In this study, thyme essential oil nanoemulsions stabilized by clean-label emulsifiers (tea saponin, TS; soy lecithin, SL; soy protein isolate, SPI), with Tween 60 (TW) as a synthetic benchmark, were fabricated via ultrasonic emulsification (573 W/cm2, 5 min). Ultrasonication effectively reduced droplet sizes to 120-220 nm, producing narrow size distributions (PDI < 0.22) and high encapsulation efficiencies, which were associated with cavitation-induced droplet disruption and enhanced emulsifier adsorption at newly generated interfaces. Among the formulations, TS- and SL-stabilized nanoemulsio

Early thymic progenitors (ETPs) retain the potential for alternative fates but commit to the T-cell lineage as they progress through the CD4-CD8- double-negative (DN) thymocyte stages early in T-cell development. Current evidence suggests that T-lineage commitment occurs during the transition from the DN2a to DN2b stage, a process characterized by downregulation of c-KIT and CD44 expression. Moreover, Bcl11b activation in DN2a thymocytes closely coincides with T-lineage commitment. Despite this heterogeneity in Bcl11b-YFP expression within DN2a thymocytes, these cells have predominantly been s

The pathogenesis of the immune dysregulation in porcine reproductive and respiratory syndrome (PRRS) virus (PRRSV) infection is not fully understood yet. In this context, immune checkpoints have been identified as pivotal regulators of inflammatory and immune responses. This study evaluated the expression of CTLA-4 at early and late stages of PRRSV-1 infection in piglets with two strains of different virulence (3249 and Rosalía). Animals infected with the highly virulent Rosalía strain showed more severe clinical signs and a significantly reduced average daily gain. In addition, they presented

BACKGROUND: Uterine aging constitutes a pivotal factor contributing to fertility decline and miscarriage among advanced maternal age women. Regulatory T (Treg) cells play a critical role in establishing maternal-fetal immune tolerance during embryo implantation, and have been shown to correlate significantly with early pregnancy loss in advanced maternal age women. Previous studies have demonstrated that Jianwei Shoutai Pill (JSP) effectively reduces early pregnancy loss; however, its regulatory mechanisms on Treg cells in the uterine aging-associated pregnancy loss remain elusive.
OBJECTIVE:

INTRODUCTION: Immunosuppression induced by chemotherapeutic agents such as cyclophosphamide (CTX) is frequently associated with gut microbiota dysbiosis. Sanghuangporus sanghuang polysaccharide (SSP1) possesses immunomodulatory potential; however, its effects on mucosal immunity and the gut microbiota remain unclear.
METHODS: In this study, we investigated the immunomodulatory activity of SSP1 in CTX-induced immunosuppressed mice. Mice were treated with different doses of SSP1 orally. We assessed immune organ indices, histopathology of immune organs and colon, oxidative stress, splenocyte pro

The thymus is a critical organ for T cell generation. It is composed of distinct layers of thymic epithelial cells (TECs) that coordinate a complex process of antigen recognition and self-tolerance checks that result in mature populations of T cells. However, it undergoes progressive and permanent involution and loss of functionality which peaks in adolescence, rendering the tissue largely non-functional. Induced pluripotent stem cells (iPSCs) have emerged as promising candidates to generate organoid models of the thymus because of their autologous potential and indefinite self-renewal. Previo

A 1,2,5-tetrazole derivative (DTE) was synthesized via a ZnSO4-catalyzed [3 + 2] cycloaddition reaction between succinonitrile and sodium azide, and its structure was confirmed through experimental and theoretical approaches. The compound exhibited favorable biocompatibility in HUVEC cells, with half-maximal inhibitory concentration (IC50) values of 764.2 and 567.9 µg/mL after 24 and 48 h, respectively, indicating relatively low cytotoxicity. In vitro antibacterial assays showed moderate activity against both Escherichia coli and Staphylococcus aureus, with minimum inhibitory concentration (MI

Tregs play an essential role in immune tolerance, and Treg-promoting therapies are in development for the treatment of many inflammatory disorders. Interleukin-2 (IL-2)-based therapies increase Treg frequency, but little is known about impacts on Treg heterogeneity and function. We extended analyses of an IL-2 mutein (MK-6194) single-ascending-dose trial in healthy human participants by comprehensively defining Treg subsets and gene expression changes in vitro and in vivo. We found highly specific and dose-dependent activation and expansion of Tregs in clinical and pre-clinical studies. Follow

BACKGROUND: Approximately 10% of patients with Severe Combined Immunodeficiency (SCID) phenotype lack a known genetic cause. In particular, the molecular basis of thymic defects is poorly understood. Homeobox (HOX) genes encode conserved transcription factors that control spatial body development. The function of human HOXA3 is undefined.
OBJECTIVE: We characterize the first human with HOXA3 deficiency and investigate the role of HOXA3 in immune system development.
METHODS: We performed clinical and immunological phenotyping, as well as genetic sequencing. A homozygous nonsense variant in HO

Arachidonic acid released from phospholipids in the cell membrane, is used to synthesize lipid mediators known as leukotrienes (LTs). LTs play a key role in numerous leukocyte functions including leukocyte migration and inflammation. However, their function in the avian central nervous system is poorly understood. Thus, this study investigated whether lipopolysaccharide (LPS) a cell wall component of Gram-negative bacteria modulates LT synthesis via 5-LOX, an LT synthase, in chicks (Gallus gallus). Intraperitoneal LPS injection substantially increased 5-LOX mRNA expression in the diencephalon,

BACKGROUND: Stress-induced thymic involution is characterized by rapid thymocyte loss and is largely attributed to glucocorticoid signaling. However, the local immune mechanisms involved remain unclear. We investigated the role of the CX3CL1-CX3CR1 axis in restraint stress (RS)-induced thymic involution.
METHODS: Wild-type (WT) and Cx3cr1-deficient mice were subjected to repeated RS. Thymic morphology, CX3CL1/CX3CR1 expression, apoptosis-related pathways, and stress-related hormonal responses were examined. The effects of CX3CL1 on corticosterone (CORT)-induced thymocyte apoptosis in vitro an

Vitamin D has pleiotropic physiological roles including immune system regulation. Notably, there are links between vitamin D deficiency and risk of autoimmunity. Hormonal vitamin D binds to the vitamin D receptor (VDR) to exert its genomic effects. In immune cells, these include regulation of activation, inflammatory responses, and cytokine production. However, vitamin D supplementation is not efficacious in treatment of existing autoimmune disease. Conversely, vitamin D intake during childhood is associated with reduced risk of developing autoimmune type 1 diabetes. This supports a prophylact

Erythroderma is a severe, chronic condition that can affect more than 90% of the body surface area, occurring either idiopathically or secondarily to various underlying diseases. There have been several reports of erythroderma from outside Japan, while reports from inside Japan are limited. In this study, we retrospectively reviewed the clinical characteristics, underlying diseases, and treatments of patients diagnosed with erythroderma at our department between 2008 and 2024. A total of 147 patients were identified (122 males, 25 females; mean age, 69.2 years). Idiopathic erythroderma was the

BACKGROUND: Traditional Moroccan herbal teas are widely consumed and have been traditionally used for managing chronic metabolic disorders, particularly type 2 diabetes mellitus (T2DM). Although these preparations have a long history of ethnomedicinal use, their phenolic composition and biological activities remain insufficiently characterized. Scientific validation is needed to support their traditional application and provide evidence for their potential relevance to T2DM management. This study aimed to characterize the phenolic profile, antioxidant potential, and carbohydrate-digestive enzy

Post-translational modifications (PTMs) chemically diversify the ~20,000 genomically encoded proteins into millions of functional variants, and this diversity has profound consequences for T cell-mediated immune responses. PTM-reactive T cells have been identified across numerous autoimmune diseases and, in several cases, implicated directly as pathogenic drivers. Mechanistically, PTMs promote autoimmunity through two intertwined routes: by altering peptide-MHC and TCR binding to favor recognition of self-epitopes, and by generating neoepitopes in peripheral and/or inflamed tissues that are ab
