Cross-Species Translation of Drug-Induced Toxicity in the Small Intestine Using Mathematical Modeling.

We previously detailed a quantitative systems toxicology modeling framework for early prediction of chemotherapeutic-induced clinical intestinal adverse effects. The model describes stem and progenitor cell dynamics in the small intestinal epithelium and integrates epithelial-related processes, such as transcriptional profiles, citrulline kinetics, and the probability that the organism will have diarrhea. This protocol describes a step-by-step guide to the examples detailed in the original manuscript, namely 5-FU- and doxorubicin-induced intestinal toxicity.

Animal-Free Intestinal Epithelial 2D "Organoid-Sheet" Cultures Using Invasin.

The intestinal epithelium consists of a single layer of polarized cells that serves as a crucial barrier against pathogens while mediating food digestion and nutrient absorption. The differentiated villus cells, important for the intestine's function, originate from rapidly dividing Lgr5+ stem cells located in the intestinal crypts. These stem cells can be expanded in vitro and form three-dimensional (3D) "mini-organs" known as intestinal organoids, which closely phenocopy the intestinal epithelium. The development and maintenance of these 3D organoids depend on a defined growth factor cocktai

Primary Cultures of Fetal Epithelial Progenitors.

In this chapter, we present a protocol for isolating fetal epithelial progenitors from the murine small intestine and colon, and for culturing them ex vivo using the three-dimensional organoid system. We also describe cellular approaches to characterize Lgr5-negative fetal progenitors and compare their identity and differentiation potential to those of Lgr5-positive adult intestinal stem cells. This system provides a valuable tool for studying the biology of fetal progenitors and exploring the developmental origins of adult intestinal stem cells.

Updated Protocols for 3D and 2D Culture and Histological Analysis of Intestinal Organoids.

The intestinal epithelium maintains continuous self-renewal and multi-differentiation capacities through the integrated regulation of multiple signaling pathways, including Wnt, bone morphogenetic protein, epidermal growth factor (EGF), and Notch signaling pathways. Based on this principle, a defined combination of EGF, Noggin, and the Wnt agonist R-spondin has been shown to support long-term expansion of mouse intestinal stem cells and the formation of organoids with indefinite self-renewal and full differentiation capacity. In human intestinal organoid cultures, the addition of small molecul

Human Small Intestinal Organoids Differentiation Protocols Towards All Mature Epithelial Lineages.

The small intestine plays a crucial role in nutrient uptake and metabolism regulation while maintaining a barrier that protects against the harsh environment of the lumen. These different tasks are carried out by a multitude of cell types, continuously generated by the resident stem cell population, and whose specification is under tight regulatory control. Organoids are self-organizing 3D structures derived from tissue-resident stem cells, which retain the ability to differentiate into all other epithelial lineages. Protocols to enrich for the various cell subtypes in the intestine have great

Stemness in Colon Cancer.

Cell plasticity is a major driver of colorectal cancer (CRC), as tumors hijack intestinal stem cell regenerative programs to create a dynamic, bidirectional axis between complementary stem-like states. Cancer stem-like cells (CSCs) display hallmark stem cell properties, including self-renewal, multipotency, controlled differentiation, and quiescence, and are now recognized as key orchestrators of CRC biology. They can sustain tumor initiation and long-term growth, seed metastases, enable periods of dormancy, and promote resistance to chemotherapy. As a result, therapeutically dismantling CSC p

Effects of in ovo feeding of arginine, tryptophan, and threonine on hatch performance, hatchability, and morphometry of broiler chickens.

The objective of this experiment was to investigate the effects of in ovo feeding of arginine (Arg), tryptophan (Trp), and threonine (Thr) on hatch performance, hatchability, and morphometry of newly hatched broiler chickens. A total of 400 fertile eggs from 28-week-old Arbor Acres broiler breeder flocks, with an average fertile egg weight of 52 ± 0.7 g, were collected for the experiment. The eggs were randomly assigned to five treatment groups (80 eggs per group, eight replicates of 10 eggs each): non-injected control (CON); phosphate-buffered saline (PBS) injection (100 μL of 1% PBS); Arg in

Mucosal attachmentand catalytic functionality of β-galactosidase-loaded polyelectrolyte microcapsules on porcine small-intestinal tissue ex vivo.

A key challenge in the development of localized intestinal enzyme systems is to retain enzyme carriers at the mucosal interface while preserving their catalytic functionality. To our knowledge, retention of enzyme-loaded layer-by-layer polyelectrolyte microcapsules (PMCs) on native small-intestinal mucosa has not previously been demonstrated. In this study, we investigated the ex vivo retention of β-galactosidase-loaded PMCs on porcine small-intestinal mucosa and determined whether the retained capsules preserved catalytic activity. FITC-labelled PMCs were incubated with mucosal explants from

Methods and Techniques for Closed-Loop Gastrointestinal Pacing.

PURPOSE: Gastrointestinal slow waves are rhythmic electrical events that contribute to the coordination of motility. Functional motility disorders such as gastroparesis are linked to abnormal slow wave activity, and current treatments do not target the underlying electrical dysfunction. Gastrointestinal electrical pacing restores normal rhythms, but open-loop pacing lacks adaptation to underlying slow wave activity, leading to reduced energy efficiency and potential overstimulation. METHODS: A novel closed-loop pacing system was developed to detect gastric and intestinal slow waves and dynami

Do wind energy installations alter gastrointestinal parasite infection patterns in roe deer (Capreolus capreolus L.) in central Poland?

The European roe deer (Capreolus capreolus L.) is a widespread cervid in Europe and an important reservoir of gastrointestinal helminths. Although environmental and host-related drivers of parasite communities have been extensively studied, the effects of anthropogenic infrastructure on host-parasite interactions remain poorly understood. This study aimed to characterize gastrointestinal helminth diversity in roe deer from central Poland and to evaluate the influence of wind farms on parasite infections. A total of 102 roe deer collected between 2022 and 2024, hunted in accordance with local m

Comparative genomics and transcriptome analysis reveal unique features of xenobiotic-metabolizing enzymes in elephants.

Elephants, the largest terrestrial mammals, have distinctive metabolic features that may explain their increased sensitivity to xenobiotics, including environmental pollutants and pharmaceuticals. While previous in silico studies have identified unique patterns in elephant cytochrome P450 (CYP), Phase I xenobiotic-metabolizing enzymes, much less is known about their Phase II enzyme systems. This study combines comparative genomic, phylogenetic, and transcriptomic analyses to thoroughly examine Phase I and Phase II xenobiotic-metabolizing enzymes in African (Loxodonta africana) and Asian elepha

Macroscopic and Microscopic Organization of the Intestine in the Arabian Jerboa (Jaculus loftusi): Adaptive Significance for Desert Survival.

The Arabian jerboa (Jaculus loftusi) able to thrive in one of the harshest conditions on the earth, where water is scarce and food is often dry and fibrous. To understand how this small desert rodent manages such challenges, we examined the structural and functional modifications of the hindgut in eight adult male individuals collected from Aljouf desert in northern Saudi Arabia. The digestive tract was about 62.7 cm long, nearly five times the jerboa's body length with the small intestine making up the greatest share (55.9%). The large intestine was characterized by a sacculated cecum serving

A comparative assessment of brush border membrane vesicles from porcine jejunum and small intestine powders from rat to study (poly)phenol hydrolysis and bioaccessibility.

Dietary (poly)phenols are predominantly present as sugar-conjugates, and the nature of the sugar influences their metabolic fate and biological activity. These compounds generally escape luminal digestion and undergo hydrolysis by intestinal brush-border α- and β-glycosidases, particularly by lactase-phloridzin hydrolase (LPH). This process is often overlooked in in vitro digestion models, limiting their physiological relevance for predicting the gastrointestinal fate of dietary (poly)phenols. In this study, brush-border membrane vesicles (BBMV) were isolated and purified from porcine jejunum

The resveratrol prodrug triacetylresveratrol targets the intestinal tract to alleviate PEDV infection via the AMPK-ULK1 autophagy axis.

Enteric coronaviruses, such as Porcine Epidemic Diarrhea Virus (PEDV), cause devastating mucosal infections in the swine industry. While host-directed natural compounds like resveratrol exhibit broad-spectrum antiviral potential in vitro, their in vivo translation is severely hindered by poor bioavailability. Here, we demonstrate that a lipophilic resveratrol prodrug, triacetylresveratrol (TCRV), overcomes these pharmacokinetic limitations to confer in vivo protection against PEDV infection. Utilizing a physiologically relevant suckling piglet model, we found that TCRV achieves targeted tissue

Underweight Versus Obesity and Gastrointestinal Cancer Mortality in Type 2 Diabetes: A Nationwide Cohort Study.

BACKGROUND: Underweight is a well-established risk factor for cancer mortality, yet its impact relative to obesity remains unclear in individuals with type 2 diabetes mellitus (T2DM), where current guidelines focus primarily on obesity management with limited guidance on underweight-related risks. We evaluated the association between body mass index (BMI) and gastrointestinal (GI) cancer mortality across the full BMI spectrum in individuals with T2DM. METHODS: This nationwide population-based cohort study utilized the Korean National Health Insurance Service database. We included 2 507 976 ad

High persistence of hepatitis A and E viruses during simulated gastrointestinal digestion across diverse food matrices.

Hepatitis A virus (HAV) and hepatitis E virus (HEV) are major causes of foodborne viral infections worldwide. Although both viruses are known to be transmitted via the fecal-oral route, their persistence during passage through the human gastrointestinal tract, particularly in the presence of different food matrices, remains poorly characterized. In this study, the persistence of HAV and HEV was evaluated using two complementary in vitro digestion models, the static INFOGEST 2.0 protocol and the dynamic TIM-1 system, simulating conditions of the stomach and small intestine. Different food matri

Giant Cavernous Hemangioma in the Mesentery of the Descending Colon: A Case Report.

Gastrointestinal hemangiomas are rare, and those involving the mesentery are even more infrequent. The majority of cases reported in the literature had mesenteric cavernous hemangioma of the small intestine. To the best of our knowledge, this is the first documented case of a hemangioma originating from the mesentery of the descending colon. Preoperative diagnosis is exceedingly difficult, and various imaging modalities may be inconclusive. The patient was a 71-year-old Asian male presenting with recurrent dizziness and fatigue over several months. Physical examination revealed a mass that was

Peritoneal neuroendocrine carcinoma of unknown primary with markedly elevated alpha-fetoprotein: a case report.

BACKGROUND: Peritoneal neuroendocrine carcinoma(NEC) is primarily metastatic, with primary cases being exceedingly rare. The relationship between alpha-fetoprotein(AFP) and neuroendocrine carcinoma remains unclear. In this report, we present a patient with peritoneal neuroendocrine carcinoma who exhibited significantly elevated AFP levels. Extensive evaluation revealed no obvious primary site outside the peritoneum; however, occult gastrointestinal origin could not be entirely excluded. The tumor was thus classified as NEC of unknown primary, with a presumed primary peritoneal origin. The find
