Salmonella infection and biofilms drive anti-C1q and anti-dsDNA autoantibodies in systemic autoimmunity.

Systemic lupus erythematosus (SLE) is characterized by autoantibody production alongside inflammatory manifestations, including nephritis. In SLE, infections are frequent and associated with disease flares, increasing morbidity and mortality. Additionally, dysbiosis, often marked by overgrowth of Pseudomonadota, is a prominent feature of SLE and lupus nephritis. We previously demonstrated that complexes of DNA and curli, an amyloidogenic protein from biofilms of Pseudomonadota, including Salmonella Typhimurium and E. coli, can induce anti-dsDNA and anti-chromatin autoantibody responses. Here,

Effect of oral gavage or intracloacal administration of Escherichia coli or Salmonella at hatch on enteric colonization and early performance in broiler chicks.

Minimal research has evaluated the efficacy of challenging chicks with Enterobacteriaceae via intracloacal gavage. The objectives of these studies were to: (1) evaluate day-of-hatch (DOH) colonization by Escherichia coli or Salmonella Enteritidis (SE), (2) determine the effects of bacterial challenge route on broiler performance, and (3) assess oral and intracloacal inoculation methods in broiler challenge models. In Exp 1, chicks were randomized, weighed, and challenged at DOH via oral or intracloacal gavage with saline (negative control), E. coli, or SE at doses of 103, 104, or 105 CFU/0.25

Antimicrobial resistance and genomic characterization of Salmonella Minnesota, ESBL-producing Escherichia coli, and ESKAPE pathogens isolated from Brazilian broiler chicken carcasses.

INTRODUCTION: Brazil is the world's largest exporter of chicken meat, and poultry products are recognized as important reservoirs of foodborne pathogens. Salmonella enterica serovar Minnesota has emerged as the predominant serovar in the poultry industry and is frequently associated with multidrug-resistant strains carrying multiple antimicrobial resistance genes. In addition, Escherichia coli and ESKAPE pathogens have been increasingly isolated from poultry products worldwide, representing a growing public health concern due to their antimicrobial resistance and dissemination potential. This

Switching apo- and holo-IscR control of LcrF alters how iron and oxygen regulate Yersinia virulence factors.

Iron and oxygen availability fluctuate spatially across mammalian tissues as well as temporally during bacterial infection. The [2Fe-2S] cluster-coordinating transcription factor IscR senses changes in iron and oxygen levels and plays a pivotal role in how Yersinia, Salmonella, and Vibrio regulate critical virulence genes, like the type III secretion system (T3SS). We previously showed that clusterless apo-IscR promotes expression of the Yersinia pseudotuberculosis T3SS master regulator LcrF by binding a type 2 IscR-binding motif in the lcrF promoter, thereby facilitating T3SS expression and d

Genomic structure of stx2i genes and prophages determined in Shiga toxin-producing Escherichia coli isolated from food, animal and human samples in Germany.

Shiga toxin-producing Escherichia coli (STEC) are important foodborne pathogens and a threat to human health. Their main virulence factor, the Shiga toxin (Stx), is encoded in two subunit genes (stxA and stxB) and occurs in various subtypes. Here, we report the occurrence of strains harbouring the new subtype stx2i. Isolates from food (n=8), animal (n=3) and human (n=2) samples were subjected to whole-genome sequencing for detailed molecular analyses. All strains harboured an identical stx2i variant independent of the genoserotype and differed from the first detected variant in 2016. In five o

Utilizing machine learning and MALDI-TOF MS platform for rapid detection of ceftriaxone-resistant nontyphoidal Salmonella.

UNLABELLED: Ceftriaxone (CRO) is a first-line antibiotic for pediatric nontyphoidal Salmonella (NTS) infections, but rising resistance threatens public health. Conventional antimicrobial susceptibility testing remains time-consuming, delaying treatment. Although MALDI-TOF MS enables rapid microbial identification, it lacks inherent resistance detection. This study integrates MALDI-TOF MS with machine learning to predict CRO resistance in NTS. Using 472 isolates (1,414 spectra) from three medical centers, we developed and validated a model based on the Light Gradient Boosting Machine (LGB) algo

Genomic Detection of Multidrug-Resistant H58 ST1 Salmonella Typhi in a South African Urban Waterway Reveals Epidemiological Links With Clinical Isolates.

Typhoid fever remains a major public health concern in South Africa. However, the role of urban waterways as environmental reservoirs and contributors to the spread of resistant Salmonella Typhi remains poorly understood. This study characterized four S. Typhi isolates recovered from the Jukskei River in Johannesburg, South Africa, using whole-genome sequencing. Genomic DNA was extracted and sequenced on the Illumina NextSeq. 2000 platform, and assembled genomes were analyzed for sequence types (STs), AMR determinants, plasmids, virulence factors, and phylogenetic relatedness using 124 clinica

Phenotypic and Genotypic Characterization of Escherichia coli O157:H7 Harboring stx2 and stx1&stx2 Genes Isolated from Frozen Raw Beef Meat and Beef Liver Samples.

Shiga toxin (Stx) is the main virulence factor of enterohemorrhagic Escherichia coli. Between its two types-Stx1 and Stx2-E. coli O157:H7 produces Stx2, which causes more severe infections compared with Stx1 or both Stx1 and Stx2. In this study, we isolated and identified E. coli O157:H7 from frozen raw beef samples and both Stx1 and Stx2 from frozen raw beef liver samples collected from the Al-Riyadh market in Saudi Arabia. Phenotypic detection was conducted using suitable and specific media-sorbitol MacConkey agar and Salmonella chromogenic agar-with E. coli O157:H7 appearing as colorless an

Functional mechanism of SptP from Salmonella Typhimurium in modulating environmental adaptability, intracellular proliferation and intestinal inflammation in Wenchang chickens.

Salmonella enterica subsp. enterica serovar Typhimurium (S.Typhimurium) is a critical foodborne zoonotic pathogen that impairs poultry production and threatens public health. The virulence effector SptP encoded by Salmonella Pathogenicity Island 1 (SPI-1) exerts unclear pathogenic functions in indigenous Wenchang chickens, a high-susceptible local broiler breed in China. Here, wild-type (WT) S. Typhimurium and an isogenic ΔSptP knockout mutant were combined with in vitro stress assays, HD11 chicken macrophage infection, in vivo oral challenge of Wenchang chicks, and ileal RNA sequencing (RNA-s

Longitudinal Clinical and Minimally Invasive Readouts Enable Early Assessment of Disease Trajectory in Preclinical Infection Models.

Murine infection models are essential for investigating host-pathogen interactions, disease progression, vaccine-induced immune responses and protection. Early identification of disease trajectories is important for both animal monitoring and high-quality biological sample collection. Although clinical and microbiological readouts such as body-weight changes, clinical scores, and circulating bacterial burden are routinely used to monitor disease progression, individual parameters often fail to capture short-term disease trajectory when analysed separately. Here, four longitudinal clinical and

Impact of dosing interval on immunogenicity following a heterologous typhoid conjugate vaccine booster in Nepalese children: An observational study.

OBJECTIVES: Typhoid conjugate vaccines (TCVs) are essential for controlling endemic typhoid fever in low- and middle-income countries. However, evidence suggests that TCV-induced protection wanes 3-5 years post-vaccination, particularly among children vaccinated before two years of age, raising the question of whether a booster dose is needed. This study leverages a unique context in Nepal, where the nationwide rollout of the Vi-CRM197 TCV occurred four years after a large-scale TCV efficacy trial using Vi-TT, allowing the study to assess the immunogenicity of a heterologous prime-boost schedu
