Engineering a hybrid chitinase-sonorensin fusion protein for enhanced antibacterial and antifungal activity.

The main objective of the study was to attain improved effectiveness against various bacterial and fungal infections, tackling the widespread problem of multidrug resistance. The study presents the development and evaluation of a hybrid antimicrobial protein created by combining chitinase and sonorensin (HyChiSono). The hybrid protein HyChiSono, a 49.3 kDa chitinase-sonorensin fusion protein, was engineered, expressed in E. coli BL21(DE3), and purified to > 95 % homogeneity. Independent folding of each domain linked by a flexible (GGGGS) spacer was revealed by AlphaFold modelling; a highly ord

Antimicrobial peptide resistance in Salmonella AMR: the role of surface binding and lipopolysaccharide remodelling: one health implications.

Salmonella spp. are the most important foodborne and zoonotic bacteria in the world, with severe implications for public health, food safety, and the economy. Antimicrobial peptides (AMPs) targeting the innate immune system and new therapeutic targets for conventional antibiotics are largely mediated through electrostatic adsorption onto microbial surfaces with membrane disruption or intracellular interference. However, Salmonella has also evolved complex mechanisms of resistance to reduce the effectiveness of AMPs, among which attachment to surfaces and lipopolysaccharide (LPS) modifications

In vitro activity of the antimalarial, antibiotic drugs fosmidomycin and clindamycin against clinical isolates of bacterial bloodstream infections in febrile, hospitalized Ghanaian children.

BACKGROUND: The clinical and laboratory distinction between malaria and bacterial blood stream infections in patients with undifferentiated fever remains challenging. Misclassification may result in ineffective treatment with the risk of progression of disease and subsequent morbidity and mortality. A pragmatic solution is the use of multi-disease treatments effective against both malaria and the clinically most relevant bacterial pathogens. Fosmidomycin and clindamycin are two antibiotics that have demonstrated high efficacy in treating malaria. A multi-drug combination treatment of fosmidomy

A booster vaccination for Salmonella spp. minimally affects feeding behavior in beef × dairy calves fed with automated milk feeders.

Farmers can use automated milk feeders (AMF) to identify changes in the behavior of individual calves before they are clinically sick. However, other management factors, such as routine procedures, may affect group- and individual-level feeding behavior patterns in calves. This matched cohort study (n = 44; 22 pairs of booster-vaccinated calves matched to controls) evaluated the effects of a booster vaccination comprised of inactivated Salmonella Typhimurium and Salmonella Dublin on feeding behavior and relative changes in behavior of Angus × Holstein calves reared at one calf-rearing facility

Rapid molecular diagnostics for detection of extensively drug-resistant Salmonella Typhi.

BACKGROUND: Salmonella enterica serovar Typhi results in >160 000 deaths annually. In endemic regions, infections disproportionately affect infants and children. Current diagnostics rely on culture-based approaches that lack sensitivity and require infrastructure and skills that are not always possible in developing regions. OBJECTIVES: We developed a suite of rapid, sensitive and specific molecular diagnostic tools to detect S. Typhi, and markers of extensive antimicrobial resistance (AMR), with the capacity to be used in low- and middle-income countries (LMIC). METHODS: Singleplex and mult

Prevalence and factors associated with concomitant bacteremia among adults admitted with severe malaria at Kayunga Regional Referral Hospital, Uganda.

BACKGROUND: Malaria-bacteremia co-infection significantly increases mortality and the risk of ICU admission. Diagnostic overlap with bacterial infections often results in misdiagnosis, impacting outcomes. While pediatric data exists, adult studies in Uganda are limited. This study aimed to determine the prevalence, bacterial isolates, and associated factors of bacteremia in adults with severe malaria at Kayunga Regional Referral Hospital. METHODS: A cross-sectional study enrolled 207 adults with severe malaria. Blood samples were cultured, and isolates tested for antimicrobial susceptibility.

Experimental Salmonella Typhimurium infection in pigs.

The genus Salmonella, particularly Salmonella enterica subsp. enterica serovars Choleraesuis and Typhimurium, poses significant challenges to swine production and leads to economic losses from conditions such as septicemia and enterocolitis. We evaluated the effects of experimental infection with Salmonella Typhimurium on clinical signs and anatomopathologic outcomes in pigs. Twenty 90-d-old pigs were divided into 2 groups: G1 received an oral inoculum of 108 cfu of Salmonella Typhimurium; G2 served as a control. Pigs were monitored clinically for 30 d; postmortem examinations and microbiologi

Impact of the COVID-19 pandemic and typhoid conjugate vaccine introduction on typhoid fever in Nepal.

BACKGROUND: While typhoid conjugate vaccines (TCV) offer promise for reducing risk in endemic settings, their population-level impact remains unclear. In 2022, Nepal introduced TCV nationally on the heels of the COVID-19 pandemic, which disrupted healthcare services, surveillance, and potentially typhoid transmission dynamics, complicating vaccine impact evaluation. We investigated the impact of TCV introduction amid shifting typhoid burden during the pandemic. METHODS: We analyzed blood culture data from four Kathmandu Valley health facilities, comparing culture positivity for Salmonella Typ

Primary Salmonella Osteomyelitis of the Sternum.

Osteomyelitis is a rarely encountered diagnosis in children. It typically occurs due to hematogenous inoculation following transient bacteremia, with bacterial infection ultimately establishing in areas of microtrauma. Prevalence is highest in long bones due to active marrow, and the most common organism isolated from affected tissue is Staphylococcus aureus. Patients with hemoglobinopathies, however, are more susceptible to unusual pathogens, particularly Salmonella species. This report summarizes the successful management of a unique presentation of primary Salmonella osteomyelitis of the st

Commensal microbe-derived butyrate enhances T follicular helper cell function to boost mucosal vaccine efficacy.

BACKGROUND: The gut microbiota plays an essential role in mucosal immunity, with secretory immunoglobulin A (IgA) acting as a key effector in neutralizing pathogens and maintaining host-microbiota homeostasis. IgA production occurs via T cell-dependent (TD) and -independent pathways, with T follicular helper (Tfh) cells driving high-affinity, antigen-specific IgA responses. However, the specific microbial taxa and metabolites that regulate Tfh-mediated IgA responses under steady-state conditions remain poorly understood. This study investigated how gut microbiota-derived signals shape Tfh resp

Bacteriophage vB_SalS_KY05 controls Salmonella in poultry without disrupting cecal microbiota composition.

Salmonellosis is a major food safety challenge in poultry production, requiring effective antibiotic alternatives. We isolated a polyvalent Tequintavirus, vB_SalS_KY05, from a Taiwanese poultry farm and evaluated its biological properties and in vivo efficacy against Salmonella enterica serovars Typhimurium and Enteritidis, and Escherichia coli. The objective of this study was to comprehensively characterize vB_SalS_KY05 and assess its potential as a biocontrol agent for poultry farming. Genome analysis confirmed a lytic lifestyle with no detectable virulence or antimicrobial resistance genes.

Salmonella relies on siderophore exploitation at low pH.

Salmonella enterica, a prominent enteric pathogen, employs sophisticated iron acquisition mechanisms to overcome host-imposed iron limitation, notably through the production and uptake of siderophores-small, high-affinity iron-chelating compounds that scavenge iron from the host environment. In this study, we investigate how environmental pH influences Salmonella's preference for its endogenous siderophores versus exogenous siderophores within the physiological range of the gastrointestinal tract. Through competition assays, gene expression analysis, and siderophore quantification, we demonstr

Chick paper sampling: a One Health approach to inform public health action during salmonellosis outbreaks linked to backyard poultry in the United States, 2023.

INTRODUCTION: Every year, cases of salmonellosis associated with backyard poultry (BYP) contact are reported, despite public health prevention efforts. METHODS: We sampled chick shipping materials ("chick paper") at agricultural retailers to detect Salmonella and to identify supply hatcheries to share mitigation information. RESULTS: Six states collected 149 samples, and 55 (37%) were positive for Salmonella. These yielded 107 Salmonella isolates; 47 (44%) of the isolates were Salmonella serotypes that matched a subset of the outbreak strains identified in the 2023 multistate BYP-associated

Comparative pan-genomics and in vivo validation identify genetic factors important for virulence of Salmonella enterica serovar Gallinarum and serovar Enteritidis in the avian host.

Salmonella enterica subspecies enterica serovar Gallinarum biovar Gallinarum (SGa) and Pullorum (SPu) are avian-specific pathogens causing systemic disease, while S. Enteritidis (SEnt) is a broad host range serovar causing gastroenteritis. The genomic mechanisms underlying this difference in host range and pathogenicity remain incompletely understood. Here, we performed a large-scale pan-genome analysis of 5440 poultry-derived genomes (4927 SEnt, 106 SGa, 407 SPu) integrated with functional chicken and macrophage experiments. Compared with SEnt, avian-specific SGa and SPu exhibited extensive p

An infected aneurysm in the internal carotid artery caused by Salmonella enterica subsp. enterica serovar Enteritidis.

An infected aneurysm in the carotid artery is a rare but life-threatening condition. The most common cause of such infections is Staphylococcus aureus; however, Salmonella is a possible but uncommon cause. We report the case of an elderly man with an infected aneurysm in the left internal carotid artery caused by Salmonella enterica subsp. Enterica serovar Enteritidis despite the absence of gastrointestinal symptoms. Following confirming the absence of neurological symptoms after blood flow disruption through balloon test occlusion (BTO), the patient underwent coil embolization of the aneurysm

Pathogen-specific predicting factors of childhood diarrhoea and their seasonality: evaluation from Rohingya refugees and host population in Cox's Bazar, Bangladesh.

BACKGROUND: Bangladesh observed a sudden massive influx of Rohingya refugees in August 2017. This large migrant population relative to a smaller host community placed a burden and threat on the public health sector. Due to the lack of pathogen-specific predicting factors and the influence of seasonal variation on childhood diarrhoeal pathogens in a densely populated area, we aimed to explore the same among Rohingya refugees and the host population. METHODS: We collected data from under-five children of Rohingya refugees and hosts between 2018 and 2023 from the Diarrhea Treatment Center (DTC)-
