Immune Responsiveness Induced by Administration of Heat-Inactivated M. bovis and BCG Prior to Experimental Infection in African Buffalo.

African buffalo (Syncerus caffer) are the primary wildlife reservoir for bovine tuberculosis (BTB) in Africa. Infected buffalo populations spread BTB to numerous species in conservation areas as well as to cattle at park boundaries. Quarantine requirements for BTB complicate wild animal metapopulation management. Considerable interest has arisen in the vaccination of cattle and indigenous wildlife species against BTB as an additional control strategy. This pilot trial aimed to evaluate cell-mediated and humoral immune responses in African buffalo immunised against BTB. Twenty-one sub-adult Afr

Utilization of the CRISPR-Cas system for investigating tuberculosis drug resistance.

Drug-resistant tuberculosis(TB) presents a significant challenge to global efforts to control the disease. Addressing this issue requires a comprehensive understanding of drug resistance in Mycobacterium tuberculosis (M.tb). The clustered regularly interspaced short palindromic repeat (CRISPR) and CRISPR-associated protein (Cas) system are efficient genome-editing tools for various organisms and have facilitated new research directions regarding drug resistance in M.tb. This review summarizes recent progress in applying CRISPR/Cas technology to investigate drug resistance mechanisms in M.tuber

A quasi-experimental study evaluating the effectiveness of a male-specific tuberculosis screening and diagnosis intervention in Uganda (IGNITE).

BACKGROUND: Despite men having a higher burden of TB, they are frequently missed by TB services due to social, behavioural, and health system factors that limit healthcare access. In Uganda, where there are no gender-specific guidelines for TB, we implemented and evaluated a gender-specific intervention to find and treat men with TB. METHODS: We conducted a quasi-experimental study comparing TB notifications before (January-June 2023) and after (July-December 2023) the intervention at both intervention (Gombe and Mityana hospitals) and control (Luwero and Kiboga hospitals) sites. The interven

Antimicrobial activity of twelve plant-derived compounds against fish-associated non-tuberculous mycobacteria.

Nontuberculous mycobacteria (NTM) are important pathogens of fish, causing chronic infections that are difficult to control in aquaculture. In this study, we evaluated the in vitro antimicrobial activity of twelve plant-derived compounds against six fish-associated NTM strains using a broth microdilution assay. Minimum inhibitory concentrations (MICs) ranged from 0.25 to >32 µg/mL. Our results demonstrated that cinnamaldehyde exhibited the strongest antimycobacterial effect, followed by sesamol and farnesol, while several other phytochemicals showed moderate activity and salicylic acid was ina

Nano-NucleOTAC-mediated Z-DNA transformation disrupts mycobacterium tuberculosis biofilms and activates antibacterial immunity.

The biofilms of Mycobacterium Tuberculosis (Mtb) use extracellular DNA (eDNA) as a protective structural scaffold that shields resident bacteria from host immunity and antibiotics, promoting persistent infection. An FDA-approved DNase can partially hydrolyze eDNA but has limited affinity and nucleolytic activity against Z-form DNA (Z-DNA). After confirming the abundance of Z-DNA in Mtb biofilms, we develop Nano-NucleOTAC, a nano-nucleolysis targeting chimera that combines an eDNA-binding ligand (lactoferrin), a DNA intercalator (chloroquine) and DNase. Nano-NucleOTAC anchors to Z-DNA in Mtb H3

Prognostic utility of immune biomarkers for identifying individuals at risk of unfavourable treatment outcomes in pulmonary tuberculosis.

INTRODUCTION: Tuberculosis remains a major global health challenge, with many patients expe- riencing unfavourable treatment outcomes despite standard therapy. Host immune biomarkers, including cytokines, chemokines, and matrix metalloproteinases (MMPs), have shown promise in predicting disease severity and treatment response, but require validation across independent cohorts. METHODS: We validated previously identified immune biomarkers using baseline plasma samples from a nested case-control cohort within the RePORT India consortium (cases: n=74; controls: n=161). Cytokines (IL-2, IFN-γ, TN

Tuberculous spinal epidural abscess: a cautionary case report of permanent paraplegia due to delayed diagnosis and management.

BACKGROUND: Spinal epidural abscess caused by tuberculosis is a rare but serious complication of spinal tuberculosis. Its atypical presentation predisposes to diagnostic delay and a poor prognosis. This case not only documents the clinical and radiologic evolution of tuberculous spinal epidural abscess but also suggests that delays in diagnosis and treatment may jointly contribute to serious neurological outcomes. CASE PRESENTATION: A 39-year-old man initially underwent acupotomy for recurrent low back pain with recent exacerbation. Subsequent imaging revealed a paraspinal abscess within the

KLRG1+CD28⁻CD57+ Ag85A-specific CD8+ T cells mark a severity-associated terminal-differentiation imprint in pulmonary tuberculosis.

BACKGROUND: Pulmonary tuberculosis (PTB) is characterized by prolonged host-pathogen interaction, chronic granulomatous inflammation, and sustained exposure to Mycobacterium tuberculosis (Mtb) antigens. How Mtb antigen-specific CD8+ T cell differentiation states relate to clinical disease activity and severity remains incompletely defined. This study investigated the clinical relevance of a terminal differentiation-associated phenotype within Ag85A-specific CD8+ T cells across clinically defined states of Mtb infection and disease. METHODS: A total of 124 participants were enrolled, including

Self-reported prevalence of tuberculosis and selected associated vulnerability factors among rural populations in six districts of India: A community-based cross-sectional study.

BACKGROUND: Tuberculosis is a major public health challenge in India, contributing to one-fourth of global burden of TB disease. Comorbidities in uninfected individuals make them vulnerable to TB disease. This study estimates the self-reported prevalence of tuberculosis and selected vulnerability factors (HIV, current smoking, silicosis, occupational silica-dust exposure, diabetes mellitus and persistent cough >2 weeks) among the rural demography of six districts in India, with the aim of informing targeted active case finding under the National Tuberculosis Elimination Programme (NTEP). Comparative serum cytokine and chemokine profiles across tuberculosis, latent TB, HIV, and COVID-19 infections in Namibian adults.

BACKGROUND: The devastating pandemic of coronavirus disease 2019 (COVID-19) highlighted the importance of early detection and global collaboration in combating infectious diseases. Namibia is one of the countries that bears a high burden of tuberculosis (TB) and human immunodeficiency virus (HIV) infection. The overlap of these diseases underscores the need to better understand host immune responses, which play a critical role in disease pathogenesis and may differ across infections. This study aimed to profile selected cytokines and chemokines among Namibian individuals with TB, latent TB inf

Predictors of tuberculosis preventive therapy initiation among people living with HIV in the Volta region, Ghana.

BACKGROUND: Tuberculosis (TB) remains a leading cause of morbidity and mortality among people living with HIV (PLHIV) despite the availability of effective preventive interventions. Tuberculosis Preventive Therapy (TPT), strongly recommended by the World Health Organization, substantially reduces the risk of active TB among PLHIV, however, uptake of TPT remains suboptimal in many settings. This study, therefore, assessed the predictors of TPT initiation, beginning the course of anti-TB medications among PLHIV. METHODS: This was a health facility-based analytical cross-sectional study among 80

Tuberculous Meningitis Alters the Proteomic Landscape of Brain-Derived Extracellular Vesicles.

Tuberculous meningitis (TB meningitis), the deadliest form of Mycobacterium tuberculosis infection, causes devastating mortality and severe neurological disability despite standard therapy. Brain injury and microglial activation are major determinants of outcome, yet the molecular mechanisms linking infection, inflammation, and neuronal injury remain poorly understood. Extracellular vesicles (EVs), critical mediators of cell-to-cell communication, have been investigated in pulmonary TB, but their role in TB meningitis remains unexplored. We used a young New Zealand White rabbit model of TB men

Identification of a spatially organized TREM2-associated permissive macrophage niche in human pulmonary tuberculosis alveolar pneumonia.

Pulmonary tuberculosis (TB), caused by lung infection with Mycobacterium tuberculosis (Mtb), often manifests both alveolar pneumonia and aggregation of immune cells containing Mtb, termed granulomas, within the same lung. Here, we integrate spatial transcriptomics, single-cell RNA sequencing, high-resolution imaging, and functional assays of lung biopsies from four men with pulmonary TB to compare alveolar pneumonia with adjacent granulomas. We find that alveolar TB pneumonia is enriched for TREM2⁺ lipid-laden macrophages characterized by increased lipid metabolism gene expression, reduced T c

Carbonic Anhydrases in Mycobacteria: Linking Metabolic Adaptation to Phenotypic Drug Tolerance.

Tuberculosis (TB) treatment is increasingly challenged by both antimicrobial resistance (AMR) and phenotypic drug tolerance, the latter representing a reversible physiological state that contributes to persistence and prolonged therapy. While AMR is driven by heritable genetic mutations, drug tolerance arises from reversible physiological adaptations that enable bacterial survival during prolonged antibiotic exposure. Increasing evidence suggests that metabolic remodeling plays a central role in establishing these drug-tolerant states. Carbonic anhydrases (CAs) are zinc-containing metalloenzym
