Ca2+ Signaling Enables Growth Hormone Secretion from Cultured Mice Pituitary Somatotrophs, Facilitating Outgrowth in Hypothalamic Neuron.

The hypothalamus regulates anterior pituitary (AP) hormone release via the hypophyseal portal system by secreting specific releasing and inhibiting peptide hormones. Growth hormone (GH) secretion from pituitary somatotrophs (PS) is regulated by a cholinergic mechanism that induces calcium (Ca2+) signaling, which in turn triggers vesicular exocytosis and GH release. Given that the transient receptor potential canonical channel 3 (TRPC3) regulates vesicle exocytosis in endocrine and neuroendocrine cells, we investigated its role in GH release from somatotrophs and whether GH release could suppor

A bloody surprise: rare presentation of pituitary apoplexy with epistaxis-a case report.

Pituitary apoplexy (PA) is an endocrine emergency caused by haemorrhage or infarction of the pituitary gland, often occurring in patients with known tumours. Classical presentation includes headache, visual disturbances, and cranial nerve palsies. We describe a rare presentation of PA with epistaxis in a 71-year-old man with a known non-functioning pituitary macroadenoma. On admission, he had acute confusion, worsening headache, and left-sided epistaxis with signs of Cranial Nerve (CN) III palsy and reduced consciousness. Initial Computerized Tomography (CT) showed a stable macroadenoma withou

Association of serum biomarkers, immunohistochemical profiles, and preoperative clinical manifestations in patients undergoing endoscopic endonasal resection of pituitary neuroendocrine tumors.

This study examined the correlation between clinical presentation, serum biomarkers, and immunohistochemical markers in patients with pituitary neuroendocrine tumors (PitNETs). This study aimed to evaluate the associations between preoperative clinical presentation, serum hormone levels, and IHC findings, and to describe postoperative endocrine outcomes in a surgically treated cohort. A cross-sectional analysis was conducted on sixty-five patients who underwent endonasal endoscopic surgery between 2015 and 2023 at our institution. Clinical data were retrospectively analyzed, and Spearman corre

Pituitary Apoplexy with the Initial Presentations Similar to Trigeminal Autonomic Cephalalgia: A Case Report.

Pituitary apoplexy, defined by an acute ischemic or hemorrhagic event of the pituitary gland, can lead to neurological deficits. We present the case of a 43-year-old male who initially exhibited symptoms resembling trigeminal autonomic cephalalgia (TAC) as sudden-onset headache, ptosis, and autonomic symptoms. He later developed abducens nerve palsy and magnetic resonance imaging revealed a pituitary macroadenoma with intratumoral hemorrhage. Endocrine evaluation showed multiple hormonal deficiencies. The patient was diagnosed with pituitary apoplexy and referred for surgical intervention. Thi

Generation of a prop1 knock-in zebrafish enables single-cell transcriptomics of early pituitary development.

The adenohypophyseal placode is a developmental structure that is conserved among vertebrates and gives rise to the anterior pituitary. The earliest steps of its formation remain poorly understood. Here, we present a powerful new genetic tool: a zebrafish GAL4 knock-in reporter line for prop1. The targeted integration disrupts prop1 function, generating the first prop1-/- zebrafish, which exhibits dwarfism, impaired sexual maturation, and pigmentation defects. The generation of this new reporter line enabled the study of early pituitary development at single-cell resolution, using both scRNA-s

Systematic review of pituitary gland and pituitary adenoma automatic segmentation techniques in magnetic resonance imaging.

Accurate segmentation of both the pituitary gland and adenomas from magnetic resonance imaging (MRI) is essential for diagnosis and treatment of pituitary adenomas. This systematic review evaluates automatic segmentation methods for improving the accuracy and efficiency of MRI-based segmentation of pituitary adenomas and the gland itself. We analysed 34 studies that employed automatic and semi-automatic segmentation methods out of 353 reviewed studies. We extracted and synthesized data on segmentation techniques and performance metrics (such as Dice overlap scores). The majority of reviewed st

Integrative analysis of corticotrophs and folliculostellate cells: aging and paracrine regulation.

BACKGROUND: Aging of the human anterior pituitary is accompanied by structural remodeling of hormone-producing and non-endocrine cellular components. The coordinated morphometric relationship between adrenocorticotropic hormone (ACTH)-producing corticotrophs and folliculostellate (FS) cells during aging remains insufficiently characterized. MATERIALS AND METHODS: Twenty-nine human anterior pituitaries (33-95 years, both sexes) obtained at autopsy were examined using immunohistochemistry for ACTH and S-100 protein. Volume densities (VVACTH, VVFS) were quantified using calibrated digital morpho

Gonadotrope remodeling in sustained low estrogen states: single-cell transcriptomic analysis reveals gonadotrope subtypes and activation of stem cell populations†.

To study the effect of low-estrogen and the menopause state upon female pituitary function, we used the 4-vinylcyclohexene diepoxide (VCD)-induced ovarian failure model to gradually reduce serum estradiol (E2) levels. We utilized single-cell RNA sequencing transcriptomics analysis (scRNA-seq) to determine how reduced E2 levels influence pituitary gonadotrope gene expression and cell state, with a focus on potential contributions from pituitary stem cells to gonadotrope population growth. VCD-treated mice were acyclic and exhibited 8-15-fold increases in serum levels of the gonadotropins follic

Icariin ameliorates gonadotropin dysregulation in an ovariectomy model: Therapeutic effect and mechanism.

BACKGROUND: MicroRNA-7 (miR-7) suppresses pituitary gonadotropins, while icariin, an active compound from Epimedium brevicornu Maxim., exhibits estrogen-like properties. However, whether icariin regulates gonadotropins via miR-7 remains unclear. PURPOSE: This study investigated whether icariin regulates gonadotropins through miR-7 and elucidated the underlying molecular mechanisms. STUDY DESIGN: An integrated approach combining ovariectomized mouse model, primary pituitary cell cultures, and molecular techniques was employed. METHODS: In vivo, ovariectomized C57BL/6 mice received intragastr

MRI findings of an incidental ectopic posterior pituitary gland in an adult: Case report and review of T1 hyperintense lesions in the literature.

The ectopic posterior pituitary represents a rare congenital malformation attributable to aberrant embryogenesis of the neurohypophysis. Although it may occur as an isolated anomaly, it is frequently concomitant with other congenital malformations of the central nervous system. Despite extensive investigations, the precise etiology remains elusive; however, accumulating evidence suggests that its pathogenesis may parallel that of septo-optic dysplasia, thereby implicating a genetic predisposition. Earlier accounts predominantly attributed this anomaly to traumatic mechanisms. In contrast, cont

Erg currents support electrical bursting in murine anterior pituitary corticotrophs.

The regulation of electrical excitability of anterior pituitary corticotrophs is critical for an appropriate response of the hypothalamic-pituitary-adrenal (HPA) axis in the face of diverse physiological challenges in health and disease. However ion channels that control corticotroph excitability remain poorly characterised. Members of the mammalian ether-à-go-go (EAG) channel family are voltage-gated potassium channels with diverse functions in the endocrine, cardiovascular and nervous systems. Expression of Kcnh2 mRNA, which encodes for the Eag-related potassium channel Erg1, is enriched in

Irisin modulates the transcriptomic profile of porcine anterior pituitary cells isolated from gilts on days 15-16 of pregnancy.

Irisin, an adipomyokine primarily known for its role in metabolic regulation, has recently emerged as a potential regulator of reproductive function. While its ovarian actions have been partially characterised, its influence on the anterior pituitary remains largely unexplored. This study aimed to investigate the in vitro effects of irisin on the transcriptome of porcine anterior pituitary cells (APc) isolated from gilts on days 15-16 of pregnancy. Primary APc were isolated from pigs on days 15-16 of pregnancy and treated with 300 ng/mL of irisin. RNA sequencing (NovaSeq 6000, Illumina) was pe

NR3C1 is required for normal somatotrope differentiation and Foxo1 expression in pituitary.

Glucocorticoids are an important signal for the differentiation of many types of cells. Consistent with this, several studies have demonstrated that glucocorticoids promote the somatotrope differentiation program and functionality. Interestingly, we previously found that loss of the forkhead factor, FOXO1, results in delayed emergence of somatotropes and prevents glucocorticoid-induced premature differentiation of somatotropes. In the present study, we find that pituitary-specific deletion of Nr3c1, the gene encoding the glucocorticoid receptor, impairs somatotrope differentiation and increase

Structural and Functional Basis of SSTR5 Homodimerization in Controlling Receptor Signaling and Pasireotide Response in Corticotroph Adenomas.

Somatostatin receptor subtype 5 (SSTR5), a G protein-coupled receptor (GPCR) highly expressed in the pituitary gland, plays a pivotal role in regulating adrenocorticotropic hormone (ACTH) secretion. While SSTR5 homodimerization has been observed in heterologous expression systems, its in situ existence, structural dynamics, and functional relevance in clinical pathology remain elusive. In this study, we provide the first evidence of endogenous SSTR5 homodimers in human corticotroph adenomas. Further analysis reveals that SSTR5 dimerization is mediated by multiple transmembrane (TM) interfaces

Recombinant FSH as an alternative to pituitary-extracted FSH for superovulation and embryo production in sheep.

The aim of this study was to compare recombinant bovine FSH (rbFSH) with a conventional multiple-dose purified porcine FSH (pFSH) for in vivo embryo production in sheep. Forty multiparous Santa Inês ewes were randomly assigned to two groups: the GPUR group received 333 IU of pFSH in six decreasing doses at 12-hour intervals; the GREC group received a single 100 μg dose of rbFSH. Both groups underwent SOV following a short estrous synchronization protocol and were naturally mated. Estrus tended to last longer in GPUR than GREC ewes (44.5 ± 3.9 h vs. 35.6 ± 3.9 h; P = 0.08). No significant diffe

Case Report: Homozygous KISS1R mutation associated with congenital hypogonadotropic hypogonadism in two siblings: pulsatile GnRH therapy restores pituitary architecture and induces pubertal development.

Congenital hypogonadotropic hypogonadism (CHH) is a rare disorder characterized by deficient production, secretion, or action of gonadotropin-releasing hormone (GnRH), the central regulator of the reproductive axis. We report two siblings from a consanguineous family with CHH caused by a homozygous nonsense mutation (c.182C > A; p. Ser 61Ter) in exon 1 of KISS1R gene. The 15-years-old male proband presented with absent puberty, micropenis, and gynecomastia. Endocrine evaluation revealed low gonadotropin levels and brain magnetic resonance imaging (MRI) showed pituitary hypoplasia. His 12-years

Third-trimester estradiol is associated with isolated maternal hypothyroxinemia and attenuated TSH-FT4 coupling: evidence from rats and human astrocytes.

INTRODUCTION: Isolated maternal hypothyroxinemia (IMH), defined by low free thyroxine (FT4) with thyroid-stimulating hormone (TSH) within the reference range, is a clinically relevant thyroid phenotype in late pregnancy, when estradiol (E2) rises sharply. We tested whether third-trimester E2 is associated with IMH and whether complementary animal and cell data are compatible with a proposed model of pituitary adaptation. METHODS: In 200 women at ≥28 gestational weeks (IMH, n = 100; euthyroid, n = 100), multivariable regression assessed associations of log2(E2) with IMH and FT4 after adjustmen

Endoscopic Versus Microscopic Transsphenoidal Surgery in the Treatment of Pituitary Adenoma: A Systematic Review.

Pituitary adenomas are the most prevalent pituitary pathology and originate from the neoplastic proliferation of anterior pituitary lobe cell lineages. Two primary surgical approaches are employed: microscopic transsphenoidal surgery (MTS) and endoscopic transsphenoidal surgery (ETS). Both approaches aim to optimize tumor resection while reducing the risk of complications. The objective of this review is to compare the outcomes of ETS and MTS in the treatment of pituitary adenomas. Studies from 1974 to March 20, 2026, from PubMed, ScienceDirect, and Cochrane databases were included. The inclus
