Skin lesions in patients treated with growth hormone and those with growth hormone excess: a current overview.

Growth hormone (GH) is an ancestral hormone secreted from the anterior pituitary gland. In adulthood, it is essential to regulate metabolism. GH synthesis and secretion are regulated in a complex manner, primarily through the actions of hypothalamic neuropeptides (GHRH and somatostatin) that integrate hormonal, metabolic, and neurogenic signals. Currently, recombinant human GH is widely used to treat growth hormone deficiency (GHD) and numerous non-GHD disorders, such as short stature and catabolic conditions. Conversely, an excess of GH may lead to different and severe conditions, such as acr

Morphometric Analysis of the Pituitary Gland and Sella Turcica Using Computed Tomography Images in Hamdani Sheep: Evaluation of Sexual Dimorphism.

BACKGROUND: This study aimed to obtain morphometric data of the pituitary gland and sella turcica in Hamdani sheep using computed tomography (CT) and to assess sexual dimorphism. METHODS: CT scans of skulls from 16 healthy adult Hamdani sheep (8 males, 8 females) were analysed using RadiAnt DICOM Viewer (64-bit). Morphometric measurements of the pituitary gland were obtained from transverse (length, height, width), dorsal (length, width), and sagittal (length, height) sections, and these values were used to calculate the volume of the gland. Additionally, using the pituitary gland measurement

Facts and news about growth hormone replacement therapy in adults.

Adult growth hormone deficiency (GHD) is associated with increased adiposity, reduced lean mass, adverse lipid profile, decreased bone density and impaired quality of life. Growth hormone (GH) replacement therapy provides significant metabolic, cardiovascular, musculoskeletal and psychological benefit. Traditional stimulation tests, such as the insulin tolerance and glucagon tests, are gold standards for diagnosis, but the oral macimorelin test offers a safe, reliable and well-tolerated alternative. GH replacement should be titrated to achieve age-appropriate insulin-like growth factor 1 (IGF-

Gigantism.

Gigantism is characterized by a height greater than two standard deviation (SD) scores above the mean height for age, sex and ethnicity or more than 2 SDs above the calculated mid-parental height. Patients with accelerated growth/tall stature are usually referred to endocrinologists or pediatric endocrinologists for the exclusion of hormonal disorders, most notably growth hormone (GH) and insulin-like growth factor 1 (IGF-1) excessive levels leading to pituitary gigantism. Endocrine disorders associated with accelerated growth/tall stature have characteristic manifestations and specific treatm

TRH can stimulate the release of two POMC-derived pituitary hormones, ACTH and MSH, in medaka.

Anterior pituitary hormone secretion is generally considered to be under the strong regulation of hypothalamic neuropeptides. In mammals, adrenocorticotropic hormone (ACTH), which plays a crucial role in the stress response, is secreted from corticotropes and is regulated primarily by corticotropin-releasing hormone (CRH). In teleosts, although the pharmacological effects of hypothalamic factors have been demonstrated, their relative importance in regulating ACTH release remains controversial. One reason for this is the lack of methods for evaluating ACTH release at cellular resolution. Using

Electrophysiological recording of hypothalamic brain regions in vivo using the transpharyngeal surgical approach in the rat.

Transpharyngeal (ventral) surgery in urethane-anaesthetised rats allows in vivo electrophysiological recording and/or imaging from superficial hypothalamic brain regions and from the pituitary gland. This surgical approach leaves the whole brain intact, providing a stable platform to study single or multiple identified cells over several hours with all central and peripheral inputs intact and the endocrine system functioning, which allows repeated drug application and stimulation of afferent inputs, as has been done for the arcuate nucleus, organum vasculosum of the lamina terminalis (OVLT) an

Emerging human cell-based in vitro models for studying pituitary development and disorders.

The pituitary gland plays a central role in maintaining physiological homeostasis by secreting multiple hormones that regulate diverse body functions. Dysregulation of pituitary hormones can arise from various conditions, such as pituitary tumors and autoimmune diseases, leading to a broad spectrum of symptoms. During embryonic organogenesis, the pituitary gland originates from the oral ectoderm in close contact with the adjacent hypothalamus. This tissue interaction is essential for proper development. Although the molecular mechanisms underlying pituitary development and related disorders ha

PitVQA++: Vector Matrix-Low-Rank Adaptation for Open-Ended Visual Question Answering in Pituitary Surgery.

Vision-Language Models (VLMs) in visual question answering (VQA) offer a unique opportunity to enhance intra-operative decision-making, promote intuitive interactions, and significantly advance surgical education. However, the development of VLMs for surgical VQA is challenging due to limited datasets and the risk of overfitting and catastrophic forgetting during full fine-tuning of pretrained weights. While parameter-efficient techniques like Low-Rank Adaptation (LoRA) and Matrix of Rank Adaptation (MoRA) address adaptation challenges, their uniform parameter distribution overlooks the featur

Incidental detection of sellar spine on a CBCT volume: clinical and radiographic considerations.

Sellar spine (SS) is a developmental anomaly of the pituitary fossa resulting in a bony spur projection anteriorly from the dorsum sellae. Etiology remains unclear; however, the most prevalent hypothesis is that it is a remnant of the notochord. SS is a rare entity with a few reported cases exclusively in neuroradiological and neurosurgical literature. It can be asymptomatic in many patients; however, it may cause various endocrinological and ophthalmological symptoms because it impinges on the pituitary gland. The current case report presents an incidental finding of SS in a 12-year-old male

Late recurrence of Cushing disease with ubiquitin-specific protease 8 mutation 19 years after initial surgery.

Recurrence after successful transsphenoidal surgery (TSS) for Cushing disease (CD), caused by adrenocorticotropic hormone (ACTH)-secreting pituitary adenomas, remains a major clinical concern. Somatic mutations in the ubiquitin-specific protease 8 (USP8) gene have been implicated in corticotroph tumorigenesis and may contribute to recurrence risk. Here, we report a rare case of delayed CD recurrence 19 years after initial surgical remission. A female aged 36 years underwent TSS for CD with typical clinical and biochemical evidence of hypercortisolism. Magnetic resonance imaging showed a 10 mm

Urine-to-Serum Osmolality Ratio as a Prognostic Marker in Traumatic Brain Injury.

Background/Objectives: Prognostication in traumatic brain injury (TBI) remains challenging. The urine-to-serum osmolality (U/S) ratio may reflect hypothalamic-pituitary axis integrity, a critical but underexplored prognostic domain. We investigated whether the U/S ratio provides early prognostic value and enhances prediction when combined with conventional severity markers. Methods: This retrospective study included 128 adult TBI patients admitted to a neurosurgical intensive care unit (ICU) with simultaneous osmolality measurements within 6 h of admission. The primary outcome was ICU mortalit

The influence of galanin receptor agonist during the seasonal recrudescence and nonbreeding phases of the ovarian cycle in the gekkonid lizard Hemidactylus frenatus.

Galanin (GAL), a neuropeptide produced in both the central and peripheral nervous systems, is implicated in various physiological processes, including reproduction in vertebrates. Yet, the role of GAL is not understood in reptilian reproduction. This study aimed to investigate the impact of the galanin receptor agonist (GALR-A) on the reproductive axis during both the breeding and non-breeding phases of the ovarian cycle in the gecko Hemidactylus frenatus. During the recrudescence phase, administration of either a low (0.2 μg) or high dose (2 μg) of GALR-A did not affect either the follicular

Cholecystokinin coordinates gonadotropin-dependent and independent pathways to orchestrate zebrafish gonadal development.

Cholecystokinin (CCK) is established as a critical regulator of teleost gonadal development, functioning not only as a follicle-stimulating-hormone-releasing hormone (FSH-RH) but also stimulating gonadal development through three distinct mechanisms. Specifically, (1) CCK directly activates pituitary lhb transcription via CCKBRb in a specific bi-hormonal subpopulation, and indirectly enhances hypothalamic gnrh3 expression to regulate LH synthesis and secretion. However, CCK's physiological role in LH regulation requires further investigation, and its effect may involve both direct and indirect

Thyroid hormones are associated with pituitary and thyroid volume in a population-based study.

There is increasing evidence that even a small variation in thyroid function, even within the reference range, is associated with adverse clinical outcomes. Furthermore, there exists a substantial interindividual variation of thyroid function even within euthyroid individuals suggesting an individual hypothalamus-pituitary-thyroid (HPT) axis setpoint. Here we assess potential contributions of the HPT axis organ volumes to the variation of thyroid hormones in the general population. Using linear regression models, we analyzed the association of circulating free T3, free T4, and log-transformed
