Beyond the Rumen: Current Evidence and Knowledge Gaps in Microbial Diversity and Function Across the Bovine Gastrointestinal Tract.

The bovine gastrointestinal tract (GIT) is a spatially organised microbial ecosystem, but the evidence supporting a whole-tract interpretation is highly uneven. This critical review distinguishes well-established rumen biology from emerging observations in the reticulum, omasum, abomasum, small intestine and hindgut. Comparative studies show strong regional filtering of microbial communities, yet most available datasets are cross-sectional, use digesta rather than mucosa, and infer function from DNA. Consequently, the presence of genes or taxa cannot be equated with active metabolism. Early li

Mucosal calprotectin is associated with severity of aGI-GVHD and poor outcomes after allogeneic stem cell transplantation.

Calprotectin, a calcium- and zinc-binding protein that comprises the subunits S100A8 and S100A9, has been extensively studied as a biomarker of gastrointestinal (GI) inflammation through fecal and serum analyses. However, its role in intestinal tissue remains poorly understood because of the limited availability of biopsy specimens. In this study, we analyzed S100A8 and S100A9 messenger RNA (mRNA) expression in 579 intestinal biopsy specimens from allogeneic stem cell transplantation recipients and observed a strong association with acute GI graft-versus-host disease (aGI-GVHD; P< .001). Adult intestinal malrotation: imaging findings.

Intestinal malrotation is the most common congenital anomaly of the small intestine. However, it is associated with delayed diagnosis due to the lack of specificity of its symptoms, which can lead to devastating consequences such as intestinal volvulus or massive intestinal necrosis. We present a clinical case in which we highlight the importance of abdominal computed tomography and the detection of its characteristic signs for the early identification of this pathology.

Purification and characterization of an IgG Fc gamma binding protein from the mouse intestine that interacts with lactoferrin.

Lactoferrin is a multifunctional protein mainly involved in the immune defence mechanisms against various pathogens. It has been reported that intestinal inflammation was reduced by lactoferrin administration. However, the precise mechanism underlying lactoferrin's involvement in intestinal inflammation is not yet fully understood. In this study, we purified a mouse intestinal lactoferrin-binding protein with a molecular mass of ~400 kDa that was expressed in the small intestine and colon. Sequence analysis revealed that the intestinal lactoferrin-binding protein represented an ortholog of rat

Characterization of the inflammatory response of a canine intestinal epithelial cell line challenged with lipopolysaccharides and/or butyrate.

Cell culture models are used to assess the impacts that dietary changes have on host health; however, there is limited knowledge about canine intestinal cell models compared to those for humans. The objective of this study was to use a canine intestinal epithelial cell (cIEC) line to investigate responses to pro- and anti-inflammatory treatments relevant to health in the dog. It was anticipated that lipopolysaccharides (LPS) would cause a pro-inflammatory response, while butyrate would cause an anti-inflammatory response and mitigate the pro-inflammatory response induced by LPS. cIEC were stim

Calcium-Sensing Receptor Regulation of Gastrointestinal Hormone Secretion.

The interaction of dietary nutrients with chemoreceptors in the gastrointestinal tract after a meal stimulates the secretion of gut hormones, which trigger the key processes of digestion and absorption, and also regulate energy intake and postprandial glycemia. One of these receptors, first recognized for its capacity to gauge extracellular calcium (Ca2+), is the calcium-sensing receptor (CaSR). Subsequent to its cloning, the CaSR was found to sense not only Ca2+, but also L-amino acids (AAs) and, based on solved protein structures, distinct binding sites have been reported for Ca2+ ions and t

Tissue signatures of human macrophages during homeostasis and activation.

Human macrophages (MΦs) reside in tissues and develop tissue-specific identities. While studies in mice have identified molecular signatures for site-specific MΦ differentiation, less is known about the transcriptional profiles of human MΦs in distinct sites, including mucosal tissues and lymphoid organs during homeostasis and activation. Here, we use multimodal single-cell sequencing and ex vivo stimulation assays to define tissue signatures for populations of human MΦs isolated from lungs, small intestine, spleen, bone marrow, and lymph nodes obtained from individual organ donors. Our result

Influence of dietary supplementation with chestnut-derived phytobiotics on growth performance, visceral organs, jejunal morphology, cecal microflora, and blood biochemicals in broiler chickens.

BACKGROUND: The broiler industry is facing major challenges following the prohibition of antibiotic growth promoters, with phytobiotics being a promising potential alternative. This study aimed to evaluate the effects of dietary chestnut burs and flower phytobiotics on performance, jejunal morphology, cecal microflora, visceral organs, and blood biochemicals in broiler chickens. Day-old male Ross broiler chickens (n = 384) were assigned to eight replicates, which were randomly allocated to six treatment groups for a 42-day feeding trial. The treatments were as follows: BD - basal diet (without

Modulation of Inflammatory and Apoptotic Pathways by Tamarix Aphylla Essential Oil in TNBS-Induced Colitis: A Focus on Cytokine Balancing, NF-κB, p38 MAPK, and Nrf2 Activation.

Inflammatory Bowel Disease (IBD) refers to the inflammatory disorders of the colon and small intestine. The effect of the essential oil from Tamarix aphylla (TAEO) on inflammation and apoptosis has been investigated using a rat TNBS-induced colitis model. The study utilized macroscopic and histopathological evaluations, cytokine profiling by ELISA, and protein expression assays to assess the effects of TAEO on ulceration, cytokine levels, apoptotic proteins, and anti-apoptotic proteins. Sound experimental groups received several dosages of TAEO (10, 30, and hundred mg/kg) and dexamethasone ser

T-bet-expressing Tr1 cells driven by dietary signals dominate the small intestinal immune landscape.

Intestinal immunity defends against enteric pathogens, mediates symbiotic relationships with the resident microbiota, and provides tolerance to food antigens, safeguarding critical nutrient absorption and barrier functions of this mucosal tissue. Despite the abundance of tissue resident activated T cells, their contributions to these various roles remain poorly understood. Here, we identify a dominant population of IL-10 producing, T-bet-expressing Tr1 T cells, residing in the small intestinal lamina propria at homeostasis. Remarkably, these intestinal Tr1 cells emerge at the time of weaning a

Di-n-pentyl phthalate exposure alters intestinal structure and gut microbiota composition and characteristics in mice.

BACKGROUND: Di-n-pentyl phthalate (DnPP), a ubiquitous plasticizer structurally analogous to the gut toxicant di-(2-ethylhexyl) phthalate (DEHP), poses environmental persistence and human exposure risks, yet its gastrointestinal toxicity remains poorly characterized. We hypothesized DnPP disrupts intestinal homeostasis via gut microbiota dysbiosis, mirroring mechanisms of other phthalates. METHODS: C57BL/6 mice (n = 10 per group) were orally gavaged with DnPP (1-100 mg/kg/d) for 21 days. Intestinal tissues and microbiota were analyzed using histomorphometry and metagenomic sequencing with fun

Moringa oleifera ameliorates oxidative damage caused by sodium valproate in the small intestine.

This study aimed to investigate the protective effects of Moringa oleifera (MO) extract against valproic acid (VPA)-induced small intestine damage in rats. Forty-six adult female Sprague-Dawley rats were divided into four groups: Control (saline), MO (300 mg/kg), VPA (500 mg/kg), and VPA + MO. All treatments were administered orally for 15 days. Biochemical oxidative stress analyses revealed that MO treatment mitigated oxidative stress in VPA-treated rats. Molecular docking studies demonstrated that bioactive compounds in MO leaves exhibited potential inhibitory activity against oxidative stre

Expression of gastrointestinal clock genes and associated hormones levels in hibernating and non-hibernating Daurian ground squirrels.

Extensive evidence indicates that the gut phenotypes of hibernating animals undergo periodic seasonal changes. However, the rhythmic mechanisms underlying this seasonal plasticity remain poorly understood. As an obligate hibernator, the Daurian ground squirrel (Spermophilus dauricus) displays seasonal feeding behaviors accompanied by pronounced seasonal plasticity in its gastrointestinal (GI) tract. This study compared the expression of clock genes in the stomach and small intestine of ground squirrels across different seasons (autumn, winter and spring), as well as the levels of serum arginin

Advances in Dietary Modulation of the Intestinal Barrier: Mechanistic, Structural, and Functional Insights.

The intestinal barrier maintains host health through the coordinated integration of mechanical, chemical, immune, and microbial defenses across intestinal segments. Dietary factors are key regulators of barrier integrity, yet the mechanisms by which nutrients act during digestion, absorption, and microbial metabolism from the small intestine to the colon remain incompletely understood, with segment-specific functions often overlooked. This review summarizes recent advances and elucidates how dietary proteins, carbohydrates, lipids, micronutrients, and bioactive compounds shape intestinal barri

Hyperglycemia alters the gene and protein expression of CDC42 in small and large intestine of Sprague-Dawley rats.

Diabetes mellitus (DM) is associated with gastrointestinal complications, including structural and functional changes in both small and large intestine. CDC42, a Rho GTPase, plays a critical role in maintaining epithelial integrity through regulation of tight junctions and cytoskeletal organization. Moreover, CDC42 expression has been reported in inflammatory bowel disease (IBD). However, its expression patterns and regulatory mechanisms in the diabetic gut remain poorly defined, particularly in the context of DM - IBD comorbidity. Our study aimed to evaluate histological changes and CDC42 gen

Gastrin for the treatment of acute graft-versus-host disease of the stomach.

Acute graft-versus-host disease (aGVHD) is a major cause of death after allogeneic hematopoietic cell transplantation, and patients with steroid-refractory aGVHD have a dismal prognosis. We have previously shown that the enteroendocrine hormone glucagon-like peptide-2 has tissue regenerative activity in the lower gastrointestinal tract in mice and patients with steroid-refractory aGVHD. Here, we explored the tissue protective effect of the enteroendocrine hormone gastrin for aGVHD of the stomach. We observed that aGVHD caused a loss of gastrin-producing G-cells and parietal cells (PCs) and an
