Profiling the cell-specific small non-coding RNA transcriptome of the human placenta.

The human placenta is the composite of multiple cell types, each which contributes uniquely to placental function. Small non-coding RNAs (sncRNAs) are regulators of gene expression and can be cell-specific. The sncRNA transcriptome of individual placental cell types has not yet been investigated due to difficulties in their procurement and isolation. Using a custom sequencing method, we explored the expression of seven sncRNA species (miRNA, piRNA, rRNA, scaRNA, snRNA, snoRNA, tRNA) from whole chorionic villi and four major sample-matched FACS-sorted cell type (cytotrophoblast, stromal, endoth

Hypoxia-Induced O-GlcNAcylation of GATA3 Leads to Excessive Testosterone Production in Preeclamptic Placentas.

The maintenance of endocrine homeostasis in the placenta is crucial for ensuring successful pregnancy. An abnormally elevated production of placental testosterone (T0) has been documented in patients with early-onset preeclamptic (E-PE). However, the underlying mechanisms remain unclear. In this study, we found that E-PE placentas exhibited significantly increased expressions of 3β-HSD1 (3β-Hydroxysteroid Dehydrogenase 1) and 17β-HSD3 (17β-Hydroxysteroid Dehydrogenase 3), the rate-limiting enzymes for T0 synthesis. This was strongly correlated with an elevated level of O-linked N-acetylglucosa

The anti-inflammatory effect of the amniotic PPARγ pathways is not dysregulated by the alternative plasticizer DINCH and its metabolite MINCH in human fetal membranes.

Plasticizers, particularly phthalates, are widely used to enhance the properties of plastics, yet their harmful effects on human health, especially reproductive health, have raised concerns. This has led governments to promote the use of non-phthalate substitutes. Preterm premature rupture of fetal membranes (PPROM), affecting 3-4% of pregnancies and contributing to 40-50% of preterm births worldwide, has been associated with traditional phthalate exposure. However, no clear link has yet been established between alternative plasticizers and preterm births. Among these substitutes, DINCH (used

The regulatory role of placental extracellular vesicle on trophoblast and endothelial cell functions.

Extracellular vesicles (EVs) are cell-derived, membrane-bound vesicles that carry molecular cargo to facilitate communication between cells. During pregnancy, EVs are secreted by the syncytiotrophoblast layer of the placenta villi, where they mediate the functions of resident leukocytes and invading extravillous trophoblasts (EVTs) in the decidua. This study aims to isolate placental EVs (pEVs) from placental explant to examine their regulatory roles on EVT and endothelial cell functions. pEVs were successfully isolated from ex vivo cultured placental explant, which were capable to be internal

Placenta Praevia with Abnormal Adhesion-A Retrospective Study.

Background: Placenta accreta spectrum (PAS) refers to abnormal placental attachment, categorized into placenta accreta, increta, and percreta, with varying severity. The incidence of PAS has risen alongside the increasing rate of caesarean sections. PAS is a significant cause of maternal complications, including bleeding, hysterectomies of necessity and intestinal or urinary surgical complications, and of foetal complications, preterm birth or foetal anaemia. Early diagnosis is crucial for its management and for improving its outcomes. Materials and Methods: This retrospective study, conducted

Seasonal Influences on Human Placental Transcriptomes Associated with Spontaneous Preterm Birth.

Demographic studies have revealed a strong association between exposure to high ambient temperatures during pregnancy and increased risks of preterm birth (PTB). The mechanism underlying this association is unclear, but it is plausible that altered placental function may contribute to it. In this study, we conducted differential gene expression analysis, gene set enrichment analysis (GSEA), and gene ontology (GO) analysis on bulk RNA-seq data from human placentas delivered at term and preterm during the warmer months compared to placentas delivered at term and preterm during the colder months

ZINC FINGER PROTEIN2 suppresses funiculus lignification to ensure seed loading efficiency in Arabidopsis.

The plant funiculus anchors the developing seed to the placenta within the inner dorsal pod strands of the silique wall and directly transports nutrients to the seeds. The lignified vasculature critically supports nutrient transport through the funiculus. However, molecular mechanisms underlying lignified secondary cell wall (SCW) biosynthesis in the funiculus remain elusive. Here, we show that the transcription factor ZINC FINGER PROTEIN2 (ZFP2) represses SCW formation in the cortex cells that surround the vasculature. This function is essential for efficient nutrient loading into the seeds.

The Significance of Selected Collagens and Their Connection with Relevant Extracellular Matrix Proteins in Bovine Early-Mid-Pregnancy and Parturition with and Without Retained Foetal Membranes.

Appropriate placental structure and function assure foetal development, delivery of nutrients, and removal of waste. Collagens, as structural proteins, are crucial for the maintenance of placental growth and function. The aim of this study was to describe the profile of collagen 1 and 4 in the placental tissues of cows and to correlate it to previously described activities of collagenases and adhesive proteins. Placental samples were collected from pregnant cows in the slaughterhouse (2nd, 4th, and 6th month; n = 12) and during parturition after caesarean section. Samples taken during caesarea

Clock Proteins Have the Potential to Improve Term Delivery Date Prediction: A Proof-of-Concept Study.

Our ability to accurately predict the delivery date of term pregnancies is limited by shortcomings of modern-day clinical tools and due date estimation methods. The pregnancy clock is a series of coordinated and harmonized signals between mother, fetus, and placenta that regulate the length of gestation. Clock proteins are thought to be important mediators of these signals, yet few studies have investigated their potential utility as predictors of term delivery date. In this study, we performed a cross-sectional proteome analysis of 2648 serum samples collected between 18 and 28 weeks of gesta

Human Placenta MSC-Derived DNA Fragments Exert Therapeutic Effects in a Skin Wound Model via the A2A Receptor.

PDRN, polydeoxyribonucleotide, which is used as a tissue-regeneration material, is present in human cells under physiological conditions and stimulates regeneration and metabolic activity. PDRN can be used as a biomaterial for several types of regeneration, including wound healing, to promote cell growth and growth-factor production. The aims of this study were to determine the effect of PDRN derived from human placenta-derived mesenchymal stem cells (hPD-MSCs) on cellular regeneration through A2A receptor signaling and to investigate its therapeutic effects in a mouse model of wound healing.

Evaluation of the effect of exogenous melatonin on the placenta and inflammatory cytokines in rats submitted to chronic alcohol consumption.

Global alcohol consumption by pregnant women is approximately 9.8%. This consumption can hinder the interaction of the placenta with the fetus and increase levels of oxidative stress by altering and deregulating the expression of cytokines, leading to complications such as slow intrauterine development, such as low birth weight, premature babies and even stillbirths. It is known that melatonin is a natural antioxidant that acts mainly on the liver and also on various organs such as the ovary, placenta and the fetus itself. Melatonin can freely cross the placenta and exert effects on the mother

MicroRNA profiling in umbilical cord plasma: links to maternal metabolism and neonatal metabolic and inflammatory traits.

MicroRNAs (miRNAs) are regulators of mRNA translation and play crucial roles in various physiological and pathological processes. In this study, we profiled miRNAs in umbilical cord plasma (UCP) to explore the association of neonatal circulating miRNAs with maternal metabolic parameters and neonatal anthropometric, metabolic and inflammatory characteristics in healthy pregnancies. Data and UCP samples were collected from 16 pregnancies, equally divided between normal-weight and overweight mothers and between male and female newborns. Using next-generation sequencing, we identified and quantifi

Controversies in chronic histiocytic intervillositis.

Understanding the"paradox" of pregnancy remains a challenging field of investigation especially when immunological dysregulation is suspected in pathological pregnancies. Chronic histiocytic intervillositis (CHI) is an example of a rare placental inflammatory disease that can occur during any trimester of pregnancy. The pathogenesis of CHI involves an abnormal immune response characterized by an inflammatory infiltrate of maternal CD68+ mononuclear immune cells in the intervillous space. CHI may be associated with villous and intervillous fibrinoid deposits. The precise immunological mechanism

Advanced multi-modal mass spectrometry imaging reveals functional differences of placental villous compartments at microscale resolution.

The placenta is a complex and heterogeneous organ that links the mother and fetus, playing a crucial role in nourishing and protecting the fetus throughout pregnancy. Integrative spatial multi-omics approaches can provide a systems-level understanding of molecular changes underlying the mechanisms leading to the histological variations of the placenta during healthy pregnancy and pregnancy complications. Herein, we advance our metabolome-informed proteome imaging (MIPI) workflow to include lipidomic imaging, while also expanding the molecular coverage of metabolomic imaging by incorporating on
