activity and reduced bacterial culture turbidity in preliminary

β-Hydroxybutyrate Exacerbates Klebsiella pneumoniae-Induced Inflammatory Injury and Milk Synthesis Impairment via the GPR109A/NF-κB Signaling Axis.

Ketotic cows exhibit a heightened mastitis risk through an unknown mechanism. This study investigated the influence of β-hydroxybutyrate (BHBA) on mastitis development. Mammary tissues from ketotic cows with mastitis showed more severe pathological damage and suppression of lactation-related genes. BHBA pretreatment further increased NF-κB pathway activation, facilitated expressions of pro-inflammatory cytokines, and suppressed lactation proteins in Klebsiella pneumoniae (K. pneumoniae)-infected BMECs, thereby exacerbating inflammatory injury and milk synthesis impairment. Selenium Supplementation Attenuates NET DNA-Mediated Intestinal Epithelial Pyroptosis and Intestinal Barrier Injury in MRSA Enteritis via Upregulating Selenoprotein S.

It is widely believed that neutrophils exert an antibacterial defense through NETs during MRSA infection. However, this study found that during MRSA enteritis, NET DNA released by neutrophils under low-selenium (Se) conditions directly triggers intestinal epithelial pyroptosis and disrupts tight junctions. Following sodium selenite intervention, intestinal selenoprotein transcriptome screening identified selenoprotein S (SelS) as the most significantly downregulated selenoprotein after the MRSA challenge and the most sensitive to Se supplementation recovery. Notably, MRSA alone did not directl

The BP26-OMP16Es Nanoparticle Vaccine Elicits Protective Immunity against Brucella.

Brucellosis, a major global zoonosis causing over 2.1 million human infections annually, poses a significant threat to public health and livestock economies. Conventional vaccines face substantial limitations, including residual virulence and diagnostic interference in live attenuated vaccines, and poor immunogenicity in subunit vaccines. To address these issues, we developed BP26-OMP16Es, a self-assembling nanoparticle vaccine constructed by fusing immunodominant B- and T-cell epitopes from OMP16 to the self-assembling antigen BP26 via flexible linkers, yielding single-copy (1×) and double-co

Development of Novel Antimicrobial Peptides and Their Engineering into Nanoparticles to Improve Performance.

Antimicrobial peptides (AMPs) are promising alternatives to antibiotics, but discovering potent, low-toxicity candidates and improving their delivery remain challenging. In this study, novel AMPs were identified by constructing and screening a synthetic random peptide library using a bacterial surface display system. Rational design generated derivative peptides, among which WP-4 and WP-6 showed high antimicrobial activity, good biocompatibility, rapid bactericidal effects, and low propensity for resistance development. WP-6 also showed good in vivo therapeutic potency in a murine Escherichia

Biopolymer-Antigen Conjugate Vaccines for Prevention of Tularemia.

Francisella tularensis, the causative agent of tularemia, is a facultative intracellular pathogen categorized as a tier I bioterrorism agent. The current live vaccine strain (LVS) vaccine poses safety and virulence concerns, advocating the need for a safer alternative. In this study, we demonstrated a combination of antigens FopA, Tul4, SucB, and DnaK conjugated to biopolymer particles (BPs) as a multivalent subunit vaccine that not only induces LVS and SchuS4-specific antibody titers but also confers protective immunity with differential cytokine profiles upon LVS challenge. This multivalent

Mycobacterium tuberculosis Rv3248c Inhibits Type I Interferon Response and Promotes Intracellular Survival by Mediating Ubiquitin-Proteasome-Dependent Degradation of TBK1.

Mycobacterium tuberculosis (M. tb) is a leading cause of infectious disease mortality worldwide, and M. tb subverts host immunity to persist intracellularly. Type I interferon (IFN-I) modulates host defense during M. tb infection, but the mechanisms controlling their induction remain incompletely defined. In this research, a previously uncharacterized mycobacterial factor, M. tb Rv3248c, which regulates IFN-I response, was identified using pull-down and mass spectrometry. Overexpression of Rv3248c suppresses IFN-β expression in macrophages at both the transcriptional and protein levels. In add

Bacteriophage Therapy in Cardiovascular Medicine: A Paradigm Shift in the Prevention of Device-Related Infections.

Cardiovascular implant infections significantly impact morbidity and mortality. Bacteriophage therapy has emerged as a potential strategy for device-related infection, particularly in biofilm-associated settings where conventional antimicrobial therapy is limited, often necessitating device removal. This review outlines phage treatment principles, including biofilm invasion, phage-antibiotic interaction, phage resistance, and infection protection mechanisms. We explore innovative approaches for using bacteriophages to treat infections around prosthetic vascular grafts, heart valves, left ventr

Collateral Sensitivity in Bacteria: Phenomenon, Mechanisms, Modulators and Therapeutic Prospects.

The rapid proliferation and infection of multidrug-resistant bacteria pose a significant threat to global public health. The gradual increase in drug resistance, primarily driven by the inappropriate use of antibiotics, underscores the urgent need for developing novel therapeutic approaches for drug-resistant bacterial infections. Collateral sensitivity (CS) describes the phenomenon whereby bacteria resistance to a specific antibiotic increases susceptibility to other antibiotics. In some cases of antibiotic resistance, CS can provide rapid and effective guidance for clinical drug use by detec

Differentially Expressed Infection-Associated Genes in a Shewanella algae isolate From a Deceased Northern Bottlenose Whale.

Diseases are an important but poorly understood influence on marine ecosystems. However, many pathogens of wild marine organisms remain poorly characterized, limiting our understanding of their ecology and potential impacts. This study investigates the potential virulence mechanisms of a Shewanella algae strain isolated from the brain tissue of a deceased northern bottlenose whale (Hyperoodon ampullatus). Using long-read Oxford Nanopore RNA Sequencing, differential gene expression analysis (DGEA) was conducted comparing growth in standard media, Tryptic Soy Broth (TSB) and TSB supplemented wit

Comparative genomic and phenotypic analysis of human- and dairy cattle -derived Streptococcus agalactiae reveals potential cross-species infection capability.

BACKGROUND: Streptococcus agalactiae is an opportunistic Gram-positive pathogen that can infect humans, dairy cattle, and other animals, posing a potential zoonotic risk. In this study, whole-genome sequencing was performed on 40 GBS strains isolated from dairy cows. The results showed high homology among strains collected from the same farm. Based on these findings, two representative dairy cattle-derived strains and one human-derived strain were selected for comparative whole-genome analysis to identify differences in antibiotic resistance and virulence genes. Additionally, we characterized

Context-dependent effects of antimicrobial coatings on microbial load and bacterial community composition on public high-touch surfaces.

BACKGROUND: Antimicrobial surfaces incorporated into high-touch public areas are used as a passive intervention to reduce surface microbial load and reduce the spread of infections. However, usually such surfaces lack proof of their antimicrobial activity in real-use conditions and against a wider variety of microbes. This study evaluated the real-world performance of copper, TiO2-, silver- and quaternary ammonium compound (SiQAC)-based surfaces, which are commercially available and have proven antibacterial activity in lab tests. The surfaces were introduced to five study sites in diverse env

Two-component system VraSR senses host defense peptide PMAP-36 to promote Streptococcus suis pathogenicity through regulating accD expression.

Streptococcus suis (S. suis) is an important zoonotic pathogen that causes severe infections in humans and animals worldwide. Upon infection, the host innate immune system produces host defense peptides (HDPs) to resist S. suis invasion. PMAP-36, a porcine-derived HDP, has been shown to fail to kill S. suis, suggesting that this pathogen has evolved strategies to evade PMAP-36-mediated immune defense. However, the exact mechanism by which S. suis responds to PMAP-36 remains unclear. Here we showed that PMAP-36 significantly enhanced S. suis adhesion, cytotoxicity, and virulence gene expression

[A delayed threat: S. aureus sepsis 4 years post-IUD insertion in a healthy young woman].

BACKGROUND: Intrauterine devices (IUDs) are highly effective long-acting reversible contraceptives with an established safety profile. Infectious complications are rare and usually occur shortly after insertion, typically involving polymicrobial pathogens. Staphylococcus aureus is an uncommon pathogen in this context, but its biofilm-forming ability may contribute to delayed and severe presentations. OBJECTIVES: A healthy 25-year-old primiparous woman developed septic shock four years after the insertion of a levonorgestrel-releasing intrauterine system (Mirena®). She was admitted to the inte
