Cholinergic interneurons (CINs) in the nucleus accumbens (NAc) play a key role in regulating motivated behaviors. Here we examine the connectivity and functional impact of cortical and thalamic inputs onto CINs in the NAc core of mice of either sex. We first use cell type-specific retrograde anatomy to identify the prefrontal cortex and thalamus as putative afferents. We then combine ex vivo slice physiology and optogenetics to characterize the properties of synapses onto CINs. We demonstrate that thalamic inputs strongly facilitate, whereas cortical inputs exhibit marked depression. We also s

OBJECTIVES: Emerging evidence shows that sodium-glucose cotransporter 2 (SGLT2) inhibitors improve glycemic control and reduce the risk of cardiovascular-related mortality. This is contingent on adherence, yet evidence of SGLT2 inhibitor adherence and associated diabetes-related clinical outcomes in real-world settings is lacking. Therefore, we examined SGLT2 inhibitor longitudinal adherence trajectories and their association with the risk of diabetes-related hospitalizations.
STUDY DESIGN: SGLT2 inhibitor new-user retrospective cohort study using a 25% random sample of IQVIA PharMetrics Plus

BACKGROUND: Extensive or atypical dermal melanocytosis (EDM) is abnormal macular blue/black skin pigmentation that can be associated with other congenital abnormalities and an increased risk of melanoma. It is probably underdiagnosed due to phenotypic overlap with common transient dermal melanocytosis (so-called 'blue spots'), and is frequently misdiagnosed as congenital melanocytic naevi. The genetic cause has remained largely unknown, with mosaic variants in only one gene, GNAQ, described in only a handful of cases to date.
OBJECTIVES: To improve our understanding of EDM by combining deep p

Background/Objectives: This cross-sectional study aimed to evaluate the relationship between nutritional intake (energy, protein, calcium, number of meals, number of dairy products) and other factors (physical activity, past fractures, body composition) with forearm bone parameters in adolescent boys with myelomeningocele (MMC). Methods: This study included 63 boys with MMC aged 11.9 ± 1.8 years, 30 active boys with MMC and 33 inactive boys with MMC. Bone mineral density (BMD) and bone mineral content (BMC) in the distal (dis) and proximal (prox) parts of the forearm were measured by densitome

Gustatory cortical (GC) and basolateral amygdalar (BLA) taste responses consist of an inter-regionally coherent three-part state sequence. This coherence suggests that reciprocal BLA-GC connectivity is important for taste processing, but it remains unknown 1) whether BLA-GC coherence actually reflects a reciprocal "conversation" (as opposed to one region simply driving the other); and 2) whether such a "conversation" has anything to do with the taste processing observed within GC response dynamics. Here, we address these questions using network and single-neuron analysis of simultaneously reco

Body size estimation—the ability to judge the size and shape of one’s own body—is a key perceptual component of body image. However, its neural basis, and the basis for inter-individual differences in accuracy, remain poorly understood, partly due to limitations in existing assessment tools. We used an adapted, fMRI-compatible version of Somatomap 3D, an interactive task in which participants manipulate a rotatable 3D avatar by adjusting the size and shape of 26 individual body parts to match their perceived body. Twenty-eight healthy male and female adults completed the task during fMRI. Brai

OBJECTIVE: We investigated the transport of gadolinium-based contrast agent (GBCA) across the blood-brain barrier (BBB) along the perivascular spaces as part of the glymphatic drainage in patients with iatrogenic BBB disruption following digital subtraction angiography (DSA).
MATERIALS AND METHODS: A retrospective analysis was conducted on patients who underwent DSA for diagnosis and/or treatment of intracranial aneurysms and received a 3-T magnetic resonance imaging (MRI) within the following day. Exclusion criteria included states with a suggested impairment of BBB integrity, such as neurod

BACKGROUND: Intrathecal contrast-enhanced magnetic resonance imaging (MRI), utilizing the contrast agent gadobutrol as cerebrospinal fluid (CSF) tracer is emerging as a useful method to study glymphatic function in the human brain. A consistent finding with this technique is large inter-individual variability regarding tracer propagation. In this study, we outline an approach which predicts the distribution of tracer in the parenchyma based only on geometric information from brain tissue as captured by MRI, addressing one possible explanation for this variability. METHODS: Registrations are co

BACKGROUND: Filopodia are thin and dynamic membrane protrusions that play a crucial role in cell migration, axon guidance, and other processes where cells explore and interact with their surroundings. Historically, filopodial dynamics have been studied in great detail in 2D in cultured cells, and more recently in 3D culture as well as living brains. However, there is a lack of efficient tools to trace and track filopodia in 4D images of complex brain cells. RESULTS: To address this issue, we have developed a semi-automatic workflow for tracing filopodia in 3D images and tracking the traced fil

In many parts of the brain, population tuning to stimuli and behaviour gradually changes over the course of days to weeks in a phenomenon known as representational drift. The tuning stability of individual cells varies over the population, and it remains unclear what drives this heterogeneity. We investigate how a neuron's tuning stability relates to its shared variability with other neurons in the population using two published datasets from posterior parietal cortex and visual cortex. We quantified the contribution of pairwise interactions to behaviour or stimulus encoding by partial informa

Neurons in the macaque superior temporal sulcus respond selectively to faces and headless bodies, but their selectivity for individual body parts is not well understood. We recorded from the middle superior temporal sulcus and anterior superior temporal sulcus regions activated by whole monkeys in functional MRI to examine tuning for body parts, including heads. In experiment 1, body parts were shown in color, achromatic, or with scrambled internal features to test contributions of internal structure. In experiment 2, parts were presented at the same location and size as in whole-body images a

The lateralization of cortical activations and functional connectivities was analyzed when 833 Human Connectome Project (HCP) right-handed participants were viewing faces, spatial scenes, body parts, and tools, using the HCP-Multimodal Parcellation atlas. Spatial scenes produce stronger activations (Bonferroni corrected) in the right hemisphere, especially in the ventromedial visual cortical stream from early visual cortical regions via ventromedial visual cortical regions (VMV1-3) and medial parahippocampal regions (PHA1-3) to the hippocampus, and in inferior parietal visual cortical regions

BACKGROUND: Cognitive processes are essential for efficient daily functioning. Demographic factors such as age and education influence cognitive performance. However, the impact of sex on cognition is less understood and previous research has reported inconsistent findings. We investigated sex differences in cognitively unimpaired adults in three cohorts, using two complimentary approaches: a univariate approach to compare direct performance across cognitive domains and the multivariate approach of graph theory to compare global and nodal features as well as the modular organization of cogniti

AIM: This study was aimed at comparing the effects of a driving-related combined exercise and cognitive training system intervention on frontal lobe function in older adults with an active control group that performed aerobic exercises.
METHODS: In this randomized controlled trial, 90 community-dwelling older adults were randomly allocated (1:1) to the intervention (n = 45) or control groups (n = 45). The intervention group participated in a combined exercise-cognitive multitask training program for six consecutive weeks, whereas the active control group performed matched-intensity aerobic ex

Social behavior and the neural circuits that regulate it are organized by sex steroid hormones during critical periods in development (i.e., sexual differentiation). Much is known about how exposure to testosterone and estradiol during the critical period drive lasting changes to the brain. However, relatively little is known about how the timing of critical periods for sexual differentiation is regulated. Given that many parts of the brain (e.g., sensory and cognitive circuitry) are shaped during critical periods, it is not surprising that the regulation of plasticity has been extensively add

Implantable biohybrid systems with computer-controlled actuation offer the capacity to modulate biological forces, but require biocompatible, self-sustaining, and scalable actuators. Engineering biological muscles can fulfill this need. However, muscle fatigue limits the fundamental capabilities of muscle-actuated systems. Here we present a fatigue-resistant myoneural actuator (MNA) with engineered recruitment biophysics in a rodent model. The MNA is based on manipulating native axonal composition through sensory reinnervation. This regenerative approach establishes functional neuromuscular ju

BACKGROUND: Multisite pain is common during adolescence and is influenced by sex-related neurobiological and developmental factors, but its developmental neural mechanisms are unclear.
METHODS: Utilizing data from the Adolescent Brain and Cognitive Development Study, we investigated the relation between cortical brain structure and multisite pain (assessed by youth self-reports of painful regions on a body map) in male (N = 3,299) and female (N = 2,844) adolescents aged 11-12 years. We focused on brain regions functionally linked to multisite pain (i.e., bilateral sensorimotor, cingulate, fro

BACKGROUND: Altered stress responses are closely linked to mental disorders, but the role of brain structure in acute cortisol responses to psychosocial stress remains underexplored, particularly in healthy individuals. Previous studies, with predominantly small samples, primarily focused on selected limbic regions and functional measures. Thus, this study investigates associations between brain structure and cortisol responses to psychosocial stress, exploring if hypothalamic-pituitary-adrenal axis reactivity can be predicted from brain morphology.
METHODS: Our study included 291 subjects (1

Twitching, an abundant and spontaneous infant behavior and a defining feature of rapid eye movement (REM) sleep, supports development of the sensorimotor system. For unknown reasons, twitching persists into adulthood. Whereas infantile twitches are distributed across all body parts, adult twitching appears to occur more selectively in those appendages used for active sensing, such as elephant trunks, rat whiskers, pig snouts, and raccoon hands. Notably, these specialized appendages also have disproportionately large representations in sensorimotor cortex, a phenomenon known as cortical magnifi
