Severe Headache and Acute Blindness: A Case of Pituitary Apoplexy.

Pituitary apoplexy is a rare, life-threatening syndrome that commonly occurs in the adenomatous pituitary gland. It presents with a myriad of severe neuroendocrine and ophthalmological signs and symptoms. Early diagnosis and prompt treatment are vital in the management of pituitary apoplexy. This case report describes a 56-year-old woman who reported to the neurology clinic with 1 week's history of visual loss, and a severe headache, on the background of a 2-year history of pituitary adenoma with poor follow-up. The patient underwent transsphenoidal surgery. She regained vision in the right ey

Secretogranin II-derived peptide secretoneurin differentially stimulates luteinizing hormone secretion by age and sex in ex vivo murine pituitaries†.

Secretoneurin (SN) is a small, bioactive proteolytic product of secretogranin II (SCG2), a chromogranin protein involved in secretory vesicle packaging. Secretogranin II co-aggregates with luteinizing hormone (LH) in pituitary gonadotrope secretory granules and thus has long been associated with gonadotropin-releasing hormone-regulated LH secretion. Previously, the SCG2-derived peptide SN has been identified as a regulator of LH production and secretion in fish and a murine gonadotrope-like cell line (LβT2). Though high levels of SCG2/SN are present in the mammalian pituitary, the role of pitu

Intranasal povidone-iodine concentration and olfactory recovery after endoscopic endonasal pituitary surgery.

PURPOSE: The endoscopic endonasal approach (EEA) is a widely accepted, safe, and effective technique to treat pituitary lesions. Among postoperative considerations, olfactory function plays a key role in the patient’s quality of life. Although povidone-iodine (PVP-I) is widely used for intranasal antisepsis, its optimal concentration remains unclear. This study evaluated whether the PVP-I concentration during antisepsis influences postoperative olfactory recovery in patients undergoing EEA. METHODS: We retrospectively reviewed patients with sellar tumors who underwent endoscopic endonasal pitu

The relationship between pituitary size and the response to recombinant human growth hormone therapy in children with isolated growth hormone deficiency.

BACKGROUND: Growth hormone deficiency (GHD) is one of the major endocrine causes of short stature in childhood. Pituitary size may reflect growth hormone secretory capacity; and children with hypoplastic pituitary exhibit more severe GHD. Given this relationship, pituitary size may also serve as a valuable predictor of the growth response to recombinant human growth hormone (rhGH) therapy. This study aimed to investigate the relationship between pituitary height and volume measured on MRI and the growth response to rhGH therapy in children with GHD. METHODS: This retrospective, single-center

GABA Enhances Growth Hormone Expression by Modulating Somatotroph Pit-1 Transcription via Activation of Calmodulin-Dependent Kinases.

Background: Gamma-aminobutyric acid (GABA), the principal inhibitory neurotransmitter in the central nervous system (CNS), is also a potent modulator of peripheral endocrine function. We previously demonstrated that dietary GABA supplementation improves growth and fatty acid metabolism in male mice while elevating pituitary growth hormone (GH). However, the mechanisms by which GABA regulates the somatotropic axis remain unclear. Methods: Adolescent mice (3-4 weeks old) were treated with or without GABA in drinking water. Cultured pituitaries and GH3 somatotroph-derived cells were exposed to GA

Establishment of an ELISA system to measure pituitary hormones in teleosts.

We developed and validated a fluorescence-based competitive enzyme-linked immunosorbent assay (ELISA) for the parallel quantification of four key pituitary hormones-growth hormone (GH), prolactin (PRL), somatolactin alpha (SLα), and somatolactin beta (SLβ)-in grass carp (Ctenopharyngodon idella). The assay utilizes recombinant grass carp hormones as standards and corresponding rabbit polyclonal antibodies, with biotin-avidin-HRP amplification and HPPA-based fluorescence detection. Standard curves demonstrated high linearity and half-maximal inhibitory concentrations (ID50) ranging from 8.004 t

Discovery of Potent and Brain-Penetrant Inverse Agonists for GPR61, an Orphan G Protein-Coupled Receptor.

GPR61 is a class A orphan G protein-coupled receptor (GPCR) that is predominantly expressed in the pituitary gland and appetite-regulating centers of the brain. Genome-wide association analysis, epigenetic analysis, and animal model data have suggested that GPR61 could be a potential therapeutic target for appetite and body weight modulation. Herein, we describe our medicinal chemistry efforts in discovering a class of potent, selective, and brain-penetrant GPR61 inverse agonists. The cryogenic electron microscopy structure of GPR61 bound to compound 15 shows that this class of inverse agonist

Endocrine effect of ghrelin on histological, hormonal and morphometric parameters of the pituitary gland and the possibility of its clinical application.

BACKGROUND: Ghrelin, a brain-intestinal hormone, is a growth hormone (GH) secretagogue. Because it regulates appetite and is secreted most prominently before meals, it is often described as a "hunger hormone". It consists of 28 amino acids. Given the connection between nutritional status and energy metabolism, it suggests that disorders in these areas can lead to anorexia, especially during aging. This review article aims to demonstrate ghrelin's influence on the histological, hormonal, and stereological characteristics of pituitary cells, as well as its potential clinical applications. METHO

Old and new pituitary biomarkers: evidence for sex-specificity.

PURPOSE: To review established and emerging biochemical pituitary biomarkers, examining how biological sex influences their concentrations and diagnostic interpretation. METHODS: We performed a literature review of recent studies and guidelines on all pituitary hormone axes, emphasizing sex-specific differences, assay variability, and new biomarkers of pituitary function. RESULTS: Sex significantly modulates all major pituitary axes. For example, women experience cyclical or estrogen-driven variations in gonadotropins, cortisol binding, and growth hormone (GH) secretion, whereas men have more

The potential roles of androgen and estrogen receptors during reproductive cycle in Pampus argenteus.

Steroid hormones are indispensable for regulating the typical reproductive processes during the development of gonads. This study aimed to clarify the expression patterns of androgen receptors (Ars) and estrogen receptors (Ers) in Pampus argenteus to better understand gonadal development and improve artificial breeding. In this study, the androgen receptors (arα and arβ) and estrogen receptors (esr1, esr2a, and esr2β) of P. argenteus were cloned and the expression profiles were detected by qPCR. The results showed that the highest expression of arα was found in hypothalamus, while arβ was main

C/EBPβ dictates postmenopausal FSHβ transcription and blockade of AEP/C/EBPβ pathway alleviates osteoporosis.

Follicle-stimulating hormone (FSH), a gonadotropin that rises in post-menopausal females, activates its receptor FSHR to trigger bone loss via increasing bone resorption by osteoclasts. FSH stimulates CCAAT/enhancer binding protein beta (C/EBPβ) /asparagine endopeptidase (AEP) pathway, facilitating neural degeneration in the brain of mouse models with Alzheimer's disease (AD). However, whether C/EBPβ/AEP pathway feeds back and modulates FSHβ bone resorption action remains elusive. Here we show that C/EBPβ acts as a transcription factor for fshb gene and directly binds its promoter, mediating i

Biochemical and metabolic tissue discrimination using a dual-modality Raman-fluorescence endonasal spectroscopy probe for pituitary adenoma surgery.

PURPOSE: A major challenge in pituitary adenoma surgery is distinguishing adenoma from normal pituitary gland and adjacent structures. Accurate intraoperative identification is critical to optimize tumor resection, hormonal remission, and preservation of pituitary function. Building on our previous preclinical validation of a bayonet-style endonasal Raman probe, we evaluated a dual-modality Raman spectroscopy and tissue fluorescence system for ex vivo discrimination of pituitary adenoma, normal gland, and surrounding sellar tissues. METHODS: A prospective study was performed in 32 patients und

Morphological changes and lateralization of the thalamic nuclei in children with growth hormone deficiency.

INTRODUCTION: Childhood growth hormone deficiency (GHD) is an endocrine disorder characterized by reduced secretion of growth hormone (GH), leading to impaired linear growth and delayed developmental milestones. Recent studies suggest that GHD is associated with cognitive, socio-emotional, and behavioral impairments, potentially via altered neurodevelopment, with neuroimaging studies revealing changes in brain morphology and broader GH-related effects on the central nervous system. The thalamus, a major subcortical relay integrating sensory, motor and cognitive information, has received limite

Growth Hormone Association With Testicular Recrudescence in a Free-Living Passerine, Amandava amandava.

Emerging evidence confirms the expression of functional growth hormone (GH) receptors in the male and female reproductive systems, and suggests that the GH signaling might influence gonadal function by acting at different levels of the hypothalamic-pituitary-gonadal (HPG) axis in animals and mammals. Despite this, the role of GH in the modulation of the gonadal cycle in seasonal breeders has been less explored. This study focused on a free-living passerine, Amandava amandava (Red Munia), a distinct seasonal breeder. It aimed at elucidating the plasticity in pituitary expression of the GH, and

The Effect of Mild Traumatic Brain Injury on the Hypothalamic-Pituitary-Ovarian Axis: A Narrative Review.

Mild traumatic brain injury (mTBI) is the most common type of traumatic brain injury (TBI), with 2.5 million cases reported annually in the United States. mTBI involves diverse signs, symptoms, and functional impairments that typically resolve within 1 month, but may persist. Female sex is one of the most consistent predictors associated with an increased risk of mTBI and worse outcomes, including greater symptom severity and prolonged recovery. Emerging evidence also suggests that females are at greater risk of menstrual cycle dysfunction following mTBI, which could have downstream consequenc
