Sir Wilfrid Edward Le Gros Clark (1895-1971) was a pioneering British anatomist, physician, and evolutionary biologist whose legacy spans the foundational development of twentieth century neuroanatomy, primate biology, and medical education. From his formative experiences as a young medical officer in the First World War to his later academic ascendancy at the University of Oxford, Le Gros Clark exemplified a rare fusion of clinical insight, anatomical precision, and philosophical curiosity. His tenure in Borneo, where he studied primates in their natural habitats, enriched his evolutionary pe

Melatonin (MT), a neurohormone secreted by the mammalian pineal gland, has been demonstrated to exert hepatoprotective effects. In this study, we employed exogenous MT intervention in both in vivo and in vitro models of chronic liver injury induced by D-galactose (D-gal) in mice to investigate the specific mechanisms underlying its hepatoprotective action, aiming to provide a reference for the prevention of liver injury by MT. Preliminary network toxicology and network pharmacology analyses revealed that D-gal possesses potential hepatotoxicity, whereas MT exerts hepatoprotective effects throu

INTRODUCTION: The placenta produces melatonin during pregnancy, a hormone essential for circadian regulation and maternal-fetal metabolic, hormonal, and immunological modulation. Throughout pregnancy, melatonin production increases progressively, being synthesized by both the pineal gland and the placenta. Since exposure to artificial light at night (ALAN) reduces pineal melatonin secretion, it is hypothesized that light pollution may negatively impact women's health, maternal health, and fetal development due to the inhibition of both pineal and placental synthesis.

Cetylpyridinium chloride (CPC) is a prevalent disinfectant utilized as an antimicrobial agent at millimolar concentrations in numerous products directly applied to humans. Recently, CPC has been documented to have detrimental effects on female reproduction. Nevertheless, the precise mechanisms through which CPC impairs mammalian oocyte quality and the corresponding protective interventions remain inadequately elucidated. In this study, we identified that CPC can hinder oocyte maturation in vitro at nanomolar concentrations. Mechanistically, CPC disrupts spindle morphology and chromosome alignm

Cardiovascular disease remains the leading cause of mortality globally, with a disproportionate burden in low- and middle-income countries. Melatonin, a hormone primarily produced by the pineal gland, exhibits notable antioxidant, anti-inflammatory and immunomodulatory properties that may confer cardiovascular benefits. Importantly, melatonin is also found in a wide range of dietary sources, raising the possibility that regular consumption of melatonin-rich foods could offer a low-cost, accessible strategy to support cardiovascular health. This review summarises knowledge on dietary melatonin

We describe a unique case of papillary tumour of the pineal region (PTPR) arising in the fourth ventricle without any demonstrable anatomical continuity with the pineal region, in a 2-year-old male patient harbouring a germline PTEN pathogenic variant. This case arisen in the fourth ventricle shows epigenetic characteristics consistent with PTPR, suggesting the possibility that PTPR develops outside the pineal region. The tumour was composed of epithelioid-looking papillary structures (A, Haematoxylin and eosin [H&E]), intermingled with more densely cellular areas displaying perivascular pseud

The vertebrate brain is protected from infection by tight barriers. However, several barrier structures, including circumventricular organs, can be breached by pathogens. Here, we show that Cochlin, an extracellular matrix-binding protein with anti-bacterial properties, is produced in barrier structures and contributes to immune defence. Transcriptome analysis and in situ hybridisation indicate that cochlin is expressed in the pineal gland, area postrema, choroid plexus and discrete regions of the meninges of zebrafish, mice and humans. The protein is present in the cerebrospinal fluid, and on

The hormone melatonin, secreted by the pineal gland, has attracted considerable interest due to its potential to prevent liver disorders by its strong antioxidant characteristics. This review examines the diverse functions of melatonin in maintaining liver health, with a specific emphasis on its mechanisms of action in different liver diseases. Melatonin counteracts the harmful effects of alcohol on the liver by reducing oxidative stress and maintaining the proper functioning of mitochondria. Melatonin improves insulin sensitivity and regulates lipid metabolism in metabolic liver diseases, suc

Epidermoid cysts comprise about 1% of intracranial tumors, they are benign and extra-axial, arising from ectopic ectodermal tissue. Pineal region localization is rare. Surgical treatment can be challenging due to critical structures encountered along surgical corridors. We aimed to synthesize the literature compiled on this disease and present an illustrative case. We performed a scoping review according to PRISMA-Sc guidelines. Studies describing pineal region epidermoid cysts reporting clinical, surgical, or radiological data were included. A total of 33 patients from 31 studies were include

Melatonin (MT) is an indoleamine primarily produced by the pineal gland in a light-dark-dependent manner; however, it is also locally synthesized in the testis, where it influences male reproductive physiology upon interaction with its receptors (MT1/MT2), the binding started the bio-signaling inside the cells mediated by MT. This review outlines the intratesticular effects of MT, addressing seasonal and non-seasonal mammals. In rodents, Leydig, Sertoli, and germ cells contain enzymatic machinery to produce MT, with MT1/MT2 signaling influencing androgen production through cAMP-dependent and i

Mutational inactivation of the DICER1 gene causes aberrant micro-RNA maturation, which in turn may have consequences for the posttranscriptional regulation of gene expression, thereby contributing to tumor formation in various organs. Germline DICER1 mutations cause DICER1 syndrome, a pleiotropic condition with an increased risk of various neoplastic conditions in the pleura, ovaries, thyroid, pituitary, pineal gland, and mesenchymal tissues. Somatic DICER1 mutations are also frequently observed in a wide variety of solid tumors, thereby highlighting the importance of this gene in tumor develo

Diabetic cardiomyopathy (DCM) is one of the most severe cardiovascular complications of diabetes mellitus (DM), yet effective treatment strategies remain lacking. Cellular senescence is a principal risk factor for multiple cardiovascular diseases, contributing significantly to cardiac dysfunction and disease progression. Melatonin, an endogenous hormone primarily secreted by the pineal gland, exerts cardioprotective effects against DCM. However, the underlying molecular mechanism remains incompletely understood, particularly whether melatonin ameliorates DCM by suppressing cardiomyocyte senesc

BACKGROUND: Melatonin is one substance that is primarily generated by the pineal gland and numerous other tissues and has vital involvement in many physiological processes, including bone remodeling. Over the last several years, another emerging molecule serving as a potential candidate for periodontal regeneration is the hyaluronic acid (HA), a key extracellular matrix component involved in cell migration. The aim of this study is the additive effect of hyaluronic acid and melatonin with GTR membrane in intrabony defects among patients with chronic periodontitis.

Melatonin is not only produced by the pineal gland but is also synthesized in extrapineal organs. It plays a key role in antioxidant defense and immune regulation during pregnancy. Two key rate-limiting enzymes are involved in the melatonin synthesis, aralkylamine N-acetyltransferase (AANAT), and acetylserotonin O-methyltransferase (ASMT). Nevertheless, it remained unclear whether pregnancy affects the expression of AANAT and ASMT in extrapineal organs. In this study, ovine maternal thymus, spleen, liver, lymph nodes, thyroid, duodenum, and endometrium were collected on day 16 of the estrous c

Deuterium (2H), a heavy isotope of protium (1H), is a naturally occurring element with a significant impact on human metabolism. Despite its natural presence, deuterium can impair mitochondrial function by damaging ATPase pumps; consequently, biological organisms have evolved sophisticated strategies to mitigate the risks of deuterium overload and protect mitochondrial integrity. Multiple enzymes have evolved to prefer hydrogen over deuterium in their reactions to protect these pumps. One class of enzymes is the cytochrome P450 (CYP) enzymes, which oxidize many substrates, mainly in the Endopl

UNLABELLED: Hepatocellular carcinoma (HCC) is the most common form of primary liver cancer worldwide, with increasing incidence and mortality rates. Although several targeted therapies are currently available, the therapeutic outcomes remain unsatisfactory due to the high heterogeneity and drug resistance of HCC. Therefore, novel molecular mechanisms and therapeutic strategies urgently need to be explored. In this study, we obtained the GSE39791 dataset from the GEO database and identified 1,186 differentially expressed genes (DEGs). Weighted gene co-expression network analysis (WGCNA) was con

Primary central nervous system lymphoma (PCNSL) is a rare extranodal lymphoma, with diffuse large B cell lymphoma accounting for most cases. T cell variants are uncommon and associated with poor outcomes. We report a case of primary CNS T cell lymphoma with simultaneous brain and spinal cord involvement. A 37-year-old Japanese man presented with impaired consciousness, and brain magnetic resonance imaging revealed multiple contrast-enhancing parenchymal lesions. Histopathological examination of a brain biopsy specimen demonstrated medium- to large-sized atypical lymphoid cells, leading to a di

This chapter investigates the clinical picture and the different clinical signs that reveal pineal region tumors or appear during the course of the follow-up. Biological malignancy and tumor extension determine the semiology and its setting up mode. Typical endocrine signs, dominated by abnormal puberty development, are frequently a part of the clinical scene. Bifocal or ectopic localization in the hypothalamic-pituitary region is accompanied by other endocrine signs such as ante- or post-pituitary insufficiencies which occur several months or even years after the first neurological signs appe

Infertility is an increasing concern for many women and can affect both physical and emotional well-being. The central nervous system (CNS) - particularly the hypothalamus, pituitary gland, and pineal gland - plays a crucial role in female reproductive health. We conducted a narrative review of relevant studies published between 2015 and 2025, sourcing data from PubMed and Scopus. Our goal was to investigate how dysregulation of hormones from the hypothalamus, pituitary gland, and pineal gland contributes to fertility-related disorders, such as impairments in ovulation, oocyte quality, and emb
