Fecal melatonin as a biomarker of emerging circadian maturity and gut microbiota in infancy.

Melatonin plays a key role in circadian regulation, and its interaction with the gut microbiota may be critical for early-life development. Beyond its circadian function, melatonin dysregulation is implicated in inflammatory, metabolic, psychiatric, and neurological disorders. While the gastrointestinal tract produces melatonin at levels far exceeding the pineal gland, its role in gut microbiota dynamics and circadian maturation remains unclear. This observational study investigates the association between fecal melatonin levels and microbial diversity, specific bacterial taxa (ZOTUs), actimet

Exploring the immune-modulatory role of melatonin in head kidney and spleen cells of freshwater snakehead teleost, Channa punctatus.

Melatonin, the ubiquitous hormone marks its presence from plants to animals with remarkable antioxidant and anti-inflammatory properties. In the teleost, melatonin is secreted from the pineal gland, which functions as a modulator of circadian rhythm, being homologous to the suprachiasmatic nuclei of mammals. The current study revolves around evaluating the role of in vitro melatonin in the immune mechanism of the snakehead fish, Channa punctatus. Reactive oxygen and nitrogen species production was assayed by respiratory burst and nitrite assay. Mitogen-stimulated lymphocyte proliferation was d

Melatonin in Integrative Oncology: Biological Mechanisms, Therapeutic Evidence and Implementation Strategies.

Melatonin, an endogenous indoleamine primarily synthesized in the pineal gland, has emerged as a promising adjunctive agent within integrative oncology due to its pleiotropic biological actions. Beyond its well-known chronobiological functions, melatonin exerts potent redox-regulatory, anti-inflammatory, oncostatic, and immune-modulating effects that are relevant across multiple stages of carcinogenesis and cancer therapy. Oxidative stress (OS), defined as an imbalance between reactive oxygen and nitrogen species (ROS/RNS) generation and antioxidant defenses, plays a central role in DNA damage

Single-cell multiomic and spatial landscape of the primate pineal gland reveals circadian and melatonin regulatory architecture.

The mammalian pineal gland maintains normal circadian rhythms and homeostasis by secreting melatonin. However, the lack of a single-cell-resolved regulatory map limits our understanding of how these neuroendocrine functions are orchestrated. Here, we constructed a multiomics atlas of the pineal gland from Macaca fascicularis by integrating snRNA-seq, snATAC-seq, and spatial transcriptomics. We identified pinealocytes as the predominant cell type, alongside six glial and vascular lineages. Chromatin accessibility analysis delineated cell-type-specific regions enriched for melatonin synthesis an

Immunohistochemical expression of pancreatic melatonin receptor (MTNR1B) under experimental sleep disturbance in rats.

Sleep amount and quality are important to maintain health, light exposure transported to the pineal gland via suprachiasmatic nucleus pineal pathway, and this affect melatonin synthesis. Melatonin exerts its action by receptors called MT1, MT2 that found in peripheral tissues including pancreas, which affect its endocrine function. In this study we investigate the effect of type of sleep disturbance on the histological structure and the expression of melatonin receptor (MTNR1B) in the pancreas. A sample of 45 adult healthy male rats had free access to water and feeding, divided into 3 groups (

History of Treatment of Pineal Tumors.

Pineal gland for a long time aroused the interest of scholars' philosophical and religious current of thought. Its history started in China in 2697 BC and in the Indu culture it was considered the centre of spiritual forces or the centre of vital energy. Herophilus (325-280) studied the pineal gland with anatomical dissection and considered the gland as the valve that permitted the flow of the vital "pnuema."Descartes (1596-1650), a French philosopher, gave a great impulsion to the studies on the role of the pineal gland. The location of the soul in the pineal gland was again proposed as it act

Incidence of Pineal Tumors.

INTRODUCTION: Although rare, the pineal region tumors are differently reported in the literature around the world. Regardless of whether a physician practices in eastern regions such as Japan, Europe, or America, the frequency with which they encounter these pathologies is distinct, as is their experience. Understanding the incidence of pineal tumors or their subtypes is crucial for their diagnosis and management in various therapeutic models. ANALYSIS OF THE DATA FROM THE LITERATURE: Comprehensive complementary research was conducted on PubMed. We use these MeSH terms: pinealoma, glioma, bra

Melatonin: Some Aspects of Biochemistry, Physiology, and Pharmacology.

Melatonin is a hormone synthesized by the pineal gland from tryptophan as a precursor and serotonin as an intermediate. It is secreted during the night and displays a peak around 3 a.m., depending on the individual chronotype. This rhythm is generated by an endogenous biological clock located in the suprachiasmatic nuclei (SCN) of the hypothalamus and entrained to the light/dark cycle (circadian rhythm). The roles of melatonin are numerous, but this hormone is described as a messenger of nighttime to the body, a modulator of circadian rhythmicity in mammals (including humans), and a regulator

Histopathology and Molecular Pathology of Pineal Region Tumors.

Tumors of the pineal region are rare neoplasms accounting for only about 1% of all central nervous system tumors. This chapter details the histopathological features and molecular alterations of these tumors. The two most common types are germ cell tumors (GCTs) and pineal parenchymal tumors (PPTs). PPTs are a group of neoplasms that exhibit varying degrees of pinealocytic differentiation and diverse prognoses. They comprise well-differentiated low-grade pineocytomas, pineal parenchymal tumors of intermediate differentiation (PPTIDs), and poorly differentiated high-grade pineoblastomas (PBs).

Decisional Tree for Pineal Tumors.

Pineal tumors are rare and it is important that they have to be treated by actors involved in their care. The first thing to do is the realization of the routine radiological studies necessary to establish if the tumor is located only in the pineal region or if there is bifocal localization or if the tumor is associated with a metastatic diffusion. Cerebral MRI represents the golden standard for diagnosis, associated, especially in children, to a spinal study. The study of pineal markers, the alpha protein, total human choroid gonadotrophin (hCG), and the free beta HCG, in the blood and in the

The Infratentorial Supracerebellar Approach: The Most Used Avenue for Pineal and Pineal Region Tumors: How We Do It.

The infratentorial supracerebellar approach is the most commonly used surgical approach for pineal and pineal region tumors in the literature (Mottolese et al. Neurochirurgie 61:176-183, 2015).The infratentorial supracerebellar approach is performed through a natural surgical corridor, which is facilitated by the use of the sitting position. This approach is particularly indicated for tumors that are strictly midline and measure no more than 2 cm in diameter (Stein. Surg Neurol 11:331-337, 1979; Stein. J Neurosurg 35:197-202, 1971; Pendl et al. Neurosurgery 26:641-648, 1990).Advances in surgica

Surgery of Pineal and Pineal Region Tumors: Alternative Approaches to Pineal Tumors: How to Do It.

Alternative approaches can be used when pineal tumors extend into the third ventricle or the lateral ventricles. For tumors that develop in the posterior cerebral fossa, a combined supra- and infratentorial approach can be proposed. An anterior avenue for lesion of the third ventricle and the pineal region using a frontal transcallosal approach was proposed by Apuzzo in 1992. A posterior parietal transventricular approach had already been proposed by Van Vagenen in 1931 and later by Jomin in 1980, through the ventricular crossroads, allowing lateral exposure of the venous structures, the tumor,

The Pinealoprive Syndrome: Myth or Reality?

Although the role of melatonin in many biological functions, foremost among them the regulation of circadian rhythms and sleep, has been widely demonstrated, the definition of a specific disorder linked to melatonin deficiency remains debated. As the main function of the pineal gland is to secrete melatonin, pineal lesions and pinealectomy provide insightful models for understanding the effect of acute and chronic suppression of melatonin secretion ("pinealoprive syndrome"). In pineal parenchymal cell tumor, melatonin levels are usually normal/high, whereas impaired secretion is thought to ref

Conclusions.

Pineal tumours represent a valuable model of multidisciplinary collaboration. Over recent years, such collaboration has led to the significant improvements in outcomes, increased cure rates and reduction in long-term sequelae. Advances in diagnostic workup, including detailed tumour investigation and the study of tumoural markers, now allow more accurate treatment strategies. When a biopsy is required, a clear increase in the use of the endoscopic techniques has been observed in recent years, especially in paediatric patients. This approach allows simultaneous treatment of intracranial hyperten

Melatonin and the Cardiovascular System.

Melatonin (N-acetyl-5-methoxytryptamine) is a neuroendocrine hormone primarily secreted by the pineal gland and also produced in extrapineal tissues such as the retina, lymphocytes, and the cardiovascular (CV) system. This hormone possesses amphiphilic properties, allowing it to penetrate most biological barriers and exert its effects at the subcellular level; it plays an important role in controlling the body's response to circadian rhythms and adjusting to internal and external environmental cues. It also has antioxidant, anti-inflammatory, and antiaging properties. Melatonin's secretion is
