Oral delivery of an ustekinumab biosimilar with bioavailability comparable to subcutaneous administration in healthy human participants.

PURPOSE: Ustekinumab, targeting interleukin (IL)-12 and IL-23, is effective in treating chronic immune-mediated inflammatory diseases, but needs to be dosed via subcutaneous (SC) injection, which impacts patient comfort and treatment adherence. To enable oral dosing of ustekinumab, the utility of a robotic pill (RP) was evaluated in humans. METHODS: A Phase 1 study was conducted to evaluate the safety, tolerability, and pharmacokinetics (PK) of single doses (0.5 and 0.75 mg) of an ustekinumab biosimilar (CT-P43) delivered via RP (RT-111) in healthy participants compared to ustekinumab (0.5 mg)

Metformin improves the impairment of cognitive function caused by total abdominal irradiation via the miR-34a-5p/BDNF/CREB axis.

The gastrointestinal system is of particular importance in radiation biodosimetry because of its constant cell renewal and sensitivity to radiation-induced injury. It has been reported that total abdominal irradiation causes distant cognitive defects in a mouse model. In this study, we demonstrated that metformin alleviated the cognitive dysfunction caused by total abdominal irradiation. No neuropathological changes were observed in hippocampal tissues in control, irradiated, and irradiated plus metformin-treated groups. However, we found that metformin treatment improved the expression of bra

Dietary muramidase leads to the downregulation of peptidoglycan biosynthesis and to caecal microbial modulation in laying hens.

BACKGROUND: In-feed muramidase enzyme has been linked to numerous advantages in several animal species. In the past years, muramidase has been shown to be effective in hydrolyzing peptidoglycan fragments, especially at small-intestine level in broilers, and to improve digestibility and performance. Moreover, previous studies also showed a possible anti-inflammatory effect of some secondary metabolites derived from the hydrolysis of peptidoglycan. Although a major effort has been carried out to unravel the in vivo mechanism of action of muramidase, there is currently little information on its m

Programmed Sequential Release Behavior and Bioavailability of Curcumin and Fucoxanthin Co-Encapsulated in Solid-In-Oil-In-Water Multilayer Emulsions.

Food-grade co-delivery systems have garnered significant attention for their ability to deliver two or more bioactive components simultaneously. Co-delivery systems possessing programmed sequential release properties allow sequential delivery of bioactive components to different sites in the gastrointestinal tract (GIT) to enhance their bioavailability. This study constructed solid-in-oil-in-water multilayer emulsions (S/O/W-E) using carboxymethyl konjac glucomannan-coated gliadin nanoparticles as the solid phase, coconut oil as the oil phase, and carboxymethyl starch/propylene glycol alginate

Proteomic and Lipidomic Atlas of Gut-Associated Lymph and Venous Depots in Female Piglets.

BACKGROUND: The venous and lymphatic outflows from organs may contain molecular signatures related to tissue function, but deep biomolecular analyses that compare outflow composition are lacking. Here, using blood and lymph samples from a piglet model system and mass spectrometry analysis, we compare protein and lipid cargo from 9 venous and 3 lymph depots centered on intestinal outflow. METHODS: We obtained venous blood and plasma from piglets using new methods for dissection and sample collection. We applied mass spectrometry-based proteomics and lipidomics, using both a low-volume method a

Long noncoding RNA GAS5 disrupts intestinal epithelial barrier function by increasing small vault RNA levels.

Disruptions in the integrity of the intestinal epithelium occur commonly in inflammatory bowel disease (IBD) and critical surgical disorders, but the underlying mechanisms remain largely unknown. Here we identified long noncoding RNA GAS5 as a repressor of intestinal mucosal growth and the function of the gut epithelial barrier. The levels of tissue GAS5/Gas5 increased in mouse intestinal mucosa after colitis and septic stress, as well as in human intestinal mucosa from patients with IBD. Transient and tissue-specific knockdown of Gas5 in mice using CRISPR/Cas9 enhanced the renewal of the muco

Molecular and Chromatin Accessibility Programs Underlying Epithelial Injury and Impaired Regeneration in Neonatal Necrotizing Enterocolitis.

BACKGROUND & AIMS: Neonatal necrotizing enterocolitis (NEC) is a severe gastrointestinal disorder with high mortality, characterized by epithelial cell injury and compromised epithelial repair. The mechanisms underlying defective epithelial regeneration remain poorly understood despite advances in single-cell omics. Addressing these challenges is essential for elucidating the pathogenesis of NEC and identifying therapeutic targets to restore epithelial regeneration and replace the damaged epithelial layer. METHODS: Using a well-established neonatal mouse model of NEC induced by formula feedin

Konjac glucomannan enhances the freeze-thaw stability of Pickering emulsions with different oil phases stabilized by double-induced whey protein isolate nanoparticles and influences their digestion characteristics.

Firstly, the present study was conducted to characterize the freeze-thaw stability of Pickering emulsions (PEs) stabilized by double-induced whey protein isolation nanoparticles (DI-WPINPs), using corn oil, soybean oil, and flaxseed oil phases as distinct oil phases, while also elucidating the underlying mechanisms involved in this process. Results revealed that DI-WPINPs-stabilized PEs with the addition of 0.04% konjac glucomannan (KGM) maintained excellent freeze-thaw stability across different oil phases. Incorporating KGM did not significantly alter the particle size of DI-WPINPs, but it d

Synthetic mucus biomaterials enable localized therapeutic antibody delivery in inflammatory bowel disease.

Inflammatory bowel disease (IBD) is a chronic condition characterized by recurrent gastrointestinal inflammation that requires long-term therapeutic intervention. While anti-TNF-α monoclonal antibodies (mAbs) are effective in maintaining remission in IBD, systemic delivery is associated with immunosuppression, poor targeting efficiency, and high cost. To address these limitations, we developed a synthetic mucin-based hydrogel for localized delivery of TNF-α-targeting mAbs. Mucins are heavily glycosylated biopolymers that naturally bind antimicrobial and anti-inflammatory proteins, making them

The Multifaceted Role of Cytochrome P450 Isoenzymes in Alcoholic and Metabolic (Dysfunction) Fatty Liver Diseases.

The current review explores the alterations in cytochrome P450 (CYP) activity and expression during alcoholic liver disease (ALD) and metabolic (dysfunction)-associated fatty liver disease (MAFLD), formerly known as non-alcoholic fatty liver disease (NAFLD). CYP is a major family of enzymes involved in the metabolism of numerous endogenous and exogenous compounds. Thus, any change in CYP activity or expression could disrupt metabolic pathways. Alterations in hepatic CYP have been shown to contribute to the development of ALD and MAFLD, and vice versa. CYP isoforms also participate in fatty aci

Electronically switchable dual-band capsule antenna for wireless endoscopic applications.

This paper presents a highly compact linearly polarized planar dual annular ring antenna designed for wireless capsule endoscopy. The focus of this work on the small intestine is motivated by clinical practice in capsule endoscopy. Most capsule endoscopy systems are specifically designed to examine the small intestines, which are difficult to access with conventional wired endoscopes and are the primary region of interest for many gastrointestinal pathologists. The antenna is derived from a conventional annular ring structure and supports dual-band operation, covering both Wi-Fi frequencies at

Alfalfa Polysaccharide Improves Rabbit Growth by Modulating Gut Microbiota and Suppressing Inflammation Through PPARγ/NF-κB Pathway.

Meat rabbits are ideal meat-producing animals. However, weaning-induced intestinal inflammation often leads to growth delays, and severe cases impair breeding efficiency. Alfalfa polysaccharides (APSs) have antioxidant and anti-inflammatory properties, making them potential natural alternatives to antibiotics. To date, relatively limited research has been conducted on APS in meat rabbits. This research investigated the effects of APS on growth performance, intestinal inflammation, and meat quality in rabbits. Eighty healthy rabbits were randomized into four treatment groups, each group consist

Oyster Peptide-Ferrous Chelate Preparation Optimization Structural Characteristics and Enhanced Bioavailability.

Iron deficiency anemia remains a global nutritional challenge due to the low bioavailability and side effects of conventional inorganic iron supplements. A novel organic iron supplement, oyster peptide ferrous chelate (OP-Fe), was prepared using oyster peptides as ligands. Its preparation process was optimized via single-factor experiments and statistical methods with the optimal conditions identified as 1% peptide concentration, 35 °C chelation temperature, 3.91:1 peptide-to-iron ratio, 1.49% ascorbic acid concentration and pH 6.05. Under these conditions, the chelate's iron content reached 1

A Human β-Defensin-Based Recombinant Protein DF2-HSA Ameliorates Cytokine Storm.

Cytokine storm is a critical driver of acute respiratory distress syndrome and multiple organ failure. Human β-defensin 2 (HBD-2) is the first inducible defensin discovered in human body. Defensin can resist pathogenic microorganisms invading the body through direct bactericidal effect and also modulates acquired immune response. Albumin exhibits immunomodulatory properties and can reduce the level of inflammatory cytokines to improve the systemic inflammatory response. We previously engineered a recombinant fusion protein, DF2-HSA, comprising two HBD-2 molecules linked to human serum albumin.
